        Porcine epidemic diarrhea virus (PEDV) is a porcine coronavirus that causes enteric diseases characterized by watery diarrhea and dehydration in suckling piglets. Concentrated and highly purified viruses are required for the preparation of vaccines, diagnostics, and virus research. Currently, most protocols for virus purification require ultracentrifugation, which can be an instrumental barrier to routine operations in a laboratory. In this study, the efficacy of low-speed centrifugation for virus concentration was examined. The SM98 strain of PEDV was propagated in Vero cells and pelleted by centrifugation for 3 h at high speed (100,000 × g) or for 18 h at low speed (10,000 × g). The efficacy of virus concentration was analyzed by virus titration and western blotting. The amounts of infectious viruses and viral proteins in the pelleted samples obtained by low-speed centrifugation were comparable to those obtained by high-speed centrifugation. Interestingly, the pelleted sample impurity level was lower in low-speed than in high-speed centrifugation. In summary, we describe an efficient, easy-to-perform protocol for the preparation of purified and concentrated PEDV.

        본 연구는 LMO 유전산물의 위해성평가를 위해 바실러스로부터 증폭된 Vip3Aa 유전자를 이용하여 대장균에서 단백질 순수분리 하였으며, MALDI-TOP 분석법을 통해 기존의 알려진 살충성 Vip3Aa 단백질과 동등한 단백질임을 증명하였다. 순수 분리한 Vip3Aa 단백질을 이용하여 꿀벌 과독성 급성섭식독성평가를 수행하였다. 그 결과 무처리군, Hepes buffer, Vip3Aa 단백질 처리군 모두 치사 및 일반 중독증상을 보이는 개체는 발견되지 않았다. 이 결과를 통해 Vip3Aa 단백질은 꿀벌에 위해성을 나타내지 않는다는 결론을 얻을 수 있었다. 본 연구 결과는 향후 국내 LMO 유전자산물 위해성평가에 유용하게 활용될 것이라 사료 된다.

        Recently, the advancement of information and communication technology(ICT) is expanding the connectivity through Internet of Things(IoT), and the media of connection is also expanding from wire/cable transmission to broadband wireless communication, which has significantly improved mobility. This hyperconnectivity has become a key element of the fourth industrial revolution, whereas the supervisory control network of purification plants in korea is operated as a communication network separated from the outside, thereby lagging in terms of connectivity. This is considered the best way to ensure security, and thus there is hardly any consideration of establishing alternatives to operate an efficient and stable communication network. Moreover, security for management of a commercialized communication network and network management solution may be accompanied by immense costs, making it more difficult to make new attempts. Therefore, to improve the conditions for the current supervisory control network of purification plants, this study developed a industrial security L2 switch that supports modbus TCP(Transmission Control Protocol) communication and encryption function of the transmission section. As a result, the communication security performance improved significantly, and the cost for implementing the network management system using Historical Trend and information of HMI(Human Machine Interface) could be reduced by approximately KRW 200 million. The results of this study may be applied to systems for gas, electricity and social safety nets that are infrastructure communication networks that are similar to purification plants.

        We develop a purification process of Hg2Br2 raw powders using a physical vapor transport(PVT) process, which is essential for the fabrication of a high performance acousto-optic tunable filter(AOTF) module. Specifically, we characterize and compare three Hg2Br2 powders: Hg2Br2 raw powder, Hg2Br2 powder purified under pumping conditions, and Hg2Br2 powder purified under vacuum sealing. Before and after purification, we characterize the powder samples through X-ray diffraction and X-ray photoelectron spectroscopy. The corresponding results indicate that physical properties of the Hg2Br2 compound are not damaged even after the purification process. The impurities and concentration in the purified Hg2Br2 powder are evaluated by inductively coupled plasma-mass spectroscopy. Notably, compared to the sample purified under pumping conditions, the purification process under vacuum sealing results in a higher purity Hg2Br2 (99.999 %). In addition, when the second vacuum sealing purification process is performed, the remaining impurities are almost removed, giving rise to Hg2Br2 with ultra-high purity. This high purification process might be possible due to independent control of impurities and Hg2Br2 materials under the optimized vacuum sealing. Preparation of such a highly purified Hg2Br2 materials will pave a promising way toward a high-quality Hg2Br2 single crystal and then high performance AOTF modules.

        반도체 제조공정 중 포토리소그래피는 반도체 제조 중 핵심 기술이며 웨이퍼위에 나노급의 패턴을 제조하는 공정이다. 웨이퍼상의 패턴이 나노급으로 작아지면서 더욱 정밀한 공정을 위하여 photo-resist 용액의 정제 기술이 매우 중요하다. photo-resist 용액 내 soft particle은 포토리소그래피 공정 중 불량을 유발하는데, 현재 개발되어진 멤브레인은 soft particle 제거 성능이 낮다. 따라서 photo-resist 용액 내 soft particle을 제거하기 위하여 PE 지지체 위에 Nylon 고분자를 함침하여 soft particle 정제 특성을 살펴보았다. 또한 팔라듐 나노입자를 합성하여 투과 특성실험에 사용하였다.

        Currently, the commercialization of the 5th Generation (5G) service is becoming more prevalent in domestic communication network technology. This has reduced communication delay time and enabled large-capacity data transmission and video streaming services in real-time. In order to keep pace with these developments, K-water has introduced a smart process control system in water purification plants to monitor the status of the water purification process. However, since wireless networks are based on the public Long Term Evolution (LTE) network, communication delay time remains high, and high-resolution video services are limited. This is because communication networks still have a closed structure due to expense and security issues. Therefore, with 5G in its current form, it is very difficult to accommodate future services without improving the infrastructure of its communication networks. In recognition of these problems, this study implemented the authentication and management function of wireless networks on a wired network management system in the K-water Bansong water purification plant. The results confirmed that wired Local Area Network (LAN) services give a higher security performance than an expensive commercial wireless LAN system. This was achieved by using an Internet Protocol (IP) address management system of wired networks and the packet filtering function of the Layer2 (L2) switch. This study also confirmed that it is possible to create a wireless LAN service that is 3.7 times faster than the existing LTE communication network.

        1970년대 중반 석유 파동 이후 대체 에너지 개발에 대한 관심이 커지면서 그 중에서 혐기성 공정에서 발생하는 바이오가스 생산과 활용 기술 개발에 대한 연구가 진행되고 있다. 바이오 가스의 주성분은 50-70%CH4/30-40%CO2으로 이루어져 있고 이 때 메탄을 > 95% 순도로 농축하면 도시가스와 자동차 연료로 사용이 가능하다. 바이오가스의 정제기술로 막 분리법은 낮은 에너지 소모량, 이동성 및 쉬운 작동성 등의 장점을 가지고 있다. 본 연구에서는 바이오가스를 95% 이상의 메탄으로 농축하기 위해 폴리설폰 중공사막과 다단 분리막 공정을 이용하였다. 분리막 공정에서 압력, 온도, 막면적 등 다양한 공정 변수에서 95% 이상의 메탄 순도를 얻기 위한 연구를 진행하였다.

        포토리소그래피는 웨이퍼 위에 선폭을 나노급으로 줄인 패턴을 제조하는 반도체 제조 공정의 핵심 기술이며 선폭을 줄이고 불량을 줄이기 위하여 photo-resist 용액 제조 및 정제 기술이 중요하다. 특히 photo-resist solution 내에 함유되어 있는 soft particle의 정제가 매우 중요하다. 현재 개발되어 있는 멤브레인 필터는 soft particle 정제 성능이 상당히 미흡한 수준이다. 나일론이나 폴리에틸렌 위에 아라미드 고분자를 함침하여 soft particle 정제 특성을 살펴보았다. 또한 팔라듐 나노입자를 합성하여 투과 특성에 사용하였다.

        Biogas produced in anaerobic digestion process has been considered as a renewable energy source. To recover biogas from the anaerobic effluent and thereby maximize the energy recovery from anaerobic processes, membrane contactor employing hydrophobic porous hollow fiber membrane has been applied. By optimizing the surface hydrophobicity and the porosity of membranes, a desirable CH4 flux could be achieved. Biogas recovered can be upgraded by applying required purification processes for the injection into natural gas grid or for the use as vehicle fuel. My group has synthesized various microporous materials including zeolites, metal-organic frameworks (MOFs) and amine-appended sorbents that are selectively transport CO2 molecule and incorporated them into polymeric films to design high performance gas separation membranes.

        Owing to the development in information and communications technologies have improved the technology for high-speed transmission of massive data, which has changed closed-circuit television (CCTV) video transmission technology. In particular, digitization of the CCTV video format and streaming technology has made it possible to minimize transmission loss and integrate video transmission and camera control(pan/tilt). It has also become possible to provide additional services like remote emergency warning broadcasting with just Internet Protocol (IP). However, because of the structural problems of IP, these changes have also brought about the threat of hacking of CCTV monitoring systems. In this study, we propose a methode to optimize network management by examining cases of enhancement of operational efficiency and security by improving the structure of CCTV monitoring network.

        In this study, finite element (FE) analysis was performed to evaluate the seismic performance of the water treatment plant, which is a major state of the art water treatment plant, to predict tensile cracks and compressive failure. The FE model simulation for two facilities of the water purification plant was made considering the initial conditions, boundary conditions and water effect. For the nonlinear dynamic analysis, seismic analysis was performed using ground acceleration. Tensile cracks and compressive failure are analyzed and the effects on the structures are analyzed. As a result of the analysis, tensile cracks can be predicted to occur in the main structure.
