The low quality fresh ginseng was fermented by Phelinus linteus or Hericium erinaceum mycelium. This fermented ginseng was extracted by water at 100℃ or water with ultrasonification at 60℃. Total phenolic compounds was improved by ultrasonification extraction process, compare to conventional water extraction. All extracts enhanced the growth of human B and T cells, showing 2.68 times and 3.43 times higher, respectively, than the control. The secretion of TNF-α and IL-6 from human immune cells was enhanced as 3.53×10-4 pg/cell, 3.40×10-4 pg/cell by adding H. erinaceum mycelium fermented ginseng. H. erinaceum mycelium fermented ginseng yielded higher nitric oxide production from macrophage than Lipopolysaccharides (LPS). The cytotoxicity on human normal kidney cell (HEK293) was as low as 20.5% in adding the maximum concentration of 1.0 mg/ml of fermented ginseng. Generally, the extracts from ultrasonification extraction process showed 10% lower toxicity than that by conventional process. H. erinaceum mycelium fermented ginseng had the highest anticancer activity on human lung cancer and stomach cancer cells as 69.33% and 75.32%, respectively at 1.0 mg/ml. It can be concluded that, in general, H. erinaceum mycelium fermented ginseng has relatively better immune and anticancer activities than P. linteus fermented ginseng. Expecially, the extracts treated with ultrasonification had higher activities than that from conventional extraction process.

Sunlight, and in particular its UV component, is the major environmental trigger that underlies the major signs of human skin and skin cancer in general. Therefore, this study was carried out to investigate the UV protection effects of R. coreanus. R. coreanus was extracted by ultra high pressure extraction process at 500 MPa and 30℃ for 5 and 15 minutes. The cytotoxicity of the extracts extracted by ultra high pressure process on human dermal fibroblast cell CCD-986sk, human kidney normal cell HEK293, and human lung normal cell HEL299 was measured as 17.5%, 16.5% and 14.0%, respectively in adding 1.0 mg/ml of the samples, which was much lower than that from conventional water extraction method at 100℃ as 23.2%, 22.5%, 21.2%. The secretion of NO- from macrophage showed 15.9 μM on the R. coreanus extract from this process, which was higher than others. Prostaglandin E2 (PGE2) production from UV-induced human skin cells was also greatly decreased down to 510 pg/ml, compared to the control. From the results, we considered that the extracts from R. coreanus could be potent natural materials for skin anti-inflammation agent, and could be used as a potential anti-aging for the photo-damaged skin.

Experimental purpose was to evaluate growth characteristic and abscisic acid (ABA) responses against salt/drought stresses. In the shoot biomass, creeping thyme was tolerated in mild NaCl stress, ranging 0 to 100 mM, while it was severely reduced in higher salinity. Under constant drought stress, the shoot biomass of creeping thyme showed a worse value compared to that of 100 mM NaCl treatment. Chlorophyll degradation was more severe in immature leaf than mature leaf under salt and drought stresses. In salt stress, immature leaf produced much amounts of ABA compared to mature leaf and also immature leaf showed faster increase of ABA than that of mature leaf. In drought stress, immature leaf responded to stress within 24 hours by the increase of ABA, while mature leaf responded to at 72 hours. Our results recommended that the optimal salinity level of creeping thyme was 50~100 mM NaCl.

To evaluate the availability of Cudrania tricuspidata Bureau as a natural source of antimicrobials, the antimicrobial activity of methanol extracts of harvested parts was investigated using the paper disc diffusion method. The extracts from leaves and root bark had broad antimicrobial activity against various bacteria, including Bacillus subtilis, Bacillus cereus, Staphylococcus aureus, Streptococcus mutans, Listeria monocytogenes, Enterobacter aerogenes, Enterococcus faecalis, Salmonella choleraesuis subsp. choleraesuis, Vibrio vulnificus, and Pseudomonas aeruginosa and inhibited Bacillus cereus, Staphylococcus aureus, and Listeria monocytogenes, agents of food poisoning especially well. The extract from ripe fruit had a very high antimicrobial activity against Staphylococcus aureus with a 20.2 mm of clear zone at 50 mg/mL sample concentration. These results indicated that Cudrania tricuspidata could be used as new source for developing natural antimicrobial agents.

Cerebral ischemia results from a transient or permanent reduction in cerebral blood flow that decreases oxygen and glucose supply. When the cellular oxygen supply is reduced to critical level, damage to cells and induction of cell death are occurred by excitotoxicity, oxidative stress and inflammation. Ischemia remains one of the leading causes of death, but there is no effective treatment that might protect neurons gainst ischemia by interrupting the cascade of cell death. In this study, human neuroblastoma SH-SY5Y cells are exposed to oxygen and glucose deprivation (OGD) followed by reoxgenation. OGD can mimic the acute restriction of metabolite and oxygen supply caused by ischemia and is widely used as a model of ischemic conditions. SH-SY5Y cells are treated samples at the commencement of OGD to achieve different final concentrations, and cell viabilities were quantified using the measurement of flow cytometry analysis. Of those tested, the extracts of Polygala tenuifolia (roots), Dictamnus dasycarpus (barks), Polygala tenuifolia (roots), Eucommia ulmoides (branches), Eucommia ulmoides (barks), Poria cocos (whole), Sophora flavescens (roots) showed neuroprotective effects, with EC50 values of 4.5±0.6, 7.9±1.5, 10.5±0.7, 18.4±1.9, 19.6±0.3, 21.6±1.9, and 30.7±3.9μg/ml, respectively.

To study the optimal cultivation for paddy soil, growth characteristics and ginsenoside content was investigated by both of poor drainage class (PDC) and imperfect drainage class (IDC) in three-year-old ginseng of varieties, Cheonpoong (CP), Yeonpoong (YP), Hwangsookjong (HS), and Jakyeongjong (JK). Root yield of IDC was higher than that of PDC by 3.6 times because stem length, leaf area, and chlorophyll contents were increased, while discolored leaf ratio was decreased. Root yield of HS in PDC was highest among four varieties because chlorophyll contents, leaf area, and survived plant ratio were relatively high. Root yield of CP in IDC was highest among four varieties because of high leaf area and survived plant ratio, and low discolored leaf ratio. Ratio of rusty-colored root showed significant difference by varieties, which was the highest in HS and the lowest in CP among four varieties irrespective of drainage classes. Total ginsenoside contents showed significant difference by drainage classes, which were high in IDC of good growth and low in PDC of poor growth. Total ginsenoside contents were high in JK and CP, while low in HS and YP both of drainage classes.

In this study, raw ginseng water extract solutions were analyzed to set up the functional saponin content and quality optimization condition. The highest saponin content among the total raw ginseng water extracts was 74.6 mg/100 ml which was extracted at 75℃ for 24 hours. In addition, the saponin content decreased according to the increased extraction temperature and time. The highest total content of Rb2 and Re was 19.9 mg/100 ml at 75℃ for 12 hours which decreased according to the increased extracted temperature and time. The highest prosapogenin (Rg2 + Rg3 + Rh1) content among the total raw ginseng water extracts was 28.6 mg/100 ml which was extracted at 85℃ for 36 hours. The reducing sugar content, sweetness and turbidity were increased according to the increased extraction temperature and time. But pH were decreased according to the increased extracted time.

Compounds of isolated from roots extract of Pueraria thunbergiana were tested their inhibitory effects on α-glucosidase and α-amylase. Inhibitory activity of methylene chloride (MC) fraction and ethyl acetate (EA) fraction against α-glucosidase showed more than 60% at a concentration of 500μg/ml. Among the nine compounds tested on α-glucosidase, biochanin A, (-)-tuberosin and calycosin from MC fraction and daidzein from EA fraction were stronger inhibitors than acarbose (IC50=530μg/ml), and their IC50 were 9, 144, 328 and 20μg/ml, respectively. Biochanin A and (-)-tuberosin also inhibited α-amylase activity as like as acarbose IC50=20.5μg/ml), and their IC50 were 22 and 348μg/ml, respectively. Although daidzein was already known α-glucosidase inhibitory effects, it was newly evaluated that biochanin A and (-)-tuberosin inhibited α-glucosidase as well as α-amylase, and that calycosin did α-glucosidase.

To fulfill the increasing demand for a high quality of flower, we investigated the effects of nitrogen application on plant growth, yield and bioactive compounds of Chrysanthemum indicum L.. C. indicum L. was cultivated in a pot scale, and nitrogen applied with the level of 0 (N0), 50 (N50), 100 (N100), 150 (N150), 200 (N200) and 300 (N300) kg ha-1 to suggest optimum rate of nitrogen fertilization. Phosphate and potassium applied the same amount of 80-80 kg ha-1 (P2O5-K2O) in all treatments. Growth characteristics and yields of C. indicum L. were significantly affected by nitrogen application. Maximum yield achieved in 265 and 295 kg ha-1 N treatment on the whole plant and the flower parts, respectively. The nitrogen content and uptake of whole plant significantly increased by the increase of nitrogen application. Five major components of essential oil, α-pinene, 1,8-cineol, chrysanthenone, germacrene-D, and α-curcumene in flowerheads of C. indicum L. occupied approximately 40% of peak area, germacrene-D decreased by the increase of nitrogen application among them. However, cumambrin A contents in the flower parts of C. indicum L. were affected negatively by the increase of nitrogen application, but total yields of cumambrin A in flower part significantly increased. Conclusively, nitrogen fertilization could increase the yield of flowerheads. The optimum application level of nitrogen fertilizer might be on the range of 265-295 kg ha-1 in a mountainous soil.
