        The characteristics and spore production of Gonji7ho, Bunhong, and Sunjung fruiting bodies were assessed at different growth stages. The shape of the Pleurotus species fruiting body starts out short and small, then takes on a typical mushroom shape as it grows. Gonji7ho has a long stalk, Bunhong has a short stalk and a wide cap, and Sunjung's cap and stalk dimensions are intermediate. Each variety displayed deep color at the beginning of growth but became steadily lighter with continued growth. The shape of the linkage between the mushroom stalk and cap changed from an initial central position to a lateral position after the growing stage. Gonji7ho cap diameter increased 7-fold from 15.5 mm (5 days of growth) to 37.9 mm (9 days of growth). Growth rates for each growth day measured using the growth percentage of the previous day were 285.5% (5 → 6th day), 182.2% (6 → 7th day), 129.4% (7 → 8th day), and 103.8% (8 → 9th day). This trend was also observed in Bunhong and Sunjung, but Bunhong’s growth rate was more rapid (4.9 fold on day 6, 2.7 fold on day 7) and continued to increase through day 9. Harvest yield, which is of greatest interest to farmers, displayed a similar trend spanning the growth period, as did cap diameter. Gonji7ho harvest yield increased rapidly until day 7 of growth (more than 177%), then growth slowed down beginning around day 8, and further decreased on day 9 (98%). Similar trends were observed in Bunhong and Sunjung. Bunhong showed characteristic rapid growth in harvest yield (4.9 fold compared to the previous day on day 6 and 2.7 fold on day 7), and the increase continued through day 9. A decrease in mushroom harvest yield commonly seen in the late growth stage is thought to be due to the death of some mushrooms and decomposition of cap tissue. Basidiospore content increased with number of growth days but decreased after day 8. Gonji7ho yielded the highest production on day 7 of growth, coinciding with harvest time, with 209,000,000 spores. This trend was also observed in Bunhong and Sunjung. These results will provide researchers with basal data and guide farmers in selecting the optimal harvest day.

        Korean Native Pig (KNP) has a uniform black coat color, excellent meat quality, white colored fat, solid fat structure and good marbling. However, its growth performance is low, while the western origin Yorkshire pig has high growth performance. To take advantage of the unique performance of the two pig breeds, we raised crossbreeds (KNP × Yorkshire to make use of the heterotic effect. We then analyzed the liver transcriptome as it plays an important role in fat metabolism. We sampled at two stages: 10 weeks and at 26 weeks. The stages were chosen to correspond to the change in feeding system. A total of 16 pigs (8 from each stage) were sampled and RNA sequencing was performed. The reads were mapped to the reference genome and differential expression analysis was performed with edgeR package. A total of 324 genes were found to be significantly differentially expressed (|log2FC| > 1 & q < 0.01), out of which 180 genes were up-regulated and 144 genes were down-regulated. Principal Component Analysis (PCA) showed that the samples clustered according to stages. Functional annotation of significant DEGs (differentially expressed genes) showed that GO terms such as DNA replication, cell division, protein phosphorylation, regulation of signal transduction by p53 class mediator, ribosome, focal adhesion, DNA helicase activity, protein kinase activity etc. were enriched. KEGG pathway analysis showed that the DEGs functioned in cell cycle, Ras signaling pathway, p53 signaling pathway, MAPK signaling pathway etc. Twenty-nine transcripts were also part of the DEGs, these were predominantly Cys2His2-like fold group (C2H2) family of zinc fingers. A protein-protein interaction (PPI) network analysis showed that there were three highly interconnected clusters, suggesting an enrichment of genes with similar biological function. This study presents the first report of liver tissue specific gene regulation in a cross-bred Korean pig.

        Although assisted reproductive technology is very useful to develop novel and therapeutic biomaterials for reproduction, research on molecular mechanism of folliculogenesis in pig is not clear. Therefore, the alteration of gene expression during follicular development in pigs was examined in this study. The expression of folliculogenesis-related genes was quantified in preantral (250~300 μm) and antral (>300 μm in diameter) follicles, and overall gene expression was evaluated by a genome-wide microarray. The microarray results showed that 219 genes were differentially expressed, and of those, 10 and 22 known genes showed higher and less expression at the preantral stage than at antral stages, respectively. Among them, the expression of NR0B1, PPARG, GATA4, and ANXA2 genes related to folliculogenesis was validated by quantitative real-time PCR analysis. The expression of PPARG and GATA4 genes were increased at antral stages, but a significantly stage-specific increase (p<0.05) was only detected in annexin A2 (ANXA2) in antral-stage follicles. The expression of NR0B1 genes was increased at preantral stage and these patterns of gene expression were comparable to the results obtained by microarray analysis. We propose that the systematical regulation of genes supporting specific follicle stage should be employed for improved in-vitro folliculognesis.

        The miniature pig is considered to be a better organ donor breed for xenotransplantation than other pig breeds because the size of the organs of the miniature pig is similar to that of humans. In this study, we aimed at identifying differentially expressed genes in the miniature pig ovary during pregnancy. For this, we used the miniature pig ovary model, annealing control primer‐based reverse transcription polymerase chain reaction (PCR), quantitative real‐time PCR (qRT‐PCR), and northern blotting analysis. We identified 13 genes showing differential expression on the based of pregnancy status and validated 8 genes using qRT‐PCR. We also sequenced the full‐length cDNA of ephrin receptor A4 (EphA4), which had a significant difference in expression level, and validated it by northern blotting. These genes may provide a better understanding of the cellular and molecular mechanisms during pregnancy in miniature pig ovary.

        The pinewood nematode, Bursaphelenchus xylophilus (Steinner & Buhrer) Nickle, has two different life stages according to several environmental factors: dispersal stage and propagative stages. The dispersal stage is closely related to the migration to other host pines, whereas the propagative stage is coupled to the direct cause of pine wilt. To establish expressed sequence tag (EST) database of two life cycles of B. xylophilus, subtractive EST libraries were constructed using suppressed subtractive hybridization (SSH). From 3,072 and 3,840 sequences of dispersal- and propagative-specific stage cDNA libraries, 1,927 and 2,604 clusters were generated, respectively, which were annotated by BLASTx and Gene ontology (GO). A total of 1,112 (57.7%) and 1,215 (46.7%) clusters from the dispersal- and propagative-specific stage cDNA libraries respectively had the matched BLASTx hits (E≤10-2), among which 913 (47.4%) and 960 (36.9%) were classified into three categories in Gene ontology. From GO database, some respective stage-specific genes were searched and estimated the relative transcripts level according to stages using the quantitative real-time PCR.

        The diamondback moth, Plutella xylostella parasitized by its endoparasitoid Cotesia plutellae undergoes various physiological alterations which involves immunosuppression and developmental arrest. Its symbiotic virus, C. plutellae bracovirus (CpBV) is highly essential for their successful parasitization which possesses more than 136 genes encoded. CpBV15βunique in CpBV genome is expressed at low levels in early and at higher levels during late parasitization period. This gene product alters the hemocyte-spreading behavior through inhibition of protein synthesis under in vitro conditions. In the current study, we investigated its specific involvement in physiological functions in the host by transient expression and RNA interference techniques. The open reading frame of CpBV15βwas cloned into a eukaryotic expression vector and this recombinant CpBV15β was transfected into healthy non-parasitized 3rd instar P. xylostella by microinjection. CpBV15βwas expressed as early as 24 h and was consistent up to 72 h. Due to the expression of this gene, the hemolymph storage protein levels were significantly reduced and the ability of the hemocytes to adhere and spread on extracellular matrix was altered or reduced, wherein CpBV15βwas detectable in the cytoplasm of hemocytes based on indirect immunofluorescence assay. To confirm the role of CpBV15β, its double stranded RNA could efficiently recover the functional efficacy of hemocytes towards non-self and synthesis of storage proteins. Thus these results clearly demonstrate the role of CpBV15βin altering the host physiology by involving in cellular immune response and host protein synthesis.

        Hemolin is a member of the immunoglobulin (Ig) superfamily and contains four Ig domainsthat are similar to neural cell adhesion molecules. It has been regarded as a recognition molecule at immune challenge in insects. This study showed that hemolin of Plutella xylostella was expressed during pupal and adult stages but absent in all larval instars without any immune challenge. It is, however, strongly induced by the injection of Escherichia coli or its lipopolysaccharide in hemocytes, fat body and gut. A double-stranded RNA (dsRNA) interference experiment revealedits role in activation of prophenoloxidase (PPO) in the hemolymph during bacterial infection. Also its involvement in cellular defense was investigated in its mediation of the adherence of hemocytes to rat blood erythrocytes which was knocked down by its dsRNA. Finally, its physiological significance in pupal stage was confirmed by using dsRNA, which significantly prevented adult development. Therefore, it is concluded that hemolin plays roles in both immune and adult development in P. xylostella.

        TCR subunits are members of membrane-bound receptors which allow the fast and efficient elimination of the specific fish pathogens have regulated function in adaptive immunity. Sequence structure of TCR subunits have been reported for various teleosts, but the information of each TCR subunit functional characterization through expression analysis in fish was unknown. In this study, we examined the gene expression of TCR subunits in the early developmental stages and observed transcript levels in various tissues from healthy adult olive flounder by RT-PCR. The mRNA expression of alpha subunit was already detected in the previous hatching step. But the transcripts of another TCR subunit were not observed during embryo development and increased after hatching and maintained until metamorphosis at the same level. It was found that all TCR subunits mRNAs are commonly expressed in the immune-related organ such as spleen, kidney and gill, also weak expressed in fin and eye. TCR alpha and beta subunit were expressed in brain, whereas gamma and delta were not expressed same tissue. The sequence alignment analysis shows that there are more than 80% sequence homology between TCR subunits. Because it has a high similarity of amino acid sequence to expect similar in function, but expression analysis show that will have may functional diversity due to different time and place of expression.

        Rice is an important model species and one of the most staple crops of the world. The use of rice appropriate promoters suitable for a specific target transgene is important for the control of spatial and temporal transgene expression. To isolate rice tissue-specific promoters, we exploited the potential of whole genome microarrays in 17 stages: callus, germinating seed, leaf, root, the size of the panicles before heading (1, 3, 5, 8, 10, 15, 20, and 22 cm), and the number of days after pollination (1, 3, 5, 11, 21 DAP) using a 300 K Rice Genome Microarray, covering 31,439 genes of the rice. Eight candidate genes for tissue-specific expression were selected in various organs and stage of reproductive development in rice: Histone H4 for constitutive expression, Dehydrin DHN1 for callus-specific expression, germinating seed-specific hypothetical protein, root-specific hypothetical protein, DNA topoisomerase and Retinoblastoma for expression at panicles before heading, heading-specific profiling, and invertase for expression at seed after pollination. Promoter regions of the selected genes were isolated and fused to the β-glucoronidase (GUS) reporter gene, and the constructs were introduced into rice plants. These promoters are highly active in the tissue-specific manner of rice and can be useful for the spatial and temporal enhancement of target gene(s).
