        Roridin E, a constituent of Podostroma cornu-damae, has excellent medicinal properties and exhibits powerful anticancer effects. However, being a poisonous mushroom, P. cornu-damae is difficult to find in its natural environment. Therefore, only a limited amount of roridin E is isolated from P. cornu-damae. We isolated roridin E from mycelial culture medium of P. cornu-damae and examined its effects on five breast cancer cell lines. The purity of the isolated roridin E was 96%. It exhibited anticancer effects on all five breast cancer cell lines, with an IC50 of at least 0.0006 mg/L. To the best of our knowledge, this is the first report on the investigation of effects of roridin E isolated from the mycelium of P. cornu-damae mushrooms on five breast cancer cell lines. However, safety issues should be investigated and addressed in future studies before roridin E is used in clinical settings.

        Roridin E, produced by Podostroma cornu-damae, is a mycotoxin with anticancer activity. To increase the content of roridin E, submerged culture conditions were optimized using response surface methodology. Three factors, namely, medium initial pH, incubation time and agitation speed were optimized using a Box–Behnken design. The optimum submerged culture conditions to increase the content of roridin E included a medium with an initial pH of 4.0, an incubation time of 12.90 days, and an agitation speed of 63.03 rpm. The roridin E content in the submerged culture, under the aforementioned conditions, was 40.26 mg/L. The findings of this study can help lower the current price of roridin E and promote its related research.

        The choice of the culture medium is an important factor for the mass production of mycelia in submerged cultures. The influence of liquid medium on the mycelial dry weight of Paecilomyces tenuipes was investigated in this study. The regression equation is expressed as Y=-1292.94187+17.78612X1+18.92425X2+2.11464X3-0.019375X1X2-0.006276X1X3+0.008177X2X3- 0.070169X1 2-0.292175X2 2-0.008818X3 2, where Y represents the value of the mycelial dry weight (g/L), X1 is the particle size of wood sawdust in liquid medium (mesh), X2 is the concentration of the wood sawdust in liquid medium, and X3 is incubation time (h). The medium was optimized using a response surface methodology, and the optimal medium contained 30 g of wood sawdust (140 mesh), 20 g of glucose, and 10 g/L of peptone. Under these conditions, the mycelial dry weight reached 38.1 g/L (actual value). The culture medium containing wood sawdust is simple and easy to use, highly efficient, and eco-friendly, and its effectiveness in large preparations of P. tenuipes mycelia with low material costs has been demonstrated.

        황해연안 조위관측소 10 개 지점에서 2013년 11월 24일 밤부터 25일 오전까지 관측한 해수면, 해면기압, 바람, 유동 자료뿐만 아니라 일기도를 분석하여 이상조위편차의 발생 원인과 관측자료들 간의 상호상관성을 알아보았다. 이상조위편차란 최대조위편차와 최소조위편차가 나타나는 시간동안 두 편차간의 차를 의미한다. 영종도의 최대조위편차는 111 cm, 최소조위편차는 -65 cm로, 4시간 1분 동안 176 cm의 이상조위편차를 보여 10개 조위관측소 가운데 가장 크다. 반면 모슬포의 이상조위편차는 8시간 52분 동안 약 68 cm로 가장 작다. 이 같은 이상조위편차는 기압점프에 의한 기상해일이 아니라 저기압에 의한 기압배치의 영향으로 바람에 의해 발생한 것으로 확인되었다. 각 지점에서 이상조위편차에 의한 흐름은 연평균 낙조류 세기의 16 ~ 41 %로 무시할 수 없을 정도이다. 조위편차, 바람, 조류잔차의 상호상관관계로부터 저기압의 중심이 한반도 서쪽에 가까이 위치해 있을 때 인천에서 남풍계열의 바람에 의한 북향류가 해수면을 상승시켰고, 한반도 통과 후 북풍계열의 바람에 의해 남향류가 해수면을 하강시켰다.

        천연물로부터 새로운 미백 화장품 소재를 개발하기 위해 참나물(P. brachycarpa)을 선택하여 B16 멜라노마 세포에서 tyrosinase 활성 저해 효과, melanin 생성 저해 효과를 측정하였다. 먼저 참나물추출물의 4가지 극성별 용매 분획화 실험을 실시하였으며, 항산화효과와 티로시나아제 저해효과를 측정하였다. 참나물의 미백 활성 메카니즘을 알아보기 위해 Western blotting과 RT-PCR을 이용하여 tyrosinase, TRP-1, TRP-2의 단백질 발현과 mRNA 변화를 연구하였다. 또한, HaCaT 각질형성세포에서 UVB 조사 후 엔도세린-1 (ET-1)의 발현은 사람 ET-1 항체를 이용하여 quantitative enzyme immunoassay (EIA)로 측정하였다. 그 결과 참나물추출물과 4가지 분획(hexane, EtOAc, butanol and aqueous)은 100 g/mL 농도에서 각각 87.2, 2.5, 97.2, 80.5, 49.8 %의 프리라디칼 소거효과를 나타내었으며, tyrosinase 저해효과는 100 g/mL 농도에서 각각 18.3, 15.1, 55.4, 13.1, 0 %로 나타났다. 각 극성별 분획 중 EtOAc 분획 100 g/mL 농도에서 58 % 이상의 가장 우수한 멜라닌 생성 저해 효과가 나타났다. 참나물 EtOAc 분획은 B16 멜라노마 세포에서 티로시나아제 활성 및 발현을 모두 저해하였으며, RT-PCR 결과에서도 tyrosinase, TRP-1의 mRNA 발현 저해효과가 우수하게 나타났다. 또한 HaCaT 각질형성세포에서 UVB 조사 후 생성된 엔도세린-1의 생성 실험에서도 참나물 EtOAc 분획 12.5 ∼ 50 g/mL 농도에서 엔도세린-1의 생성이 컨트롤에 비해 40 % 정도로 우수하게 저해되었다. 결론적으로 참나물추출물은 멜라닌 합성과정에서 우수한 tyrosinase 저해효과와 엔도세린-1 발현저해효과를 가지는 새로운 천연 미백 소재로 적용될 수 있을 것이라 기대된다.

        Nitrate-selective ion exchange resin which have bulky tertiary amine as functional group have been synthesized by the reaction of chloromethylated polystyrene-divinylbenzene copolymer and the corresponding tertiary amine [NR3=NEt3 1, N(C2H4OH)3 2] in ethanol, while commercial resin has NMe3 as functional group. The fundamental properties such as bulk density, water content, appearance index, exchange capacity, effective size, uniformity coefficient of synthesized anion exchange resin (1) have been measured. The ion exchange resin (1) and (2) exhibited the better selectivity for nitrate than sulfate in both batch and continuous column experiments.

        A kinetic study for nitrate removal by anion exchange resin was performed using continuous column reactors. Kinetic approach from the packed bed showed the reaction rate constant k1 was 0.07∼0.17 ℓ/㎎·hr and maximum exchange quantity q0 was 27.75∼31.81 ㎎/g. The results from the continuous column well agreed with that from the batch reactor. An economic analysis of the water treatment plant by anion exchange resin with a regenerating system was performed to design plant and process. Based on the treatment of 20 ㎎/ℓ nitrate-contained wastewater of 10,000 gallons per day to 2 ㎎/ℓ , total capital cost and total annual cost are estimated to be 836 million wons and 211 million wons, respectively.

        A kinetic study for anion exchange was performed for commercially available Cl- type anion exchange resin in use to remove nitrate in water. The obtained results from the batch reactor were applied to the Langmuir and Freundlich models. The constants for Langmuir model were qmax=29.82 and b=0.202, and for Freundlich model were K=5.509 and n=1.772. Langmuir model showed better fit than Frendlich model for the experimental results. Ion exchange reaction rate was also calculated and the approximate first-order reaction, rate constant k1 was 0.16 L/㎎·hr. Effective diffusion coefficient was obtained in the range from 9.67×10 exp (-8) to 1.67×10 exp (-6) ㎠/sec for initial concentration change, and from 6.09×10 exp (-7) to 3.98×10 exp (-6) ㎠/sec for reaction temperature change. Activation energy during the diffusion was calculated as 36 ㎉/㏖.
