        In vitro digestibility and protein digestibility corrected amino acid scores (PDCAAS) were investigated to verify the availability of protein in various Rhizopus oligosporus fermented products of domestic soybean (Glycine max L.) cultivars. Danbaegkong (DBK), Daepung (DP), Daewonkong (DWK), Saedanbaek (SDB), Seonyu (SY), and Cheongja4ho (CJ4) were used as raw samples, which were fermented using commercially available Rhizopus oligosporus for 48 h. All cultivars showed increased crude protein content after fermentation. The crude protein content of DBK and SDB was significantly higher than that of the other samples (55.12% in DBK and 54.22% in SDB) (p<0.001). CJ4 had the highest alanine content of 28.88 mg/g (p<0.001), and no significant difference in cysteine content was detected among the cultivars. In most of the fermented samples, the in vitro digestibility was 0.9 or higher, indicating high protein in the fermented samples. However, it is considered that restrictions on digestion are low. In DWK, the amino acid content and PDCAAS, which together indicate protein quality, were 0.917 and 0.855, respectively, confirming that it was the best cultivar to provide the raw material for fermentation. In conclusion Rhizopus oligosporus fermented soybean products can be considered a prospective source of protein with high utility value.

        The purpose of this study was to investigate the Maillard reaction–related physicochemical properties of three maize varieties (Kwangpyeongok, Sinhwangok2ho and Gangdaok) after roasting them for different times (0, 15, 25, 40, and 55 minutes). The Maillard reaction is a non-enzymatic browning reaction involving reducing sugars and amino compounds. The content of reducing sugar, the causative agent of the Maillard reaction, decreased as roasting time increased. Gangdaok showed the lowest reducing sugar content of 1.04 mg/g after 55 minutes of roasting. In the elapsed roasting time, chromaticity ‘L’ and ‘b’ values decreased. At 55 minutes of roasting, wherein the Maillard reaction occurred most actively, Gangdaok showed the lowest ‘L’ value of 56.37 and the highest ‘a’ value of 7.60. Gangdaok had superior conditions for inducing the Maillard reaction compared to other varieties, and it is consider that 'flint–type', an endosperm characteristic, may have been the influencing agent. This study detected a total of 52 types of volatile aroma compounds (VACs), of which 28 were produced after roasting. Of the total VACs detected, 2-Formyl-5-methylfuran and 2-Furancarboxaldehyde accounted for 43.8~45.5% and have been confirmed to be the major VACs present in roasted maize. Most of the correlations between the Maillard reaction–related characteristics showed high correlation coefficients.

        The effect of 16 cultivars on the quality of the rice porridge was investigated. The ‘Geunnun’ had the highest water absorption rate, but the ‘Segyejinmi’ yield (w/w) was the highest. The total sugar content of the rice porridge was 0.29~8.10%, showing significant variation among the cultivars. High amylose ‘Dodamssal’ and ‘Hwaseonchalbyeo’ glutinous rice displayed rotational viscosities of <20,000 cP. Rotational viscosities for boiled rice cultivars were 30,000~40,000 cP, representing an intermediate level, and the rotational viscosities of ‘Geonyang2’ and ‘Hanareum4’ were over 50,000 cP. These results suggest that the viscosity of rice porridge varies significantly among raw material cultivars. Among other variables affecting the texture profile of rice porridge, there were significant differences in hardness and gumminess among the cultivars. As a raw material, ‘Baekokchal’, a kind of glutinous rice, is known to be whiter than the non-glutinous rice, but after processing to porridge, it showed the lowest L value (71.1). Starch degrading enzyme activity was not significant in most types of rice porridges within 30 or 60 minutes. Therefore, enzymatic starch degradation is thought to be completed within 30 minutes. Among the tested raw materials, ‘Miho’ was 73.5 μg/mg, indicating the best digestibility in vitro.

        This study was conducted to secure basic information for corn processing by comparing the quality characteristics according to maize cultivars and kernel types (dent, intermediate, flint-like). As a result of analyzing 15 cultivars, a range of measurements were observed: 100-kernel weight, 22.89~35.63 g; moisture, 7.57~8.42%; crude protein, 8.46~11.45%; crude lipids, 3.26~4.83%; Hunter’s L-value, 83.70~86.79; a-value, 2.61~5.49; b-value, 22.01~28.15; and total carotenoids, 6.74~17.07 μg/g. Significance among the cultivars was shown in all quality characteristics (p<0.001), but the significance among the kernel types was found only in crude protein (p<0.005), crude fat (p<0.001), and Hunter’s L-value (p<0.05). The hardness of maize was decreased proportionally to the soaking time for all maize cultivars (p<0.001). In particular, with the same soaking time for different kernel types, the hardness difference was shown in the order of flint-like > dent ≒ intermediate. It was confirmed that the decrease in the hardness of flint-like kernel of close to that of hard-type starch was slowed compare dent and intermediate kernels. So it is expected the some characteristic of kernel types will contribute to the appropriate customized use of the developed cultivars.

        This study was performed to select the high-yield soybean cultivar suitable for the mid part of Korea and to provide as a basic data for improving productivity of the farmhouse. To this, Daewon as a control and four soybean cultivars such as Uram, Taeseon, Saegeum and Seonpung soybean seeds known that high-yield varieties developed after 2010 were selected. Growth characteristics and functional isoflavone contents of five soybeans were field tested at the field in Goesan, Chungbuk during 2014 and 2015. In comparison with Daewon, maturity date was recorded the earliest at Taesun with 15th Oct. Height of the first pod for the mechanical harvesting have a benefit to Uram (16.8 cm) and Taeseon (14.6 cm), however; shattering percentage of Taeseon were showed the high value of 11.2%. Productivity of Uram, Seonpung and Taesun were increased with 20, 9, and 5 %, respectively, compare to that of Daewon. Total isoflavones contents (μg/g) of five soybeans were ordered as Saegeum (1222.2) > Uram (1111.9) > Seonpung (897.2) > Daewon (885.8) > Taesun (829.5). Total isoflavones contents when soil mulching compare to the non-mulching were increased 206.6 μg/g at Daewon and 460.8 μg/g at Uram soybean.

        고구마 칩의 최적 제조 조건 확립과 적합품종을 선정하기 위하여 신자미, 주황미 및 하얀미를 시료로 세절 두께(1~3㎜), 침지 당액 농도(15~30%), 데치기 시간(15~60 sec), 굽기온도(110~140℃) 및 굽기 시간(23~31 min)을 달리하여 칩을 제조하고, 각 조건별 관능평가와 파괴강도를 측정하였다. 품종 별 최적 제조 조건은 신자미는 각각 1 ㎜, 20%, 45 sec, 120℃ 및 31 min이었으며, 주황미는 1 ㎜, 25%, 45 sec, 130℃ 및 29min이었고, 하얀미는 1 ㎜, 25%, 30 sec, 130℃ 및 27 min이었다. 칩 제조 후 유리당 함량은 증가하였으며, maltose는 원료에서 검출되지 않았으나, 칩에서 3.85~13.50% 범위에서 증가하였고, sucrose 함량은 10.31~20.67% 범위에서 25.24~34.06% 범위로 칩 제조 시 증가하였다. 칩의 관능특성에 영향을 미치는 요인은 fructose, maltose, sucrose 함량, b-value 및 failurestress로 나타났다. 이러한 결과는 칩의 제조 시 적합한 품종의 선택과 품종에 따른 최적 제조 조건의 설정이 필요할 것으로 판단되며, 주황미가 칩의 제조에 가장 적합한 것으로 판단되었다.

        한국의 농업유전자원센터로부터 분양받은 40점과 국내에서 수집되어진 28점을 포함한 총 68점의 아주까리 유전자원에 대한 종실특성과 지방함량 및 지방산 조성을 구명하고, 그 상호관계를 파악하여 육종 및 재배기술의 기초자료로 이용고자 수행하였으며 요약한 결과는 다음과 같다. 1. 종실 100립중의 평균은 26.6 g이었으며, 그 범위는 최소 20.1 g부터 최대 37.1 g이었다. 종실 1 l중의 평균은 481.1 g이었으며, 범위는 407.2~531 g이었다. 2. 종실길이의 평균은 12.3 mm이었으며, 범위는 10.1~14.9mm이었고, 종실넓이는 평균 5.7 mm이었으며, 범위는 4.8~6.4mm이었다. 3. 유전자원의 지방함량의 평균은 47.8%이었으며, 그 범위는 최소 41.6%에서 최대 57.2%이었고, 지방함량이 50%이상인 자원은 6종으로서 IT032142, IT104010, IT104993, IT162783, CCB056 및 CCB057이었다. 4. 포화지방산인 palmitic acid(16:0)와 stearic acid(18:0)의 함량의 범위는 각각 0.9~1.8%와 1.0~2.1%이었다. 불포화지방산인 oleic acid(18:1)의 함량의 범위는 3.6~6.5%이었고, linoleic(18:2) 및 linolenic acid(18:3) 함량의 범위는 각각 4.7~7.3% 및 0.5~1.2%이었다. Ricinoleic acid(18:1-OH) 함량의 평균은 86.2%로 아주까리의 지방산에 있어서 대부분을 차지하였고, 범위는 82.5~88.7%이었다. 5. 종실의 지방함량, palmitic, stearic, oleic, linoleic 및 linolenic acid는 ricinoleic acid 및 불포화지방산과는 부의상관, 포화지방산과는 정의 상관이었다. ricinoleic acid는 포화지방산과는 부의 상관, 불포화지방산과는 정의 상관이었고, 포화지방산과 불포화지방산과는 부의 상관이었다.

        This study was conducted to establish the type and method of fertilization for no-tillage during the third year of No-tillage (NT) and Conventional-tillage (CT) practices, towards different kinds of fertilizers. In this experiment, the livestock manure showed higher in response to fertilizer effects of no-tillage. Comparing growth characteristics and yield in NT and CT. Regarding yield, there is no significant between livestock fertilizer and chemical fertilizer, but between livestock fertilizer and chemical fertilizer in conventional fertilization has significant differences. Based on the result, livestock fertilizer is effective way on the quantity of the crop. Nitrogen absorption of plant in livestock of no-tillage is more effective than conventional fertilization. In case of the phosphorus absorption and potassium absorption of plant, fertilizer effect has no significant. Nitrogen is highly absorbed in livestock fertilization of NT. Absorption of phosphorus and potassium are similar.

        The present study was conducted to establish the optimal fertilization type and method for no-tillage during the first year of No-tillage (NT) and Conventional-tillage (CT) practices for soybean, using different types of fertilizers. In this experiment, the culm length and stalk diameter showed a greater response to fertilization with surface irrigation than to conventional fertilization. The fastest flowering period (July 28) occurred using chemical fertilization applied via subsurface irrigation. Comparing maturation based on growth characteristics and flowering date revealed that fertilization with subsurface irrigation was more effective for the growth of crops than other methods. Regarding yield, there was no significant difference between livestock and chemical fertilizers in subsurface irrigation, but there were significant differences between these fertilizers when using conventional fertilization methods. Based on the results, livestock fertilizer with subsurface irrigation effectively enhanced crop quantity. Nitrogen absorption of plants using subsurface irrigation was more effective than that using conventional fertilization. Regarding phosphorus absorption of plants, chemical fertilizers showed higher absorption than did livestock fertilizers for both subsurface irrigation and conventional fertilization. Unlike nitrogen, phosphorus was highly absorbed using conventional fertilization. Absorption of phosphorus and potassium were similar but phosphorus was not absorbed using livestock fertilizers applied either using subsurface irrigation or with conventional fertilization.

        Breeding and cultivation techniques are being treated very severely regarding ecological and physiological development in buckwheat. This study was conducted to focus on the diversity occurring in the cultivated and tartary buckwheat and provide an overview of the characteristics and genetic resources activities. Morphological results showed that the height of common buckwheat ranges from 82-90cm, common buckwheat induced by 200Gy ranges from 52-75cm, common buckwheat induced by 300Gy ranges from 43-56cm, common buckwheat induced by 400Gy ranges from 33-60cm whereas the tartary buckwheat hight ranges from 65-87cm, and while it exposed to various radiation (200Gy, 300Gy and 400Gy), the obtained height ranges from 73-92cm, 55-80cm and 60-75cm respectively. However, the stems from the both cultivar are hollow and that’s why, the plant is very prone to lodging. The leaf color of common buckwheat was green, 200Gray, 300Gy 400Gy common buckwheat light green and green, whereas the tartary buckwheat green and bottle-green, 200Gray 300Gy 400Gy tatary buckwheat bottle-green, common buckwheat (control, 200Gy, 300Gy, 400Gy) stem color is light green and pink, flower color is white, tartary buckwheat (control, 200Gy, 300Gy, 400Gy) flower color is light green. The stem color from tartary buckwheat showed (200Gy, 300Gy, 400Gy) light green and light red color. The results revealed that the two buckwheat cultivars showed diversified characteristics.

        This study was carried out to investigate the effect of pre-germination soaking on germination in 90 Korean soybean varieties and identification of protein in seeds of 7 soybean varieties. The results obtained that germination rate of soybean seeds was decreased as amount of soaking water and soaking duration in number of days. Difference in germination rate of soybean seeds was significant at three days soaking with water volume of 90 ml. Water absorption of seeds was rapidly increased during the first 6 hours, followed by slow increase until 24 hours and then decreased 24 to 48 hours after soaking soybean varieties for bean sprout soaked the lowest amount of water, while soybean varieties for cooking with rice showed the lowest seed water content. Dissolved oxygen (DO) in soaking water was rapidly decreased during the first 3 hours after soaking, and then slowly decreased. Soybean varieties for vegetable and early maturity showed the lowest DO during early soaking periods, but showed higher DO after 24 hours than other groups of soybean varieties. Electrical conductivity and Total Dissolved Solid (TDS) were increased as number of soaking days increased. Soybean varieties for vegetable and early maturity showed the highest electrical conductivity and TDS, followed by those for sauce and paste or cooking with rice, while showed the lowest electrical conductivity and TDS, varieties for bean sprout. Among 90 Korean soybean varieties, varieties which showed the highest germination rate were Jangsu-kong for sauce and paste, Sobaegnamul-kong for bean sprout, Seonheuk-kong for cooking with rice, Seunnokkong for vegetable and early maturity. On the ather hand varieties which showed the lowest germination rate were Iksan and Songhak-kong for sauce and paste, Pangsa-kong for bean sprout, Jinyeul-kong for cooking with rice, Sinlok-kong for vegetable and early maturity. Germination rates of soybean seeds were higher when electrical conductivity, TDS and water absorption of seeds were lower. There were negative correlations between electrical conductivity, TDS and water absorption of seeds and germination rate, while there were positive correlations among electrical conductivity, TDS and soybean seed weight.

        Soybean is very useful crop to supply vegetable protein for human. However, cultivation arear of this economically important crop is gradually diminished in upland field. Hence, cultivation area of soybean is increased in paddy field. During the growth duration of soybean, excessive moisture injury is serious problem for sustainable production and supply. We investigated protein expression according to different period of seed soaking and germination after seed soaking. For comparison on expression of protein according to different condition, we performed two-dimensional electrophoresis. After electrophoresis analysis, we selected differentially expressed protein spots according to different condition such as soaking period and germination after soaking to identify protein function by using MALDI-TOF. Results revealed that pattern of expression of protein according to soaking period and germination after soaking were generally not different in major spots. However, degree of expression of protein in some protein spots was increased in accordance with decrease of soaking period. Especially, in Hwangkeum-Kong, Danyeop-Kon, and Pecking, the degree of expression of protein was remarkably increased for 4 days after soaking. But, according to germination after soaking, degree of expression of protein in germinated seeds of all cultivars was higher than un-germinated seeds. In results of MALDI-TOF analysis, specific proteins were identified by different soaking period such as Allergen Gly m Bd 28K, P24 oleosin isoform B. Also, in accordance with germination, degree of protein expression of the related protein, Gibberellin was increased in un-germinated seeds of Iksan-Kong. In ungerminated seeds of Sinpaldal-kong, proteins were identified as down-regulated by soaking such as ATP binding and Inhibitor II', proteinase.

        In the present study, different expression of protein from Taekwang was revealed by 2-DE, and expressions of protein on each week after flowering was investigated. After analysis of expression of protein, MALDI-TOF was executed to identify expected protein function. Results revealed that there were three patterns of expression of protein during the maturing. The first pattern was that proteins were gradually expressed as up-regulation from 1 week to 6 week. The second pattern was that proteins were expressed gradually from 1 week to 5 week and then it started down-regulation in 6 week. The last pattern was that proteins were gradually as up-regulation from 1 week to 3 week and then down-regulation until 6 week. This phenomenon suggests that young stage has more protein related to correspondence mechanism against disease and growth and then maturing stage has more expression of protein related to storage protein. In MALDI-TOF analysis, p24 oleosin isoform A protein was identified that relates oleosin which is synthetic product in oil body. This protein spot increased gradually until 5 week and then decreased after 5 week. It explained that the protein is active until maturing stage to protect oil in seed and then its activity has gradually degraded. This result may be expected that a protein, related to growth of a seed has increased until maturing and then a seed fills up with a storage protein

        Soybean is very useful crop to supply vegetable protein for human. Supply of soybean is increased because it has useful ingredient. Recently, cultivation of soybean in paddy field is increasing due to the increase of rice stockpile in Korea. Hence, in this study, expression of protein was identified regarding different environment for cultivation to investigate the effect of different environment on protein expression. Two-dimensional electrophoresis was performed to investigate the expression of protein using image analysis program to measure degree of protein expression in numerical value. Hannam-kong, Beakcheon-Kong, Hwangkeum-Kong, and Danwon-Kong were used as plant material. 2-DE combined with image analysis revealed that each degree of protein expression of Hannam-Kong and Hwangkeum-Kong in upland field was higher than degree of protein expression in paddy field. However, in case of Beackcheon-Kong, the phenomenon was opposite. In Danwon-kong, the degree of protein expression was not different between up-land field and paddy field. To this end, major protein spots were not different between paddy field and upland field among all cultivars. It could be suggested that protein expression is not severely different by various environment, but different environment affects degree of protein expression.

        Soybean seed is a good source of plant protein in human consumables such as baby formula and protein concentrate. The seeds contain an abundance of storage proteins, namely β-conglycin and glycinin that account for ~ 70-80% of the total seed protein content. Proteome profiling has been proved to be an efficient way that can help us to investigate the seed storage proteins. In the present study, the seeds were removed from the pods and the cotylendonary tissues were separated from the testa for proteome analysis in order to investigate the seed storage proteins. A systematic proteome profiling was conducted through one-dimensional gel electrophoresis followed by MALDI-TOF-TOF mass spectrometry in the seeds (cotyledonary tissue) of soybean genotypes. Two dimensional gels stained with CBB, a total of 10 proteins were identified and analyzed using MASCOT search engine according to the similarity of sequences with previously characterized proteins along with the UniProt database. A total of ten proteins such as glycinin Gy4 precursor, glycinin G3 precursor, glycinin G1 precursor, glycinin chain A2B1a precursor, glycinin chain A2B1a precursor were identified in our investigation. However, the glycinin subunit may be considered to play important roles in soybean breeding and biochemical characterization. In addition, the improved technique will be useful to dissect the genetic control of glycinin expression in soybean.

        Common buckwheat has the sporophytic self-incompatibility mechanism and that’s why it has the ability to cross pollinate between two plants with different styles (thepin type and thrum type). The S supergene is thought to govern self-incompatibility, flower morphology and pollen size in buckwheat. Already, we have produced self-compatible buckwheat lines by an interspecific hybridization between Fagopyrum esculentum and F. homotropicum by embryo culture. The pollen size of F1 plants produced by a cross between a pin type plant and the self-compatible plant was similar to that of the self-compatible lines and segregated together with flower morphology without exception. The pollen tubes of the self-compatible plants were compatible with styles of the pin plants but incompatible with the styles of thrum plants. But, the pollen tubes of thrum flowers were compatible with the styles of self-compatible plants. Also, the pollen tubes of pin flowers were incompatible with the styles of self-compatible plants. Already, from these results, we have reported a tentative genotype for heterostyle and homostyle flower types. Homomorphism was controlled by a single allele Sh, while the pin/thrum-complex gene was governed by a single genetic locus S, with two alleles, S and s, which control Ss (thrum-type) as well as the ss (pin-type), respectively. Corresponding represents the case of a single locus S with three alleles, Sh, S and s, and the phenotypes, homomorphic, pin and thrum. It can be characterized by relationship of dominance, S>Sh>s. Using the two self-fertile lines, one is considered as the long-homostyle flowers and the other is considered as the short homostyle flowers. If the short-homostyle trait had arisen by recombination in the S supergene, its genotype would be considered to be GIs ip a/GIs ip a. The pollen tubes of the short-homostylous plant should be compatible with the styles of thrum plants. Also, the pollen tubes of short-homostylous plants should be incompatible with the style of long-homostylous plants, and the reciprocal cross also should be incompatible, because the genotype of long homostyle is gis Ip PA/gis Ip PA. Furthermore, the flower morphology of F1plants produced by the cross between cross and short homostyle flowers should be thrum or short homostyle and only short-homostylous plants should be produced by the cross between pin and short homostyle flowers. However, the compatibility or incompatibility of short homostyle flower was not clarified. So, we need to clarify the compatibility or incompatibility of the style of short homostyle flowers for the next step.

        콩 관련 제품을 생산하는 과정에서 발생되는 부산물은 가축의 사료 및 퇴비 등에 이용되고 있으나 일부는 폐기물로처리되어 추가 비용 및 각종 환경오염을 유발하는 등 사회적인 문제점으로 지적되고 있다. 따라서 본 연구에서는 가공 부산물로 다량 발생되는 콩 배아를 활용하여 생리활성물질인 isoflavone과 soyasaponin을 동시에 분리하는 방법을개발하고자 실시하였다.1. 콩 배아 methanol 추출물을 preparative C18 column에 주입하고, 210 nm의 파장에서 0.5% 초산 용액 30%로부터100% acetonitrile까지 분당 15mL의 유속으로 53분간흘려주어 isoflavone과 soyasaponin 분획을 동시에 분리하였다.2. Preparative C18 column으로 분리된 isoflavone 및 soyasaponin분획은 동결건조시켜 isoflavone 분말 ISF-1과 soyasaponinSAP-1, SAP-2, SAP-3 및 SAP-4의 분말을 얻었다.3. Isoflavone 분획 ISF-1의 재분리는 젤투과 컬럼에서100% acetonitrile을 분당 5 mL가 되도록 흘려주면서254nm 파장에서 관찰하여 2종의 분획 ISF-1-1 및ISF-1-2을 분리하였다.4. 분리된 2종의 isoflavone 중 ISF-1-1은 그 조성이daidzin, glycitin 및 genistin 이고, ISF-1-2는 genistin 단일물질이 주성분인 것으로 나타났다.5. 분리된 4종의 soyasaponin 중 SP-1은 soyasaponin A계열인 Aa(MW: 1364), Ab(MW: 1436), Ac(MW: 1420),Ae(MW: 1202), Af(MW: 1274), SAP-2는 B계열인 Ba(MW: 958), Bb(MW: 942), Bc(MW: 912)와 E계열인Bd(MW: 956), Be(MW: 940), SAP-3는 B계열인 Ba,Bb, Bc, E계열인 Bd, Be와 DDMP계열인 βg(MW:1068), SAP-4는 B계열인 Ba, Bb, Bc, E계열인 Bd, Be와 DDMP계열인 βg, βa(MW: 1038)가 주성분임을 알 수 있었다.

        This study investigated saponin contents according to collection area, seed size and seed color in 293 Korean soybean landraces. Results showed that total group A was 149.8 to 1279.0㎍/g with an average of 484.9㎍/g while total group B saponin content was 2160.1 to 7868㎍/g with an average of 3670㎍/g. Total groups A and B saponin content were ranging from 2502.7 to 8763.9㎍/g with an average of 4154.8㎍/g. The landraces with high group B saponin content were IT226841, IT226761, IT226844 and IT226826. The landraces with low group A saponin content were IT228580, IT228319, IT228308 and IT226841. According to collection areas, the highest amount of saponin content was observed in the landraces collected from North Korea. High total of saponin content was showed by seeds from Gyeonggi-do, followed by Chungcheongbuk-do, Gangwon-do, Gyongsangbuk-do, Chungcheongnam-do Jeollabuk-do and Jeollanam-do. According to seed size, small seeds have the higest saponin contentfollowed by the medium and large size of seeds. For seed color, there was no significant difference in saponin content. The total of saponin content was postively correlated with group A saponin, group B saponin and total of saponin cntent. Among each components, group A saponin showed postive correlations with total of saponin and group B saponin content.

        We improved the separation of the basic proteins from the soybean cotyledon, Glycine max L. Merr. by searching N-terminal sequences data in proteins isolated by two-dimensional electrophoresis (2-DE). After removed Hexane, proteins were extracted from cotyledon with a urea/Triton/2-mercaptoetanol solution. Using this method, the highly reproducible isoelectric focusing (IEF) can formed with polyacrylamide gels with pH 4.0-9.8. The IEF tube gels were used as the first dimension, and proteins were visualized by second-dimensional gel electrophoresis, and identify a number of soybean cotyledon proteins using mass spectrometry in the proteome analysis. These instruments of 2-DE and IEF tube gels were used 27 cm and investigate under various conditions. The total number of spots and features was obtained by PDQuest software (Bio-Rad). In this experiments performed, the IEF tube gels and instruments afforded good reproducibility in the number of PDQuest-detected spots from gel to gel while IPG offered better reproducibility in the total number of manually detected spots from gel to gel. In conclusion, we have separated of the basic 13 proteins in soybean. The glycinin subunit separations are also considered to play important roles in soybean breeding and biochemical characterization. The improved technique will be useful to dissect the genetic control of glycinin expression in soybean.

        1913년부터 2006년까지 국내에서 육성된 콩 106 품종의 종실 isoflavone 함량의 변이를 구명하였다. 총 isoflavone의 함량은 평균 1489.0 μg/g이었고, 527.9~3436.5 μg/g의 범위이었다. 구성성분의 함량은 daidzein은 평균 650.5 μg/g이었고, 153.0~1710.4 μg/g의 범위이었으며, glycitein은 평균 146.2 μg/g이었고, 32.2~308.2 μg/g의 범위이었다. genistein은 평균 692.3 μg/g이었고, 203.4~1502.4 μg/g의 범위이었다. 총 isoflavone 함량이 높은 품종은 백천(2553.9 μg/g), 보석콩(3436.5 μg/g), 대황콩(2709.2 μg/g), 대망콩2호(2595.0 μg/g), 장기콩(2769.9 μg/g), 풍원콩(2872.8 μg/g), 소진콩(2521.8 μg/g) 및 소록콩(2956.1 μg/g)이었으며, 국내 콩 품종 중에서 보석콩이 가장 높았다. 국내에서 육성된 콩 품종들의 isoflavone 함량은 육성연대, 용도, 종실크기 및 육성모지에 따라서 유의적인 차이가 인정되었다. 육성연도에 따른 총 isoflavone 함량은 2000년대에 육성된 품종이 가장 높았고, 다음으로 1980년대와 1990년대에 육성된 품종이 높았으며, 1980년 이전에 육성된 품종이 가장 낮았다. 용도에 따른 총 isoflavone 함량은 나물용 품종이 가장 높았고, 다음으로 장류용 품종이, 그 다음으로 밥밑용 품종이 높았으며, 풋콩 및 올콩용 품종이 가장 낮았다. 종실크기에 따른 총 isoflavone 함량은 소립종이 가장 높았고, 다음으로 중립종이 높았으며, 대립종이 가장 낮았다. 육성모지에 따른 총 isoflavone 함량은 익산이 가장 높았고, 다음으로 밀양이 높았으며, 수원이 가장 낮았다.
