        Immunoprecipitation-based high performance liquid chromatography (IP-HPLC) is a type of modified enzyme-linked immunosorbent assay (ELISA) that uses protein A/G (or antibody)-conjugated beads instead of the antibody-conjugated wells used in ELISA. In order to determine the fidelity of IP-HPLC, the author used 83 antisera to identify protein expression changes caused by cisplatin treatment in KB human oral cancer cells. KB cells were cultured for 12 or 24 hours with 10 ug/mL cisplatin. The results obtained by IP-HPLC were comparable with published cisplatin data, although ELISA was not conducted in the present study. Cisplatin dominantly reduced the levels of proteins associated with cell proliferation, transcription factors, growth factors, cytoskeletal proteins, and cellular differentiating factors, but on the other hand, apoptosis-related factors, oncogenes, and protective proteins were usually up-regulated, presumably to address cisplatin-induced DNA damage. In particular, cisplatin directly inactivated genomic DNA by down-regulating histone H1 and demethylase and by up-regulating deacetylase. Cisplatin also rapidly induced p53 overexpression and mitochondria-mediated endogenous apoptosis occurred after 12 hours of cisplatin treatment, although this was almost completely replaced by FASL/FAS-mediated exogenous apoptosis after 24 hours. This preliminary study was conducted to investigate the anticancer effect of cisplatin on the KB human oral cancer cells and to determine the fidelity of IP-HPLC data. It was concluded that IP-HPLC is useful for identifying profile changes of genome wide essential proteins and signaling changes of major molecular pathways.

        This study was conducted to investigate the distributional characteristics of ground beetles and to provide basis information for biodiversity management including the ground beetles in the Naejangsan National Park area. Pitfall traps were installed throughout 20 sites within Naejangsan National Park during 2008 to 2011 to collect ground beetles. A total of 2,409 collected ground beetles were identified with 35 species belonging to 19 genera of 8 subfamilies. Coptolabrus jankowskii jankowskii, Eucarabus sternbergi sternbergi, and Pterosticus audax were dominant at the core area, while Pheropsophus jessoensis, Synuchus nitidus, Synuchus cycloderus, and Chlaenius naeviger were dominant at the border of the National Park and adjacent to the road or grassland. These differences of dominant species also affected to the similarity of species composition between core and border area, and caused increasing dissimilarities between sites with cluster analysis. Although the result of the present study was a case study using ground beetles, it will be helpful to develop a management strategy of biodiversity conservation in Naejangsan National Park and its surroundings.

        This study was conducted to assess the environmental risk of CMV resistant GM pepper and to evaluate the effects of the transgenic pepper (Line: H15) with CMV resistant gene and non-transgenic peppers (Line: P2377, PM Singang, Berota) without gene introduction on the insect community in the three pepper cultivation areas. Sampling was conducted using yellow sticky traps in Deokso, Anseong and Ochang fields, respectively. The total number of insect individuals caught by sticky trap was not shown significant differences between GM and non-GM pepper at Deokso, Anseong and Ochang fields, respectively. Also, there were no significant differences in density of aphids caught on sticky traps between GM and non-GM pepper, and the species richness and species diversity was no statistical difference between the types of crops, GM and non-GM at three experimental pepper fields.

        Hanok recently is getting attention by Korean wave and the eco-friendly construction. However it has the not a few . weak points to live in modem and been recognized as a big problem such as efficiency problem, hight cost to construct and maintenance difficulty to be solved. The solution are the modularity and weight lightening of Hanok but the roof is one of the most difficult parts of it, the roofrepresents the beauty of Hanok with graceful catenary line, filling timber and soil should be improved to the modem materials and construction methods as an alternative. As the result, this study conceives the improvement figuring out the problem and the case study review as well as suggests the valuation of feasible plan and the potential for modularity by application of the actual field. The roof is consisted of the framework as the constructable role on the upper part of roof board, the insulation and air layer as well as additional roof board should be installed to make easy the roof construction and attach the vapor permeability water proofed paper. Accordingly, Additional rafter, Diaphragm board and Panel can be used as the alternatives. and this reports the possibility of workability, modularity applied in the field. The results of this study are expected to improve the efficiency of the waterproofing of roofs, insulation, maintenance, etc and so on contribute to the pervasiveness of the hanok.

        Spider community was surveyed at Nakdonggang River which is the longest river (525 km and 23,384 km2) located East-south part of Korea. Nakdonggang river is developing for water management and environmental conservation including restoration of river ecosystem under “The Four-River Restoration Project”. Sampling was made 3 occasions from June to August in 2009. Eight survey sites were selected from both sides of river in Andong, Sangju, Gumi and Seongju prefectures. Spiders were collected with sweep net for on the plants above ground and pitfall traps for on the ground. A total of 94 species of 71 genera in 24 families were identified from 805 collected spiders. Species richness was high in Salticidae (18 species, 19.1%), Araneidae (12 species, 12.8%), Lycosidae (9 species, 9.6%) in order. Wandering spiders (55 species occupied 58.5%, 493 individuals occupied 61.2%) were higher than webbing spiders (39 species occupied 41.5%, 493 individuals occupied 61.2%) in species richness and abundance. Dominant species were Tetragnatha praedonia (Tetragnathidae) and Neoscona adianta (Araneidae) in webbing spiders, and Anahita fauna (Ctenidae), Pirata procurvus (Lycosidae) and Gnaphosa kompirensis (Gnaphosidae) in wandering spiders with abundance. Collectively, most spiders collected from this study were water preferring spiders and showed different community structure compared to other reports from terrestrial spider fauna. Present result will bevaluable to compare the effect of development on the arthropod fauna after completion of the project.

        This study was conducted to estimate the control thresholds (CTs) at different larval densities of Oides decempunctatus Billberg (Coleoptera: Chrysomelidae) of Campbell early in the vineyard and investigated life cycle. Each stage of O. decempunctatus was sampled 18 times from May to September in 2010~2012. The seasonal occurrence of O. decempunctatus showed the highest peak in mid-late June and mid-late August. Overwintered O. decempunctatus's eggs were hatched from late May to early June. Larva period was from late May to mid July and adults appeared in mid July. The percentage of leaf damage (Y) of Campbell early inoculated by different densities of O. decempunctatus (X, no. of larvae/fruiting mother branch) for six weeks was estimated by Y= 0.498X+2.041 (R2=0.988) during vegetation period. The decreasing rate of soluble solid (Y) after grape harvest of Campbell early damaged by different densities of O. decempunctatus (X) was estimated by Y= - 0.046X+15.3 (R2=0.8543). Based of the relationships between the densities of O. decempunctatus larvae and the index of reducing soluble solid of Campbell early, the number of larvae (2nd to 3rd instar) which decreased less than 15°Bx loss of soluble solid was determined as the injury level of 7/fruiting mother branch.

        운간초의 분화 품질을 향상시키기 위하여 ‘Kumoma’ 와 ‘Kumoma-Gusa’ 두 품종의 생장과 개화에 미치는 영향을 구명하고자 식물생장억제제 (PGRs) 4종을 처리 하였다. 식물생장억제제 종류와 처리농도는 paclobutrazol (10, 20, 40, 80 mg·L-1), flurprimidol (5, 10, 20, 40mg·L-1), daminozide (500, 1000, 2000, 4000mg·L-1), chlormequat (50, 100, 200, 400 mg·L-1)이고 엽면살포 와 토양관주 방법으로 처리하였다. Paclobutrazol 40 mg·L-1 처리는 운간초의 두 품종에서 초장과 화경 장을 줄이는데 효과적이었다. ‘Kumoma’ 품종에서, paclobutrazol 40 mg·L-1 엽면살포와 토양관주 처리방 법은 초장을 각각 12.6, 12.5 cm로, 화경장은 3.4, 3.3 cm로 줄일 수 있었다. ‘Kumoma-Gusa‘ 품종에서 는 paclobutrazol 토양관주는 처리 농도가 증가함에 따라 초장은 13.2-10.7 cm, 화경장은 3.9-2.0 cm까지 줄일 수 있었지만 꽃의 수는 대조구와 비교했을 때 현 저하게 감소하여서 20개까지 감소하였다. Flurprimidol 20mg·L-1 스프레이와 토양관주 처리는 두 품종 모두 초장과 화경장을 줄일 수 있었다. ‘Kumoma’ 품종에 서 flurprimidol 10 mg·L-1 스프레이 또는 토양관주 처리는 개화 품질을 향상시킬 수 있었고 대조구와 비 교해 꽃의 수는 44.7개 이상, 출하 일수는 5일 정도 빨리 출하 할 수 있었다. ‘Kumoma-Gusa‘ 품종에서 PGRs처리에 의한 초장 과 화경장 억제효과가 가장 큰 것은 paclobutrazol 80mg·L-1과 flurprimidol 40mg·L-1 이었고 paclobutrazol 과 flurprimidol 스프레이와 토양관주 처리에서 각각, 초장은 10.7, 9.9 cm, 화경장은 2.0, 1.5 cm 로 생장 억제효과가 가장 큰 것을 알 수 있었다. 그러나 flurprimidol 40 mg·L-1 토양관주 처리에서 출하일수는 농도에 따라 3-13일 정도 대조구보다 늦어졌고 꽃의 수는 15.6개로 가장 적은 것을 관찰 할 수 있었다. 두 품종 모두에서 daminozide 처리는 초장과 화경장 이 줄어들지 않았지만 ‘Kumoma’ 품종에서는 대조구 와 비교했을 때 꽃의 수가 증가하였다. Chlormequat 처리는 paclobutrazol 과 flurprimidol 처리와 비교했 을 때 화경장 감소효과는 작았고 처리 식물의 잎은 다른 처리에 비해 연한 녹색을 보여주었다. 두 품종 모두에서 chlormequat 과 daminozide 처리는 생육과 개화 품질에 효과적으로 영향을 주지는 않았지만 꽃의 수는 모든 처리농도에서 41개 이상을 보여주었고, ‘Kumoma-Gusa’ 품종에 chlormequat 토양관주 처리는 최대 꽃의 수 63개를 볼 수 있었다. 이번 결과들은 paclobutrazol 과 flurprimidol 처리가 운간초 분화의 품질 향상을 위한 화경장의 생육을 조 절하는 생장억제제로서 사용 될 수 있음을 보여 주었 고, ‘Kumoma’ 품종에서는 paclobutrazol 40 mg·L-1 스프레이는 생육특성을, flurprimidol 10 mg·L-1 토양관 주 방법은 개화관련 특성의 품질을 향상시키는 생장억 제제의 농도로 추천 될 수 있고, ‘Kumoma-Gusa’ 품종 에서는 flurprimidol 20mg·L-1 스프레이와 flurprimidol 5mg·L-1 토양관주 방법이 운간초 분화의 생육특성과 개 화품질을 향상시키는 생장억제제의 농도로 추천 될 수 있다.

        The pear psylla, Cacopsylla pyricola Foerster (Homoptera: Psyllidae), is a serious insect pest of commercial pear crops. The species, which resides on pear trees throughout its life cycle, is rapidly spreading in some regions of the world. Given the life cycle, it is unclear how such a rapid spread has been facilitated. Presently, the population genetic structure of the species including genetic diversity and gene flow was studied to understand the nature of dispersal and field ecology of the species. Pear psylla was collected from several pear orchards in Korea. The 658-bp region of mitochondrial COI gene and the 716-bp long complete internal transcribed spacer 2 (ITS2) of the nuclear ribosomal DNA were sequenced. Unlikely other previously studied insect pests, the COI-based genetic diversity of the pear psylla was extremely low (maximum sequence divergence of 0.15%). This finding allowed us to conclude that the species may have been introduced in Korea relatively recently, possibly with the phenomenon of genetic bottlenecks. ITS2 sequence-based analyses of phylogeny, population differentiation, gene flow, and hierarchical population structure all concordantly suggested that the pear psylla populations in Korea are neither genetically isolated nor hampered for gene flow. These genetic data are concordant with the dispersal of an overwintering winterform morph outside the non-pear habitat in the fall and the possibility of subsequently longer distant dispersal.

        Diamondback moth (Plutella xylostella L.) belonging to genus Lepidoptera is a notorious pest of cruciferous crops worldwide. We evaluated the bioinsecticidal activity of the liquid cultures (LB and NB) of a bacterial strain, Serratia sp. EML-SE1, isolated from a diseased diamondback moth. The pathogenicity of a bacterial strain to diamondback moth was confirmed by the following procedures: treatment of liquid culture on cabbage leaves, ingestion of inoculated cabbage and mortality response. For the test, twenty 3rd instar larvae of diamondback moth were placed on the Chinese cabbage leaf in a round plastic cage (Ø 10 × 6 cm) and sprayed with the liquid cultures. After 72 hours, insecticidal activity of LB and NB cultures of Serratia sp. against P. xylostella larvae showed 91.7% and 88.3%, respectively. In addition, the bioinsecticidal activity on potted cabbage with 14 leaves in a growth cage (165 × 83 × 124 cm) also was similar to that of plastic cage experiment. Summarized, the Serratia sp. EML-SE1 may be a potent candidate as a bioinsecticidal agent to control diamondbac kmoth.

        We have determined the complete mitochondrial genome of the yellow-spotted long horned beetle, Psacothea hilaris (Coleoptera: Cerambycidae), an endangered insect species in Korea. The 15,856-bp long P. hilaris mitogenome harbors gene content typical of the animal mitogenome and a gene arrangement identical to the most common type found in insect mitogenomes. As with all other sequenced coleopteran species, the 5-bp long TAGTA motif was also detected in the intergenic space sequence located between tRNASer (UCN) and ND1 of P. hilaris. The 1,190-bp long non-coding A+T-rich region harbors an unusual series of seven identical repeat sequences of 57-bp in length and several stretches of sequences with the potential to form stem-and-loop structures. Furthermore, it contains one tRNAArg-like sequence and one tRNALys-likes equence. Phylogenetic analysis among available coleopteran mitogenomes using the concatenated amino acid sequences of PCGs appear to support the sister group relationship of the suborder Polyphaga to all remaining suborders, including Adephaga, Myxophaga, and Archostemata. Among the two available infraorders in Polyphaga, a monophyletic Cucujiformia was confirmed, with the placement of Cleroidea as the basal lineage for Cucujiformia. On the other hand, the infraorder Elateriformia was not identified as monophyletic, thereby indicating that Scirtoidea and Buprestoidea are the basal lineages for Cucujiformia and the remaining Elateriformia.

        The complete nucleotide sequences of the mitochondrial genome (mitogenome) from the white-spotted flower chafer, Protaetia brevitarsis (Coleoptera: Cetoniidae) was determined. The 20,319-bp long circular genome is the longest among the completely sequenced arthropods. This extraordinary length of the genome stemmed from 5,654-bp long A+T-rich region composed of twenty-eight 117-bp tandem repeats, seven 82-bp tandem repeats, and each two 19-bp and 38-bp tandem repeats. The P. brevitarsis contains a typical gene complement, order, and arrangement identical to most common type found in insects. The P. brevitarsis COI gene does not have typical ATN codon. Thus, we also designated it as AAC (asparagine), which is found in the start context of all sequenced Polyphaga within Coleoptera. All tRNAs showed stable canonical clover-leaf structure of other mt tRNAs, except for tRNASer (AGN), DHU arm of which could not form stable stem-loop structure. The 5bp-long motif sequence (TAGTA) that has been suggested to be the possible binding site for the transcription termination peptide for the major-strand also was found betweent RNASer (UCN) and ND1, as have been detected in all sequenced coleopteran insects.

        The 15,389-bp long complete mitogenome of the endangered red-spotted apollo butterfly, Parnassius bremeri (Lepidoptera: Papilionidae) was determined. This genome has a gene arrangement identical to those of all other sequenced lepidopteran insects, which have the gene order of tRNAMet, tRNAIle, and tRNAGln at the beginning. Due to the uncertainty the start codon for COI gene in insect has been discussed extensively. We propose the CGA sequence as the start codon for COI gene in lepidopteran insects, based on complete mitogenome sequences of lepidopteran insects including our P. bremerii and additional sequences of the COI start region from a diverse taxonomic range of lepidopteran species (a total of 51 species belonging to 15 families). As has been suggested in other sequenced lepidopteran insects the 18 bp-long poly-T stretch and the downstream conserved motif ATAGA that were previously suggested to serve as a structural signal for minor-strand mtDNA replication also was found at the 3’-end region of the P. bremerii A+T-rich region. In an extensive search to find out tRNA-like structure in the A+T-rich region, each one tRNATrp-like sequence and tRNALeu (UUR)-like sequence were found in the P. bremeri A+T-rich region, and most of other sequenced lepidopteran insects were shown to have tRNA-like structure within the A+T-rich region, thereby indicating that such feature is frequent in the lepidopteran A+T-rich region. Phylogenetic analysis using the concatenated 13 amino acid sequences and nucleotide sequences of PCGs of the four macrolepidopteran suferfamilies together with Tortricoidea and Pyraloidea well recovered a monophyly of Papilionoidea and a monophyly of Bombycoidea. However, Geometroidea and Noctuoidea were unexpectedly clustered as one group and placed this group to the sister group to Bombycoidea, instead of Papilionoidea in most analyses.

        Invertebrate mitochondrial genome contains 13 protein-coding genes and major start codons for them are ATA (Met) and ATG (Met). However, alternative start codons such as ATT (Ile), ATC (Ile), TTG (Leu), and GTG (Val) also have been suggested from a diverse organism. Approximately 120 complete mitochondrial genome reported showed that the start codon for COI gene evidences an array of diverse designation of COI start codon such as typical ATN, tetranucleotide TTAG and ATAA, newly proposed AAT and AAC and so on. In the case of Lepidoptera, many completely sequenced species showed no typical start codon at the start context of COI and even within the neighboring tRNATyr. In order to clarify, we newly sequenced the beginning context of COI gene, encompassing the neighboring tRNATyr and start region of COI gene from 39 species belonging to eight lepidopteran families. We found the newly sequenced 39 species and 14 available complete lepidopteran mitochondrial genomes all possessed CGA (arginine), which is the first non-overlapping in-frame codon in COI gene. Furthermore, this CGA is highly well aligned in terms of both nucleotide and amin o acid sequences with neighboring region. Thus, the CGA (arginine) may be synapomorphic character for Lepidoptera, functionally constrained. We, therefore, propose the CGA sequence as the start codon for COI gene in lepidopteran insects.

        We analyzed a portion of mitochondrial COI gene sequences (658 bp) to investigate the genetic diversity and geographic variation of the swallowtail butterfly, Papilioxuthus L., and the cabbage butterfly, Pieris rapae (Lepidoptera: Papilionidae). P. xuthus showed a moderate level of sequence divergence (0.91% at maximum) in 15 haplotypes, whereas P. rapae showed a moderate to high level of sequence divergence (1.67% at maximum) in 30 haplotypes, compared with other relevant studies. Analyses of population genetic structure showed that most populations are not genetically differentiated in both species. The distribution pattern of both species appears to be consistent with category IV of the phylogeographic pattern sensu Avise (Avise et al. 1987): a phylogenetic continuity, an absence of regional isolation of mtDNA clones, and extensive distribution of close clones. The observed pattern of genetic diversity and geographic variation of the two butterfly species seems to reflect the abundant habitats, abundant host plants, and flying abilities in connection with the lack of historical biogeographic barriers.

        The 15,338-bp long complete mitochondrial genome (mitogenome) of the Japanese oak silkmoth, Antheraeayamamai (Lepidoptera: Saturniidae) was determined. This genome has a gene arrangement identical to those of all other sequenced lepidopteran insects, but differs from the most common type, as the result of the movement of tRNAMet to a position 5’-upstream of tRNAIle. No typical start codon of the A. yamamai COI gene is available. Instead, a tetranucleotide, TTAG, which is found at the beginning context of all sequenced lepidopteran insects was tentatively designated as the start codon for A. yamamai COI gene. Three of the 13 protein-coding genes (PCGs) harbor the incomplete termination codon, T or TA. All tRNAs formed stable stem-and-loop structures, with the exception of tRNASer(AGN), the DHU arm of which formed a simple loop as has been observed in many other metazoan mt tRNASer(AGN). The 334-bp long A+T-rich region is noteworthy in that it harbors tRNA-likestructures, as has also been seen in the A+T-rich regions of other insect mitogenomes. Phylogenetic analyses of the available species of Bombycoidea, Pyraloidea, and Tortricidea bolstered the current morphology-based hypothesis that Bombycoidea and Pyraloidea are monophyletic (Obtectomera). As has been previously suggested, Bombycidae (Bombyxmori and B.mandarina) and Saturniidae (A.yamamai and Caligula boisduvalii) formed a reciprocal monophyletic group.

        We determined the complete mitochondrial genome of the yellow-spotted long horned beetle, Psacothea hilaris (Coleoptera: Cerambycidae) that is an endangered insect species in Korea. The genome was sequenced from four overlapping fragments: two short fragments and two long fragments. The 15,857-bp long P. hilaris mitochondrial genome has the gene content typical of animal mitochondrial genome: 13 protein-coding genes, 22 tRNA genes, 2 ribosomal genes, and one non-coding A+T-rich region. The gene arrangement of the molecule is identical to the most common type found among insect mitochondrial gene arrangement that is regarded as ancestral for insects. Like several other coleopteran species the P. hilaris COI gene has typical mitochondrial start codon ATT. The 1,190-bp long A+T-rich region contains 57-bp long seven identical repeat sequences and at least seven stem-and-loop structures, composed of stems with perfect matches and loops with variable size. All P. hilaris tRNAs can be folded into the typical clover-leaf structure, with the exception of tRNASer(AGN), the dihydrouridine (DHU) arm of which forms a simple loop. After more genomic and phylogenetic analyses are performed, further detailed information will be available.

        The purpose of this study was to evaluate the elementary students’ knowledge, attitudes, and practice for food hygiene and safety. A self-administered questionnaire was developed and offered to 375 elementary school students throughout the Seoul region. The results of the survey were as follows: First, the knowledge, attitudes, and practice were not at satisfactory level. The scores of children's knowledge, attitudes, and practice were 67.42%, 3.83, and 3.81 out of 5 point scale, respectively. Second, knowledge, attitudes and practice levels were significantly different according to demographic characteristics. The lower grade-students had significantly higher scores for knowledge (p<.05), attitudes (p<.05) and practice (p<.01) than higher gradestudents, suggesting that food hygiene and safety education is more effective in younger students or that repeated education is needed for higher grade-students. Third, knowledge, attitudes and practice levels were significantly correlated each other. These results suggest that the education programs for food hygiene and safety should be developed for, as well as implemented in, lower elementary school grades.

        We determined the complete mitochondrial genome (mitogenome) of the Japanese Oak Silkmoth, Antheraea yamamai (Lepidoptera: Saturniidae) from two overlapping fragments and subsequent shotgun sequencing. The 15,601-bp long A. yamamai mitogenome contains gene arrangement and content identical to the most common arrangement found in lepidopteran insects. Most individual A. yamamai mitochondrial (mt) genes were well within the range found in the respective genes of other insects, except for small ribosomal RNA (1,037 bp). The 336-bp A+T-rich region is relatively smaller than that of other lepidopteran insects. The region is interesting in that it contains tRNA-like structures as found in the A+T-rich regions of other insect mitogenomes. The start codon of A. yamamai COI gene is unusual in that no typical one (ATN) is available. Three of the 13 protein-coding genes have incomplete termination codon T or TA. All tRNA formed stable stem-and-loop structure, except for tRNASer(AGN), the DHU arm of which formed a simple loop as seen in many other metazoan mt tRNASer(AGN).

        We determined the complete mitogenome of the oriental mayfly, Ephemera orientalis (Ephemeroptera: Ephemeridae) and the dragonfly Davidius lunatus (Odonata: Gomphidae). The 16,463-bp long E. orientalis and the 15,912 bp long D. lunatus mitogenome contains gene arrangement and content identical to the most common type found in a diverse insect order. Most individual E. orientalis and D. lunatus mt genes were well within the size found in the respective genes of other insects. The initiation codon for the D. lunatus COI gene was typical as ATA, whereas no typical start codon was found in the start region of E. orientalis COI gene. The A+T-rich regions of both mitogenomes have a few unusual feature. The A+T-rich region of E. orientalis contains a tandem repeat composed of two identical copies of 55 bp long, whereas that of D. lunatus contains a tandem repeat composed of duplicated identical 261-bp copies and one partial copy of the repeat. Also, the A+T-rich region of E. orientalis contains a single sequence and that of D. lunatus contains nine sequences, along with the tandem triplicate sequences, that has the potential to form stem-and-loop structures, flanked by the conserved sequences, “TA(A)TA” at the 5’ end and “G(A)nT’ at the 3’ end. Furthermore, the A+T-rich region of D. lunatus contains two tRNA-like structures, tRNALeu(UUR)-like sequence and tRNATyr-like sequence that have proper anticodon TAA and clover-leaf structure that were previously found in the hymenopteran insects.
