Background: Mahonia Nepalensis DC. (Hoang lien o ro), the specie of the family Berberidaceae, is widely distributed in the high mountainous areas at altitudes 1700 – 1900 m of Vietnam. It is found that the stem of Mahonia nepalensis indicated anti-inflammatory, anti-bacterial and antifungal activities and they are used particularly for the treatment of eczema, psoriasis, and other skin conditions. However, no study on the antioxidant and anti-cancer activities of Mahonia Nepalensis stem has been previously reported. The aim of this study was to evaluate anti-oxidant and anti-cancer activities of Mahonia Nepalensis stem. Methods and Results: The stem pieces of Mahonia Nepalensis were dried and extracted three times with 100% methanol. After that, the extract was suspended in distilled water and then partitioned with n-hexane, ethyl-acetate (EtOAc) and butanol (water saturated BuOH) fractions were then evaporated using a vacuum rotary evaporator. Evaluation of the anti-oxidative activity of Mahonia Nepalensis was carried out using a DPPH (2,2-diphenyl-1-picrylhydrazyl) radical-producing system. The results revealed that the ethyl acetate fraction of M. nepalensis possessed higher potential DPPH radical scavenging activity (IC50, 81.88 ± 1.33㎍/㎖) than other fractions as well as BHT (2,6-Di-tert-Butyl-4-methylphenol) (IC50, 250.49 ± 1.60㎍/㎖). The reducing power assay was also investigated and EtOAc fraction showed higher absorbance values than other fractions. At 1.0 mg/ml concentration, EtOAc fraction showed absorbance of 1.72, be higher than Ascorbic acid. Cell viability was evaluated according to the MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyl tetrazolium Bromide) assay. By MTT assay, all fractions showed a significant reduction in cell viability on COLO 205 (Human colon carcinoma cell) at the highest concentration tested (200㎍/ ㎖) with over 70% decrease in cell viability was obtained, and the highest significantly inhibiting effect occurred in butanol fraction with approximately 90% reduction in cell viability. Conclusion: We demonstrated that Mahonia Nepalensis stem extract has highly potential in anti-cancer activity. Further studies are necessary in order to explore the variety of Mahonia Nepalensis stem to be applied as a valuable natural material.

Optimum concentrations of anesthetic clove oil and anesthetic lidocaine-HCl were determined for a species of adult marine medaka, Oryzias dancena, over a range of salinity conditions, and investigated in a transport simulation experiment by analyzing various water and physiological parameters. Research indicated that the higher the concentration of anesthetic at each salinity, the shorter the anesthesia time at each salinity. At each concentration, fish were anesthetized slower at water salinities over 10 ppt (P<0.05). Anesthesia time at 10 ppt was faster than any other salinity. In 10 ppt salinity, the dissolved oxygen (DO) concentrations and respiratory frequencies of the clove-oil-administered groups decreased until 48 hours (P<0.05), whereas the NH4 + and CO2 concentrations increased until 48 hours (P<0.05). In same period, the DO, NH4 +, and CO2 concentrations and respiratory frequencies all decreased as the clove oil concentration increased (P<0.05). The trends in the DO, NH4 +, and CO2 concentrations and respiratory frequencies in the lidocaine-HCl-administered groups were similar to those in the clove-oil-administered groups. In conclusion, clove oil and lidocaine-HCl are effective anesthetics, improving the transportation of the marine medaka. The results from this study will contribute to safe laboratory handling of the marine medaka, which are commonly required by many research studies and experiments.

The marine medaka, Oryzias dancena is a suitable sample as a laboratory animal because it has a small size and clearly distinguishes between female and male. Data on the growth and maturity of the diploid and triploid sea cucurbit species suitable for laboratory animals are very useful for studying other species. Triploidy was induced in the marine medaka by cold shock treatment (0°C) of fertilized eggs for 45 min, applied two minutes after fertilization. The diploid and triploid male fish were larger than their female counterparts (P<0.05), and the concentrations of thyroid stimulating hormone (TSH) and thyroxine (T4) were higher in the induced triploids over 1 year (P<0.05). In both the diploid and tri-ploid groups the concentrations of TSH and T4 were higher in the male fish than in the females (P<0.05), while the testo-sterone and estradiol-17ß concentrations in the induced triploids were lower than in the diploids (P<0.05). The gonadosomatic index (GSI) of the triploid fish was lower than that for the diploids, and the GSI for females in each ploidy group were higher than that for the males. For both groups the GSI was highest at 4 months of age, and decreased thereafter to 12 months. Analysis of the gonads of one-year-old triploid fish suggested that the induction of triploidy probably causes sterility in this species; this effect was more apparent in females than in males.

The optimum concentrations of clove oil as an anesthetic for olive flounder (Paralichthys olivaceus) and the stress response of the fish to clove oil anesthesia were determined over a range of water temperatures, and investigated in a simulated transport experiment using analysis of various water and physiological parameters. While the time for induction of anesthesia decreased significantly as both the concentration of clove oil and water temperature increased, the recovery time increased significantly (P<0.05). The plasma cortisol concentration in fish at each temperature increased significantly up to 12 h following exposure (P<0.05), then decreased to 48 h (P<0.05). The DO dissolved oxygen concentrations, pH values, and the fish respiratory frequencies decreased over 6 h following exposure to clove oil in all experimental groups (P<0.05), whereas the NH4 + and CO2concentrations in all experimental groups increased up to 6 h (P<0.05). The pH values and DO concentrations increased with increasing clove oil concentration (P<0.05) in the 6 h following exposure, and the CO2 and NH4 + concentrations and the respiratory frequencies decreased with increasing clove oil concentration (P<0.05). The results of this experiment suggest that clove oil reduced the metabolic activity of olive flounder, thus reducing NH4 + excretion and O2 consumption. In conclusion, clove oil appears to be a cost-effective and efficient anesthetic that is safe for use and non-toxic to the fish and users. Its use provides the potential for improved transportation of olive flounder.

The Science and Technology process focuses on a number of areas where science and technology development provides a major opportunity to innovate water resources management. A number of areas have been carefully selected and are described below. Main focus 1: Efficient water management Today’s water management continues to be highly inefficient in many respects, seriously hampering the sustainable management of water resources, delivery of services and the protection of human health and the environment. In the years to come, major efforts will need to be made to improve water efficiency as a means to secure water resources, adapt to climate change, or operate water services systems at lower costs. Sub Focus: Urban water efficiency, Agricultural water efficiency, Industrial water efficiency, Energy efficiency in water and waste water systems Main focus 2: Resource recovery from water and waste-water systems Increasingly, water and waste-water systems are recognized as potential sources of recovering water, energy, nutrients and other materials. At the same time, the re-use of waste water in agricultural, industrial and urban applications is gaining momentum. The development of re-use and recycling within the water and waste-water sectors provides major opportunities for improving environmental performance, creating climate benefits and reducing costs. Sub Focus: Water re-use and recycling technologies, Energy recovery and production from water and waste water cycles, Nutrients recovery from waste water Main focus 3: Water and Natural disasters As the devastating impact of recent natural disasters such as the flood occurred in Thailand in 2011, indicates, mankind is vulnerable to extreme weather events in developing and even in developed countries. Clearly, such extreme events have always been part of our life and may be caused by climate change. This makes water related problems, especially, such as floods and droughts, more difficult to analyze and predict. Climate change is predicted to have a range of serious consequences, some of which will have impact over the longer term, like droughts, while some have immediate and obvious impacts, such as intense rain and flooding. Floods and droughts are major natural disasters involving loss of life and the destruction of property. So, we may need advanced technologies and measures in order to cope with natural disasters. Sub Focus: Climate change : impact assessment and adaptation, Drought analysis and management, Urban floods and damage reduction studies, RS and GIS applications for natural hazards Main focus 4: Smart technology for Water Water management is increasingly influenced by developments in Smart Technology(ST). Better use of this ST in measuring, monitoring and distributing water can lead to a significant contribution in overcoming water management challenges in the 21st century. Bridging the ‘divide’ between the water and ST sectors will be important in the development and (wide-scale) application of new smart technologies in the water sector. Sub Focus: Urban and Irrigation water management and ST, Integrated and intelligent river basin water management and ST, Design and implementation of smart water grid, Water resources management and Big Data Main focus 5: Understanding and managing ecosystem services for water Ecosystems provide valuable services underpinning water security and the sustainable management of water resources. In recent years, major advances have been made in understanding the role of ecosystems in, for example, reducing floods, recharging aquifers, or protecting and improving water quality. Sub Focus: The science and technology of natural and green infrastructure management, The economic valuation of ecosystem services for water, The payment for ecosystem services for water quality and delivery
