본 연구는 돼지 수정란의 체외 성숙 및 체외 배양액의 retinol 첨가 효과를 규명하기 위하여 체외 성숙 및 체외 배양액에 retinol을 첨가하여 수정란의 체외 발달에 미치는 영향을 구명하고자 수행되었다. 체외 성숙 배양액에 retinol을 첨가한 결과 성숙율은 으로 각 처리구 간의 유의적인 차이가 없었다(p>0.05). 체외 수정 후 배반포로의 발달율은 첨가구에서 의 발달율을 나타내어 타 처리구에 비하여 유의적으로(p<0.05) 높게 나타났으며,

Mitis-salivarius sucrose bacitracin(MSB) medium is widely used in the selective isolation of mutans streptococci(MS), a designation for a group of oral cariogenic species. Recently, we have isolated three bacterial strains grown on MSB agar from human dental plaques. The three strains exhibited biochemical characteristics similar to those of the biotype IV of MS, with the exception that they manifested a positive reaction for arginine deaminase. The objective of this study was to identify and characterize these three clinical isolates. The bacteria were identified with biochemical tests as well as by 16S rDNA cloning and sequencing. In order to compare the antibiotics susceptibility of the clinical isolates with that of type strain, the minimum inhibitory concentrations of 9 antibiotics were determined using broth dilution assays. The results identified all of our three clinical isolates as Enterococcus faecalis. All E. faecalis strains were found to be susceptible to penicillin G, amoxicillin, augmentin, and vancomycin, but were resistant to ciprofloxacin, cefuroxim axetil, and clindamycin. Our findings indicate that E. faecalis is capable of growing on MSB agar, and suggest that the MSB medium be improved so that only MS should be recoverable on the medium, as originally devised for their selection.

결정질 암반에 위치한 가상의 방사성폐기물처분장에 대한 정확한 안전성 평가를 수행하기 위해서는 다공암반으로 대표되는 공학방벽 및 결정질 암반으로 대표되는 자연방벽을 모두 고려한 매질에서의 물질 거동 특성을 정확하게 모사하는 것이 중요하다. 본 논문에서는 단열-다공암반내 유동 및 물질전달의 정확한 해석을 위한 연결망 구축방법에 대하여 서술하였다. 실제 단열암반을 사실적으로 모사하기 위하여 단열 물성자료는 확률밀도함수를 이용하여 생성하였다. 2차원의 사각격자로 모사된 단열암반과 육면체로 모사된 다공암반간 교차선의 원활한 검색을 위하여 단열암반과 연결전 다공암반의 교차면에 가상의 단열암반을 추가적으로 도입하였다. 전체유동경로를 구성하기 저하여 생성된 단열들 간의 교차선을 효율적으로 검색할 수 있는 방법 및 단열암반 및 단열-다공암반간의 연결도를 신속히 확인하는 알고리즘을 제안하였다. 이러한 방법들은 추후 방사성폐기물처분장에서의 단열-다공 암반을 통한 핵종 이동 특성을 모사할 수 있는 수치코드 개발에 많은 도움을 줄 것으로 사료된다.

Quality system management adapted by small and medium enterprises in Kangwon province to enhance the competitiveness was studied. Variance analysis on several questionnaire answers was performed. Motives for acquiring the accreditation, such as product export, adjustment to international trend, enhancement of brand/product recognition, CEO's mind change, and management innovation, have been changed significantly among business types. Mind changes after the accreditations were setting company's first priority on quality, enhanced recognition on compliance of in-house standards and regulations, employee's performance with the recognition of quality. Amongst service problems to maintain the ace reditations were difficulties in maintaining the recognition of the company's finality management, labor increase to maintain the ISO 9000 enforcement team, and financial burden to keep the accreditation. Quality recognition after the accreditations was significantly improved in setting company's first priority on quality, enhanced recognition on compliance of in-house standards and regulations, employee's performance with the recognition of quality.

The present study was conducted to investigate gelatinolytic activities in HAM and to determine whether there are any changes in gelatinolytic activity profiles when the cells are cultured in hepatogenic medium. Placenta was obtained during caesarean section of the volunteers, with informed consent. HAM were isolated from amniotic membrane using collagenase type A HAM were cultured in hepatogenic medium for 3 weeks and the conditioned media were obtained at day 7, 14 and 21. The zymographic pattern of gelatinolytic activity of the HAM did not undergo a change during passages. When the HAM were cultured in a fibronectin-coated dishes in a hepatogenic medium, there was no significant difference of the gelatinase pattern between before and after culture. However, when bFGF was added to the culture, a dramatic increase of 62kDa and 59kDa gelatinases was observed. Interestingly, when ITS instead of FN was present, HAM-conditioned medium also showed a similar increase of both gelatinases. Immunoblotting analysis demonstrated that both 62kDa and 59kDa gelatinases were the active form of MMP-2 resulting from the turnover of MMP-2 proform. Futher study will be necessary to determine the relationship between bFGF and active MMP-2 during hepatogenesis of HAM.

Porcine oviduct epithelial cells (POEC) are widely used in co-culture experiments to improve early embryonic development, in vitro fertilization in embryo transfer programs for domestic animals and in vitro maturation of immature germ cells. POEC were mechanically isolated and cultured in tissue culture medium 199. Cells grew continuously, and confluent monolayers were formed after 7 days. After forming confluent monolayer of epithelial cells, supernatant was collected as the condition medium for maturing round spermatids in vitro. Round spermatids were also separated mechanically and cultured in the POEC condition medium. In this study we observed that 20% of round spermatid cultured were matured into elongating spermatid after 24 h, and about 10% of round spermatid cultured showed complete elongation (elongated spermatid) within 24~48 h of in vitro culture. No further development was observed within 50~72 h and transformed cells lost their viability after 72 h. These preliminary findings suggest that the condition medium from POEC may be possible to overcome the round spermatid block by improving the milieu of culture system.

본 연구는 재활용 PET-Bottle-Particles(PBT)가 함유된 4가지 생육배지를 이용하여 플러그묘 재배의 가능성을 실험하기 위해 유리온실에서 수행되었다. 토마토 '서광'의 플러그묘를 PBT 100%, peat moss 100%, 그리고 PBT 50%+peat moss 50%가 함유된 배지 및 상업적으로 이용되는 배지(토실이)를 대조구로 이용하여 육묘하였다. 128공 플러그 트레이에서 2004년 7월 12일에 파종하여 플러그묘의 생육은 파종 후 30일째 측정하였다. 모든 처리에서 95% 이상 발아하였다. 초장, 지상부 생체중과 건물중, 엽면적, 그리고 하배축 길이는 대조구에서 가장 컸다. 뿌리 길이와 무게는 PBT 100%처리에서 가장 컸으며, 이 배지의 pH는 7.3이었다. PBT 50%+peat moss 50%처리에서 엽면적과, 지상부 건물중이 PBT 100% 처리에서 보다 컸으며, peat moss 100%처리에서 가장 작았다. PBT를 사용함으로 인한 독성증상은 관찰되지 않았다. 이 결과는 원예 묘종 생산에 있어 배지의 물리성 개선을 위한 구성요소로써 재활용 가능한 PBT의 이용 가능성을 제시해 준다.

We investigated the situation of an accident occurred in the company which entrusted the specialized organization with his own safety control notwithstanding it is free from an obligation to have any safety manager for aiming at the comparison and analysis of the outcome. The economic and preventive effect of industrial accident has been compared and analyzed in order to strengthen the obligation of selecting a safety manager.
