Wolsong unit 1, the first PHWR (Pressurized Heavy Water Reactor) in Korea, was permanent shut down in 2019. In Korea, according to the Nuclear Safety Act, the FDP (Final Decommissioning Plan) must be submitted within 5 years of permanent shutdown. According to NSSC Notice, the types, volumes, and radioactivity of solid radioactive wastes should be included in FDP chapter 9, Radioactive Waste Management, Therefore, in this study, activation assessment and waste classification of the End shield, which is a major activation component, were conducted. MCNP and ORIGEN-S computer codes were used for the activation assessment of the End shield. Radioactive waste levels were classified according to the cooling period of 0 to 20 years in consideration of the actual start of decommissioning. The End shield consists of Lattice tube, Shielding ball, Sleeve insert, Calandria tube shielding sleeve, and Embedment Ring. Among the components composed for each fuel channel, the neutron flux was calculated for the components whose level was not predicted by preliminary activation assessment, by dividing them into three channel regions: central channel, inter channel, and outer channel. In the case of the shielding ball, the neutron flux was calculated in the area up to 10 cm close to the core and other parts to check the decrease in neutron flux with the distance from the core. The neutron flux calculations showed that the highest neutron flux was calculated at the Sleeve insert, the component closest to the fuel channel. It was found that the neutron flux decreased by about 1/10 to 1/20 as the distance from the core increased by 20 cm. The outer channel was found to have about 30% of the neutron flux of the center channel. It was found that no change in radioactive waste level due to decay occurred during the 0 to 20 years cooling period. In this study, activation assessment and waste classification of End Shield in Wolsong unit 1 was conducted. The results of this study can be used as a basis for the preparation of the FDP for the Wolsong unit 1.
The bacterial soft-rot disease is one of the most critical diseases in vegetables such as Chinese cabbage. The researchers isolated two bacteria (Pseudomonas kribbensis and Pantoea vagans) from diseased tissue samples of Chinese cabbages and confirmed them as being the strains that cause soft-rot disease. Lactic-acid bacteria (LAB), were screened and used to control soft-rot disease bacteria. The researchers tested the treatments with hypochlorous acid water (HAW) and LAB supernatant to control soft-rot disease bacteria. The tests confirmed that treatments with the HAW (over 120 ppm) or LAB (Lactobacillus plantarum PL203) culture supernatants (0.5 mL) completely controlled both P. kribbensis and P. vagans.
We present a BV I optical photometric study of the old open cluster Ruprecht 6 using the data obtained with the SMARTS 1.0 m telescope at the CTIO, Chile. Its color-magnitude diagrams show the clear existence of the main-sequence stars, whose turn-o point is located around V 18:45 mag and B - V 0:85 mag. Three red clump (RC) stars are identied at V = 16:00 mag, I = 14:41 mag and B - V = 1:35 mag. From the mean Ks-band magnitude of RC stars (Ks = 12:39 0:21 mag) in Ruprecht 6 from 2MASS photometry and the known absolute magnitudes of the RC stars (MKS = - :595 0:025 mag), we obtain the distance modulus to Ruprecht 6 of (m -M)0 = 13:84 0:21 mag (d = 5:86 0:60 kpc). From the (J - Ks) and (B - V ) colors of the RC stars, comparison of the (B - V ) and (V - I) colors of the bright stars in Ruprecht 6 with those of the intrinsic colors of dwarf and giant stars, and the PARSEC isochrone ttings, we derive the reddening values of E(B - V ) = 0:42 mag and E(V - I) = 0:60 mag. Using the PARSEC isochrone ttings onto the color-magnitude diagrams, we estimate the age and metallicity to be: log(t) = 9:50 0:10 (t = 3:16 0:82 Gyr) and [Fe/H] = -0:42 0:04 dex. We present the Galactocentric radial metallicity gradient analysis for old (age > 1 Gyr) open clusters of the Dias et al. catalog, which likely follow a single relation of [Fe/H] = (-0:034 0:007)RGC + (0:190 0:080) (rms = 0.201) for the whole radial range or a dual relation of [Fe/H] = (-0:077 0:017)RGC + (0:609 0:161) (rms = 0.152) and constant ([Fe/H] -0:3 dex) value, inside and outside of RGC 12 kpc, respectively. The metallicity and Galactocentric radius (13:28 0:54 kpc) of Ruprecht 6 obtained in this study seem to be consistent with both of the relations.
Activated carbons (ACs) have been used as electrode materials of electric double-layer capacitors (EDLC) due to their high specific surface areas (SSA), stability, and ecological advantages. In order to make high-energy-density ACs for EDLC, petroleum pitch (PP) precarbonized at 500–1000°C in N2 gas for 1 h was used as the electrode material of the EDLC after KOH activation. As the pre-carbonization temperature increased, the SSA, pore volume and gravimetric capacitance tended to decrease, but the crystallinity and electrode density tended to increase, showing a maximum volumetric capacitance at a medium carbonization temperature. Therefore, it was possible to control the crystalline structure, SSA, and pore structure of AC by changing the pre-carbonization temperature. Because the electrode density increased with increasing of the pre-carbonization temperature, the highest volumetric capacitance of 28.4 F/cc was obtained from the PP pre-carbonized at 700°C, exhibiting a value over 150% of that of a commercial AC (MSP-20) for EDLC. Electrochemical activation was observed from the electrodes of PP as they were pre-carbonized at high temperatures above 700°C and then activated by KOH. This process was found to have a significant effect on the specific capacitance and it was demonstrated that the higher charging voltage of EDLC was, the greater the electrochemical activation effect was.
Activated carbons (ACs) were prepared by activation of coal tar pitch (CTP) in the range of 700°C-1000°C for 1-4 h using potassium hydroxide (KOH) powder as the activation agent. The optimal activation conditions were determined to be a CTP/KOH ratio of 1:4, activation temperature of 900°C, and activation time of 3 h. The obtained ACs showed increased pore size distribution in the range of 1 to 2 nm and the highest specific capacitance of 122 F/g in a two-electrode system with an organic electrolyte, as measured by a charge-discharge method in the voltage range of 0-2.7 V. In order to improve the performance of the electric double-layer capacitor electrode, various mixtures of CTP and petroleum pitch (PP) were activated at the optimal activation conditions previously determined for CTP. Although the specific capacitance of AC electrodes prepared from CTP only and the mixtures of CTP and PP was not significantly different at a current density of 1 A/g, the AC electrodes from CTP and PP mixtures showed outstanding specific capacitance at higher current rates. In particular, CTP-PP61 (6:1 mixture) had the highest specific capacitance of 132 F/g, and the specific capacitance remained above 90% at a high current density of 3 A/g. It was found that the high specific capacitance could be attributed to the increased micro-pore volume of ACs with pore sizes from 1 to 2 nm, and the high power density could be attributed to the increased meso-pore volume.
The leaf beetle, Chrysolina aurichalcea (Coleoptera: Chysomelidae), is a pest damaging plants of Compositae. In order to understand the genetic diversity and geographic variation of the species we sequenced a portion of mitochondrial COI gene (658 bp) and complete nuclear internal transcribed spacer 2 (ITS2) collected from seven Korean localities. A total of 18 haplotypes (BARCA01 ~ BARCA18), with the maximum sequence divergence of 3.04% (20 bp) were obtained from COI gene sequence, whereas 17 sequence types (ITS2CA01 ~ ITS2CA17), with the maximum sequence divergence of 2.013% (9 bp) were obtained from ITS2, indicating substantially larger sequence divergence in mitochondrial gene sequence. Phylogenetically, the mitochondrial DNA has shown several haplotypes formed independent groups with substantially high node support (≥ 90%), whereas no such grouping was evidenced for ITS2, indicating different behaviors of the two molecules. Such difference may reflect a diverse dynamics of the species such as biogeographic history, mating behaviors, and also possibly different mode of inheritance of the two molecules, but requires further scrutinized examination of the dataset. In terms of population genetic perspective, overall no population subdivision was detected from both molecules, except for locality 7 (Eocheong islet) from mitochondrial DNA. As more scrutinized analysis is performed, further fruitful inference on the geographic contour of the species might be available.
A new species, Hatchiana n. sp., is confirmed by morphological and molecular data. The genus Hatchiana Fender, 1966 belonging to Podabrini LeConte, 1881 is distinguished by the shape of tarsal claws: all tarsal claws with blunt tooth in both sexes. The genus was recognized as a subgenus of Podabrus Westwood, 1838. Recently, however, Hatchiana was suggested as separated genus. The genus Hatchiana was known as 12 species in Palaeartic region including three species from Korea.
In this study, we found Hatchiana n. sp. from several areas of Taean-gun, Chungcheongnam-do, Korea. This new species is different from closely relative species, Hatchiana jirisanensis (Kang & Kim, 2000) by following morphological characters: size of compound eye, length of antennae, shape of pronotum, shape of scutellum, and shape of aedeagus. Also, we compare the DNA barcoding region (the former region of CO1 gene) between these two species as molecular characters. In result, Hatchiana n. sp. is distinct from Hatchiana jirisanensis Kang & Kim by discrepancy of three percents in CO1 sequence. Therefore, the Korean Hatchiana is confirmed as four species, adding Hatchiana n. sp.
Lectins belong to the pattern-recognition receptors (PRRs) class and play important roles in the recognition and elimination of pathogens via the innate immune system. Recently, it was reported that lily-type lectin-1 is involved when a pathogen attacks in the early immune response of fish. However, this study is limited to information that the lectin is involved in the innate immune response against viral infection. In the present study, the lily-type lectin-2 and -3 of Oplegnathus fasciatus (OfLTL-2 and 3) have been presented to be included B-lectin domain and two D-mannose binding sites in the amino acid sequence that an important feature for the fundamental structure. To investigate the functional properties of OfLTLs, the tissue distribution in the healthy rock bream and temporal expression during early developmental stage analysis are performed using quantitative real-time PCR. OfLTL-2 and 3 are predominantly expressed in the liver and skin, but rarely expressed in other organ. Also, the transcripts of OfLTLs are not expressed during the early developmental stage but its transcripts are increased after immune-related organs which are fully formed. In the challenge experiment with RBIV (rock bream iridovirus), the expression of OfLTLs was increased much more strongly in the late response than the early, unlike previously known. These results suggest that OfLTLs are specifically expressed in the immune-related tissues when those organs are fully formed and it can be inferred that the more intensively involved in the second half to the virus infection.
To successful molecular breeding, identification and functional characterization of breeding related genes and development of molecular breeding techniques using DNA markers are essential. Although the development of a useful marker is difficult in the aspect of time, cost and effort, many markers are being developed to be used in molecular breeding and developed markers have been used in many fields. Single nucleotide polymorphisms (SNPs) markers were widely used for genomic research and breeding, but has hardly been validated for screening functional genes in olive flounder. We identified single nucleotide polymorphisms (SNPs) from expressed sequence tag (EST) database in olive flounder; out of a total 4,327 ESTs, 693 contigs and 514 SNPs were detected in total EST, and these substitutions include 297 transitions and 217 transversions. As a result, 144 SNP markers were developed on the basis of 514 SNP to selection of useful gene region, and then applied to each of eight wild and culture olive flounder (total 16 samples). In our experimental result, only 32 markers had detected polymorphism in sample, also identified 21 transitions and 11 transversions, whereas indel was not detected in polymorphic SNPs. Heterozygosity of wild and cultured olive flounder using the 32 SNP markers is 0.34 and 0.29, respectively. In conclusion, we identified SNP and polymorphism in olive flounder using newly designed marker, it supports that developed markers are suitable for SNP detection and diversity analysis in olive flounder. The outcome of this study can be basic data for researches for immunity gene and characteristic with SNP.
The immune system in teleost fish is not completely developed during embryonic and larval stages, therefore effective innate mechanisms is very important for survival in such an environment. However, the knowledge of the development of immune system assumed to be restricted. In many species, lysozymes have been considered as important genes of the first line immune defense. The early detection of lysozyme mRNA in previous reports, led to the investigation of its presence in oocytes. As a result, c-type lysozyme mRNA transcripts were detected in unfertilized oocytes indicating maternal transfer. Therefore, we investigated the expression patterns of lysozymes in flounder, including the matured oocyte. In our results, c-type lysozyme mRNA was first detected in unfertilized oocyte stage, observed the significantly decreased until hatching stage, and was significantly increased after hatching stage. On the other hand, g-type lysozyme mRNA transcripts were first detected at late neurula stage, and the mRNA level was significantly increased after 20 dph. It may be suggest that maternally supplied mRNAs are selectively degraded prior to the activation of embryonic transcription. This study will be help in understanding the maturation and onset of humoral immunity during development of olive flounder immune system.
