        In this study, the effects of heat-moisture treatment (boiling or steaming for 45 min) prior to hot air drying (70 o C for 8 h) on water activity (Aw), moisture (MC), Brix, color, and texture of dried sweet potato slices were investigated to identify the best pretreatment condition for producing dried sweet potato with the best eating quality. Dried sweet potato slices pretreated by boiling (BL) showed significantly lower Aw, MC, and hardness while Brix was significantly higher than with steaming (ST) treatment. There were no significant differences for the color, springiness, cohesiveness, gumminess, and brittleness indexes. At 8 h drying, the Aw, MC, Brix, and hardness of the BL and ST samples were 0.81 and 0.82, 19.71% and 25.53%, 53.80% and 49.40%, and 20.49 kg/cm 2 and 31.98 kg/ cm 2 , respectively. This information will be useful for manufacturers in the production of dried sweet potato slices. These findings provide evidence of the feasibility of heat-moisture treatments in the production of dried sweet potato.

        The culture of the intestinal epithelium into three dimensional (3D) structures typically termed organoid culture. Organoid culture is based on the ability of intestinal stem cells (ISCs), at the base of the crypt, perpetually to divide and produce a fully differentiated, polarized epithelium. Leucine-rich-repeat-containing G-protein-coupled receptor 5 (Lgr5) positive ISCs isolated from the intestine can form organoids in long-term culture. Thus, when cultured under the appropriate 3D conditions, single Lgr5+ ISCs undergo cycles of self-renewal, differentiation and morphogenesis, and self-organize into crypt-villus domains that house cycling ISCs and differentiated intestinal epithelial cells (IECs). In this study, we performed isolation, characterization and consecutive subculture of small intestinal crypts from BALB/c-nude mouse. Briefly, isolated mouse crypts were embedded in matrigel, cast into 40 μL droplets at the bottom of well in a 48-well plate. Following polymerization, the gels were overlaid with ISCs expansion medium containing B27, N2, N-acetylcysteine, epidermal growth factor, noggin, and R-spondin 1. As a result, mouse crypt-derived ISCs had enteroids and spheroid morphologies. We also confirmed by quantitative real-time RT-PCR that expression of ISCs-related specific genes (Lgr5, sox9) and IECs-related specific genes (chromogranin A, defensin-5, mucin-1, mucin-2, and villin) was maintained at eight passages or more. Thus, we observed that expression of specific markers and consecutive self-renewing in the mouse small intestinal crypt-derived organoids.

        Particulate matter (PM) has recently been considered one of the most harmful air pollutants to public health. Plants have been known to degrade and deposit particle pollutants with epicuticular wax (EW), and this capacity can be influenced by environmental conditions including relative humidity (RH). The present study examined the effects of RH on EW generation and PM deposition upon leaf surfaces within Asplenium nidus ‘Avis’. The plants were treated in growth chambers with two levels of RH (low: 30% - 40% and high: 80% - 90%) for a period of four weeks, and subsequently exposed to a 30 μg・m-3 concentration of TiO2 particles as a PM resource for 72 hours. The EW ultrastructure on the leaf surface was observed as the thin films type, which was not morphologically changed in the condition of low or high RH treatment. For four weeks of RH treatment, the fresh weight and leaf area per plant were not significant between low and high RH treatment, while dry weight was significantly higher in the high RH condition. We also found that greater amounts of EW per fresh weight, dry weight and leaf area were generated in high RH. However, the total amounts of PM deposition (surface PM + in-wax PM) of the plants were higher within the low RH treatment with a higher proportion of surface PM. In contrast the proportion of in-wax PM was 15% higher within the high RH. These results suggest that EW generation is affected by air humidity and that proportion of PM deposition in the EW layer were influenced by the amount of total wax load.

        Natural killer (NK) cells are innate lymphocytes that aid in the protection of the host from infectious diseases and cancer. Regulation of cytotoxicity in NK cells is mediated by inhibitory receptors that bind major histocompatibility complex class I (MHC-I) molecules on target cells. Human myelogenous leukemia K562 cells are readily attacked by NK cells, because K562 cells expressed very low levels of MHC-I molecules for inhibitory NK cell receptors. In this study, we compared the ability of γ-irradiated- or mitomycin C (MMC)-treated K562 feeder cells to support expansion and activation of canine NK cells. We isolated CD5 negative cells from canine peripheral blood mononuclear cells by immunomagnetic separation and co-cultured with γ-irradiated (100 Gy)- or MMC (20 μg/mL)-treated K562 cells in the presence of interleukin (IL)-2, IL-15 and IL-21 for 21 days. As a result, number of CD5 negative cells, co-cultured with γ-irradiated- or MMC-treated K562 cells (56.72 ± 13.77 fold or 32.99 ± 10.83 fold), was increased than those of CD5 negetive cells (2.99 ± 1.42 fold). Also, we found that gene expression markers of activated NK cells such as NKp30, NKp44, NKp46, Ly49, NKG2D, CD244, perforin, and granzyme B and production of interferon gamma were similarly upregulated in NK cells co-cultured with γ-irradiated- or MMC-treated K562 cells, respectively. Next, we observed that cytotoxicity of NK cells co-cultured with γ-irradiated K562 cells was more sensitively reacted to canine mammary carcinoma cells than those of MMC-treated K562 cells. These results revealed that γ-irradiated K562 cells are more efficient feeder cells than MMC-treated K562 cells for enhancing NK cells expansion and activation.

        Purpose: Despite the effort to bridge the disconnect between educational preparation of competent nurses and employers' expectation of new graduates, there is little known about what nursing competencies are required for new graduates. This paper aims to identify the nursing competencies expected of new graduates, both globally and in the specific Korean context. Methods: An integrative literature review was undertaken. International and Korean computerised databases were searched and 72 relevant papers were appraised using the Mixed Methods Appraisal Tool and the Data Relevance. Twenty-eight studies were selected for final data analysis. Results: Three themes with eleven competency areas were discovered: new graduates are expected to be competent in delivering an evidence-based practice with a patient-centered approach as professional nurses. Competency in evidence-based practice involves being able to integrate theory and skills, utilise contemporary evidence, and undertake a systematic approach. Patient-centred care involves ensuring patient safety, educating the patient and their family, and demonstrating sound interpersonal communication skills with a collaborative approach. New graduates are also expected to practice legal and ethical nursing care, undertake continuing professional development, and demonstrate a capacity for clinical management and appropriate professional attributes. Conclusion: Identifying nursing competencies for new graduates can add a level of detail to the current nursing competencies required for new graduates in the global and the Korean context. The findings can be used not only to inform education providers of potential gaps in preparation for practice of new graduates, but also be used to identify support needed when graduates enter nursing workforce.

        Jeju Haenyeo have been playing a crucial role in Jeju economy for a long time. However, the number of Jeju Haenye has decreased since the 1970s, despite the efforts from the government and Jeju Provincial Office including a variety of welfare and benefits. In order to understand the unique culture of Jeju Haenyeo, and to provide a good program to help transfer their muljil skills, a qualitative research including in-depth interviews and participant observation in combination of an ethnographic method was performed. For the qualitative research, the interview preparation was reviewed, the language and culture of local communities were understood, and the location of the researcher was determined. Then, the following procedures were carried out; selecting informants, gaining their confidence, forming a rapport with them, and collecting data. For a preliminary survey, The first author visited Jeju alone for 10 days from late June to early July in 2015. The first author walked around the area and observed the dialects and life of local people from the viewpoint of an outsider. For interviews, four still active and three former Jeju Haenyeo were selected from skilled workers with diving experience of at least 20 to 60 years. The transfer of Jeju Haenyeo’s muljil skills, and their education and training vary, depending on the community culture of each village. As a result, the transfer of Jeju Haenyeo’s muljil skills is carried out not just at the level of maternal succession, but it is also performed by the village community that participate in co-childcare and joint training of diving skills.

        Cu/PET composite films are widely used in a variety of wearable electronics. Lifetime of the electronics is determined by adhesion between the Cu film and the PET substrate. The formation of an anisotropic nanostructure on the PET surface by surface modification can enhance Cu/PET interfacial adhesion. The shape and size of the anisotropic nanostructures of the PET surface can be controlled by varying the surface modification conditions. In this work, the effect of Cu/PET interface nanostructures on the failure mechanism of a Cu/PET flexible composite film is studied. From observation of the morphologies of the anisotropic nanostructures on plasma-treated PET surfaces, and cross-sections and surfaces of the fractured specimens, the Cu/PET interface area and nanostructure width are analyzed and the failure mechanism of the Cu/PET film is investigated. It is found that the failure mechanism of the Cu/PET flexible composite film depends on the shape and size of the plasmatreated PET surface nanostructures. Cu/PET interface nanostructures with maximal peel strength exhibit multiple craze-crack propagation behavior, while smaller or larger interface nanostructures exhibit single-path craze-crack propagation behavior.

        This study aimed to examine the effect of a mild elevation in serum cholesterol level in a porcine coronary overstretch restenosis model using a balloon angioplasty catheter or drug-eluting coronary stent. Pigs were divided into two groups and were fed a commercial normal diet (CND, n = 4) or a high-fat diet (HFD, n = 4) for 5 weeks. Coronary overstretch injury by balloon angioplasty or stent implantation was induced in the left anterior descending and left circumflex artery after 1 week of feeding. Histopathological analysis was performed at 4 weeks after coronary injury. During the experiment, the total cholesterol level in the HFD group increased by approximately 44.9% (from 65.9 ± 3.21 mg/dL at baseline to 95.5 ± 9.94 mg/dL at 5 weeks). The lumen area in the CND group was reduced in comparison with that in the HFD group after balloon angioplasty. After stent implantation, the injury score showed no significant difference. There were significant differences in the neointimal area (2.7 ± 0.33 mm2 in the CND group vs. 3.3 ± 0.34 mm2 in the HFD group, p<0.05), lumen area (2.6 ± 0.54 mm2 in the CND group vs. 2.0 ± 0.33 mm2 in the HFD group, p<0.05), and percent area stenosis (52.0 ± 7.96% in the CND group vs. 62.4 ± 5.15% in the HFD group, p<0.05). Body weight change was not different between the two groups. Increased serum cholesterol level activated vascular smooth muscle cell proliferation in the porcine coronary overstretch model.

        We conducted a survey of open clusters within 1 kpc from the Sun using the astrometric and photometric data of the Gaia Data Release 2. We found 655 cluster candidates by visual inspection of the stellar distributions in proper motion space and spatial distributions in l 􀀀 b space. All of the 655 cluster candidates have a well de ned main-sequence except for two candidates if we consider that the main sequence of very young clusters is somewhat broad due to di erential extinction. Cross-matching of our 653 open clusters with known open clusters in various catalogs resulted in 207 new open clusters. We present the physical properties of the newly discovered open clusters. The majority of the newly discovered open clusters are of young to intermediate age and have less than 50 member stars.
