This study analyzed the effect of time of trot on hematology and blood chemistry values of the Jeju Pony crossbreed horses that are commonly used for riding (14.1±1.4 years old, Gelding). A total of 28 parameters including vital signs as well as stress hormones such as cortisol and lactic acid levels were examined as the time of the trot exercise progressed. Vital signs such as heart rate (38.0→81.0 times/min) and respiratory rate (11.7→35.7 times/min) increased significantly within 30 minutes of exercise. However, difference in the body temperature was not observed before and after exercise. The hematology including white blood cell count (8.03→9.52×103 cells/μL), red blood cell count (5.94×103→7.23–7.32×103 cells/μL), hemoglobin levels (11.82→14.65–14.78 g/dL), and hematocrit levels (25.04→30.27%) significantly increased 30 minutes after the start of the exercise (p<0.05). The blood chemistry value of albumin (3.25→3.47 g/dL) (p<0.05) only showed a significant increase after the exercise. However, the other blood chemistry levels such as, Na+, K+, Ca2+, total CO2, creatine kinase, glucose, blood urea nitrogen, creatinine, aspartate transaminase, total bilirubin, gamma–glutamyl transpeptidase, and total plasma protein did not change. Also, cortisol and lactic acid levels did not show significant difference. The middle-aged Jeju pony crossbreed horses were not stressed by the 30-minute exercise; therefore, it can be concluded that there is no problem regarding the safety of both the rider and the animal.
본 연구는 Canada에서 수입한 동결정액으로 인공수정된 모돈의 번식성적을 조사하여 동결정액을 이용한 인공수정(동결정액 인공수정)의 효율을 개선할 수 있는 통찰력을 얻기 위해 수행되었다. 이를 위해 A(GGP) 농장에서 2016년 5월과 9월 사이 총 626회의 동결정액 인공수정과 B(GGP) 농장에서 2015년부터 2017년 기간 동안 수행한 총 2,429 동결정액 인공수정의 결과 기록을 분석하였다. A종 돈장에서 동결정액을 썼을 때 총산자수 및 실산자수는 9월 보다 5월에 높았다(p<0.05). B종돈장의 분만율, 총산자수 및 실산자수는 조사연도 간에 차이가 없었다. A종돈장 결과와 B종돈장 결과를 통합하여 분석했을 때, 분만율은 A종돈장이 높았으나(p<0.01) 총산자수와 실산자수는 두 종돈장간에 차이가 없었고, 동결정액 인공수정을 했을 때가 액상정액 인공수정을 했을 때보다 낮았다(동결정액:액상정액 인공수정에 대한 총산자수와 실산자수는 각각 10.9±0.3:13.4±0.1두 및 10.0±0.3:12.0±0.1두 이었음; p<0.01). 결론적으로, 이상의 결과는 동결정액 인공수정이 액상정액 인공수정보다 번식 효율이 낮긴 하지만 연중 적절한 시기에 동결정액 인공수정을 실시하면 번식 효율이 증가될 수도 있음을 시사한다.
The aim of this study is to examine current status of swine AI and boar stud in South Korea using survey and data analysis. This survey included 48 boar studs registered as ‘semen processing business’. The survey data were collected by direct visitation, FAX and/or telephone conversation for 7 months from June through December in 2013. 48 boar studs owned a total of 3,537 boars and the Duroc breed accounted for the highest rate (75.3%) of all boar breeds. In case of ownership, agricultural management corporations was the highest (50.0%) and followed by individual ownership (33.3%). Large-scale boar studs in terms of own over 151 boar were surveyed as 4.2% and most boar studs owned less than 100 boars (77.1%). The amount of liquid semen provided by 48 boar studs were 1,889,000 doses and each boar stud provided average of 39,000 does, which is represented for 90% consumption by sows in South Korea.
This study was conducted to determine the relationship between elapsed time after semen preservation on the changes of bacteria and semen quality. Semen was diluted with BTS(Beltsville Thawing Solution) extender without antibiotic for 7 days and sperm parameter and fertility were measured. Sperm motility was measured by CASA and total bacteria number was counted after 22~24 hr incubation from counting agar plate in which sperm dilute to 10 ~106 in 0.9% saline solution and inoculate to agar. Acrosomal integrity was measured by Chlortetracycline (CTC) staining. CTC patterns were uniform fluorescence over the whole head (pattern F), characteristic of incapacitated acrosome- intact spermatozoa; fluorescence-free band in the post-acrosomal region (pattern B), characteristic of capacitated acrosome-intact spermatozoa; and almost no fluorescence over the whole head except for a thin band in the equatorial segment (pattern AR), characteristic of acrosome reacted spermatozoa. Total number of bacteria was significantly increased (p<0.0001) 3 days after preservation. Sperm motility, viability, and morphological abnormality on elapsed time after preservation were lower from 5 (77.24±6.47, p<0.001) and 7 days (77.24±6.47, p<0.001) after preservation compared to 1 (15.71±7.18) and 3 days(18.39±7.22) after preservation, respectively. Sperm viability was significantly lower (53.25± 35.03, p<0.0001) at 7 days after preservation. Morphological abnormality of sperm was lower (p<0.001) at 1 (15.71±7.18) and 3 (18.39±7.22) days compared to 5 (21.84±7.91) and 7 (22.59±9.93) days after preservation. Acrosomal integrity and capacitation rate (pattern F) were significantly lower (p<0.001) from 5 days after preservation. Based on the data we obtained from this study suggested that semen preserved more than 5 days without antibiotic would not recommend use for artificial insemination.
발정주기와 상관없이 CIDR을 삽입하는 날에 50 mg progesterone, 2.5 mg estradiol benzoate를 근육주사하였다. CIDR 삽입 후 4, 5일에 28 AU FSH (Antorin R10)을 4일 동안 감량법으로 주사하였다. 6, 7회 FSH 주사 후 25 mg, 15 mg 를 각각 주사한 다음, CIDR는 7회 FSH 주사 후 제거하였다. 1회째 주사 후 48시간에 GnRH를 주사하였다. 공란우는 발정확인 후 12시간 간격
돼지 정액 채취과정은 무균적으로 진행되기가 어려우므로 채취된 정액의 세균오염 (bacteriospermia)은 일반적인 것으로 알려져 있다. 돼지 정액에서 주로 분리되는 오염 세 균은 그람음성균이고, 장내세균과(Enterobacteriaceae)에 속하는 것으로 조사되고 있다. 정액 내 세균오염은 오염 정도에 따라 정액의 품질과 수명에 부정적인 영향을 미치는 것 으로 보고되고 있다. 본 실험에서는 돼지 정액에서 순수분리한 대장균을 실험적으로 농 도별로 오염시켜 돼지 정액의 활력, 생존율 및 pH에 대한 대장균 오염의 영향을 확인코 자 하였다. 정자 농도와 세균오염 정도를 조절하기 위하여 항생제가 첨가되지 않은 시판 돼지 정 액 희석제(Seminark Pro)를 이용하였고 돼지 정자수 대비 대장균수가 4,000 : 1(T1), 400 : 1(T2), 40 : 1(T3), 4 : 1(T4)이 되도록 액상정액 시료를 제조하였다. 모든 시료는 17℃ 저온배양기에 보존하면서 유세포분석기(flow cytometer)를 이용하여 0일차, 1일차, 3일차 및 5일차에 각 시료별 정자 활력(미토콘드리아 함량 측정)과 생존율(Live/DeadⓇ 염색)을 측정하였고, 산도측정기(pH meter)로 정액 pH를 측정하였다. 정자의 활력은 T3, T4에서 0일차(17℃, 4시간 배양)부터 대조군(C)에 비하여 유의성 있 게 감소하였고, 모든 시료에서 3일차 이후부터 활력이 감소(p<0.05)하였다. 정자 생존율 의 경우에도 T3, T4에서 0일차부터 C에 비하여 유의성 있게 감소하였고, 모든 시료에서 3일차부터 생존율이 감소(p<0.05)하였다. 보존일 경과에 따른 정액의 pH 변화의 경우, C, T1 및 T2에서는 0일차에 pH 7.00 정도에서 5일차(p<0.05)에 pH 7.10 이상으로 높아진 반면, T3에서는 3일차(p<0.05)부터 pH 6.90 이하로 낮아져 5일차에는 pH 6.86이었고, T4 에서는 1일차(p<0.05)부터 pH 6.86 이하로 낮아져 3일차(p<0.05)와 5일차(p<0.05)에는 pH 6.65 이하로 낮아졌다. 본 결과로부터 돼지 정자수 대비 대장균수가 40 : 1(20×106 sperm cells/ml : 5×105 cfu/ ml) 이상으로 오염되었을 경우 오염 당일부터 5일차까지 대조군에 비하여 정자의 활력과 생존율이 유의성 있게 감소되었으며 40 : 1의 경우는 3일차부터, 4 : 1의 경우는 1일차부 터 정자의 사멸 혹은 세균오염에 의한 산패로 정액의 pH가 낮아지는 경향을 확인할 수 있었다. 이는 정액 내 세균오염이 정액의 품질과 수명에 부정적인 영향을 미친다는 보고 와 일치하며 위생적인 정액 채취를 위한 노력과 고품질 정액 공급을 위한 정기적인 모니 터링 검사 및 보존액 내 유효한 항생제 첨가가 중요할 것으로 사료된다.
This survey was conducted to investigate the current status of swine artificial insemination(AI) centers registered as 'semen processing business' in Korea. The survey responses were collected by direct visitation or telephone conversation for 5 months from May through September in 2008. The survey showed that sixty-four AI centers were enrolled in local government and those of fifty-two AI centers were under operation. Forty-nine AI centers surveyed owned a total of 3,334 boars and the Duroc breed accounted for the highest rate(73.1%) of all boar breeds. In type of ownership, agricultural management corporations was the highest(42.3%) and followed by private ownership (34.6%). Large-scale AI centers in terms of own over 151 boar were surveyed as 5.9% and most AI centers own less than 100 boars(86.5%). The average number of boars per AI center was 68. The amount of liquid semen provided by 52 AI centers were 1,791,000 doses and each AI center provides average of 39,000 does, which is represented for 90% consumption by sows in Korea.
The objective of this study was to determine the effect of semen extenders on the motility, viability and fertility in vitro of spermatozoa during storage of fresh boar semen diluted in different commercial extenders used for pig artificial insemination (AI). In this experiment, semen were diluted in Androhep plus, Beltsville Thawing Solution (BTS), Modena, Seminark and Vitasem LD. Five ejaculates were collected from three Duroc boars and sub-samples were diluted (30×106 spermatozoa/ml) in different extenders. Semen was stored at 17℃ for 10 days. Sperm motility and viability was assessed using Computer-Assisted Semen Analysis (CASA) and flow-cytometry on 1, 3, 5 and 10 day post collection. The motility of spermatozoa stored in different extenders was gradually decreased by increasing the duration of storage of semen. However, there was not significantly different in the sperm motility and viability among other extenders. On the other hand, the in vitro-matured oocytes were fertilized and cultured in vitro to assess the fertility of boar spermatozoa stored for 3 and 10 days in different extenders. The percentage of morula and blastocyst were taken as indicators of fertility in vitro of spermatozoa. Therefore, there were no differences in the rate of embryos developed to the molular and blastocyst stage. There were no differences in the motility and fertility in vitro among 5 kinds of commercial boar semen extenders.
Though food safety efforts of the government has been increasing, but the health hazards resulting from food does not stop and thus increases consumers' anxiety and distrust of government. For more reliable safety management, developing analysis and intake measurement methods and strengthening regulatory control with strict criteria is necessary. In addition, as not only the known hazards by now, but also unknown but obvious hazards are simultaneously ongoing in current environments, the unified risk management and health assessment for food, health food and medicines from the media is critical. As risks and damage to health may show up after a long time after exposure,preventive measures and strategies for safety control is also needed.
Various functional foods, marketing health and functional effects, have been distributed in the market. These products, being in forms of foods, tablets, and capsules, are likely to be mistaken as drugs. In addition,non-experts may sell these as foods, or use these for therapy. Efforts for creating health food regulations or building regulatory system for improving the current status of functional foods have been made, but these have not been communicated to consumers yet. As a result, problems of circulating functional foods for therapy or adding illegal medical to such products have persisted, which has become worse by internet media. The cause of this problem can be categorized into (1) product itself and (2) its use, but in either case, one possible cause is lack of communications with consumers.
Potential problems that can be caused by functional foods include illegal substances, hazardous substances,allergic reactions, considerations when administered to patients, drug interactions, ingredients with purity or concentrations too low to be detected, products with metabolic activations, health risks from over- or under-dose of vitamin and minerals, and products with alkaloids. (Journal of Health Science, 56, Supplement (2010)). The reason why side effects related to functional foods have been increasing is that under-qualified functional food companies are exaggerating the functionality for marketing purposes. KFDA has been informing consumers, through its web pages, to address the above mentioned issues related to functional foods, but there still is room for improvement, to promote proper use of functional foods and avoid drug interactions. Specifically, to address these issues, institutionalizing to collect information on approved products and their side effects, settling reevaluation systems, and standardizing preclinical tests and clinical tests are becoming urgent. Also to provide crucial information, unified database systems,seamlessly aggregating heterogeneous data in different domains, with user interfaces enabling effective one-stop search, are crucial.
In modern diet relying heavily on processed food, it is difficult to live without food additives, added to food regardless of whether consumers want or not. This poses the following questions: Are food additives really safe? Are they toxic? Are they cancerous? Are they still good to eat? Are there any concerns on excessive intake and interactions? Is toxicity testing adequate? Is ADI evaluation being set up properly? In this article, I discuss a desirable management strategy for food additives, by addressing these questions and concerns and analyzing and reviewing current status of food additive management.
The brilliant cresyl blue (BCB) has been used to select the developmental competent oocytes in pigs, goats and cows. Growing oocytes have a higher level of active glucose-6-phosphate dehydrogenase(G6PDH) compare to mature oocytes and are rarely stained compared to mature oocytes, because G6PDH converts BCB to colorless. First polar body extrusion regard as a guideline of meoisis completion. Selection of polar body extrude oocyte is more developmental competent to blastocyst than unselected. This study was conducted to compare the BCB test to the polar body extrusion on selection of developmental competent porcine oocytes for the production of blastocyst. Cumulus-Oocytes complex were exposed to 26uM BCB stain diluted in NCSU-23 for 90 min. There was no significant difference embryo development to blastocysts between BCB treated and not treated(19.58±1.99 vs 18.75±2.27 %), which means there was no detrimental effect of BCB exposure to oocytes. Normal fertilization is not differed among treatment groups from 70.0 to 78.4% development to blastocyst, beside polyspermy did not. To compare two different selection methods, BCB test and polar body extrusion, evaluate the developmental competent of IVP embryos. BCB+PB+(blue stained and polar body extruded, 20.71±0.45%) and BCB-PB+(colorless and polar body extruded, 20.04±1.29%) groups are significantly (p<0.05) higher developed than those of BCB+PB-(blue stained and no polar body, 13.24±0.73%) and BCB-PB-(colorless and no poladbody, 7.25±0.77%). These results showed that selection of polar body extruded oocytes method is more efficient than that of BCB test.
To provide the research strategy for protection of children’s health from hazardous chemical, we reviewed the hazardous chemicals can be exposed through maternity, children's life style and living environment. Recently, diseases related with children’s living condition were focused as asthma, atopy, childhood developmental disability, congenital malformations and obesity. Children can be exposed to hazardous chemicals through an ambient air, water, soil, food, toys and other factors such as floor dust. Also children’s health was deeply related with a wrong life style and neglectful caring by a lack of knowledge and information of harmful ones at parents and child care center’s nursers. According to the previous study, the chemical risk factor of children’s health were identified as inorganic arsenic, bisphenol A, 2,4-D, dichlorvos, methylmercury, PCBs, pesticide, phthalates, PFOA/PFOS, vinyl chloride, et al. Domestic studies for identification of causality between children exposure to chemicals and resulted hazardous effects were not implemented. The confirmation of chemical risk factors through simultaneously performing toxicological analysis, human effect study, environmental/human monitoring, and risk assessment is needed for good risk management. And also, inter-agency collaboration and sharing information can support confirming scientific evidence and good decision making.
유해물질의 식품기준은 위해성이 인정될 때 정하는 것을 원칙으로 한다. 위해평가의 절차에 거쳐 정량적 수치로 인정되는 위해성은 정부의 관리방향을 설정하는데 중요한 정보를 제공한다. 대표성 있는 모니터링자료가 확보되어야 정확한 위해평가를 수행할 수 있으므로 만성적 건강영향을 나타내는 유해물질의 경우 장기적 모니터링결과를 확보하는 것이 필요하다. 과거에는 대다수 인구집단의 안전에 초점을 두었으나, 최근에는 고섭취에 의한 유해물질 고노출그룹, 동일노출 수준에서도 유해영향을 크게 나타내는 민감그룹, 노약자, 임산부 등의 안전까지도 검토 후 소수그룹까지 안전할 수 있는 기준을 설정하는 추세이다. 유해물질 기준검토를 위하여 최신의 독성정보, 통계정보, 분석기술정보, 낮출 수 있는 한 낮출 수 있는 ALARA(as low as reasonably achievable)정보들이 필요하다.
