The antioxidant activity, total phenol content, and inhibitory effect of xanthine oxidase (XOase) for six different medicinal plant complexes such as Achyranthes japonica, Acanthopanax sessiliflorus, Carthamus tinctorius, Eucommia ulmides, Viscum album, and Caragana koreana were investigated in this study. The free radical scavenging activity values using DPPH(2,2'-diphenyl-1-picrylhydrazyl) and ABTS[2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical cation] analyses were found to be 532.03±86.60 μg/mL and 1376.50±35.01 μg/mL, respectively, in 95% of ethanol extracts among six medicinal plant mixed compositions (6MPMC). The total phenolic content in 70% ethanol extract of 6MPMC was 125.19±1.34 mg GAE/g. The inhibitory activity against XOase was highest at 33.20±0.17% in the 70% ethanol extract of 6MPMC. In the antimicrobial activity test of 6MPMC, the minimum inhibitory concentration (MIC) value showed activities against V. litoralis and E. coli in 70% and 95% ethanol extracts, and these two microorganisms were created in clear zones by method of paper disc diffusion. These results suggest that the 6MPMC, composed of Achyranthes japonica, Acanthopanax sessiliflorus, Carthamus tinctorius, Eucommia ulmides, Viscum album, and Caragana koreana, have both antioxidant and antimicrobial activities and, therefore, may serve as a functional health food product.
This paper concerned with design of constant stress accelerated lift test(ALT). Becase a major factor to affect a life of automotive V-ribbed belts is a temperature, we selected a temperature as a accelerated fator to perform a test. Acclelerated life test plans were developed with the assumption of weibull lifetime distribution. The genreal value of beta parameter was accepted in this life test (β=1.1). The sample number was calculated by alpha parameter which was induced by beta.
Background : Korea's agriculture has been forced to change due to the decline in agricultural population, the aging of the population and the conclusion of the Convention on Biological Diversity. Thus, agriculture is seeking to develop into a sixth industry. For this reason, it is necessary to find high-function alternative plant resources for health promotion to meet the changes in national needs for agricultural products. The adventitious roots culture of wild mountain ginseng is the raw material of food code that produced by the biotechnology technique. introduced in the culture of the plant, which is the only way to use a raw material of food. Therefore, in order to increase the yield of ginsenoside in adventitious roots culture of wild mountain ginseng, ginsenoside precursor was treated and ginsenoside contents were analyzed.
Methods and Results : In order to investigate effect of precursor treatment on the production of ginsenoside in adventitious roots culture of wild mountain ginseng, 5 g of adventitious roots culture of mountain ginseng were cultured in liquid SH media supplemented with 2 ㎎/l IBA, 3% sucrose for 8 weeks, which was co-cultured with β-sitosterol and Squalene (0.0625 to 1 mM) or without. Determination of 17 ginsenosides contents of each extract was carried out by HPLC. Rb3 was accumulated by only β-sitosterol, also it increased production of total ginsenoside in the cultured ginseng roots at a concentration of 0.125 mM, which was 2.47-fold higher than that in the control (78.13 ㎎/g of extract).
Conclusion : These results are an important to improve the production yield of ginsenoside in adventitious roots culture of wild mountain ginseng, and they provide an opportunity for development of adventitious roots culture of wild mountain ginseng in dietary health supplement and pharmaceutical industries.
Background : This study was carried out to investigate the antioxidative activity and active ingredients of Glehenia littoralis through purification process.
Methods and Results : Above-ground and below-ground parts of Glehenia littoralis, dried in Gangneung, were purchased, crushed, sonicated for 2 hours in 100% ethanol, filtered and concentrated. Above-ground part of G.littoralis which is more effective higher antioxidant effect than below-ground part in 2,2-diphenyl-1-picrylhydrazyl (DPPH) assays. Was used to extract the above-ground part of the solvent fraction according to polarity (hexane, ethyl acetate, water) and HP20 column separation (0, 30, 60, 100% EtOH). Using the fraction was the DPPH assay with high-performance liquid chromatography (HPLC) analysis. Comparing the antioxidant efficacy with that of fraction isolated from Glehenia littoralis extract. Hexane, water layer and 100% EtOH fraction showed lower efficacy than Glehenia littoralis extract. 60% EtOH fraction showed more than 8 times higher efficacy. In order to compare the components according to their efficacy, HPLC analysis was carried out. The fraction (hexane, water layer, 100% EtOH fraction) which showed low antioxidative activity confirmed imperatoin and nonpolar compound, the fraction (ethyl acetate, 60% EtOH fraction) showed a higher antioxidant activity was confirmed 2 flavonoid and scopoletin.
Conclusion : Glehenia littoralis extract showed low antioxidant activity of 893 ㎍/㎖ with IC50 of DPPH assay. However, it showed an increase of antioxidant activity by IC50 of 115㎍ /㎖ of DPPH assay of 60% EtOH fraction by fractionation and separation. Through HPLC analysis, the active ingredient, scopoletin and two flavonoids were identified.
Background : Collagen and elastin contribute to form a network under the epidermis and reduce the wrinkles. Collagen and elastin are degraded by collagenase and elastase. Therefore, inhibition of collagenase and elastase activity could be an excellent method for anti-wrinkle. Pinus Koraiensis leaves contain various flavonoid compounds such as quercetin and kaempferol. According to previous reports, kaempferol has increases procollagen synthesis and inhibitory effect on MMP-1 activity. This study was performed to investigate the anti-wrinkle effect of Pinus Koraiensis leaves extract and fractions.
Methods and Results : The active components of Pinus Koraiensis leaves extracts and fractions were determined. Total phenol and total flavonoid contents measured using the Folin-Ciocalteu reagent and according with aluminum chloride colormetric method. The kaempferol content analysis using HPLC system. The total phenol, flavonoid and kaempferol contents of ethyl acetate fraction was the highest. The antioxidant activity measured in according with DPPH, ABTS and RP assay. Pinus Koraiensis leaves extract and factions were the highest ethyl acetate fraction in antioxidant activity. The inhibitory activity on collagenase and elastase of Pinus Koraiensis leaves extract and factions were determined. The results of inhibitory activity on enzymes associated with wrinkle formation was the highest of n-butanol fraction, the next higher was 80% ethanol extract. As a result of using MMP-1 content and procollagen type-I C-peptide content assay kit, the anti-wrinkle effect of n-hexane fraction was highest.
Conclusion : These results indicate that Pinus Koraiensis extract may help anti-wrinkl
