This study was performed to compare effect of anticancer activities on Rhodiola sachalinensis by ultrasonifi-cation process and solvent. Compared the yield to the water extracts (WE), wafer extracts with ultrasonification (WEU) at60, loot and ethyl alcohol extracts (EE), ethyl alcohol extracts with ultrasonification (EEU) at 60 ˚C from Rhodiola sachalinensis. Experimental studies were progressed through the anticancer activities such as cell cytotoxicity, inhibition activities of cell growth. The cell cytotoxicity using human embryonic kidney cell (HEK293) was showed cytotoxicity of below 26.26%by extracts of Rhodiola sachalinensis in 1.0 mg/ml concentration, The anticancer activities were increased in over 70% by extracts of Rhodiola sachalinensis in A549, AGS, Hep3B and MCF-7 cells. From the results, the extract Rhodiola sachalinensis were showed useful anticancer activities.
Green tea is of particular source as it has been found to have strong antioxidant activities. The extracts of green tea during the commercial harvest seasons from April, 2003 to August, 2003 were compared. The purpose of this study was to determine the correlation between the polyphenol content of green teas and its antioxidant activities. The con-tent of total polyphenols was analyzed and several antioxidant testings were performed. The levels of total polyphenols were higher in the green teas (e.g. Woojeon, Sejak) harvested during very early spring and lower in the green teas harvested late(eg. Ipha, Yepcha). In particular, the free radical scavenging, the inhibition of LDL oxidation, the cytoprotective effect and the inhibition of DNA damage were correlated with the total polyphenol contents of green tea extracts harvested early spring such as Woojeon, Sejak and Jungjak. The results obtained here show that all extracts of green teas including purified green tea catechin, GTC, have strong antioxidant activities on oxidative stress in vifrθ. The variation in polyphenol content and antioxidant activities among various types of green tea by the harvesting time may provide critical information for investigators and consumers using tea in purposes of nutrition and chemoprevention.
The present study was designed to investigate aging-related effects on the activities of xenobiotic metabolizing enzymes of rat liver by dietary supplementation of Korean red ginseng water-extract. Rat did not show any discernible signs of the rejection symptoms, and blood GOT and GPT levels were not influenced by ginseng water extracts, Cytochrome 450 levels and NADPH cytochrome P45O reductase, p-450 dependent ethoxycoumarin O-deethylase, and benzphetamine N-demethylase activities were decreased with aging, however, these phase I system enzymes activities in the ginseng group of24 months were well maintained compared with normal group. But, Levels of cytochrome bs and NADH-cytochrome b5 reductase activities were also decreased with aging and were not found a clear difference between two groups. Glutathione-s-transferase activity, phase II enzyme system, in liver cytosols was also decreased in old ages, but the degree of decrease was higher in normal group than in giuseng supplemented group. These results indicate that long-term supplementation of red ginseng water extracts from weaning to 24 months do not show any side effects to rats, and retard age-related deteriorations of xenobiotic metabolizing enzymes activities in old ages.
The endophytic bacteria were isolated from the rusty-root ginseng. This isolated bacteria were occurred the rusty-root ginseng with artificial inoculation. For the suppressing of rusty-ginseng, disinfectants, antibiotics, kitosan, micro-organisms and metabolites were tested to isolated endophytic bacterium. All of the isolated bacteria strains were sensitive sodium hypochlorite, however, some of isolated bacteria lines were sensitive to other tested materials. For example, D (didecyl dimethyl ammonium bromide), CIO2, ODDA (octyldecyl dimethyl ammonium chloride + diocyul dimethyl ammonium chloride + alkyl diethyl benzyl ammonium chloride), GD (glutaraldehyde + dimethy cocobenzyl ammonium chloride) suppressed some of bacteria strains. Otherwise, some of antibiotics (e.g. ampicillin, chloramphenicol, erythromycin, kanamycin, neomycin, rifampin, streptomycin, tetracycline) were sensitive to the isolated bacteria strains. All of isolated bacteria strainswere inhibitive to the mixed formation with neomycin and streptomycin, and neomycin and tetracycline. Both sodium hypochlorite and antibiotic mixing of neomycin and tetracycline were effective to prevention of rusty-root ginseng of sub-merging ginseng in the ginseng field.
Anatomical characters of the Bupleurum latissimum Nakai, an endemic species of Korea, were investigated to confirm its phylogenetic relationships. Compare to other species with anatomical characters, B. latissimum is very similar with B, euphorbioides and B, longeradiatum in point of lacking of pith in the stem, shape of involucres, number of vascular bundles in radical leaf and cauline leaf, and lacking stomata in adaxial leaf surface. The other hand, protruded pollen aperture character appears in B. latissimum and B. euphorbioides. On the based of anatomical characters, therefore, B. latissimum has closest relationships with B. euphorbioides and B. longeradiatum. It also needs molecular study including Asian species in order to confirm phylogenetic position and speciation process apparently.
This study was conducted to evaluate the vegetation structure and ordination of the Peucedanum japonicum Thunb. community by the Phytosociological method, floristic composition table on east coast of South Korea. The vegetation of Peucedanum japonicum Thunb. community was classified into 2 communities such as the Aster Spathulifolius community and the calystegia soldanella-Artemisia princeps community. Vegetation of the Peucedanum japonicum Thunb. community with the Aster spathulifolius community was shown southward, whereas vegetation of Peucedanum japonicum Thunb. community with the calystegia soldanella-Artemisia princeps community was shown northward from the base line of Pohang at North latitude of 36" 05'. Accordingly, the Peucedanum japonicum Thunb. community was grown commonly with the Aster spathulifolius community southward of the base line of Pohang, whereas it was grown commonly with the calystegia soldanella-Artemisia princeps community northward on the east coast. All environmental conditions of habitat taken together, the optimum habitat of Peucedanum japonicum Thunb. was at Ulsan geographically and the middle region of sea cliff topography, and was suitable for alkali. sandyloam.
Chromosome analysis using mitosis, meiosis and bicolor FISH were carried out in Bupleurum latissimum Nakai, which is one of the endemic plants in Ulleung island of korea. The somatic methaphase chromosomes number of this plant was 2n = 2x = 16 and the chromosome complements consisted of six pairs of metacentrics and two pairs of submetacentrics. The size of chromosomes ranged 2.40~4.20 μm and NOR (nucleolus organizer region) chromosome did not observed using conventional staining. In meiosis chromosomes, metaphase-I and anaphase-I were observed. Metaphase-I anaphase-I showed 8 bivalents and chromosomes migration to make two daughter cells. Using bicolor FISH, one pair of 5S and 45S rDNA signals were detected on the centromeric region of chromosome 3 and the end of short of chromosome 2,respectively. We also observed the NOR using 45S rDNA probe.
To investigate the effect of culture conditions on growth and ginsenosides accumulation, we cultured the ginseng hairy root under three different media, white light or ultra-violet irradiation. The MS/B5 medium containning MS basal salt and B5 vitamin was good for the growth and ginsenoside accumulation. The light during the culture period of ginseng hairy root was irradiated. The growth was abundant in the ginseng hairy root cultured in dark. But the ginsenosides accumulation was higher than in the ginseng hairy root cultured in the light irradiation. When the ginseng hairy root was cultured in 20 L bioreactor, the ginsenosides accumulation was observed at 34% higher than the hairy root cultured in dark. UV irradiated the ginseng hairy root during the culture period. The long time irradiation of UV was caused decreasing the growth of ginseng hairy root, but the accumulation of ginsenosidess was increased as to the irradiated time.
The study was carried out to establish an improved protocol for shoot organogenesis and plant regeneration from leaf explant cultures of Scrophularia buergeriana M. with the treatment of ethylene inhibitors [silver nitrate (AgNO3), aminoethox-yvinylglycine (AVG), Cobalt chloride (CoCl2)]. The regenerated shoots obtained from leaf explant cultures on MS medium containing 2 mg/l BAP, The additions of AgNO3. AVG and CoC12 substantially improved the shoot regeneration frequency, at the optimal concentration of 7 mg/L, 7 mg/L, and 3 mg/L respectively, The regenerated shoots could be easily rooted with 0.1 mg/L IBA treatment. The noted plants were hardened and transferred to vermiculite with a 85% survival rate where they grew normally.
To investigate effects on the growth and ginsenosides accumulation in ginseng hairy root, potassium phosphate was supplemented with 1.25, 2.5, and 5.0 mM concentration in 1/2 MS medium, respectively. Potassium phosphate supplement was increased the biomass and ginsenosides accumulation when it was dose at the concentration of 1.25 mM. And the growth rate of hairy root in the light condition was higher than in dark condition. The highest contents and productivity of ginsenosides were observed at the supplement of 1.25 mM potassium phosphate at the 7th day after the culture onset. Ginseng hairy root cultured in 20 L bioreactor supplemented with 1.25 mM KH2PO4 was increased the growth with 1,609 g (F·W) and ginsenosides content with 11.09 mg than those in control.
