        Insect antimicrobial peptides (AMPs) have been characterized more than 150 peptides since identification of cecropin in the hemolymph of pupae from Hyalophora cecropia in 1980. Therefore, it is considered that insects are good species of AMPs selection. Insect AMPs are small (below 10 kDa), cationic, and amphipathic with variable length, sequence, and structure. They perform a critical role on humoral immunity in the insect innate immune system against invading pathogens such as bacteria, fungi, parasites, and viruses. Most of insect AMPs are induced rapidly in the fat bodies and other specific tissues of insects after septic injury or immune challenge. Then the AMPs subsequently released into the hemolymph to act against microorganisms. These peptides have a broad antimicrobial spectrum against various microorganisms including anticancer activities. Insect AMPs can be divided into four families based on their structures and sequences. That is α-helical peptides, cysteine-rich peptides, proline-rich peptides, and glycine-rich peptides/proteins. For instance, cecropins, insect defensins, proline-rich peptides, and attacins are common insect AMPs, but gloverins and moricins have been identified only in lepidopteran species. In this presentation, we focus on AMPs from insects and discuss current knowledge and recent progresses with potential application of insect AMPs.

        The dynastid beetle Allomyrina dichotoma has been used as a herbal medicine. Recently, we performed de novo RNAsequencing of Allomyrina dichotoma and identified several antimicrobial peptide candidates based on in silico analysis.Among them, cationic antimicrobial peptide, Allomyrinasin, was selected and we assessed the anti-inflammatory activitiesof Allomyrinasin against mouse macrophage Raw264.7 cells. The results showed that Allomyrinasin decreased the nitricoxide production of the lipopolysaccharide-induced Raw264.7 cells. In addition, quantitative RT-PCR, ELISA and Westernblot analysis revealed that Allomyrinasin reduced cytokine expression levels in the Raw264.7 cells. Taken together, thesedata indicated that Allomyrinasin had anti-inflammatory activity in the lipopolysaccharide-induced Raw264.7 cells.

        We have analyzed the transcriptome of Scolopendra subspinipes mutilans using RNA sequencing and identified severalantimicrobial peptide candidates. Among the peptides, named scolopendrasins, were selected based on the physicochemicalproperties of antimicrobial peptides via an in silico analysis. As a result, we evaluated the antimicrobial activities ofscolopendrasins against Gram positive and negative bacteria including Candida albicans by radial diffusion assay and colonycount assay. We also investigated the cytotoxicity of scolopendrasins through hemolysis assay. We found that the actionof scolopendrasins involved binding to the surface of microorganisms via a specific interaction with lipopolysaccharides,lipoteichoic acid, and peptidoglycans, which are components of the bacterial membrane. These results will provide a basisfor developing therapeutic agents such as peptide antibiotics.

        The objective of this study was to evaluate the effect of the Allomyrina dichotoma larva (AD) on allergy and inflammation.We examined inhibitory effect of AD on allergic reactions in mast cells (RBL-2H3) activated by Compound 48 / 80and inflammatory response in macrophages (Raw 264.7) activated by LPS. Anti-allergy and anti-inflammatory actions ofAD water extract had no cytotoxicity. At these concentrations AD inhibited ẞ-hexosaminidase, tumor necrosis factor- α(TNF- α), interleukin-4 (IL-4), interleukin-6(IL-6) and cyclooxygenase-2(COX-2). AD also inhibited the expression of inducibleNO synthase (iNOS). AD reduced the release of inflammatory cytokines including IL-4, IL-6, TNF-α, and ẞ-hexosaminidase.These results suggest that AD may be potential anti-allergy and anti-inflammatory therapeutic agent.

        We recently reported the in vitro and in vivo antiobesity effects of Tenebrio molitor larvae, a traditional food in manycountries, but it remains unknown how the larvae affect appetite regulation in mice with diet-induced obesity. We hypothesizedthat the extract of T. molitor larvae mediates appetite by regulating neuropeptide expression. We investigated T. molitorlarvae extract's (TME's) effects on anorexigenesis and endoplasmic reticulum (ER) stress–induced orexigenic neuropeptideexpression in the hypothalami of obese mice. Central administration of TME suppressed feeding by down-regulating theexpression of the orexigenic neuropeptides neuropeptide Y and agouti-related protein. T. molitor larvae extract significantlyreduced the expression of ER stress response genes. These results suggest that TME and its bioactive components arepotential therapeutics for obesity and ER stress–driven disease states.

        Previously, we performed de novo RNA sequencing of Scolopendra subpinipes mutilans using high-throughput sequencing technology and identified several AMP candidates. Among them, a synthetic peptide (CP112) was designed based on the physicochemical properties of antimicrobial peptide such as length, charge, isoelectric point. Here, we have assessed the antimicrobial activities of CP112 against various microbes and the antioxidative effects. The results showed that CP112 had antimicrobial activities in radial diffusion assay and colony count assay. In addition, we found that CP112 bound to the surface of microorganisms via a specific interaction with lipoteichoic acid, lipopolysaccharide and peptidoglycan, which is one of bacteria cell wall components. Furthermore, CP112 has shown significant DPPH radical scavenging activity. Taken together, the results would be provided the basis for developing of peptide antibiotics and antioxidants.

        The purpose of this study was performed to determine the whitening effect of organic solvent extracts from the centipede, Scolopendra subspinipes mutilans. We prepared different concentrations (50%, 70% and 100%) of ethanol, methanol, 100% ethyl acetate and water extracts. We tested melanin inhibitory effect and tyrosinase activity using B16/F10 melanoma cell. As a result, treatment of organic solvent extracts is decreased the biosynthesis of melanin and tyrosinase activity to 30~60%. Especially the 70% ethanol extracts was the most effective in B16/F10 melanoma cells. In the study on melanogenic protein expression, 70% ethanol extracts of Scolopendra subspinipes mutilans blocked glycosylation of tyrosinase. Therefore this result suggests that 70% ethanol extracts could be developed as a skin whitening agents.

Among them, a synthetic peptide (scolopendrasin I) was designed based on SVM algorithm. In this study, we reported that the synthetic peptide scolopendrasin I had an antimicrobial and anticancer activity. As a result, scolopendrasin I showed antibacterial activities against Gram positive and Gram negative bacteria strains in radial diffusion assay and colony count assay without hemolytic activity. In addition, we confirmed that scolopendrasin I bound to the surface of bacteria via a specific interaction with lipoteichoic acid and lipopolysaccharide, which is one of bacteria cell membrane components. In addition, we found that scolopendrasin I had anticancer activities in the human leukemic T lymphocyte cell line Jurkat using MTS assay. In conclusion, our results suggested that scolopendrasin I could be useful for developing peptide antibiotics and anticancer agents.

        The centipede Scolopendra subspinipes mutilans has been a medically important arthropod species by using it as a traditional medicine for the treatment of various diseases. In this study, we derived a novel lactoferricin B like peptide (LBLP) from the whole bodies of adult centipedes, S. s. mutilans, and investigated the antifungal effect of LBLP. LBLP exerted an antifungal and fungicidal activity without hemolysis. To investigate the antifungal mechanism of LBLP, a membrane study with propidium iodide was first conducted against Candida albicans. The result showed that LBLP caused fungal membrane permeabilization. The assays of the three dimensional flow cytometric contour plot and membrane potential further showed cell shrinkage and membrane depolarization by the membrane damage. Finally, we confirmed the membrane-active mechanism of LBLP by synthesizing model membranes, calcein and FITC-dextran loaded large unilamellar vesicles. These results showed that the antifungal effect of LBLP on membrane was due to the formation of pores with radii between 0.74 nm and 1.4 nm. In conclusion, this study suggests that LBLP exerts a potent antifungal activity by pore formation in the membrane, eventually leading to fungal cell death.

        An assessment was made of the toxicity of 12 insecticides, three essential oils and Bacillus thuringiensis var. israelensis (Bti) alone or in combination with the oil major constituents, (E)-anethole (AN), (E)-cinnamaldehyde (CA) and eugenol (EU), to third instars from bamboo forest collected Aedes albopictus and rice paddy field collected Anopheles sinensis resistant to various groups of insecticides. The toxicity of the test insecticides, essential oils and binary mixtures of Bti and the oil constituents (1:1 ratio) was evaluated using a direct-contact mortality bioassay. Binary mixtures of B.t.i. and CA, AN or EU were significantly more toxic against Ae. albopictus larvae (LC50, 0.0084, 0.0134 and 0.0237 mg/l) and An. sinensis larvae (0.0159, 0.0388 and 0.0541 mg/l) than either B.t.i. (1.7884 and 2.1681 mg/l) or CA (11.46 and 19.43 mg/l), AN (16.66 and 25.11 mg/l) or EU (24.60 and 32.14 mg/l) alone. Based on the co-toxicity coefficient (CC) and synergistic factor (SF), the three binary mixtures operated in a synergy pattern (CC, 140.7–368.3 and SF, 76–213 for Ae. albopictus CC, 75.1–245.3 and SF, 40–136 for An. sinensis).The binary mixtures of Bti and essential oil constituents described, particularly (E)-cinnamaldehyde, merit further study as potential mosquito larvicides for the control of malaria vector mosquito populations in light of global efforts to reduce the level of highly toxic synthetic insecticides in the aquatic environment.

        A quantitative sequencing (QS) protocol that detects the frequencies of sodium channel mutations (M815I, T917I and L920F) responsible for knockdown resistance in permethrin-resistant head lice was tested as a population genotyping method. Genomic DNA fragments of the sodium channel α-subunit gene that encompass the three mutation sites were PCR-amplified from individual head lice with either resistant or susceptible genotypes, and combined together in various ratios to generate standard DNA template mixtures for QS. Following sequencing, the signal ratios between resistant and susceptible nucleotides were calculated and plotted against the corresponding resistance allele frequencies. Quadratic regression coefficients of the plots were close to 1, demonstrating that QS is highly reliable for the prediction of resistance allele frequencies. Prediction of resistance allele frequencies by QS in several globally collected lice samples including 12 Korean lice populations suggested that permethrin resistance varied substantially amongst different geographical regions. Three local populations of Korean lice were determined to have 9.8-36.7% resistance allele frequencies, indicating that an urgent resistance management is needed. QS should serve as a preliminary resistance monitoring tool for proper management strategies by allowing early resistance detection.

        We have made semi-analytical studies to investigate the configurations of caustics and the probability distribution of the flux factor K for the binary microlensing including external shears. A parametric equation of critical curve is derived in a 4th order complex polynomial. We present the topological dependencies of the caustics for selected gamma parameters (0, 0.3, 0.6, 1.3, 2.0, and 2.5) and convergence terms (0., 0.4, 0.8, 1.2, 1.6, and 2.0). For the purpose of analyzing the efficiency of High Amplification Event (HAE) on each caustics, we examine the probability distribution of the flux factor by a Monte Carlo method. Changing the separation of the binary system from 0.8 to 1.3 (in normalied unit), we examine the probability distribution of the K-values in various gamma parameters. The relationship between gamma parameters, seperations and their probabilties of the flux factor K have been studied. Our results show that the relatively higher K values (K>1.5) are increased as increasing the separation of the binary system. We therfore conclude that, in the N-body microlensing, the probabilities of higher HAEs are inversely proportional to the star density as well. We also point out that the present research might be used as a preliminary step toward investigating heavy N-body microlensing simulations.

        Acinetobacter baumannii is usually considered an opportunistic pathogen that it responsible for a variety of nosocomial infections, such as, pneumonia, tracheobronchitis, meningitis, endocarditis, peritonitis, skin and soft tissue infections, and urinary tract infections. However, over the years, the organism has developed substantial antimicrobial resistance, and thus, the management of infections has become more difficult. The less common infective endocarditis is one of the more serious consequences of nosocomial bacteremia. Here, we report the successful treatment of the first case of multidrug-resistant A. baumannii endocarditis in a 33-year-old patient.
