        목적: 본 연구는 근대5종 선수들의 종목에 대한 인식을 알아보는 데 그 목적이 있다. 방법: 연구자는 근대5종 선수 8명을 연구참여자로 섭외하여 인터뷰를 진행하였으며, 풍부한 자료수집을 위해 연구참여자를 A그룹(대학선수)과 B그룹(국가대표)으로 나누어 초점집단 면담을 진행하였다. 인터뷰는 그룹당 1회기씩 실시하였고, A그룹은 80분, B그룹은 100분이 소요되었다. 면담 내용은 휴대 전자기기에 내장된 녹음기를 이용하여 녹취한 뒤 전사하였다. 수집된 인터뷰 원자료를 질적연구 방법인 주제분석 기법을 활용하여 분석하였다. 결과: 연구 자료를 분석한 결과는 5개의 주제와 11개의 요소로 도출되었다. 본 연구의 결과를 종합하면 다음과 같다. 주제로는 타인의 혹평, 근대5종에 대한 불만, 근대5종의 매력, 근대5종과 삶, 타인의 호평으로 나타났다. 이에 따른 요소로 낮은 인지도, 주변의 무시, 긴 경기 시간, 폭 좁은 진로, 한정된 시설, 부족한 홍보, 성취감, 만족감, 삶의 일부, 긍정적인 평가, 주변의 격려가 도출되었다. 결론: 본 연구의 결과를 활용하여 근대5종의 발전을 도모하기 위해서는 근대5종에 대한 적극적인 홍보를 비롯해 인프라 및 저변 확대, 선수들을 위한 진로 지원이 이루어져야 할 것이다.

        Background : Collagen and elastin contribute to form a network under the epidermis and reduce the wrinkles. Collagen and elastin are degraded by collagenase and elastase. Therefore, inhibition of collagenase and elastase activity could be an excellent method for anti-wrinkle. Pinus Koraiensis leaves contain various flavonoid compounds such as quercetin and kaempferol. According to previous reports, kaempferol has increases procollagen synthesis and inhibitory effect on MMP-1 activity. This study was performed to investigate the anti-wrinkle effect of Pinus Koraiensis leaves extract and fractions. Methods and Results : The active components of Pinus Koraiensis leaves extracts and fractions were determined. Total phenol and total flavonoid contents measured using the Folin-Ciocalteu reagent and according with aluminum chloride colormetric method. The kaempferol content analysis using HPLC system. The total phenol, flavonoid and kaempferol contents of ethyl acetate fraction was the highest. The antioxidant activity measured in according with DPPH, ABTS and RP assay. Pinus Koraiensis leaves extract and factions were the highest ethyl acetate fraction in antioxidant activity. The inhibitory activity on collagenase and elastase of Pinus Koraiensis leaves extract and factions were determined. The results of inhibitory activity on enzymes associated with wrinkle formation was the highest of n-butanol fraction, the next higher was 80% ethanol extract. As a result of using MMP-1 content and procollagen type-I C-peptide content assay kit, the anti-wrinkle effect of n-hexane fraction was highest.         Background: Planting vigorous cuttings that quickly develop shoots and roots is essential to the biological and economic success of producing medicinal flowers. The present study aimed to evaluate the effect of storage temperature and duration on seedling capacity in the propagation of Chrysanthemum indicum L. and to investigate the effect of rooting media on the growth of C. indicum L. after cutting.Methods and Results:Returning cuttings to supplemental cold storage (2.0 ± 1.0°C) may extend duration of cutting viability 6 weeks, returning cuttings to supplemental warm storage (25.0 ± 1.0°C) is not recommended. The treatment of the growing media experiments, which were conducted in the 2014 planting seasons, included sawdust, river sand, topsoil + sawdust, topsoil + poultry manure, sawdust + river sand, river sand + poultry manure, topsoil + river sand + poultry manure, topsoil + poultry manure + river sand + sawdust. Result indicated that the topsoil + poultry manure media performed best and supported the highest number of branches (3.47), branch length (26.39), and number of leaves (88.63).Conclusions:The results of the present study suggest that cold storage and the topsoil + poultry manure growth media was superior in supporting the early establishment of C. indicum cutting, this result will have a tremendous influence on propagation of this species.

        Background: In recent years, adjuvants have received increasing attention owing to the development of purified subunit and synthetic vaccines which are poor immunogens and require additional adjuvants to evoke an immune response. Therefore, immunologic adjuvants have been developed and tested. Plant polysaccharides have been recognized as effective biological response modifiers with low toxicity.Methods and Results: In this study, the polysaccharide from the aboveground part of Astragalus membranaceus Bunge containing immunomodulating arabino-3,6-galactan was evaluated for its hemolytic activity and adjuvant potential in the specific cellular and humoral immune responses to ovalbumin. The polysaccharide from the aboveground part of Astragalus membranaceus Bunge was co-immunized with the purified Vi capsular polysaccharide of Salmonella typhi vaccine in mice. The polysaccharide from the aboveground part of Astragalus membranaceus Bunge did not induce any hemolytic activity or side effects at doses up to 500㎍/㎖. The concanavalin A-, lipopolysaccharide-, and ovalbumin-induced splenocyte proliferation and serum ovalbumin-specific IgG, IgG1 and IgG2b antibody titers in immunized mice were significantly enhanced by AMA. Pharmacological data revealed that the polysaccharide from the aboveground part of Astragalus membranaceus Bunge increased antigen-specific antibody levels in immunized mice. The polysaccharide from the aboveground part of Astragalus membranaceus Bunge-adjuvanted purified Vi capsular polysaccharide of Salmonella typhi vaccine improved the proliferation of splenocytes and macrophages as well as stimulated cytokine production.Conclusions: These results suggest that the polysaccharide from the aboveground part of Astragalus membranaceus Bunge-adjuvanted vaccines enhanced humoral and cellular immunity and that the polysaccharide from the aboveground part of Astragalus membranaceus Bunge is a safe and efficacious adjuvant candidate suitable for use in prophylactic and therapeutic vaccines.

        Background : Perilla frutescens L. is valuable as a medicinal plant as well as a natural medicine and functional food. Limonene perilla collected from various places showed 60% limonene compounds. However biological activity of these accession has not been reported before. Therefore, this study was conducted to investigate the biological activity of limonene perilla. Methods and Results : Fractional solvent extracts were obtained by using organic solvents such as n-hexane, chloroform, ethyl acetate, n-BuOH, and aqueous solvent from different parts of limonene perilla extracted initially in 70% EtOH. We investigated the effects of limonene perilla on total phenol and flavonoid contents, FRAP (Ferric Reducing Antioxidant Power), total saponin contents and tyrosinase inhibition activity. Leaves of limonene perilla produced the highest total phenolic contents (29.88 mg·CAE/g), flavonoid (8.39 mg·QE/g) and saponin contents (47.77 mg·GIE/g) than stems and roots of limonene perilla. FRAP of leaves was 823.00±3.58 μM·FeSO4·E/mg. Tyrosinase inhibition activity rate was 40.31% in 70% ethanol extracts from leaves of limonene perilla. Conclusion : This results suggest that leaf of limonene perilla fractions has significant antioxidant activity. Also, limonene perilla could be used as a functional biomaterial in developing cosmetics and functional foods.

        Background : Plants are the rich source of antioxidants, which plays a very important role in maintaining human health. Their antioxidant property protects cells of different organs of human beings against free radicals and free radical mediated diseases. Even though, there is lack of knowledge on the antioxidant effect of lutein present in plants. In the present study, lutein was isolated from the GreenTea leaves (Camellia sinensis) which is used as a dietary source. Methods and Results : The procedure adopted for the isolation and purification of lutein using acetone extraction and preparative high performance liquid chromatography (HPLC) is simple and less time consuming. Free radicals scavenging activity of isolated lutein from acetone extract of GreenTea was assessed by DPPH radical scavenging assay and reducing power. The isolated lutein scavenged 79% of DPPH radicals at 20 ㎍/㎖ and two fold lower concentration compared to the standard antioxidants (α-tocopherol). No significant differences were found between the reducing power of the lutein and BHT when their concentrations were high. However, significant differences were observed at relatively low concentrations, the reducing power of lutein was isolated from the GreenTea leaves was stronger than those of their acetone extract and standard antioxidants (BHA). Both electron spin resonance (ESR) and in vitro assay confirmed that lutein was isolated from the GreenTea leaves, exhibited a greater capacity for scavenging superoxide (O2 •-) and hydroxyl (OH •) radicals than standard antioxidants β-carotene and α-tocopherol respectively. Conclusion : The results proven that lutein isolated from GreenTea leaves has an efficient antioxidant ability, it could serve as an antioxidant to scavenge reactive oxygen species.

        Background : Lutein, a xanthophyll, consists of chains with 8 conjugated double bounds containing closed rings on each end of the chain. This carotenoid is found in fruits and vegetables, especially dark green leafy vegetables such as green tea. In this study, we investigated the anticancer effects of purified lutein from green tea on human cancer cell lines containing prostate carcinoma cancer cells (LNCaP). Methods and Results : Prostate carcinoma cancer cells (LNCaP) were cultured and evaluated the inhibitory effect of lutein isolated from green tea compared other carotenoids (β-carotene and lycopene) on cell proliferation. Cyclin D1 and PCNA were evaluated as cell differentiation. In results, PCNA/cyclin regulates the initiation of cell proliferation by mediating DNA polymerase. Under cultural conditions, lycopene remarkably suppressed the PCNA expression prostate cancer cell line LNCaP in higher doses (20 μM - 100 μM) statistically. However, β-carotene and lutein presented the less inhibitory effects on PCNA expression. Determination of PCNA expression in control and treated cells demonstrates that lycopene did affect proliferation in LNCaP cancer cells in dose-dependent manner. However, β-carotene and lutein suppressed the cyclin D1 expression in dose-dependent manner but no in lycopene group. These results indicate that differ carotenoids presented the various suppressive ability of PCNA and cyclin D1 expression in cell proliferation. Conclusion : In conclusion, lutein suppressed the carcinogenesis of induced prostate cancer cell line by acting as a suppressor for inhibiting the expression of cyclin D.

        Background : The objective of this study was to investigate antioxidant activities, inhibitory activities against heme induced colonic epithelial cell proliferations, anti-inflammatory activities and anthocyanin profiles in the anthocyanin rich fraction (ARFAM) from fruits of Aronia melanocarpa, where these are considered functional substances and available food coloring agents in Korea. Methods and Results : Anthocyanins were identified by reversed-phase C18 column chromatography and HPLC-DAD-ESI/MS analysis. To compare the antioxidative and anti-inflammatory capacity of Aronia melanocarpa berries, recognized for their high content of anthocyanins, isolation method was developed to obtain high-purity anthocyanins in the extract. Anthocyanin-rich fractions (ARFAM) enriched in anthocyanins were found to be potent strong inhibitory activity towards heme induced colonic epithelial cell proliferations are associated with an increased risk of colon cancer than acidic ethanol extract (AME). The immunomodulation properties were assessed in growth of both human B and T cells, its cytokines secretion such as IL-6 (interleukin-6) and TNF-α (tumor necrosis factor-alpha). AME enhanced interleukin-6 and reduced tumor necrosis factor-a production, whereas ARFAM only had a effect in increasing of IL-6 expression. Conclusion : These results demonstrated that there was no major relationship between the antioxidative and immunomodulation capacities of AME and ARFAM.

        Bacillus amyloliquefaciens CGD3 was used as a starter for fermentation of amaranth and cultural characteristics and biological activities of amaranth were investigated. The viable cells in fermented amaranth was 4.54~8.01 log CFU/mL during 96 hr of fermentation period. Protease activities of amaranth showed the highest by 36.7 unit/mL after 72 hr of fermentation. The total phenolic and flavonoid contents of amaranth increased with fermentation time while its total sugar content decreased. The total protein content was the highest in amaranth fermented for 96 hr (0.25 mg/g). The DPPH scavenging activities, FRAP (ferric reducing antioxidant power), and reducing power of amaranth were highest in amaranth fermented for 72 hr, showing 84.46%, 551.91 μM, and 2.74, respectively. ACE (angiotensin converting enzyme) and α-Glucosidase inhibition rates increased with fermentation time and showed the highest after 72 hr.

        본 연구에서는 새로운 프로바이오틱스를 선발하고자 protease를 생산하는 GRAS균주를 된장 및 청국장으로부터 분리하고 그 특성을 조사하였다. 분리한 균주를 분리 동정 한 결과 각각B. amyloliquefaciens CDD5, B. amyloliquefaciens CPD4 및B. amyloliquefaciens CGD3로 명명하였다. 분리균 주의 생육은 12시간 배양 시, 7.13~7.32 log CFU/mL로 최대 생육도를 보였으며 24시간까지 유지되다가 감소하는 경향 을 나타내었고, 그 중B. amyloliquefaciens CGD3의 protease 활성이9.21 U/mL로서가장높게나타났다. B. amyloliquefaciens 이 생산하는 protease 활성은 pH 7.0~10.0, 온도는 50℃에서 최적활성을 나타내었으며, casein에서 protease활성이 가장 우수하게 나타났다. pH, NaCl, glucose 농도를 달리한 배양 조건에 따른 B. amyloliquefaciens의 생육특성으로는 pH 5.0~10.0의 넓은 범위에서 생육하였고, NaCl은 12% 농도까 지 증식하였으며 glucose 5%까지는 생육하나 10% 농도부 터는 현저하게 생육이 낮아짐을 확인하였다. 또한 혈당저 하를 유도하는 α-glucosidase의 저해활성을 측정한 결과, B. amyloliquefaciens CPD4 및B. amyloliquefaciens CGD3에 서는 각각 96.65% 및 97.85%의 저해율을 나타내었다. 이러 한 결과는 특히 protease활성이 가장 우수하고 α-glucosidase 또한 높은 저해활성을 가진 B. amyloliquefaciens CGD3 균 주의 생육특성을 고려하여 단백질 분해 분야의 프로바이오 틱 적용 및 당뇨예방용 기능성식품 소재로의 가능성이 기대 된다.

        본 연구에서는 식품산업에 적용 가능한 amylase의 발굴을 위하여 된장으로부터 amylase를 생산하는 균주를 분리하였고, 분리된 미생물들의 동정을 통하여 GRAS 미생물로 부터 생산되는 amylase에 대한 효소 특성을 규명하였다. 그 결과 균주 유래 16S ribosomal RNA 유전자 염기서열에서B. subtilis로 동정되어 본 균주를B. subtilis CBD2로 명명하였다. B. subtilis CBD2 균주의 생육에 미치는 배지 성분에 대한 특성을 검토한 결과 본 균주는 pH 7.0, NaCl 3%, glucose 10%에서 가장 높은 균 생육 특성을 나타내었다. 그리고 본 균주의 amylase 생산에 미치는 탄소원의 영향을 검토한 결과, 본 균주는 soluble starch, corn starch, maize amylopectin 및 wheat starch 중에서 soluble starch를 탄소원으로 사용하였을 때 가장 높은 amylase 생산성을 나타내었다. B. subtilis CBD2유래 amylase의 활성에 미치는 반응 조건을 검토한 결과 본 균주 유래 amylase는 pH 8.0 그리고 50°C에서 가장 높은 효소 활성을 나타내었으며 기질특이성에 대한 검토에서는 corn starch>wheat starch>soluble starch>maize amylopectin 순으로 효소 활성을 나타내었다. B. subtilis CBD2가 생성하는 amylase 효소와 corn starch 기질 반응에 대한 전분 분해능 및 당 생성율 변화에서는 24시간에서 가장 높은 전분 분해능 및 당 생성율을 나타내어 corn starch를 이용한 효소 반응은 24시간이 가장 적절함을 확인할 수 있었다. 따라서 본 연구에서 분리된 균주는 GRAS 미생물인B. subtilis이었으며 본 균주 유래의 amylase 특성을 활용한다면 향 후 식품산업에 있어서 전분 분해 관련 분야에 유용하게 이용될 것으로 기대되어진다.
