        Mushroom is cultivated as one of the major economical crops in many areas in Korea. The total production has steadily increased from approximately 186,400 M/T in 2007 to 190,104 M/T in 2011. This study was carried out to investigated optimum mixing ratio of Korean natural Juglans mandshurica for production of functional oyster mushroom. Mycelial growth of Pleurotus ostreatus showed the highest growth at the media of 20% Juglans mandshurica after culture of 34 days. But mushroom mycelial growth at Juglans mandshurica media was slower than that of the control. Total nitrogen and carbon source of Juglans mandshurica was 0.21% and 46.0%, respectively and C/N ratio was 219. Total nitrogen source and pH of substrates mixed with Juglans mandshurica were 2.8∼2.9 and 5.0, respectively. The contents of P2O5,, CaO, MgO and Na2O were no significant difference. The contents of Fe and Mn were decreased according to increasing of Juglans mandshurica, but the contents of Zn was increased. Yields and valid stipe number of fruiting body was the highest at 10% Juglans mandshurica, and diameter and thick of pileus were no significant difference to increase of Juglans mandshurica addition ratio. The L value of pileus was the highest at the 20% Juglans mandshurica during mushroom harvest, but there was no significant difference in the a-value and the b-value. Chemical contents of fruiting body were no significant difference.

The oyster mushroom, Pleurotus ostreatus, showed the 2th production to 46,598 M/T in 2011. This study was carried out to investigated optimum mixing ratio of Korean natural Acer tegmentosum for production of functional oyster mushroom. Mycelial growth of Pleurotus ostreatus showed the highest growth at the media of 10% Acer tegmentosum and was decreased to increase of Acer tegmentosum addition ratio. But mushroom mycelial growth at Acer tegmentosum media was slower than that of the control. Total nitrogen and carbon source of Acer tegmentosum was 0.19% and 44.4%, respectively and C/N ratio was 234. Total nitrogen source and pH of substrates mixed with Acer tegmentosum were 2.7∼3.0 and 4.8∼5.0, respectively. The contents of P2O5 and K2O were decreased by increasing Acer tegmentosum but the contents of CaO, MgO and Na2O were no significant difference. The contents of Fe and Mn were decreased according to increasing of Acer tegmentosum, but the contents of Zn and Mn were no significant difference. Yields of fruiting body and diameter and thick of pileus were the highest at 10% Acer tegmentosum. The L value of pileus was the highest at the 30% Acer tegmentosum during mushroom harvest, but there was no significant difference in the a-value and the b-value. The contents of P2O5,, CaO, MgO and Na2O of fruiting body were no significant difference, but contents of Cu was decreased and Fe was increased according to increasing of Acer tegmentosum.

        Runx2 and Osterix, the transcription factors for osteoblast differentiation, are known as fundamental factors to regulate the development of calcified tissues. However, the biological functions of these factors in the development of the periodontal tissues remain unclear. In this study, we investigated the distribution of Runx2 and Osterix during periodontal tissue development of the mice. Mandibles from 14-day-old mice were prepared for paraffin section. Serial sections of the mandible containing 1 st molar tooth germs were obtained as a thickness of 7 μm. Some sections were stained with hematoxylin and eosin. Others were used for immunohistochemistry for PCNA, Runx2, and Osterix. Epithelial cells in growing end of Hertwig’s epithelial root sheath (HERS) and mesenchymal cells adjacent to the growing end of HERS expressed PCNA. Undifferentiated mesenchymal cells and hard tissue forming cells like cementoblasts and osteoblasts in early stage of differentiation expressed Runx2. Fully differentiated cementoblasts and osteoblasts secreting matrix proteins expressed Osterix. However, the cells terminated the matrix formation did not express Osterix. Periodontal ligament cells expressed Runx2 and Osterix. Pulp cells expressed Runx2 only.These results suggest that Runx2 and Osterix might regulate the differentiation of cementoblasts in the same manner as osteoblasts. Runx2 might participate in the process of cementoblast differentiation in early stage, whether Osterix might regulate the maturation and matrix synthesis of the cells.

        Microsatellite loci are increasingly used as markers in the human, animal and plant genomes. Being highly mutable, microsatellite regions are able to differentiate between related taxa, even at the level of individual isolates in a single species. Studies on mushroom population structure, gene flow and dispersal between natural and cultivated species have become central in breeding programmes and the knowledge of new polymorphic, codominant markers will be a promising avenue to exploit wild genetic resources. The molecular phylogeny in 50 different commercial cultivated strains of Pleurotus eryngii using PCR amplification with URP primers and mitochondrial microsatellite primer was studed. The sizes of the polymorphic fragments obtained were in the range of 200 to 2000 bp. RAPD analysis techniques were able to detect genetic variation among the tested strains. With these isolated PCR amplification with URP primers we intend to analyse the population structure of the P. eryngii species complex and investigate the structure of the basidiomycete genome which deserves. A few single-locus microsatellite markers have been isolated in Pleurotus eryngii and Pleurotus ferulae. This technique is useful in those species where microsatellite loci are rare in the mitochondria.

        The mating-type genes control formation of the dikaryon from two haploid strains. These genes are now used in mating-type-assisted breeding programs for economically important mushrooms, especially the oyster mushroom, Pleurotus ostreatus, aiming at high-yield and high-quality standard mushroom production. However, it improves the breeding program when the breeder is able to quickly identify compatible strains in a given set of progeny. The two mating factors with their mating-type loci are used as markers for breeding and have been incorporated in a chromosome mapping investigation. The linkage maps include not only genetic markers such as the mating types that can be cored, but also molecular markers such as PCR-assisted approaches, e.g. RAPD analyses, or RFLP markers. Once mating-type genes within progeny may be more easily identified by the use of PCR-directed cloning of partial mating-type genes. We analyzed homeodomain (HD1 and HD2) and pheromone receptor(rcb1, 2 and 3) genes as molecular markers for breeding using mating type A and B of Pleurotus eryngii and Pleurotus ferulae by direct PCR.

        The susceptibility of the Blattella germcanica (L.) in the Republic of Korea (ROK) to insecticides was evaluated under laboratory conditions using 12 insecticides currently used by the local public health centers and/or pest control operators in the ROK. The insecticides included seven pyrethroids and five organophosphages. Based on their LD50values, the order of susceptibility of B. germanica adults to the insecticides was chlorpyrifos-methyl, profenofos, and chlorpyrifos with the values of 0.07, 0.29, and 0.88 ㎍/♀, respectively. The least susceptibility was obtained with tetramethrin at LD50 of 7.39 ㎍/♀. In the comparative resistance test, the resistance ratios (RR) of 12 insecticides were compared to each other using field collected B. germanica adults in Seoul between 1993 and 2007. Blattella germanica demonstrated higher RRs to pyrethroids such as λ-cyhalothrin, and low RRs among the organophosphates. Among the pyrethroids, λ-cyhalothrin had the highest RRs of 111 and 129 fold differences at LD50 and LD90 values, respectively. Among the organophosphates, profenofos was observed to have the highest RRs of 4 and 15 fold differences at LD50 and LD90 values, respectively. However, there were no significant differences in susceptibility to tetramethrin, chlorpyrifos, and fenitrothion. Blattella germanica was more susceptible to pyridafenthion showing a 0.7 fold difference in a resistance rate (RRLD50), respectively. Resistance ratio of tetramethrin was low, but susceptibility was also not high.

        Tissue stem cells are used for the regenerative medicine. In previous study we observed hard tissue formation of human dental pulp-derived cells using alginate scaffold. In this study, we explore the ability to differentiate of the 13th passage cells with glycerol 2-phosphate disodium salt hydrate (β-GP) which accelerate calcification. Reverse transcriptase Polymerase Chain Reaction (RT-PCR), transplants using alginate scaffold and histological examination were performed. We observed the expression of DSPP mRNA on day 10 cultured cells with β-GP. In conclusion, the 13th passage cells still have an ability to differentiate into odontoblast-like cells and alginate supports the differentiation of cultured cells in the transplants.

        This study was undertaken to develop PCR primers for the identification and detection of Streptococcus anginosus using species-specific forward and reverse primers. These primers targeted the variable regions of the 16S ribosomal RNA coding gene(rDNA). The primer specificity was tested against 12 S. anginosus strains and 6 different species(10 strains) of oral bacteria. The primer sensitivity was determined by testing serial dilutions of the purified genomic DNA of S. anginosus ATCC 33397T. The data showed that species-specific amplicons were obtained from all the S. anginosus strains tested, but not in the six other species. The PCR could detect as little as 0.4pg of the chromosomal DNA from S. anginosus. This suggests that the PCR primers are highly sensitive and applicable to the detection and identification of S. anginosus.

The syndrome is usually caused by the transcriptional silencing of fragile X mental retardation gene (FMR-1). In the C)π。gene디c analysis the pa디ent and his parents did not show any discontinuity in the long arm end of X chromosome, but in the PCR produαs targeω19 the CGG repeat sequence in the 5' untranslated region of FMR gene both the patient'’ s and his mother' s gDNAs produced a normal and an extra bands, sized about 400 and 800 bps, respectively, while the his father' s gDNA produced only one normal band, sized about 400 bps. 까1US , we suppose that the pa디.ent has heterozygotic alleles of FMR gene inherited matemally, and that the patient s FMR gene was in a premutated state relevant to the dentofacial abnormalities.
