        Brucellosis is the most common zoonosis worldwide, which is caused by Brucella spp. In humans, it can be mainly occurred by direct contact with infected animals or consumption of contaminated dairy products. This study focused on human brucellosis caused by B. melitensis discovered from Chinese worker in Korea in 2015. We investigated molecular epidemiological evidence to find the infection source. We first performed several PCR methods including 16S rRNA PCR, multiplex PCR and real-time PCR to identify Brucella species. We also conducted MLVA typing for epidemiological trace-back analysis. The isolate from the patient was confirmed to B. melitensis through Brucella-specific PCR. In clustering analysis with B. melitensis from foreign countries, this human isolate was correlated with B. melitensis isolates from humans and sheep in China by 99.9% similarity. Thus, we assumed the brucellosis patient has been already infected in China followed by migration to Korea according to molecular epidemiological analysis with history evidence. Moreover, we suggest it needs to take measures to reduce the risk for intercountry transmission of brucellosis due to the influx of infected people from abroad.

        Amine-functionalized graphene was synthesized via a one-step solvothermal method and used as a metal-free cathode for non-aqueous lithium–oxygen batteries. The material delivered an outstanding specific capacity of 19,789 mAh/g at a current density of 200 mA/g as well as better cycling stability than graphene without the amine functional group. This improvement was attributed to the electron-donating effect of the amine groups and appropriate mesopore volume, which can promote the penetration of oxygen, electrons, and lithium ions, as well as accommodate more discharge products, Li2O2 in Li–O2 batteries. Amine-functionalized graphene has an amine functional group on the carbon surface, which improves the electrical conductivity of carbon and provides electrochemical active sites for oxygen absorption reactions.

        High-level heteroatom, N and S, dual-doped graphene with an improved mesoporous structure was fabricated via facile in situ carbonization and used as metal-free cathode for non-aqueous lithium oxygen batteries. The prepared cathode delivered an ultrahigh specific capacity of 22,252 mAh/g at a current density of 200 mA/g as well as better cycling reversibility because of the larger and copious mesopores, which can promote the penetration of oxygen, electrons, and lithium ions and the ability to accommodate more discharge products, e.g., Li2O2, in Li–O2 batteries. The material had a high level of heteroatom co-doping in the carbon lattice, which enhanced the electrical conductivity and served as active sites for the oxygen reduction reaction.

        Sow longevity is important for efficient and profitable pig farming. Recently, there has been an increasing interest in social genetic effect (SGE) of pigs on stress-tolerance and behavior. The present study aimed to estimate genetic correlations among average daily gain (ADG), stayability (STAY), and number of piglets born alive at the first parity (NBA1) in Korean Yorkshire pigs, using the SGE model. The phenotypic records of ADG and reproductive traits of 33,120 and 11,654 pigs, respectively, were evaluated. The direct effect on ADG had a significantly negative genetic relationship with STAY, whereas the social effect on ADG had a neutral genetic relationship. In addition, the genetic correlation between the social effects on ADG and NBA1 tended to be positive, unlike the direct effects. The genetic correlation of the total effect on ADG with that of STAY was negative but non-significant, owing to the social effect. These results suggested that total genetic effect on growth in the SGE model might reduce the negative effect on sow longevity owing to the growth potential of pigs. We recommend including social effects as selection criteria in breeding programs to obtain satisfactory genetic changes in both growth and longevity.

        Cryopreservation of bovine embryos is used to efficiently implant surrogate mothers. It has been widely accepted that high lipid content in the oocyte interrupts its survival during freeze-thaw cycles. Serum component in the culture medium is thought to increase the embryo`s lipid contents. Conversely, L-carnitine stimulates lipid metabolism by transporting long chain fatty acids into the mitochondria. Objective of this study was to analyze the effect of L-carnitine supplementation in IVM medium and defined IVC medium on the development, lipid contents and the cryosurvival of bovine IVF embryos. 0.0, 1.5, 3.0 and 6.0 mM L-carnitine was supplemented in IVM medium, respectively (IVM-LC 0.0, LC 1.5, LC 3.0 and LC 6.0). Development rate from the 2cell to the morula stages was higher in IVM-LC 3.0 groups than those of IVM-LC 6.0 (p<0.05). But there were no significant differences among the other groups in the blastocyst rates and lipid content results. When 0.0, 1.5, 3.0 and 6.0 mM L-carnitine were supplemented in IVC medium (IVC-LC 0.0, LC 1.5, LC 3.0 and LC 6.0), development competence was not significantly different between those embryos. Lipid contents of embryos treated L-carnitine (IVC-LC 1.5, 3.0 and 6.0) were significantly lower than embryos of non-treated group. L-carnitine was supplemented 0.0, 1.5, 3.0, 6.0 mM during IVM and 3.0 mM during IVC (LC 0.0 - 3.0, LC 1.5 – 3.0, LC 3.0 – 3.0, LC 6.0 – 3.0) and cryosurvival of blastocysts confirmed after freezing-thawing. There were no significant differences on development, but LC 3.0 – 3.0 was significantly lower lipid contents than other groups. And LC 3.0 – 3.0 had better survival rates and hatched rates of blastocysts than LC 0.0 – 0.0. In conclusion, supplementation of L-carnitine in defined IVC medium decreases lipid contents. And L-carnitine supplementation improves cryosurvival and developmental ability of bovine IVF embryos.

        The early-onset familial Alzheimer's disease (EOFAD/ FAD), the less common type of Alzheimer's disease (AD) currently affects a vast number of individuals worldwide. This type is being inherited as an autosomal dominant fashion. Missense mutations on Amyloid precursor protein (APP) and Presenilins 1 and 2 (PSEN1 & PSEN2) are known as major genetic factors in FAD. Conversely, missense mutations on microtubule-associated protein tau (MAPT) are also thought to involve. Up to date, several triple-transgenic animal models with muted forms of the human APP, PSENs and MAPT have been reported. Compared to other animals, canines are more emotional and their disease signs can be easily diagnosed. This attempt was to develop a triple transgenic canine model for the AD. We have obtained the coding sequences of APP, PSEN1 and MAPT from Dana-Farber/Harvard Cancer Center DNA resource core at HMS and incorporated several common AD mutations. The transgenic construct is composed of hNSE (ENO2) promoter-driven three AD genes fused together with modified 2A sequences. It was transfected into the canine fetal fibroblasts which were then used to perform somatic cell nuclear transfer (SCNT). The viable transgenic embryos were obtained after in vitro culture and the GFP was detected. In this study, we have successfully produced viable triple transgenic canine cloned embryos using SCNT technique. These transgenic canine embryos will be further developed into canines with FAD. The transgenic canines will be a good candidate in the AD research field.

        Insect-killing fungi have high potential in pest management. A deeper insight into the fungal genes at the whole genome level is necessary to understand the inter-species or intra-species genetic diversity of fungal genes, and to select excellent isolates. In this work, we conducted a whole genome sequencing of Beauveria bassiana (Bb) JEF-007 and characterized pathogenesis-related features and compared with other isolates including Bb ARSEF2860. A large number of Bb JEF-007 genes showed high identity with Bb ARSEF2860, but some genes showed moderate or low identity. The two Bb isolates showed a significant difference in vegetative growth, antibiotic-susceptibility, and virulence against Tenebrio molitor larvae. When highly identical genes between the two Bb isolates were subjected to real-time PCR, their transcription levels were different, particularly in heat shock protein 30 (hsp30) gene which is related to conidial thermotolerance. In several B. bassiana isolates, chitinases and trypsin-like protease genes involved in pathogenesis were highly conserved, but other genes showed noticeable sequence variation within the same species. Given the transcriptional and genetic diversity in B. bassiana, a selection of virulent isolates with industrial advantages is a pre-requisite, and this genetic approach could support the development of excellent biopesticides with intellectual property protection.

        Insect peptides have been extensively studied due to beneficial effects in the treatment of infectious diseases. Melittin, a fundamental component of honeybee venom produced by European honeybee Apis mellifera, has applied to prevent various inflammatory disease and bacterial infections in human. However, the therapeutic application of melittin is limited due to its low stability, hemolytic activity and expensive manufacturing costs. In this study, we aimed to discovery unknown peptides from the Apis mellifera and evaluate its antibacterial activity against Escherichia coli KACC 10005. A total 15,853 peptide sequences were diciphered using Illumina HiSeq 2500 next-generation sequencing (NGS) platform and analyzed based on the Apis mellifera official Gene Set Version 3.2 (amel_OGSv3.2) and the Collection of Anti-Microbial Peptides (CAMPR3) database. All the peptide sequences and annotation data sets were combined and sorted by physicochemical features of antimicrobial peptides (AMPs), such as short peptide length <=50, positive charge, isoelectric point (8.0<=pl<=12), and aggregation propensity (in-vitro: <=500, in-vivo: –40<= Na4vSS <=60). Among the screened peptides, four unknown peptide candidates, named AMP1-4, were chemically synthesized and tested for antimicrobial activity in comparison with a reference peptide, melittin. Inhibition of bacterial growth was observed in the AMP4 treated group from 6 hours to 48 hours post-treatment against E. coli. These results suggest that honeybee-derived peptide sequences can be applied as natural resources to acquire novel AMPs and the peptide sequences derived parameters are enough to recognize antibacterial peptides. In addition, the selected novel peptide candidate, AMP4, has antibacterial activity.

        The hypopharyngeal gland (HPG) of the honeybee worker produces royal jelly (RJ) and has a developmental cycle closely related to the division of labor. In this study, we investigated to compare the HPG acini diameter of differently aged worker bees with high royal jelly producing colony (HRC) or less producing colony (LRC). Additionally, we also evaluated whether the fresh weight of the head is a reliable indicator of the developmental status of HPG. The HRC showed a significantly higher RJ production about two-times as compared with those of the LRC. We measured the HG-diameters on days 1, 3, 6, 9, 12, 15. The microscopic analysis revealed that the acini size of the HRC was significantly larger than the LRC. In addition, the acini diameter of HRC was 15% longer than the LRC on the first day after emerging. It was shown that the fastest development during 3 days which is preparing for nurse the brood. The HPG acini diameters increased in both colonies in a similar fashion until day 12 and then decreased. We also compared the fresh head weight of the experimental colonies, differences were similar to the development of HPG. Therefore, high royal jelly production may have a positive correlation between HPG acini size and the fresh head weight.

        The purpose of this study was to investigate the effect of heel off stairway walking exercise on the increase of muscle activity and balance activity of the ankle joint muscles in university students with functional ankle instability. The conservative treatment for the control group consisted of stairway walking (n=10) and the experimental group consisted of heel off stairway walking (n=11). The therapeutic intervention of the control and experimental groups was performed a total of 12 exercise sessions, 3 times per week for 4 weeks. To compare the two groups, the level of ankle disability was assessed by using the EMG, BT4 and Pedoscan in pre-treatment and post-treatment. Muscle activity increased in both the experimental groups and control groups in each group, however there was no significant difference between the groups. Balance ability did not show any significant difference. This study demonstrates that heel-off stairway walking is effective in significantly increasing muscle activity, however did not significantly improve balance ability.

        To elucidate the effect of cellular phone electromagnetic wave (EMW) exposure on the developing cerebellar cortex of neonatal Sprague-Dawley rats, animals were exposed to cellular phone electromagnetic waves for 1 hr per day for 3 weeks. At the end of the experimental period, animals were sacrificed by cardiac perfusion, after which histological samples were prepared and observed microscopically. In the EMW exposure group, external granule cells were remained partially in the external granular layer without migrating into the internal granular layer. In addition, dark stained shrunken Purkinje cells with pyknotic nuclei increased and the outline of cells became irregular and showed degenerative signs, such as mitochondrial swelling and disrupted cristae. Moreover, the cisternae of rough endoplasmic reticula and Golgi complex were severely swollen. Bergmann glial cells adjacent to the dark stained Purkinje cells were swollen and cytoplasmic organelles were scant. Dark stained shrunken granule cells were also observed and the outline of cells was irregular. The results of the present study suggest that cellular phone EMW exposure to neonatal Sprague-Dawley rats leads to a partial delay of early migration of cerebellar cortical cells and degenerative changes in Purkinje cells, Bergmann glial cells and granule cells.

        Porcine spermatogonial stem cells (SSCs) prefer three-dimensional (3D) culture systems to 2D ones for the maintenance of self-renewal. Of the many 3D culture systems, agar-based hydrogels are candidates for supporting porcine SSC self-renewal, and there are various types of agar powder that can be used. In this study, we sought to identify an agar-based 3D hydrogel system that exhibited strong efficacy in the maintenance of porcine SSC self-renewal. First, 3D hydrogels with different mechanics were prepared with various concentrations of Bacto agar, lysogeny broth (LB) agar, and agarose powder, and the 3D hydrogel with the strongest alkaline phosphatase (AP) activity and greatest increase in colony size was identified for the different types of agar powder. Second, among the porcine SSCs cultured in the different 3D hydrogels, we analyzed the colony formation, morphology, and size; AP activity; and transcription and translation of porcine SSC-related genes, and these were compared to determine the optimal 3D hydrogel system for the maintenance of porcine SSC self-renewal. We found that 0.6% (w/v) Bacto agar-, 1% (w/v) LB agar-, and 0.2% (w/v) agarose-based 3D hydrogels showed the strongest maintenance of AP activity and the most pronounced increase in colony size in the culture of porcine SSCs. Moreover, among these hydrogels, the strongest transcription and translation of porcine SSC-related genes and largest colony size were detected in porcine SSCs cultured in the 0.2% (w/v) agarose-based 3D hydrogel, whereas there were no significant differences in colony formation and morphology. These results demonstrate that the 0.2% (w/v) agarose-based 3D hydrogel can be effectively used for the maintenance of porcine SSC self-renewal.
