Astragalus membranaceus has a long history of medicinal use in Chinese herbal medicine. It has been shown to have immunostimulant, tonic, antioxidant, antiperspirant, diuretic, anti-diabetic, expectorant properties, and a supplementary medicine during cancer therapy. In this study, we investigated the effect of anti-oxidation of Astragalus membranaceus root extract. The anti-oxidative activities of water, 80% methanol, and 100% methanol extracts from Astragalus membranaceus were analyzed by DPPH free radical scavenging activity, Superoxide dismutase-like activity, reducing power, and crude ash. The water extract demonstrated to be more effective than methanol extract for a DPPH radicals scavenging activities and reducing power. Superoxide dismutase-like activity showed higher efficiency in 80% methanol extract. Our results indicate that Astragalus membranaceus extracts could be used as a source of antioxidant ingredients in the food industry.

Somatic embryos on growth regulator-free medium can be produced directly from cotyledon explants of Panax ginseng C. A. Meyer. When the embryo developmental stage was torpedo and cotyledon, the germination rate of embryos was quite high on MS medium supplemented with gibberellic acid (GA3). However, the percentage of plantlet formation at the cotyledon stage was higher than that at the torpedo stage. This result demonstrates that the embryo at the cotyledon stage was the most appropriate for increasing germination by GA3. Embryos cultured on medium including four levels of GA3 concentrations (3, 5, 10, or 20 mg/l) showed all quite high germination rates (87-91%). When the well-developed embryos were continuously cultured on media including high concentrations of GA3 from 10 to 20 mg/l, the percentage of formation of normal plantlets was lower than that seen under low concentrations from 3 to 5 mg/l. This treatment of high concentrations resulted in shoots with abnormal shape. The optimal GA3 treatment provides a basis for the efficient method obtaining healthy plantlets derived from ginseng somatic embryos.

Fifty percent ethanol extract of Lythrum salicaria Linne root (LSR) was tested in vitro on antioxidant activity, and furthermore was investigated on antioxidative and fibrosis protecting activities in CCL4-induced liver fibrosis rat model. Ratio of hepatic GSH/GSSG (reduced glutathione/oxidized glutathione) as bio-parameter of antioxidant level in CCL4 plus LSR-treated rats for 6 weeks significantly increased from 2.8- to 5.7-fold than that of CCL4-treated rats at p 〈 0.05. Thiobarbituric acid reactive substances (TBARS) contents in CCL4 plus LSR-treated rats ranged from 1.57- to 2.19-fold of normal rats and were lower than those in CCL4 plus silymarin-treated rats (1.78~2.46-fold of normal rats) (p 〈 0.05). Amounts of hydroxyproline of liver tissue showing the content of total collagen, a parameter of fibrosis, in CCL4 plus LSR-administrated rat livers were 4.9~8.8μg/mg (-47~-71%, compared with that in CCL4-treated rat livers (16.6μg/mg tissue), which were significantly lower than those in CCL4 plus silymarin-administrated rats being 8.4~11.7μg/mg (-30~-50%). This collagen reducing effect of liver tissue in CCL4 plus LSR-treated rats was supported by histological observation using microscopy assay. From the results, we conclude that the root of L. salicaria have efficient antioxidant potential and effective antifibrotic activities.

The parts of leaves, flowers and stems in ginseng were obtained for analyzing the component of saponin on 15th April, 25th April, 5th May, 25th May, which were considered as ginseng foliation stage. The total saponin content of the leaves were 97.29, 66.42, 67.61, 36.24 mg/g, respectively, in which the content of Re, Rb1 and Rd were more than 2/3 amount of total saponin. Especially, the saponin content of leaves decreased according to the sequential collection days, in which the similar results were observed from the flowers and stems of ginseng. The total saponin content of the flowers and stems were 141.09,143.84,139.25,133.47 and 13.32, 9.85, 8.00, 4.65 mg/g, respectively. Among them, the content of Re, Rd and Rb2 in flowers were more than 2/3 while the content of Re, Rg1 and Rd in stems showed more than 9/10 amount of total saponin. The total saponin content of individual leaf were 19.46, 28.56, 58.82 and 169.24 mg/plant, 2.53, 2.76, 5.20 and 12.32 mg/plant in stems, and 14.11, 30.21, 37.60 and 73.41 mg/plant in flowers. Therefore, the total saponin content of aboveground parts in ginseng were leaves 〉 flowers 〉 stems.

Phosphatidylcholine was used to encapsulate aqueous extracts of Centella asiatica, and its biological activity was compared with another aqueous extracts. Nanoparticle of C. asiatica was made by encapsulation to w/o type spherical liposome which of aqueous extracts seized with oil phase as 78.2 nm average diameter. Cytotoxicity of the nanoparticle was measured on human skin fibroblast cells, CCD-986sk, and showed lower cytotoxicity on 1.0 mg/ml of highest concentration as 28% than that of another extracts. The nanoparticle showed the highest promotion of human B and T cell growth up to 138% and 135%, respectively, compared to the control. and the NK cell growth was promoted up to 8% higher than the control in proportion to secretion of IL-6 and TNF-α from immune cell growth. Also nanoparticle showed highest inhibition activity of hyaluronidase on 1.0 mg/ml of highest concentration as 60.5%. It seems that because of enhanced biological application of aqueous extracts on cell through nano-encapsulation process.

Cinnamyl alcohol dehydrogenase (CAD, EC 1.1.1.95), catalyzes the reduction of hydroxycinnamaldehydes to give hydroxycinnamyl alcohols, or "monolignols," the monomeric precursors of lignin. Lignins are important components of cell walls and lignified secondary cell walls play crucial roles in long distance transport of water and nutrients during plant growth and development and in plant defense against biotic and abiotic stresses. Here a cDNA clone containing a CAD gene, named as PgCAD, was isolated from a commercial medicinal plant Panax ginseng. PgCAD is predicted to encode a precursor protein of 177 amino acid residues, and its sequence shares high homology with a number of other plant CADS. The expression of PgCAD in adventitious roots and hairy roots of P. ginseng was analyzed using reverse transcriptase (RT)-PCR under various abiotic stresses such as salt, salicylic acid, wounding and chilling treatment that triggered a significant induction of PgCAD at different time points within 2-48 h post-treatment. This study revealed that PgCAD may help the plants to survive against various abiotic stresses.

GATA-binding protein-3 (GATA-3) and T-box expressed in T-cells (T-bet) are now considered as master transcription factors involving Th cell differentiation, but the roles of these factors are still uncertain in vivo. This study was conducted to investigate the expression of these transcription factors in the liver damage induced by carbon tetrachloride (CCl4) in rats. In this study, liver damage were induced with Scutellaria baicalensis Georgi water extracts (SBW) and followed for 4 weeks. The expression of GATA-3 and T-bet protein in liver damage induced by CCl4 and the serum levels of immunoglobulin A (IgA), IgE were studied after 4 weeks of treatment. We found that effect of SBW on IFN-γ, STAT1, pSTAT1 and T-bet was decreased in vivo. Several genes were demonstrated to be IL-4 inducible prior to the discovery of STAT6. CCl4+SBW group was significantly lower than CCl4 group in IL-4, STAT6, pSTAT6 and GATA-3. Our data indicate that cytokine protein production were increased in CCl4 group and CCl4+SBW group. From these results, water extracts obtained from Scutellaria baicalensis Georgi may have an immunoregulatory effect in the liver induced by CCl4 of rats.

Both fresh Codonopsis lanceolata and lactic acid bacteria fermented Codonopsis lanceolata were extracted with water at 100℃, and tested for anticancer activity using several human cancer cell lines. The fermented extracts inhibited the growth of hepatocellular carcinoma cells up to 90%, compared to 75% for fresh Codonopsis lanceolata. The extracts of cytotoxicity on human normal lung cells (HEK293) were as low as 15%. Especially, human hepatocellular carcinoma cell were more efficiently inhibited than other cells. This extract also inhibited α-glucosidase activity up to 60% at 1.0mg/ml. This fermented extracts showed the inhibition potency on tyrosinase by 25% at 1.0mg/ml. From the results, the fermented Codonopsis lanceolata enhanced several biological activities up to 20~30%, compared to those from fresh Codonopsis lanceolata. It implies that fermentation process could be one of useful methods of utilizing low quality Codonopsis lanceolata. Because this process could yield high amounts of biologically active compounds by the help of microbial growth.

The aim of this study was to investigate the effect of plant growth at several different growing periods on antioxidant activities and zeatin and ABA contents of Artemisia iwayomogi. Measurements of antioxidant activities, lipid peroxidation inhibition, and superoxide radical scavenging activity were done using PMS, NBT and lipid auto-oxidation analysis, respectively. The results show that activities of antioxidants from Artemisia iwayomogi had much higher than BHT. DPPH free radical scavenging effect of Artemisia leaf extract was increased from 71.06±4.36% in April to 90.06±4.41% in October. Activities of superoxide radical scavenging and lipid peroxidation inhibition were 33.83±3.45% and 45.60±3.10 in April and then increased to 84.40±4.00% and 75.86±3.50% in October, respectively. An ELISA technique has been developed for the determination of zeatin and ABA in Artemisia leaves. By this method, the content changes of zeatin and ABA from Artemisia during the growth were investigated. The zeatin content in leaf was measured to be 186.86±1.40 pmol/g dry weight in April, however, decreased to 117.93±5.83 pmol in October. The ABA content in leaf increased from 19.00±1.26 nmol in April to 68.20±2.52 nmol in October. Relationship between antioxidant activities and plant hormone contents was indicated that antioxidant activity may depend on decreasing zeatin content or increasing ABA content.

This study was carried out to investigate the effect of Cinnamomum camphora on melanogenesis. The MeOH extract of Cinnamomum camphora inhibited mushroom tyrosinase activity in dose-dependent manner. Moreover, it significantly suppressed the melanin production in melan-a cells at the concentration of 100μ/ml. The MeOH extract was partitioned with ethyl acetate, n-butanol and water. Among them, the BuOH soluble fraction exhibited significant inhibitory effect on mushroom tyrosinase. In addition, the BuOH soluble fraction reduced the melanin production in melan-a cells. But, the BuOH soluble fraction had less inhibition effects on melan-a cell originated tyrosinase. So, it was performed western blotting for melanogenic proteins (tyrosinase, tyrosinase-related protein (TRP-2)) using melan-a cells. The BuOH soluble fraction inhibited the protein expression of tyrosinase at the concentration of 100μ/ml. The results suggested that the BuOH soluble fraction of C. camphora might be a potent inhibitor of melanin biosynthesis in melan-a cells.
