The purpose of this study is identify chemical changes in yield components of soil using organic fertilizer for cultivating rice and establish reasonable methods of fertilization. In chemical change, comparing the control plot, the soil using organic fertilizer consistently for three years for cultivating rice showed an increase in pH and available phosphate in the experiment plots provided by the organic fertilizer, increase in spikelet number and 1000 grain weight in terms of yield components, and increase in yield in the three-year experiment plots. As for the quality of brown rice, the rate of head rice showed no difference when compared to that of the control plots. As for protein content, it was difficult to identify a statistical significance between the control plots and the experiment plots in 2009, but since 2010, the protein content of the experiment plots has been lower than that of the control plot, indicating that fertilization of organic fertilizer was effective in terms of rice quality. In conclusion, long-term use of organic fertilizer improved the chemical properties of soil and increased yield when compared to practical fertilization. Reasonable fertilization was identified to fertilize the nitrogen component of organic fertilizer to be a level of 12kg/10a.

The purpose of this study was to know the seed longevity of rice (Oryza sativa L.) germplasm for effective viability monitoring. The longevity was determined via germination tests of 3,066 accessions of rice germplasm from the National Agrobiodiversity Center, Rural Development Administration, Korea. The rice germplasm accessions have been conserved at a mid-term storage (4°Csealed metal can on either sides for 25~26.5 years. The final germination percentages of 3,066 rice germplasm accessions of 6.5±1.0% seed moisture content with 94% initial germination stored at 4°C for 26.5 years declined to 47% while at -18°C for 25 years maintained high germinability as 93%. Germination time courses, which represent the average performance of rice ecotypes stored at 4°C and 30% RH, were fitted regression equation, to calculate the time at which germination characteristically declined to 50% (P50). These P50 values of Indica, Japonica, Javanica and Tongil type in rice were 39.9, 22.9, 25.4 and 31.8 years, respectively. The rice germplasm stored at 4°C could be clustered in 4 groups using quartile of final germination after 26.5 years storage. The seed longevity (P50) of each group was estimated by regression equation of changed germination percentages according to storage periods. The P50 values of group Ⅰ, group Ⅱ, group Ⅲ and group Ⅳ were 21.1, 23.6, 30.0 and 75.7 years.

The objective of this study was to evaluate quality properties including amylose, alkali digestion value (ADV), texture and GABA (Gamma Amino-Butyric Acid) of brown rice and 1~5 mm germinated brown rice of Keunnun and Samkwang culitvars. The protein content of germinated brown rice showed increasing tendency according to shoot length, however, the difference was not statistically significant. The amylose content of all samples ranged from 15.0% to 16.0% in Keunnun and 17.0% to 18.0% in Samkwang, without significant differences. Compared to regular brown rice, germinated brown rice from both cultivars demonstrated reduced gel consistency, which contributed to the improved cooked rice texture. Especially, germinated brown rice had significantly lower setback values, which is correlated with high palatability of cooked rice. However, there was no significant difference in gel consistency between 1 mm and 2~5 mm germinated brown rice. GABA content in Keunnun increased by 3~5 times after germination process. In Samkwang, GABA content increased by 50 times (from 0.005 μg/g to 0.243~0.247 μg/g) after germination. The hardness of germinated brown rice was significantly lower than that of brown rice. However, there was no significant difference in hardness according to shoot length. Consequently, when processing germinate brown rice, it is recommended to maximize functional ingredients by germination while maintaining shoot length less than 2 mm.

The purpose of this study was to develop the cost-effective and efficiency seed longevity prediction method of rice (Oryza sativa L.) germplasm for viability monitoring. To find an optimum predicting method for rice seed longevity at genebank, an accelerated ageing (AA) test, a controlled deterioration (CD) test and a dry-heat treatment (DHT) were conducted to the four groups of rice germplasm based on ecotype, such as Indica, Japonica, Javanica and Tongil type. Among the three artificial aging treatments, the dry-heat treatment of 36 hours at 90°C is suggested as a routine predictive test method of rice germplasm longevity at a genebank. The distribution of germination rate on 3,066 accessions which conserved 26.5 years at 4°C showed similar trend with the result of distribution by dry-heat treatment at 90°C on 36 hours using 106 accessions of rice selected samples which composed four ecotype groups. The results show that the dry-heat treatment affect not only predicting the rice seed longevity but also determining effective interval for monitoring germination of rice germplasm in genebanks.

Barnyard grasses are dominant weed that are not eliminated easily in the direct water seeding cultivation. So, deep water treatment can reduce their growth at the early growing stage of submerged paddy. This study was carried out to investigate the reducing growth of the barnyard grasses through flooding at seedling stages of rice plant in the green house. Under the normal condition, the plant height of rice variety, Samgwangbyeo, and 3 species of barnyard grass, E. caudata, E. pratocola and E. utilis were not showed the difference up to 10 days after seeding while the plant height of 3 barnyard grass species, especially E. utilis, was more elongated than the rice at 25 days after seeding. Plant height of the 3 barnyard grasses were not elongated largely during 3, 5 and 7 days of flooding treatment at 10 days after seeding. Interestingly, the rice seedlings was grown over 20 cm, and the flooding tolerance of rice seedling was higher than the 3 barnyard grass varieties. However, after flooding treatment for 3, 5 and 7 days, the elongation of plant height of 3 barnyard grasses, especially E. utilis was more speedy compared to rice seedlings as 6 to 9 days passes. And the protein spots from barnyard grasses were also reduced and eliminated more than the spots of rice seedling after flooding treatment.

Sorghum seed is traditionally used as health supplements and the secondary food mixed with rice in Korea. While the research of reserve protein in sorghum seed have been carried out in many countries used as major food, much less is known about reserve proteins of Korean local sorghum seeds. To obtain protein characteristics in 20 Korean local sorghum seed, quantitative content of reserve protein was determined after fractionation by modified ‘Osbone’ method and α-kafirin of prolamin was determined by SDSPAGE. Mean albumin, globulin, prolamin and glutelin contents based on total seed protein content of 20 Korean local sorghum seed were 6.2%, 0.9%, 57.9% and 35.1%, respectively. Sorghum cultivar with high prolamin were ‘Whin-susu’, ‘Whin-Chalsusu’, ‘Whanggeum-Chalsusu’, and ‘Daepungshushu’. Sorghum cultivar with high α-kafirin were ‘Whin-susu’, ‘Geumsan-Chalsusu’, ‘Whin-Chalsusu’, and ‘Jangmok-susu’. Among the 20 varieties, ‘Whin-susu’ and ‘Whin-Chalsusu’ were selected as high α -kafirin and prolamin sorghum cultivar, which showed 64.5 and 71.9% of prolamin contents, respectively.

Sorghum seed is traditionally used as secondary food sources in addition to rice in Korea. While the hypoglycemia regulating phytochemicals have been found in sorghum seed, peptides related with hypoglycemia never been studied before. To obtain the peptide characteristics and the specifically high-expressed peptides in hypoglycemic sorghum seed, peptide profiles of seven hypoglycemic and five nonhypoglycemic sorghum lines bred in RDA were determined using surface-enhanced laser desorption/ionization time-offlight mass spectrometry (SELDI-TOF MS). The twelve sorghum lines exhibited 104 peptides on CM10 protein chip array (weak cation exchange) and 95 peptides on Q10 (weak cation exchange) in the molecular mass range from 2,000 to 20,000 Da. Heat map via supervised hierarchical clustering of the significantly different peptides (p < 0.01) in peak intensity among the 12 lines effectively revealed the specifically upregulated peptides in each line and distinguished between 7 hypoglycemic and 5 non-hypoglycemic lines. Through the comparison with hypoglycemic and non-hypoglycemic lines, 10 peptides including 2231.6, 2845.4, 2907.9, 3063.5, 3132.6, 3520.8, 4078.8, 5066.2, 5296.5, 5375.5 Da were specifically high-expressed in hypoglycemic lines at p < 0.00001. This study characterized seed peptides of 12 sorghums and found ten peptides highly expressed for hypoglycemic sorghum lines, which could be used as peptide biomarkers for identification of hypoglycemic sorghum.

Transcription factors are essential for the regulation of gene expression in plant. They are binding to either enhancer or promoter region of DNA adjacent to the gene and are related to basal transcription regulation, differential enhancement of transcription, development, response to intercellular signals or environment, and cell cycle control. The mechanism in controlling gene expression of transcription can be understood through the assessment of the complete sequence for the maize genome. It is possible that the maize genome encodes 4,000 or more transcription factors because it has undergone whole duplication in the past. Previously, several transcription factors of maize have been characterized. In this review article, the transcription factors were selected using Pfam database, including many family members in comparison with other family and listed as follows: ABI3 / VP1, AP2/EREBP, ARF, ARID, AS2, Aux/IAA, BES1, bHLH, bZIP, C2C2-CO-like, C2C2-Dof, C2C2-GATA, C2C2- YABBY, C2H2, E2F/DP, FHA, GARP-ARR-B, GeBP, GRAS, HMG, HSF, MADS, MYB, MYB-related, NAC, PHD, and WRKY family. For analyzing motifs, each amino acid sequence has been aligned with ClustalW and the conserved sequence was shown by sequence logo. This review article will contribute to further study of molecular biological analysis and breeding using the transcription factor of maize as a strategy for selecting target gene.

This study conducted experiments on the reclaimed land of Saemangeum located in Jeongrabuk-do in order to gain basic information about growth characteristics and yield ability according to soil salinity. Having soil excluding salt as a control group, this study adjusted the specimens’ soil salinity to level 4 and then planted four varieties including Ilmichal Corn to investigate the growth or grain yield according to salinity. About the corn establishment rate according to soil salinity, over 97% up to 3.2 dS m-1 was established, and then, it was reduced gradually according to the increase of concentration. According to the salt concentration of soil more required growth duration from seeding to heading comparing to non-treatment salt was delayed, at 1.6 dS m-1, two days were delayed, at 3.2 dS m-1, four to six days were delayed differently by varieties, and at 4.8 dS m-1, six to 10 days were delayed. About the plant fresh weight according to soil salinity, Chalok 4 and Eolrukchal indicated 93%~97% or so compared with the salt-free one at 1.6 dS m-1, and Chalok No. 4 showed 79% at the salinity of 3.2 dS m-1, too, and it was a relatively higher growth percentage than those of the other varieties. In terms of dried seed weight according to soil salinity, compared with the corns cultivated in the control group, averagely 12.1% was lowered at the time of cultivation at 1.6 dS m-1, and 3.2 dS m-1 40% was lowered, and about 70% was lowered at 4.8 dS m-1. According to the result of examining the point of time that dried seed start to reduce due to soil salinity with the regression equation, soil salinity which starts the reduction of grain weight is 1.67 dS m-1~2.18 dS m-1, and it differs by varieties, and EC of 50% that the yield reduces in half is 2.96 dS m-1 ~4.45 dS m-1. And the degree of influence on each of the growth factors according to soil salinity is founded to be in the order of establishment rate<plant length=the ear’s fresh weight<the plant fresh weight<yield.

Fresh edible sweet corns demand relatively short period to harvest fresh ears, which can allow farmers to make a choice sweet corns for various cropping systems. For this reason, we were to find the optimum planting date of late-planted sweet corns to sell fresh ears in the autumn linked to cropping system with winter crops, investigating yield and properties of marketable fresh ears and growth traits of sweet corns (cv. 'Godangok' and cv. 'Guseulok') depending on planting dates such as 10 July, 20 July, and 30 July in Suwon 2012 and 2013, respectively. The 20 July-planted sweet corns showed the most fresh ear yield. However, the 10 July-planted and the 30 July-planted had 32% less yield caused by consecutive rainfall from 10 July through 20 July, and 15% less yield due to low air temperature during ripening than the 20 July-planted, respectively. The 10 and 20 July-planted sweet corns had average 140g of a fresh ear weight and 15% heavier ear than the 30 July-planted. For the July-planted sweet corns, silking days after planting (r=-0.80**), and harvesting days after silking (r=-0.97**) and planting (r=-0.91**) were highly negatively correlated with daily mean air temperature during the period, resulting in it takes 1,100 growing degree days (GDD) to harvest fresh ears from the July-planted sweet corns. The fresh ears of the 20 July-planted sweet corns are able to be harvested by early October. Therefore it will be a good choice for the cropping system based on winter vegetable cash crops such as temperate garlic and onion with medium or late maturity. Among three planting dates 20 July-planted sweet corns had the best field performance in every year considering fresh ear yield, ear size, and stability to grow.

We evaluated the morphological characteristics of 156 maize inbred lines, which were developed to breeding normal maize variety at Maize Experiment Station, Gangwon Agricultural Research and Extension Services, by examining 11 quantitative and three qualitative characteristics. On the evaluation of three qualitative traits for 156 maize inbred lines, most inbred lines showed yellow (85 and 84 inbred lines) at tassel color (QL1) and silk color (QL2), and showed semi erect (105 inbred lines) at plant type (QL3). While, the evaluation of 11 quantitative traits for 156 maize inbred lines, they showed the morphological variation in days of tasseling (QN1, 56.5 to 76.0 days), days of silking (QN2, 59.0 to 85.5 days), stem thickness (QN3, 12.7 to 42.9 mm), plant height (QN4, 111.8 to 239.8 cm), ear height (QN5, 48.2 to 126.5 cm), 100 kernel weight (QN6, 14.9 to 36.4 g), ear length (QN7, 10.0 to 79.0 cm), setted kernel length (QN8, 8.0 to 70.5 cm), ear thickness (QN9, 4.0 to 22.0 cm), total kernel weight (QN10, 22.0 to 490.0 kg) and water content (QN11, 9.3 to 11.9%), respectively. As a result, 11 inbred lines (00hf3, 00hf19, 00hf30, 00hf36, 02S8069, 02S8072, 02S8090, 02S8099, 05S10011, 06S8085-6, 07S8011) in the 156 normal maize inbred lines have showed comparatively high values. While, the results of PCA (principal component analysis) indicated that the ear length (QN7), setted kernel length (QN8), ear thickness (QN9) and total kernel weight (QN10) greatly contributed in positive direction on the first principal components. And also, days of tasseling (QN1), days of silking (QN2), plant height (QN4) and ear height (QN5) contributed in negative direction on the second principal component. Thus these morphological characters, which were greatly contributed in the first and second principal components, might be considered to be useful for discrimination among 156 normal inbred lines. Specifically, this study's assessment of morphological characteristics of 156 normal inbred lines will be helpful useful for normal maize breeding programs such activities as planning crosses for hybrid and line development at Maize Experiment Station, Gangwon Agricultural Research and Extension Services.

Soybean (Glycine max (L.) Merr.) is an important crop for protein, oil, carbohydrates, isoflavones, and many other nutrients to humans and animals. But, antinutritional factors in the raw mature soybean are exist. Raffinose and stachyose are main antinutritional factors in soybean seed. Both raffinose and stachyose are carbohydrates, belonging to the raffinose family of oligosaccharides (RFOs). RFOs are not readily digested in humans and cause flatulence or diarrhea. The objective of this research is to obtain the information on raffinose and stachyose content according to genotype and environment. A total of twenty two soybean genotypes (11 cultivars, 3 germplasms and 8 breeding lines) were selected. Each genotype was grown in the field for two years with two replications and harvested in bulk at natural maturity for two years. Content of raffinose and stachyose was detected by HPLC. The raffinose content (g/kg) of 22 genotypes was 2.68±0.21 - 5.87±2.43 in year 1 and was 3.24±0.37 - 9.05±0.16 in year 2. The stachyose content (g/kg) was 4.23±0.98 - 27.68±9.90 at year 1 and was 5.11±1.09 - 25.32±0.35 in year 2. Genotype and environment have highly significant effects on raffinose and stachyose content. Three genotypes (Da-7, 116-13, and RS-78) have low stachyose content at 5% significant level in two years. A positive correlation (R2=0.1985*) between raffinose and stachyose was observed in year 2. These informations are valuable in soybean genetics and breeding program related with raffinose and stachyose content.

The purpose of this study was to establish the proper sowing time and planting density of cowpeas for labor-saving cultivation. Experiments were carried out in Naju, Jeonnam Province (Latitude 35° 04' N, Longitude 126° 54' E) during 2012 and 2013. The intermediate-erect type strains used in this study were Jeonnam1 and Jeonnam2. Sowing was performed five times between June 25 and August 5 at approximately 10-day intervals in order to establish proper sowing time, and sowed at 5,000, 10,000, 15,000, and 20,000 plants per 10a to establish proper planting density. The days from sowing to first flowering was shortest (32 days) in plants sowed on July 25 and became longer for plants sowed on or around July 25. The days from sowing to first flowering was longest (41 days) in plants sowed on June 25. The days from first flowering to first maturing was shortest (8 days) in plants sowed on June 25 and, became considerably longer at later sowing dates. The days from first maturing to first harvesting ranged from 8 to 10 days, with little difference among the sowing periods. Plants sowed on August 5 harvested at the same time, and plants sowed between June 25 and July 25 were harvested either three or two times. The yield was highest in plants sowed on July 25: 209 kg/10a was harvested for Jeonnam1 and 221 kg/10a for Jeonnam2. Furthermore, harvested at the same time enabled when the harvesting was delayed for around 15 days because the share of the seeds first harvested was highest (91%). The proper planting density was estimated to be 15,000 plants/10a, showing the highest yields of 199 kg/10a for Jeonnam1 and 224 kg/10a for Jeonnam2.

The purpose of this study was to establish the optimal growth temperature and to select genetic resources for production of cowpea sprouts. Seowon was treated between 15°C and 30°C at intervals of 3°C to investigate growth temperature. Twelve resources, including Seowon, IT154149, IT154153, Tvu7426, and Tvu7778, were used for cultivating sprouts at a temperature of 27°C. The yield ratio of cowpea sprouts was highest at 27°C (657%), and was reduced when growth temperature was decreased. The hard seed rate was lower when the growth temperature was increased. Vitamin C content was highest at 24°C (2.85 mg/g), ranged between 2.15 and 2.29 mg/g at other growth temperatures, and increased with the length of the growth period. The inorganic component content of cowpea sprouts did not vary based on growth temperature, while the amino acid content increased with increasing growth temperature between 15°C and 24°C, and then subsequently decreased as growth temperature rose from 24°C to 30°C. IT154153 had the highest yield ratio of cowpea sprouts per genetic resource (647%), followed by Seowon (615%), and Tvu7426 (608%). Genetic resources with a higher yield ratio had smaller seeds, a thinner seed coat, and superior germinability. The inorganic components found at highest concentrations in the cowpea sprouts were potassium, magnesium, calcium, sodium, iron, molybdenum, and zinc (in that order). In comparison to raw seeds, the protein, calcium, zinc, molybdenum, and iron content in the cowpea sprouts was higher, while the content of aluminum and boron was lower.

Field emergence of Azuki bean is poor due to hard seed coat as compared to other legumes. In this study, an attempt was made to develop prediction method with regression analysis based on various seed vigor tests in laboratory for field emergence of azuki bean. Azuki bean seeds artificially aged to provide various levels of seed quality were evaluated by the standard germination test (SGT), cold germination test (CT), cool germination test (CGT), complex stressing vigor test (CSVT), tetrazolium(TZ) vigor test and electroconductivity test. The SGT was suitable for predicting the field emergence in the unaged high vigor seeds. The abnormal seedling percentage and shoot length in the CGT were highly correlated with field emergence of moderate vigor seeds artificially aged for 2 days. Electroconductivity, seed viability in the CSVT, and vigor and predicted germinability in the tetrazolium vigor test were also useful for predicting field emergence. Percent of ungerminated seed in the CSVT was correlated with field emergence in the low vigor seeds artificially aged for 4 days. In a stepwise multiple regression analysis, seed viability in the SGT, normal seedling percentage and dry matter weight in the CGT accounted for 86.9% of the predicted value of field emergence in azuki bean.

Soybean seed is a good source of plant protein in human consumables such as baby formula and protein concentrate. The seeds contain an abundance of storage proteins, namely β-conglycin and glycinin that account for ~ 70-80% of the total seed protein content. Proteome profiling has been proved to be an efficient way that can help us to investigate the seed storage proteins. In the present study, the seeds were removed from the pods and the cotylendonary tissues were separated from the testa for proteome analysis in order to investigate the seed storage proteins. A systematic proteome profiling was conducted through one-dimensional gel electrophoresis followed by MALDI-TOF-TOF mass spectrometry in the seeds (cotyledonary tissue) of soybean genotypes. Two dimensional gels stained with CBB, a total of 10 proteins were identified and analyzed using MASCOT search engine according to the similarity of sequences with previously characterized proteins along with the UniProt database. A total of ten proteins such as glycinin Gy4 precursor, glycinin G3 precursor, glycinin G1 precursor, glycinin chain A2B1a precursor, glycinin chain A2B1a precursor were identified in our investigation. However, the glycinin subunit may be considered to play important roles in soybean breeding and biochemical characterization. In addition, the improved technique will be useful to dissect the genetic control of glycinin expression in soybean.
