PPARα activator가 고지방 사료를 섭취한 운동하지 않은 쥐에 비해 고지방 사료를 섭취한 운 동 쥐에서 백색지방조직의 혈관신생을 보다 효과적으로 억제하는지를 조사하였다. 수컷 쥐는 무작위로 PPARα activator인 fenofibrate와 운동을 모두 처리하지 않은 대조군(Con), fenofibrate 단독처리군(FF), 운동 단독처리군(Ex) 및 fenofibrate와 운동의 조합처리군(Ex+FF)으로 나누어 8주간 고지방 사료를 섭취시 켰다. 백색지방조직의 무게와 백색지방세포의 크기는 Con에 비해 FF, Ex 및 Ex+FF 모두 감소하였으며, Ex+FF는 FF에 비해 더욱 감소하였다. 백색지방조직에서 MMPs와 혈관신생 인자의 유전자 발현은 Con에 비해 FF, Ex 및 Ex+FF 모두 감소하였으며, Ex+FF는 FF에 비해 더욱 감소하였다. 그러나 혈관신생 억제인 자의 유전자 발현은 Con에 비해 FF, Ex 및 Ex+FF 모두 증가하였고, Ex+FF는 FF에 비해 더욱 증가하였 다. 따라서 본 연구는 fenofibrate 단독처리보다는 fenofibrate와 운동의 조합처리가 효과적으로 백색지방조 직의 혈관신생을 억제함으로써 백색지방조직의 증가를 감소시키고 복부비만을 억제한다는 것을 밝혔다.
본 연구는 수영운동이 백색지방조직의 혈관신생을 조절함으로써 비만을 개선하는지를 조사하였다. 고지방식이를 섭취한 암컷 쥐는 모의 수술 군(Sham), 난소절제 수술 군(OVX) 및 수영운동을 실시한 난소절제 군(OVX + Swim)으로 나뉘었다. Sham에 비해 OVX는 몸무게, 지방조직무게 및 지방세포의 크기가 증가 되었다. 그러나 OVX + Swim의 이러한 요소들(: 몸무게, 지방조직무게 및 지방세포의 크기)은 OVX에 비해 감소 되었다. Sham에 비해 OVX는 백색지방조직에서 혈관신생 촉진인자와 MMPs의 유전자 발현이 증가하였고, 혈관신생 억제인자들의 유전자 발현은 감소하였다. 그러나 OVX + Swim은 OVX에 비해 백색지방조직에서 혈관신생 촉진인자와 MMPs의 유전자 발현이 감소하였고, 혈관신생 억제인자들의 유전자 발현은 증가하였다. 이러한 연구결과들은 고지방식이를 섭취한 난소절제 암컷 쥐에서 수영운동이 백색지방조직의 혈관신생을 억제함으로써 비만을 개선한다는 것을 제시하였다.
In this study, the inhibitory effect of Paecilomyces tenuipes extract on PSA and angiogenesis-related factor expression levels were investigated in human prostate cancer cells, LNCaP. P. tenuipes extract significantly inhibited PSA expression in a dose-dependent manner. We also investigated the inhibitory effect of P. tenuipes extract on the expression of angiogenesis-related genes including VEGF, MMP-2, MMP-9, TIMP-1, and TIMP-2. P. tenuipes extract significantly down-regulated the expression of MMP-2 and MMP-9 in a dose-dependent manner. On the contrary, P. tenuipes increased the expression of TIMP-1 and TIMP-2. Our findings indicate that P. tenuipes exhibits an inhibitory effect on angiogenesis in human prostate cancer cells.
Dental pulp is a highly vascularized tissue with high regenerative potential. Revascularization of severed vasculature in the tooth is required for pulp healing during avulsed tooth treatment. In this study, the relative expression of angiogenesis-related proteins was determined in human dental pulp cells using a human angiogenesis proteome profiler array. The proteome profiler array detected differentially expressed angiogenesis-related factors under conditions of hypoxia, which enhances the angiogenic potential of dental pulp cells. We confirmed that hypoxia regulates the mRNA expression of angiogenesis-related factors, including CXCL16 in dental pulp cells. Furthermore, conditioned media of hypoxic pulp cells induced tube-like structures of vascular endothelial cells, which were reduced by the neutralization of CXCL16 function. In conclusion, our data show that angiogenesisrelated factors are differentially expressed by hypoxia in dental pulp cells and suggest that CXCL16 may involve in the revascularization of hypoxic dental pulp.
Oil spills have occurred throughout the years of industrialization and represent a global challenge as they affect vast areas of the ocean. The toxicity of crude oil to aquatic organisms has been extensively investigated, but the potential impacts of crude oil on vertebrate development remain largely unknown. Here, we investigated the effects of dispersants used in treating a recent oil spill, as well as that of crude oil, on vertebrates by using the zebrafish (Danio rerio) model species, which has been widely used in empirical studies of both early embryonic development and adult physiology. Chronic exposure to crude oil resulted in marked developmental abnormalities, including pericardial edema, abnormal trunk vessel development, retardation of axonal branching, and abnormal jaw development. Embryonic development was affected more severely by exposure to the oil-dispersant combination than to the oil alone. Thus, the zebrafish in vivo model system suggests that dispersant treatment can have detrimental developmental effects on vertebrates and its potential impact on marine life, as well as humans, should be carefully considered in clean-up efforts at the site of an oil spill.
The effects of angiogenesis inhibitor from the extract libraries of Korean and Chinese medicinal plants were investigated using a protein microarray chip. Protein chip was constructed by immobilization of integrin α5β1 on protein chip base plates and employed far screening active extracts that inhibit the integrin-fibronectin interaction from the extract libraries. The 100 extracts of medicinal plants were obtained from extract bank of National Institute of Crop Science, RDA. The 14 extracts among 100 extract libraries were shown efficient inhibition activity for the interaction between integrin-fibronectin. The medicinal plants of 14 extracts were Vitex negundo var. incisa (Lam.) C.B. Clarke, Epimedium koreanum Nakai, Cedrela sinensis A. Juss, Ipomea aquatica Forsk, Schisandra chinensis Baill, Pulsatilla koreana Nakai, Paeonia lactiflora Pall. var.hortensis Makino, Oenothera odorata, Allium chinense, Allium victorialis var. platyphyllum MAKINO, Polygonatum odoratum Druce var. pluriflorum Ohwi, Hosta lancifolia, Agrimonia pilosa L. var. japonica Nakai and Potentilla chinensis SER. The Paeonia lactiflora, Oenothera, and Agrimonia pilosa from these 14 extracts libraries were shown strong inhibition activity of integrin α5β1.
