        Fusarium head blight (FHB) is a major disease problem on wheat and barley around the world. F. graminearum produces trichothecenes mycotoxins such as nivalenol (NIV), deoxynivalenol (DON), and zearalenone (ZEA). The objectives of this study were to survey the natural occurrence of FHB and mycotoxins of 32 Korean wheat cultivars grown in 2011-2012 seasons at the National institute of crop science, Iksan, Korea. There was great deal of rainfall and high humidity during flowering time in May 2011. FHB incidence was counted by Fusarium infected spikes per square meter. The samples of 32 wheat cultivar were collected. The grain and flour samples were to analysis for DON and NIV by gas chromatography and ZEA by high performance liquid chromatography. The result showed that the average of FHB incidence(%) per square meter in 2011 and 2012 were 4.2%, 0.5% respectively. There were significant cultivar differences for FHB incidence ranged from 0% to 24% in 2011. All of 32 wheat cultivars contained 9-2088 ng/g for NIV and ten wheat cultivars contained 5.7-8.5 ng/g for ZEA. In addition, DON concentration of Tapdong, Shinmichal1, and Hanbaek were 217, 35 and 683 ng/g respectively. However, the grain and flour sample harvested in 2012 showed that lower FHB incidence and NIV concentration. These results showed that the 32 wheat cultivars harvested in 2011 were heavily contaminated with Fusarium mycotoxins (NIV, DON, ZEA).

        Amplified fragment length polymorphism (AFLP) is one of molecular marker technique based on DNA and is extremely useful in detection of high polymorphism between closely related genotypes like Korean wheat cultivars. Six Korean wheat cultivar specific marker sets have been developed from inter simple sequence repeat (ISSR) analysis and we can identify the 13 Koran wheat cultivars form other cultivars using six that (Son et al., 2013). We used four combinations of primer sets in our AFLP analysis for developing additional cultivar specific markers in Korean wheat. Twenty-one of the AFLP bands were isolated from ACG/M-CAC primer combination and 19 bands were isolated from E-AGC/M-CTG primer combination, respectively. We used forty bands to design sequence characterized amplified region (SCAR) primer pairs for Korean wheat cultivar identification. Only one of 40 amplified primer pairs, C2, were able to use for wheat cultivar identification. The DNA band of 215bp length was amplified by C2 primer pairs in ten cultivars, Eunpa, Olgeuru, Gobun, Saeol, Milsung, Sinmichal, Jokyung, Sugang, Goso, and Joah. Then C2 primer was applied to these primer sets as newly SCAR marker, six cultivars are identifying from other cultivars, additionally. Finally, to use the C2 and six primer sets, 19 Korean wheat cultivars are identified.

        Stem rust of wheat caused by Puccinia graminis f. sp. Tritici is historically one of the most important diseases of crops worldwide. The objective of this study was to screen the growth habits and yield of wheat accessions in Korea winter condition. We collected ninety stem rust resistant germplasms from USDA-Grain and planted for the characteristics of growth and yield in 2013. We measured the growth characteristics such as the plant type, leaf color, heading date, stem length, spike length, awn length, number of spikelet, number of tiller, and 1,000 kernel weight (g). Early maturity is most important wheat breeding target in double cropping system such as Korea. Heading dates of ninety cultivars ranged from May 17 to June 7. The Keumkang control cultivar headed on May 23. Heading dates of seventeen accessions had as early as Keumkang control (May 23). The numbers of semi-erect and intermediate type for growth habit were 45 and 33 respectively. The 1000 kernel weights were also measured. Mean 1000 kernel weights ranged from 16.7g (Webster) to 41.8g (123 A1C2). The average of stem length was 100.2cm and Keumkang was 58.4cm. These results showed that A4849-60P-7P-1P (Chile), MG 31492 (Ethiopia), 1338.F.6.C.3.B (Kenya), 1203 A1C2 (Kenya), and II-90-20/3-1R-1M- 3R (Mexico) were best lines for agronomic characteristic such as early heading date and grain yield

        Characteristics of agronomy, especially good yield productivity is very important in wheat (Triticum aestivum L.) as human food crop. Development of elite cultivar having good traits is required for food stability. In 2013, a new wheat line called Iksan370 has been developed by the National Institute of Crop Science, RDA. Iksan370 has hard red winter wheat characteristics and better features like resistance to biotic, abiotic stress and higher yield ability than Keumkang. Heading and maturing date were delayed about ten days and period to flowering date was shorter three or four days than Keumkang. Culm and spike length of Iksan370 were 95cm and 15.3cm, averagely. This means that culm and spike length were longer than Keumkang (79cm and 7cm, respectively). It showed higher milling rate (75.5%), protein content (13.5%) and SDS-sedimentation volume (77.5ml) than Keumkang. Genotypes of six genes, GBSSA, B, D, PPO18, Pina and Pinb, were represented that were similar to Keumkang, respectively. Average yield of Iksan370 in the preliminary trial test was 6.61 tons/ha in upland. We expect that yield productivity and farm income were increased by Iksan370.

        Fusarium head blight (FHB), primarily caused by Fusarium graminearum is a major disease problem on wheat and barley around the world. The objectives of this study were to evaluate for FHB resistance under greenhouse and filed condition in 2012 and 2013. We screened for Type I resistance in the field using spray inoculation and for Type II resistance in the greenhouse using single floret inoculation. Sumai 3 and Ning7840 were used the FHB resistant check. Five hundred and twelve lines were evaluated for resistance to initial infection (type I) and to spread of symptoms within spike (type II). The inoculum was F. graminearum (GZ3639) prepared at a concentration of 1 X 105 macroconidai/ml with Tween 20. The field screening plots were inoculated twice and mist-irrigation was applied to facilitate FHB development. FHB severity was assessed visually 21 days and determined as the percentage of symptomatic spikelets from the total of all spikelets observed in 20 spikes. Both in the greenhouse and field test, we could observe five categories of FHB severity: resistant (R: 0-20%), moderately resistant (MR: 21-40%), moderate (M: 41-60%), moderately susceptible (MS: 61-80%), and susceptible (S: 81-100%). The results showed that seventy seven lines showed the R and MR category on FHB severity. In addition, nine lines showed similar FHB severity compared to Sumai 3 (13.3%).

        국내 밀 재배 환경에서 수발아 저항성 밀 품종 육성의 기초 자료를 제공하기 위하여 금강밀 등 31 품종을 이용하여 2년 동안 조사한 농업 형질과 종자 특성이 수발아율 및 GI에 미치 는 영향을 평가하였다. 농업형질은 출수기, 성숙기, 간장, 수 장, 망장, 경수와 1수립수를 조사하였고, 종자 특성으로는 리 터중, 천립중, 종자의 길이, 폭과 두께를 측정하였다. 수발아 관련 특성으로는 모래묻이법을 이용한 수발아율과 종자 휴면 성과 상관 있는 GI를 조사하였다. 수장과 경수는 년차간 변이 가 없었으며, 리터중은 년차간 및 품종간 차이가 없었고, 이외 나머지 농업형질과 종자 특성 및 수발아율은 년차간, 품종간 및 품종과 년차간 상호작용에서 차이가 있는 것으로 나타났 다. 농업형질은 수발아 특성과 상관이 없었지만, 출수기와 성 숙기는 1수립수 및 종자 길이와 상관을 나타냈으며, 천립중과 종자 길이는 간장과 망장과는 정의 상관을 1수립수와는 부의 상관을 나타내었다. 수장과 경수는 종자 폭 및 두께와 상관을 보였다. 천립중은 수발아율 및 GI와 정의 상관을 나타내었으 며(r = 0.618, P < 0.001, r = 0.525, P < 0.01), 종자의 길이, 폭과 두께 역시 수발아율 및 GI와 정의 상관을 나타내었다. 백립계 품종은 적립계 품종에 비하여 리터중을 제외한 모든 특성에서 높은 값을 나타내었으며, 수발아율과 GI 값도 높게 나타났다. 적립계 품종인 수안밀, 수강밀, 알찬밀과 고소밀은 수발아 저항성이면서 휴면성이 강한 품종으로, 국내 밀 육종 프로그램에서 수발아 저항성 육성 모재로 이용할 수 있을 것 이다. 국내 밀 품종의 수발아 저항성 증진을 위한 계통 선발 에서 종피색과 종실의 무게와 크기도 고려해야 한다.

Thirty-two korean wheat cultivars were evaluated for resistance to spread of symptoms within spike. The 2013 field screening with wheat cultivar was located in Kimjae-si Joeonbuk Korea. All plots were inoculated twice. Mist-irrigation was applied to facilitate FHB development. FHB severity was assessed visually 21 days after inoculation on 20 arbitrarily selected spikes per plot. FHB severity was determined as the percentage of symptomatic spikelets from the total of all spikelets. For FHB resistance, the average of FHB severity of Type I exhibited ranging from 21.9% to 77.2% and FHB severity of Type II ranging from 20.8% to 100%. Namhae, Milseong, Geuru, Joen, Anbaek and Sukang were the moderately resistant cultivars while Gobun, Alchan, Dajoong, Eunpa, Shinmichal1, Eunpa and Uri were the most susceptible cultivars for Type II resistance.

        밀의 대표적인 식이성 섬유소인 아라비노자일란에 대한 국내 재배 환경에서 국내 밀 품종의 아라비노자일란 함량 변이를 조사한 결과 품종에 따른 함량 변이도 있지만 년차간, 재배조건 및 재배지역에도 영향을 받는 것으로 나타났다. 전작 조건에서 재배된 밀 품종의 아라비노자일란 함량이 답리작 조건에서 재배된 품종 보다 높았으며, 익산 지역에서 재배된 품종이 진주에서 재배된 밀 품종 보다 아라비노자일란 함량이 낮았다. 외국 계통인 CIMMYT 계통과 비교한 결과 국내 밀 품종은 아라비노자일란 함량이 높은 것으로 나타났다. 종실의 아라비노자일란 함량은 밀가루의 아라비노자일란 함량과 정의 상관을 나타내어 아라비노자일란 함량에 따른 계통 선발에 있어서 종실 함량으로 평가가 가능할 것이다. 제빵적성이 양호한 국내 밀 품종인 금강밀과 조경밀은 다른 품종에 비하여 아라비노자일란 함량이 낮았으며, 은파밀, 진품밀, 서둔밀과 신미찰 1호는 다른 품종에 비하여 아라비노자일란 함량이 높았다. 아리비노자일란 함량은 제빵 적성에 적합한 단백질 함량, 반죽 최고 높이 및 빵 부피와는 부의 상관을 나타내어 국내에서 빵용 밀 육성을 위해서는 아라비노자일란 함량이 낮은 계통으로 육성을 해야 하는데, 이러한 결과는 국내 밀 품종의 등숙 과정과 품종의 글루텐닌 조성 등의 영향으로 보여진다. 최근 들어 밀의 영양성 증진을 위하여 통밀을 이용한 가공제품 개발에 대한 연구가 미국과 유럽을 중심으로 활발하게 진행되고 있으며, 아라비노자일란 함량 관련 표지인자 개발에 대한 연구도 이루어지고 있다. 국내 밀 품종 역시 아라비노자일란 함량이 환경이나 재배 조건에 영향을 받지만 유전적인 특성에 영향을 받기 때문에 제빵 적성이 양호하면서 식이성 섬유소인 아라비노자일란 함량이 높은 품종 개발을 위해서는 지속적인 연구가 필요할 것으로 생각한다.

        Allelic variations in glutenin and puroindolines of 26 Korean wheat cultivars were evaluated to determine their effects on the physicochemical properties of flour and quality of white salted noodles. Cultivars carrying Pina-D1b and Pinb-D1b exhibited a coarser particle size of wheat flour and a higher ash and damaged starch content than those with Pina-D1a and Pinb-D1a. Glu-B1b, Glu-D1f, Glu-B3d and Pina-D1a alleles exhibited lower protein content than other alleles. Glu-A1c, Glu-B1b, Glu-D1f Glu-B3d, Glu-B3i and Pinb-D1b alleles appeared to be related to a lower SDS-sedimentation volume than other alleles. In dough rheological properties, Glu-A1a and Glu-D1d alleles showed a longer mixing time on the mixograph and maximum dough height but Glu-A3e and Glu-B3i alleles had a lower mixing time on the mixograph and a lower maximum dough height than other alleles at Glu-1 and Glu-3 loci. Regarding the quality of white salt noodles, about 10% of the variations in the hardness of cooked noodles were explained by Glu-A1 and Glu-B3 loci. Hardness rankings of cooked noodles were Glu-A1a > Glu-A1c > Glu-A1c at the Glu-A1 locus. Glu-B3h showed higher cooked noodle hardness (5.10 N) than other alleles at the Glu-B3 locus (< 4.66 N).

        PCR-based assays with co-dominant or dominant allele-specific STS (sequence tagged sites) markers and sodium-dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE) have been used in this study to discriminate Glu-1 and Glu-3 alleles in Korean wheat cultivars. Three alleles were identified at each high molecular weight glutenin (Glu-1) homoeologous locus and 10 alleles were identified at low molecular weight glutenin (Glu-3) homoeologous locus in Korean wheat cultivars. At Glu-1 loci, wheat cultivars contained Glu-A1a, b and c alleles amplified a 1093, 1063 and 920bp fragments, respectively, Glu-B1b, c and f alleles discriminated with 707(Glu-B1b and f) and 662bp(Glu-B1c) fragments and Glu-D1a, d and f alleles amplified a 576(Glu-D1d) and 612bp(Glu-D1a and f) fragments, respectively. At Glu-3 loci, wheat cultivars contained Glu-A3c, d and e alleles amplified a 573, 967 and 158bp fragments, respectively. Glu-B3d, g, h and i alleles gave a 662, 853, 1022 and 621bp, respectively and Glu-D3a, b and c alleles amplified a 884(Glu-D3a and b) and 338bp(Glu-D3c) fragments. A multiplex-PCR assay was also established which permitted the discrimination of the major Glu-1 and Glu-3 to determine glutenin compositions in a single PCR reaction and agarose gel assay because these systems could be useful to select DNA-based identification of wheat lines carrying good breadmaking performance in Korean wheat breeding programs. Three multiplex-PCR for Glu-1 and Glu-3, Glu-A1c + Glu-B1acf + Glu-D1d, Glu-A3ac + Glu-A3d + Glu-A3e and Glu-B3d + Glu-B3fg + Glu-B3h were established in this study.

We screened for Type II resistance in the greenhouse using single floret inoculation and for Type I resistance in the field using spray inoculation. Sumai 3 was used the FHB resistant check. Three hundred and seventy lines were evaluated for resistance to spread of symptoms within spike (type II). The 2012 field screening with 300 wheat lines was located in Kimjae-si Joeonbuk Korea. All plots were inoculated twice. The first inoculation was applied at anthesis for wheat. The second inoculation was applied three days after the initial inoculation (dai) for each plot. The inoculum was F. graminearum (GZ3639) prepared at a concentration of 100,000 macroconidai/ml with Tween 20 added as a wetting agent. Mist-irrigation was applied from the first inoculation on May 7 till June 7 to facilitate FHB development. FHB severity was assessed visually 21 days after inoculation on 20 arbitrarily selected spikes per plot. FHB severity was determined as the percentage of symptomatic spikelets from the total of all spikelets observed in these 20 spikes. Based on the field test, we could observe four categories of FHB severity: resistant (R: 0-20%), moderately resistant (MR: 21-40%), moderately susceptible (MS: 41-60%), and susceptible (S: 61-100%). The results showed that forty four lines showed the resistant category on FHB severity between 2.7% and 19.8%. In addition, ten lines showed similar FHB severity compared to Sumai 3 (9.9%).
