        Purpose: The objective of this study was conducted to investigate the effects of rutin, buckwheat components on cell growth and anti-inflammation in adipocyte 3T3-L1 and human colon cancer cell SW-480. Methods: We cultured 3T3-L1 adipocyte and SW-480 colon cancer cell to confluence, at which time starvation was induced with SFM for 1 day. Cells were then cultured in medium containing 0, 25, 50, or 100 μmol/mL of rutin 3T3-L1 or 0, 10, 20, or 40 μmol/mL SW-480. Cell viability was measured using a cell viability kit. In addition, we examined the expression of mRNA related to inflammation. RT-PCR was used to quantity tumor necrosis factor (TNF-α), interleukin-1β (IL-1β), IL-6, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) mRNA levels. Results: Rutin significantly inhibited 3T3-L1 and SW-480 cell proliferation in a dose and time dependent manner. Rutin also significantly reduced the mRNA expression of IL-1β, IL-6 and TNF-α at the highest dose. In addition, rutin treatment caused a significant reduction in COX-2 and iNOS mRNA levels compared to the control group. Conclusion: Overall, our results suggest that rutin has the potential to reduce inflammation, and that these effects are greater during tissue-damaging inflammatory conditions.

        Purpose: This study examined the effects of α-lipoic acid in diluted solvents on cell growth in 3T3-L1 cells according to the treated concentration and times. Methods: Adipocyte 3T3-L1 cell were cultured. Confluent cells underwent starvation with SFM for 1 day and then were cultured in a medium containing various concentrations 0, 100, 200, and 400 μmol/L of α-lipoic acid. The cell viability was measured using the EZ Cytox assay kit. In addition, the effect of α-lipoic acid of diluted solvents on the cell growth in 3T3-L1cells was examined according to the treated concentration and times. Results: The α- lipoic acid diluted ethanol inhibited cell proliferation in a dose and time dependent manner. The α-lipoic acid diluted ethanol induced adipocyte 3T3-L1 cells proliferation with an adipocyte inducer. In addition, α-lipoic acid inhibited adipocyte 3T3- L1 growth in a dose and time dependent manner (p<0.05). Conclusion: This study showed that a treatment with α-lipoic acid diluted ethanol inhibits cell growth of, adipocyte 3T3-L1 cells induced with an adipocyte inducer, (200 μmol/L of α- lipoic acid) treated for 48 hr.

        Background : Lentinus edodes are highly utilized as food and medicine, and are known to be suitable for preventing diabetes, high blood pressure and arteriosclerosis. L. edodes were known for prevent obesity metabolisms. Also, present consumers persue the healthy and thin condition. Our research perpose is determined the prevent obesity using L. edodes extracts. Methods and Results : The effect on adipocyte differentiation and the accumulation of fat in the L. edodes extract on 3T3-L1 cell preadipocyte and cell toxicity were tested. We measurement to evaluate the inhibition effects of L. edodes extracts on the growth of 3T3-L1 cell adipocytes. The tested cells were treated with L. edodes extracts by ethanolic extracts (concetrations at 10, 50, 100 ㎍/㎖) and hot water extracts (temperature at 50℃, 60℃, 70℃, 80℃, 90℃), respectively. Cell toxicity did not affect the growth of cells growth in hot water extracts. Intracellular lipid accumulation was measured by Oil red O method. The inhibitory effect on 3T3-L1 cell by treatment different ethanolic extract concentrations and temperature of hot-waters. The optimal inhibitory conditions of lipid accumulation by ethanoic extracts and hot water extracts were determined the 32.5% ethanol extracts and 33.3% 50℃ hot water extracts. Conclusion : The results of this study showed that L. edodes extracts had an potentials for reduce lipid accumulation activities. Further more we will examine about the mechanisms of anti obesity activities from L. edodes extracts. This results provides that the basis for antiobesity foods or medicines using Lentinus edodes extracts.

        This study was executed to evaluate the phenolic content, DPPH radical scavenging rate, and the cytotoxic effect in human cancer cell, 3T3-L1 cell from C. lanceolata extracts at various ethanol concentration. Total polyphenol and flavonoid content of the C. lanceolata at various ethanol concentration showed the high amount in 70%, 100% ethanol extract. The DPPH radical scavenging activity progressively increased in a dose-dependent manner, and showed the highest in 100% ethanol extract. The cytotoxic effect against human cancer cell of the C. lanceolata was higher in 50% and 70% ethanol extracts. In particular, the cytotoxic effect in MCF-7 cell was relatively higher than in other cells. The IC50 (concentration causing 50% cell death) value showed the highest on MCF-7 cell (538.39 ㎍/㎖) in 70% ethanol extract, and exhibited significant activity against Hela cell (637.87 ㎍/㎖), Calu-6 cell (728.64 ㎍/㎖). The extract of 70% ethanol at 1,000 ㎍/㎖ exhibited a pronounced cytotoxic effect on 3T3-L1 cell comparable to that of the other extracts, and reduced in a concentration-dependent manner.

        Background : Obesity, a global health problem and a chronic diseases, is associated with increased risk of developing type 2 diabetes and coronary heart diseases. A wide variety of natural remedies have been explored for their obesity treatment potential. To elucidate the anti-obesity effect of ginsenoside Rg5 : Rk1 (Rg5 : Rk1), a mixture of protopanaxadiol type ginsenosides isolated from Panax ginseng Meyer in a 3T3-L1 adipocytes. Methods and Results : In order to determinate the anti-obesity effect of Rg5 : Rk1, Oil Red O staining and triglyceride (TG) content was assessed. Furthermore, to elucidate the possible mechanism whether Rg5:Rk1 affects lipid accumulation, mRNA and protein expression analyses of adipocyte markers such as STAT3, PPARγ, CBEPα and ap2 were carried out. Rg5:Rk1 treatment showed an inhibition of lipid droplet accumulation and decrease on TG content. In addition, expression of STAT3, PPARγ, CEBPα and ap2 were decreased in dose dependent manner. Similar to these results, Rg5:Rk1 treatment reduced PPARγ and CEBPα protein expression. Conclusion : Rg5 : Rk1 treatment exhibits anti-adipogenic activity by down-regulation of the STAT3PPARγ/CEBPα pathway in 3T3-L1 adipocyte cell line.

        Background: Hair loss related syndromes are increasing due to environmental pollution and stress. Hair care products are mainly prepared by mixing chemicals and natural extracts, such as those obtained from medicinal plants. The purpose of this study was to investigate the effects of 70% ethanol extracts from the flowers of Calendula officinalis, fruit body of Phellinus linteus, and the whole plant of Houttuynia cordata on the growth of CCD-986 cells, hair follicle dermal papilla cells (HFDPC), and 3T3-L1 cells. Methods and Results: All sample extracts at all concentrations, except for that from P. linteus fruit body at 500㎍/㎖, were cytotoxic to CCD-986 cells. However, none of the sample extracts were cytotoxic to HFDPC. The lipid differentiation of 3T3-L1 cells regulates hair regeneration via secretion of platelet derived growth factor. The 70% ethanol extract of H. cordata whole plant promoted hair growth. Adipogenesis rate significantly increased in a treatment concentration-dependent manner. Conclusions: These results suggest that 70% ethanol extracts of C. officinalis flower, P. linteus fruit body and H. cordata could be used for the development of hair care products.

        Brown algae is variety of biological compounds, including xanthophyll, pigments, fucoidans, phycocolloids, and phlorotannins. Several studies concerning these types of compounds have pointed out the variety of biological benefits associated with the algae, including antioxidant, anticoagulant, antihypertension, antibacterial, and antitumor activities. Diphlorethohydroxy- carmalol (DPHC) is a phlorotannin compound isolated from the brown algae Ishige okamurae, with various biological activities in vitro and in vivo. Numerous studies have shown that antioxidant assist inhibition of accumulation of fat. So we studied that effect of DPHC isolated from Ishige okamurae modified the accumulation of fat on preadipocyte, 3T3-L1 cells in vitro. First, the viability of cell was analyzed after 0.4, 2, 10, 50 μg/ml of DPHC treatment using MTT (3-[4,5-dimethylthiazo-2-yl]-2,5-diphenyl tetrazolium bromide) assay. Second, proliferation of cell was analyzed after 0.4, 2, 10, 50 μg/ml of DPHC treatment through measure doubling time. 3T3-L1 cell differentiation into adipocyte was analyzed after induction in the induction medium containing DPHC. The metabolic activity was suppressed by DPHC in concentration dependent manner. Doubling of 3T3-L1 was delayed by the treatment of DPHC in concentration dependent manner. DPHC also inhibit accumulation of triglyceride in the adipocyte. The expression of the marker genes for adipocyte differentiation coincided with cytochemical results. Base on them, it is suggested that DPHC suppress proliferation of adipose precursor cell and differentiation into adipocytes.
