        본 연구는 가축 개량을 위해서 사용되는 육종가의 신뢰도를 PEV (Prediction Error Variance) 방법과 MTEDC (Multiple-trait Effective Daughter Contribution) 방법으로 각각 추정하여, 한우 육종가 신뢰도 수준을 평가·비교하고자 하였다. 본 연구를 위해 혈통등록 거세우 중 결측치와 이상치를 제거하고 도축월령이 27-32개월령 1,391,141두의 도체자료를 이용하였다. 한우 농가의 도축기록이 있는 개체의 모집단(Population)에서 무작위 추출법(Random Sampling)으로 10번 반복하여 추출된 표본자료(Data 1)과 전국 한우농가의 전체 도체자료(Data 2)를 분석에 이용하였다. 한우 도체형질 추정육종가의 신뢰도는 PEV로 분석한 Data 1에서 도체중, 등심단면적, 등지방두께, 근내지방도가 각각 0.38, 0.39, 0.40, 0.42로 추정되었고 MTDEC로 분석한 Data 2에서는 0.45, 0.48, 0.48, 0.52로 분석되었다. 도체중의 후대 두수별(0-5두) 신뢰도는 Data 1에서 0.30-0.64, Data 2에서 0.42-0.52로 분석되었고 등심단면적에서 Data 1과 Data 2가 각각 0.57-0.67, 0.47-0.55, 등지방두께는 Data 1과 Data 2가 각각 0.31-0.67. 0.43-0.55, 근내지방도는 Data 1과 Data 2가 각각 0.43-0.60으로 분석되었다. 분석 결과 표본에 따라 다소 차이가 있었으나 암소 후대 수에 따라 뚜렷한 차이가 있었으며, 신뢰도는 형매 기록 수가 늘어날수록 추정육종가의 신뢰도가 향상됨을 확인할 수 있었다. 한우 암소의 후대가 대부분 0-1두 사이로 개체별 높은 신뢰도를 갖기 어려운 상황이므로 신뢰도 향상을 위해서는 형매 두수가 100두 이상 필요할 것으로 판단되며 이를 위해 육종가 추정을 위한 데이터 구성에서 강력한 혈연계수 행렬을 구현할 수 있는 전국단위 혈통자료와 그에 따른 능력검정 성적이 함께 분석되어야 할 것으로 사료된다.

        본 연구에서는 한우 암소와 씨수소의 육종가를 기반으로 연도별 유전적 개량량을 추정하여 현재 한우 축군의 개량 정도를 파악하여 향후 한우 개량 방향을 위한 기초자료로 활용하고자 본 연구를 시행하였다. 본 연구를 위하여 한우농가 4,040호에서 축산물품질평가원에 도축된 970,567두의 도축자료를 이용하여 10번 반복되어 추출된 표본 자료와 한우농가에서 2009년부터 2019년까지 사육되고 도축된 개체 중에서 한국종축개량협회에 혈통등록 되어 있는 거세우를 선별하여 결측치 및 이상치를 제거하고 도축월령이 27~32개월 이외의 기록은 분석에서 제외한 후, 1,391,141두의 도체 자료를 분석에 이용하였다. 이용된 형질은 도체중(Carcass Weight, CW), 등심단면적(Eye Muscle Area, EMA), 등지방두께(Backfat Thickness, BF) 및 근내지방도(Marbling Score, MS)의 4개 형질을 고려하였다. Random Sampling 10 반복되어 추출된 Data 1의 도체중, 등심단면적, 등지방두께 및 근내지방도의 개량추세에 대한 회귀계수는 매년 평균 0.44㎏, 0.197㎠, -0.051㎜ 및 0.034점으로 나타났으며, 도축성적을 보유한 전체 개체의 자료로 구성된 Data 2에서는 각각 0.35㎏, 0.22㎠, 0.06㎜ 및 0.04점의 개량추세를 보였다. Data 2에서 씨수소의 개량추세는 1.54㎏, 0.343㎠, -0.045㎜ 및 0.050점의 추세를 보였으며 암소와 씨수소 간에는 1.19㎏, 0.119㎠, -0.014㎜ 및 0.010점의 차이를 보였다. 유전적 개량량을 확인하기 위하여 육종가의 표준편차를 이용하였으며, 암소의 선발강도는 본 연구에서 보고한 암소의 후대 거세우 기록 빈도 및 비율을 통해 계산하였다. 한우 암소의 선발 시기를 3산차 이전을 기준으로 하는 것과 4산차 이후를 기준으로 하는 것으로 나눠서 선발강도를 적용하였다. 그 결과, 암소의 선발 시기를 3산차 이전으로 했을 때 세대당 유전적 개량량은 도체중, 등심단면적, 등지방두께 및 근내지방도에서 각각 5.04㎏, 1.31㎠, 0.71㎜ 및 0.32점으로 나타났다. 이러한 결과를 바탕으로 현재 한우 암소집단에서 정확한 육종가를 추정하고, 유전적 개량량을 높이기 위해서는 후대의 기록을 높이기 위한 육종 계획이 필요할 것으로 판단된다.

        본 연구는 한우농가에서 직접 사육되어 도축되어진 한우 거세우 전체의 도축성적 및 혈통자료를 수집하여 한우 암소의 유전능력평가를 위한 유전모수를 추정하고 농가에서 직접적으로 활용 가능한 유전능력 추정을 위한 기초자료로 제공하고자 하였다. 본 연구를 위해 검증된 970,141두의 도축기록으로부터 60,000두 이상의 표현형 표본을 추출하기 위해 전체 4,040농가 중 600농가를 단순임의추출(Simple Random Sampling)하여 선정된 농가의 도축기록을 모두 추출하는 방식으로 10회 반복하여 표본을 생성하였다. 본 연구에서 추정된 도체형질에 대한 유전력은 도체중에서 0.24-0.30, 등심단면적에서 0.24-0.31, 등지방두께에서 0.31-0.39, 근내지방도에서 0.38-0.58로 추정되었으며, 10회 반복 추정된 유전력의 평균값은 도체중, 등심단면적, 등지방두께 및 근내지방도에서 각각 0.28, 0.28, 0.35 및 0.48로 추정되었다. 추정된 결과가 기존에 한우에서 보고된 유전력에 비해 다소 낮게 추정되었으나 실제 농가에서 사육되어 도축된 전체 자료가 표본이 된 만큼 본 연구 결과는 전국단위의 도축자료를 이용한 결과로서 암소개량을 위해 직접적으로 활용 가능한 자료가 될 수 있을 것으로 기대된다.

        Recently, we published a microinjection method for generating transgenic cattle using the DNA transposon system and their analysis by next-generation sequencing (Yum et al. Sci Rep. 2016 Jun 21;6:27185). In that study, we generated transgenic cattle using two different types of DNA transposon system, sleeping beauty (SB) and piggybac (PB), carrying Yellow fluorescent protein with SB (SB-YFP, female) and green fluorescent protein with PB (PB-GFP, male) under the control of the ubiquitous CAG promoter, respectively. The female and male founder cattle have been grown up to date (the female age: 40 months old, the male age: 33 months old) without any health issues. In genomic instability and blood analysis, there was no significant differences between wild type and founder cattle. In the present study, we confirmed germ-line transmission of the transposon-mediated transgene integrations and ubiquitous and persistent expression of transgene in second generation of offspring (F1). The F1 was born without any assistance and expressed GFP in the eyes without UV light. The ubiquitous expression of GFP was detected in skin fibroblast from the ear tissue and confirmed by genomic DNA PCR, which suggest that the transgene from the PB-GFP was successfully transmitted. Unfortunately, no transgene from SB-YFP were identified. To confirm the transgene integration site, the genomic DNA from blood was extracted and performed next-generation sequencing (NGS). The GFP gene was integrated in chromosome 4 (two copies), and 6. As results, a total of two copies of paternal transgene transmitted into the F1. All the integrated position was not related with coding region and there was no significant difference in genomic variants between transgenic and non-transgenic cattle. To our knowledge, this is the first report of germ-line transmission through non-viral transgenic founder cattle. Those transgenic cattle will be valuable resource to many fields of biomedical research and agricultural science.

        CRISPR/Cas9-induced knock-out/-in can be occurred at specific locus in the genome by non-homologous end joining (NHEJ) or homology directed repair (HDR). Here, we demonstrate the targeted insertion into the specific loci of embryo fertilized by semen from transgenic cattle via CRISPR/Cas9 system. Recently, we published on the efficient generation of transgenic cattle using the DNA transposon system (Yum et al. Sci Rep. 2016 Jun 21;6:27185). In the study, eight transgenic cattle were born following transposon-mediated gene delivery system (Sleeping Beauty and Piggybac transposon system) via microinjection. In the analysis of their genome stability using next-generation sequencing, there was no significant difference in the number of genetic variants between transgenic and non-transgenic cattle. All the transgenic cattle have grown up to date (the oldest age: 33 months old, the youngest age: 15 months old) without any health issue. One of transgenic male cattle expressing GFP reached puberty and semen was collected. Over 200 frozen semen straws were produced and some were used for in vitro fertilization (IVF). On seven days after IVF, expression of GFP was observed at blastocyst stage and was seen in 80% of the embryos. Another application is to edit the GFP locus of the transgenic cattle because long-term and ubiquitous expression of transgene didn’t affect their health. In one cell stage embryos produced using GFP frozen-thawed semen, microinjection of sgRNA for GFP, Cas9, together with donor DNA that included RFP and homology arms to link the double-strand break of sgRNA target site into fertilized eggs resulted in expression of RFP. This indicated that the GFP locus of transgenic cattle shows potential candidates for stable insertion of the functional transgene. Knock-out/-in for editing GFP locus using CRISPR-Cas9 might be a valuable approach for the next generation of transgenic models by microinjection. In conclusion, we demonstrated P-112 that transgenic cattle via transposon system are healthy to date and germ-line competence was confirmed. The GFP locus will be used as the potential target site for future gene engineering via genome-editing technology. Finally, all those animals could be a valuable agricultural and veterinary science resource for studying the effects of gene manipulation on biomedical research and medicine.         On the analysis of triacylglycerol (TG) from the kernels of Acanthopanax sessiliflorus by reversed phase-HPLC, it was separated into three main fractions of PN 44, 46 and 48, according to partition number (PN). On the contrary, it could be clearly classified into seven fractions of SMM, MMM, SMD, MMD, SDD, MDD and MDT by silver ion-HPLC by the number of double bond in the acyl chains of TG species. But resolution of so-called critical pairs of TG molecular species such as molecular pairs of PeLL [C18:1Ω12/(C18:2Ω6)2] and OLL [C18:1Ω9/(C18:2Ω6)2] and OOL [(C18:1Ω9)2/C18:2Ω6], and PePeL [(C18:2Ω12)2/C18:1Ω6] was not achieved (Pe; petroselinic acid, L; linoleic acid, O; oleic acid). On the other hand, TG extracted from Aralia continentalis kernels were also fractionated into seven groups of SSM, SMM, MMM, SMD, MMD, SDD and MDD (S; saturated acid, M; monoenoic acid, D; dienoic acid) by silver ion-HPLC, although it's were classified into three groups of PN 44, 46 and 48 by reversed phase-HPLC. The fractions of SMM, MMM, MMD and MDD were divided into two subfractions, respectively; the fractions of SMM, MMM, MMD and MDD were resolved into the subfraction of PPe/Pe and POO (critical pairs from each other), that of Pe/Pe/Pe and OOO, that of Pe/Pe/L and OOL, and that of Pe/L/L and OLL.

        Fatty acid compositions of the seed oils of P. schinseng, A. continentalis and A. sessiliflorus, were analyzed by gas chromatography (GC) equipped with a capillary column. A large unusual peak was observed just before the peak corresponding to oleic acid (cis-9-C18:1). This unknown fatty acid was isolated by silver ion chromatography and then derivatized into the picolinyl ester. The mass spectrum of the picolinyl ester showed molecular ion at m/z=373 with other diagnostic ions such as m/z=178, 218, 232, 246, 274, 288, 302 and 344. Characteristic absorption peaks at 720 cm-1, 1640 cm-1 and 3010 cm-1 in IR spectrum indicated the presence of cis-configurational double bond in the molecule. The 1H-NMR spectrum of this acid gave two quintets centered at δ1.638 (2H, C-3) and δ1.377 (2H, C-4), and two multiplets centered at δ2.022~2.047 (2H, C-5) and δ2.000~2.022 (2H, C-8), and multiplet signals of olefinic protons centered at δ5.3015~5.3426 (C-6, J=9.5 Hz) and δ 5.3465~5.3877 (C-7, J=9.5 Hz). The 13C-NMR spectrum showed 18 carbon resonance signals including an overlapped signal at δ29.7002 for C-12 and δ29.6520 for C-13 (or they can be reversed), and other highly resolved signals at δ33.950, δ24.558, δ26.773 and δ27.205 due to C-2, C-3, C-5 and C-8 of a δ6-octadecenoic acid, respectively. From analysis results this unknown fatty acid could be identified as cis-6-octadecenoic acid. The seed oils of P. schinseng and A. sessiliflorus contained petroselinic acid (59.7%, 56.0%), oleic acid (18.3%, 6.1%) and linoleic acid (16.2%, 30.4%) with small amount of palmitic acid (3.0%, 3.1%) while the seed oil of A. continentalis comprised mainly oleic acid (30.2%), petroselinic acid (29.0%), linoleic acid (24.1%) and palmitic acid (13.1%).

        A mixture of methyl ester derivatives of fatty acids from the oils of pine nuts was well resolved to five fractions differing by degree of unsaturation by silver ion solid-phase extraction column chromatography (Ag+-SEC). Polyunsaturated fatty acid with non-methylene interrupted conjugated double bond (NMiDB) radical held more strongly to silver ions in the column than methylene interrupted conjugated double bond (MiDB) one when they had the same number of double bonds. Although both the picolinyl ester and DMOX derivative provided clear mass ion species powerful enough to elucidate the structure of the polyunsaturated fatty acid (PUFA) with NMiDB and/or methylene interrupted conjugated double bond (MiDB) radical in the oils, the picolinyl ester of PUFA with NMiDB radical did not provide a cluster of mass ions neighboring diagnostic mass ions induced by the double bond in the proximal to the carboxyl group. However, the DMOX derivative of PUFA with NMiDB group as well as MiDB showed abundant mass ion species differing by gaps of 12 amu, which made it possible with greater ease to locate the double bonds in the molecule. The oil contained C18:2Ω6 (46.2 %) and C18:1Ω9 (25.4 %) as main components, and considerable amounts of PUFAs with NMiDB radical such as δ5. 9. 12-C18:3 (16.0 %), δ5. 9-C18:2 (2.3 %) and δ5. 11. 14-C20:3 (0.8 %).

        Purpose – In this study, previous CSR researches was reviewed to help understand the CSR effects, and offer suggestions for future research directions. Research design, data, and methodology – To achieve the purpose of this study, previous CSR effects related researches were reviewed based on stakeholder perspectives. Results - After reviewing, some limitations were found. First, most of the CSR related studies in the management area have focused on CSR effects on individual stakeholders such as the shareholder, the employee, and the consumer. Second, it was difficult to find studies on the management aspect of CSR from both an effective and efficient perspective. Conclusions - To overcome these limitations of previous studies, several studies need to be conducted in the future. First, CSR research must be expanded into B-to-B businesses. Few studies try to figure out the effect of CSR on B-to-B businesses but only suggest the effect of CSR on B-to-B business conceptually. Therefore, empirical study needs to be conducted about CSR to show business performance in the B-to-B market. Second, the process of CSR must be developed on how to improve CSRs’ effectiveness and efficiency. Finally, a relationship between a company and individual stakeholders needs to be revised using stakeholders’ multiple identities.

        This study was conducted to obtain basic information on mainstem, branch and leaf characteristics related to canopy for development of high yielding cultivar using 70 Korean soybean cultivars developed from 1913 to 2000. Variations of canopy width, branch length, and canopy width/length ratio were higher compared to other characteristics among 12 mainstem and branch characters. Variations of petiole angle, leaflet width/length ratio and compound leaf dry weight were higher than other characteristics among eight leaf characters related to plant canopy. Three classifications of soybean cultivars were used based on usage: I)soy sauce and tofu, II)bean sprout, and III)cooking with rice. Canopy width/length ratio was higher in group III, cooking with rice than group I, soy sauce and tofu, and group II, bean sprout, and there was no difference between the two, group I and group II. The total branch length/main stem height ratio was higher in group II, bean sprout and group III, cooking with rice than group I, soy sauce and tofu. Mainstem and branch characteristics related to plant canopy were classified into four groups by ratio of canopy width/length and total branch length/main stem length, respectively. Soybean cultivars with narrow canopy and high dependence of mainstem were Danweonkong, Keumkangkong, Shelby, and Shinpaldalkong. Soybean cultivars with broad canopy and high dependence of mainstem were Kanglim, Keumkangdaelip, and Jinyulkong, and a cultivar with broad canopy and high dependence of branch were Geomjeongkong 2. Leaflet length/width ratio was lowest in cooking with rice and there was no difference between soy sauce and tofu and bean sprout. Compound leaf area was largest in cooking with rice and smallest in bean sprout. Leaf petiole length was short in bean sprout and there was no difference between soy sauce and tofu and bean sprout. Leaf petiole angle was highest in cooking with rice and lowest in bean sprout. Leaf type was classified into four groups based on leaflet width/length ratio and compound leaf area, respectivly. Buseok and Taekwangkong had an oval leaflet and largest area of compound leaf. Eunhakong and Sohokong had extreme narrow leaflet and smallest area of compound leaf. Leaf petiole type was classified into three and four groups based on leaf petiole length and angle, respectively. A soybean cultivar with the shortest petiole length and smallest petiole angle was Eunhakong and cultivars with short petiole length and large petiole angle were Alchankong, Muhankong, and Pureunkong. A soybean cultivar with long petiole length and small petiole angle was Sinpaldalkong 2. Among a total of 70 Korean soybean cultivars, Eunhakong had an extreme narrow type in leaf, smallest compound leaf area, shortest petiole length, and smallest petiole angle of compound leaf.
