This study was carried out to investigate the effect of stem-cutting (SC) length and portion on the major agronomic characters related with dry matter production and accumulation, and to examine their relation­ships in cassava. When cassava was planted with longer SCs or with older portion SCs, major canopy or source characters like leaf number and leaf area index developed excessively, while tuber yields could be reduced due to the decrease of root/shoot ratio, relative growth rate, root dry weight, and harvest index, particularly in the bitter varieties (high cyanide-level varieties). It was considered that the sweet varieties (low cyanide-level varieties) be early­bulked with higher tuber yield when they are planted with the 25-30 cm long SCs or with the SCs from young type to semi-mature portions of mother stems, while the bitter varieties with the 15-20 cm long SCs or with the SCs from semi-mature to hardwood portions of mother stems, respectively. However, a significant interaction between length and portion of SC was not observed in all agronomic characters.

Higher level of KCl application stimulated both leaf area index and leaf area duration in all cassava varieties, while the leaf and tuber number of the bitter varieties (high cyanide-level varieties) decreased in proportion to the level of KCl application. The root/shoot (R/S) ratio and harvest index (HI) were negatively related with the level of KCl application in all cassava varieties. The bitter varieties obtained the lowest R/S ratio at the level of 100 - 150 kg KCl ha-1 , while the sweet varieties (low cyanide-level varieties) acquired the highest values at the level of 50 - 150 kg KCl ha-1 . Also, the sweet varieties showed the lowest HI at the level of 250 kg KCl ha-1 , but the bitter varieties at the level of 150 kg KCl ha-1 . At 6 - 8 months after planting, the sweet varieties tended to obtain higher starch content of roots (tubers) at the level of 50 - 150 kg KCl ha-1 , while the bitter varieties at the level of 150 - 250 kg KCl ha-1 . Relatively lower level of 50 - 150 kg KCl ha-1 was more appropriate for decreasing hydrocyanic acid (HCN) content of roots (tubers) in the sweet varieties at the harvest time, and the level of 250 kg KCl ha-1 was adequate to decrease not only HCN content of leaves but also that of roots (tubers) in the bitter varieties during the growing period. To obtain higher yield and starch content of tubers, and lower HCN content of roots (tubers), it was recommended that the sweet varieties are applied with the level of 50 - 100 kg KCl ha-1 and the bitter varieties with the level of 150 - 200 kg KCl ha-1 , respectively, in Latosol soils of Bogor areas, West Java.

This study was conducted to identify the soluble carbohydrates in soybean seeds using on-line HPLC-RID-ES/MS and HPLC behavior, and to deter­mine their contents for high quality soybean breeding. The monosaccharide (glucose) and three oligosaccharides (sucrose, raffinose, and stachyose) were identified in Korean soybeans by their chromatographic behavior and results of on-line HPLC-RID-MS with Electro­spray Ionization mode. On the basis of HPLC with a RID detector, the 32 Korean major soybeans contain 0.37~pm0.26~% glucose, 4.55~pm0.91~% sucrose, 1.19~pm0.19~% raffinose, and 2.72~pm0.37~% stachyose on a dry basis. In 468 soybean germplasms, the ranges of glucose, sucrose, raffinose, and stachyose were 0.03 - 0.98~% , 2.33 - 6.96~% , 0.08 -1.87~% and 0.75 - 3.18~% , respectively. Among 500 soybean samples, oligosaccharide contents of 32 Korean major cultivated soybeans and 468 soybean germplasms were varied 5.83 - 10.06~% and 3.66 - 10.32~% , respectively. The composition of glucose, sucrose, raffinose, and stachyose in soluble carbo­hydrates of 500 soybean samples were 2.07 ~pm 1.75~% , 58.01~pm5.82~% , 10.13~pm2.28~% and 29.80~pm4.54~% , respectively. Sucrose appeared to be most prevalent in soy­bean soluble carbohydrates.

Seedlings of two rice genotyopes, cvs. Ilpumbyeo and Gancheokbyeo, were exposed to 0, 50 and 100 mM NaCl in nutrient solution for nine days. Plants were collected at the interval of 3 days and organic and inorganic solutes in leaves and roots and antioxidative enzyme activity in leaves were determined. Under salinity, the accumulation of soluble sugars occurred considerably in the older leaves of stressed seedlings compared to younger leaves and roots. The endogenous Na+ contents markedly increased at higher NaCl concentration in leaves and roots of seedlings, though it was higher accumulated in roots. Salinity resulted in an excessive proline accumulation in the stressed plants. A more pronounced increase was observed in Gancheokbyeo leaves. SOD activity in Impumbyeo cannot found any remarkable change, whereas, in Gancheokbyeo, its activity was rapidly decreased. CAT and POD activities increased with an increase in NaCl concentration in both genotypes. In sum­mary, the high capacity of rice seedlings to overcome an unfavorable growth condition such salt stress appears to be related to an adequate partition of organic solutes between shoots and roots and to changes in absorption, transport and re-translocation of salts.

The three Korean wheat cultivars with different plant types; the erect, the middle and the creeping growth habit, were studied for their utilization to solar radiation, temperature changes on the furrow and to provide optimum planting space for producing the high yield in 2003. The average solar radiation rate was lowest for creeping type (39.2~% ) and highest for erect type (75.8~% ) The correlation coefficient between the coverage rate and the solar transmission rate was r = 0.8624 which was significant at 5~% level. The relative growth of the plant, tiller rate and leaf size was increased in the erect and the middle type at lower plant density, while no change on plant growth at creeping type regardless of plant density. The increase of leaf size in the lower plant density was due to longer flag and the first leaf than those of other plant types. The temperature on the furrow of growing plants was changed by the canopy. The changes in temperature pattern on the furrow according to plant types during winter season was different compared to the non plant ground. The temperature of the nonplant ground was the lowest due to solar reduction increasing the amount of cool air flowing in the furrow.

Common thistle contains water-soluble substances that are antioxidative to foods. Antioxidant activities measured by DPPH method for the ground samples were the greatest in leaves, although was less than that of commonly used antioxidants, BHT and ascorbic acid. Methanol extracts and fractions from Xanthium occidentale plants dose-dependently increased DPPH free radical scavenging activity, in vitro test. The extracts from leaves showed the strongest antioxidant activity. DPPH scavenging activity of the individual fraction was in order of n-butanol>water>ethyl acetate>n-hexane fraction. By means of HPLC analysis, leaf samples of Xanthium occidentale had the highest amount of phenolic compounds, related with antioxidant activity, and followed by stems and roots. Total content of these antioxidant phenolic com­pounds for leaves extracts were detected in water fraction (36.7 mg 100 g-1 ) as the greatest amount, especially chlorogenic acid (39.4 mg 100 g-1 ) was the greatest component. These results suggest that Xanthium occidentale plants had potent antioxidant activity, and their activities were differently exhibited depending on plant part and fraction.

A field experiment was conducted to assess the growth characteristics, seed and oil yield of two cultivars of soybean (G max (L.) Merr.) cv. PK-416 (V1 ) and cv. PK-1024 (V2 ) in relation to sulphur and nitrogen nutrition. Six combinations (T1-T6 ) of two levels of sulphur (0 and 40 kg ha-1 ) and two levels of nitrogen (23.5 and 43.5 kg ha-1 ) were applied to the two soybean cultivars as nutrients. Results indicated significant effect of sulphur and nitrogen, when applied together, on the growth characteristics, yield components, and seed and oil yield. Maximum response was observed with treatment T6 (having 40 kg S and 43.5 kg N ha-1 ). Seed and Oil yields were increased 90 and 102~% in V1 > and 104 and 123~% in V2 , respectively as compared to the control i.e. T1 (having 0 kg S and 23.5 kg N ha-1 ). Positive responses of S and N interaction on leaf area index, leaf area duration, crop growth rate and biomass production were also observed. The results obtained in these experiments clearly suggest that balanced and judicious application of nitrogen and sulphur can improve both seed and oil yield of soybean cultivars by enhancing their growth.

To investigate the application of biochemical markers' and small-sample methods using whole-wheat flours for screening in early generation in Korean wheat breeding system, 74 Korean wheats, including cultivars, local breeding lines and experimental lines, were analyzed. Seed storage protein and amylose contents of grains were evaluated. Biochemical makers, including granule bound starch synthase (GBSS), high molecular weigh glutenin subunits (HMW-GS) and friabilin were also evaluated by using one-dimensional sodium dodecyl sulfate-polyacryla-mide gel electrophoresis with a single kernel. The small­sample methods, including modified SDS-sedimentation test (MST), micro-alkaline water retention capacity (AWRC) and whole-wheat flour swelling volume (WSV) were also tested in this study. Protein content, MST and AWRC was 11.0 - 15.8~% , 2.7 - 26.2 ml and 71.9 - 109.7~% , respectively. Apparent and total amylose content and WSV was 20.6 - 25.0~% , 26.1 - 32.4~% and 9.0 - 16.9 ml, respectively. There were highly significant correlations between MST and AWRC (r=0.592, P<0.001), but Korean wheats showed no significant difference in protein content, amylose content and small-sample methods. In the biochemical markers, Korean wheats contained all three GBSS encoded by Wx loci, except for Suwon 252. Korean wheats showed the high frequency (58.1~% ) of 1Dx2.2 + 1Dy12 subunits of HMW-GS. Friabilin band was present in 46 lines (62.2~% ) and absent in 28 lines (37.8~% ). Friabilin-absence lines showed the higher MST (14.9 ml) and AWRC (92.1~% ) value than friabilin-presence lines (8.5 ml and 82.4~% , respectively).

Mature embryos were aseptically excised with a scalpel and sliced in fragments measuring 0.5 mm in diameter (sliced mature embryo fragment; 4 ~~ 5 fragments/one embryo). Sliced mature embryo fragments of six wheat cultivars were cultured to develop an efficient method of callus induction and plant regeneration. Callus derived from sliced mature embryo fragments showed a good capacity to embryogenesis and regeneration. Furthermore sliced mature embryo fragments decreased contamination from fungi and bacteria. The high efficiency of callus induction were obtained Keumkangmil and Bob­white. For plant regeneration, selected embryogenic calli were transferred to two types regeneration media. An average number of green spots per callus was 4 to 5 in regeneration media after about one week. Percentage of plant regeneration showed high in regeneration medium containing 0.1 mg/l 2,4-D and 5 mg/l zeatin. Especially, Keumkangmil (27.5~% ) and Bobwhite (33.3~% ) showed high regeneration efficiency. This regeneration system from sliced mature embryo fragments may provide an effective and convenient explant for plant transformation studies

Single nucleotide polymorphisms (SNPs) are valuable DNA markers due to their abundance and potential for use in automated high-throughput genotyping. Numerous SNP genotyping assays have been developed. In this report, one of effective and high throughput SNP genotyping assays, which was named the template-directed dye-terminator incorporation with fluorescence polarization detection (FP-TDI) was described. Although the most of this assay succeed, the objective of this work was to deter­mine the reasons for the failures, find ways to improve the assay and reduce the running cost. Ninety F2 -derived soybean, Glycine max (L.) Merr., RILs from a cross between 'Pureunkong' and 'Jinpumkong 2' were genotyped at four SNPs. FP measurement was done on Victot3 microplate reader (perkinelmer Inc., Boston, MA, USA). Increasing the number of thermal cycles in the single-base extension step increased the separation of the FP values between the products corresponding to different genotypes. But in some assays, excess of heterozygous genotypes was observed with increase of PCR cycles. We discovered that the excess heterozygous was due to misincorporation of one of the dye­terminators during the primer extension reaction. After pyrophosphatase incubation and thermal cycle control, misincoporation can be effectively prevented. Using long amplicons instead of short amplicons for SNP genotyping and decreasing the amount of dye terminator and Acyclopol Taq polymerase to 1/2 or 1/3 decreased the cost of the assay. With these minor adjustments, the FP-TDI assay can be used more accurately and cost-effectively.
