Many manufacturers applying third party logistics (3PLs) have some challenges to increase their logistics efficiency. This study introduces an effort to estimate the weight of the delivery trucks provided by 3PL providers, which allows the manufacturer to package and load products in trailers in advance to reduce delivery time. The accuracy of the weigh estimation is more important due to the total weight regulation. This study uses not only the data from the company but also many general prediction variables such as weather, oil prices and population of destinations. In addition, operational statistics variables are developed to indicate the availabilities of the trucks in a specific weight category for each 3PL provider. The prediction model using XGBoost regressor and permutation feature importance method provides highly acceptable performance with MAPE of 2.785% and shows the effectiveness of the developed operational statistics variables.
Consumers have an increasingly active role in the marketing cycle, using social media channels to create, distribute, and consume digital content. In this context, this paper investigates the impact of user- and marketer-generated content on consumer purchase intentions and the approach to designing an effective social media marketing platform. Referencing a literature review of social media marketing and consumer purchase intentions, a case study of the social media-marketing platform, 0.8L, was undertaken using both qualitative and quantitative results through content analysis and a participatory survey. First, about 450 consumer reviews for ten sunscreen products posted on the 0.8L platform were compared with products’ marketer-generated content. Next, 55 subjects participated in a survey regarding purchase intentions toward moisturizing creams on the 0.8L platform. The results indicated that user-generated content (i.e., texts and photos) provided more personal experiences of the product usage process, whereas marketers focused on distinctive product photos and features. Moreover, customer reviews (particularly high volume and narrative format) had more impact on purchase decisions than marketer information in the online cosmetics market. Real users’ honest reviews (both positive and negative) were found to aid companies’ prompt and straightforward assessment of newly released products. In addition to the importance of customer-driven marketing practices, distinctive user experience design features of a competitive social media-marketing platform are identified to facilitate the creation and sharing of sincere customer reviews that resonate with potential buyers.
This research investigates how digital design agencies and client companies incorporate design thinking―a human-centered approach to solving difficult problems―into their website development processes. Based on a literature review on the design-thinking-driven web development process, multiple case studies of award-winning website projects were performed by in-depth interviews with key practitioners. Through analyses of these cases, their user-centered approaches for website innovation were identified according to the following four phases: web planning (discovery and defining phases) and web design and development (development and delivery phases). Moreover, distinct approaches of design thinking practices were identified according to two website types: a brand promotion website with a killer brand storytelling approach; and a service channel website with a strategic UI/UX-driven approach. Next, the key success drivers of these website projects were suggested in light of the typical themes of design thinking (i.e., human centeredness, research based, context awareness, and collaboration). Some practical limitations were also found in adopting the design-thinking-driven web development process, such as limited research methods and tools, and insufficient prototyping and experimentation. Along with these limitations, it was also discovered that current digital design agencies still face the following challenges in adopting the design-thinking-driven web development process: building a long-term, playful partnership with clients; leveraging decision-makers' design thinking awareness; and coping with limited resources (design thinking practitioners, budgets, and schedules).
Physicochemical properties of buckwheat roasted at specified temperatures (175, 200, 225 and 250°C) and times (5 and 10 min) were investigated. Appearance of roasted buckwheat showed that non-enzymatic browning reaction occurred mainly at the embryo due to the presence of sugars and proteins. Microstructure of roasted buckwheat showed the formation of air cells inside the endosperm indicating the expansion of grain during roasting. Rapid Visco Analyzer profiles revealed that the extent of starch gelatinization was depending preferentially upon the roasting time rather than temperature. Total polyphenol and flavonoid content in roasted buckwheat were not significantly different from those of unroasted grains, although polyphenols and flavonoids in ethanol extracts increased slightly after roasting. Antioxidant activities of buckwheat increased after roasting.
In order to identify effective approaches for creating more viral Facebook posts, this research conducted an empirical content analysis of leading Korean brands' Facebook fan-pages (Samsung Mobile, SK Telecom, Kia Motors, and POSCO). Their distinctive visual storytelling and communication patterns were investigated as effective user engagement triggers. Through analysis of the research results, it was statistically proved that the different industrial attributes of the four brands, which are primarily characterized by their product (or service) types, affect their Facebook posting patterns by showing different engaging rates (measured by like, comment, and share metrics). In addition, the user engagement rates of the posts were influenced by their visual storytelling factors (i.e. ad objective, value scale, and visual media types). In line with these statistical findings, the distinctive visual storytelling strategies of the four brands were identified. Moreover, competitive and uncompetitive visual storytelling tactics were suggested according to the ad objectives and visual media types on Facebook.
Zinc (Zn2+) is one of essential factors during mammalian oocyte maturation and fertilization. Previous studies showed that depletion of cellular Zn by metalion chelator impair asymmetric division of oocyte. But the detailed mechanism of these phenomena is unclear.
We found that depletions of zinc by cell-permeable heavy metal chelator N,N,N',N'-tetrakis(2-pyridylmethyl)ethane-1,2-diamine (TPEN) caused the decrease of cytoplasmic actin mesh level. Spire2-GFP is co-localized with zinc at the cortex and intracellular vesicle. By the treatment of TPEN, number of Spire2-GFP decorated vesicle is drastically decreased, indicating that Zn2+is essential for the localization of the spire in mouse oocyte. Two putative zinc-binding regions were located in the C-terminal part of Spire2. Mutations of zinc binding site on spire abolish its localization at the intracellular vesicle. Over expression of C-terminal region containing zinc binding site of spire impair oocyte maturations and decrease cytoplasmic actin mesh. Taken together, these results suggest that intracellular zinc is crucial for the proper localizations of spire in the mouse oocyte, and unraveling the novel regulatory mode of actin nucleator spire by Zn2+.
This study investigated whether people experience a correspondence between color hue-tone and the main characters’ emotions in the 2012 British musical drama film, Les Misérables through three practical experiments. Six screen images, which represent the characters’ different emotions (Parrot’s six primary types including love, joy, surprise, anger, sadness, and fear) were selected. For each screen image, participants were asked to judge the degree of the character’s dominant emotions evoked from 17 varied screen images, which consisted of original chromatic and achromatized images as well as 15 color-filtered images (5 hues X 3 tones of the IRI color system). These tasks revealed that a chromatic color scheme is more effective to deliver the characters’ positive emotions (i.e. love and joy) than an achromatic one. In addition, they proved that the hue and tone dimensions partially influence the relationships between the character emotions and colors.
The objective of this study was to investigate the efficiency of nicotinic acid on sperm cryosurvival and fertilization ability in frozen-thawed boar semen. Boar semen was collected by glove-hand method and was frozen using freezing solution treated to 0, 5, 10 and 20 mM of nicotinic acid. The frozen sperm for sperm characteristic analysis was thawed such as viability, acrosome reaction, and mitochondrial integrity. The frozen-thawed sperm was estimated by SYBR14/PI double staining for viability, FITC-PNA/PI double staining for acrosome reaction and Rhodamine123/PI double staining for mitochondrial integrity using a flow cytometry. The embryo was estimated in vitro development and DCFDA staining for reactive oxygen species assessment. As results, frozen-thawed sperm viability was significantly higher in 5 and 10 mM (61.1 ± 1.5%, 64.7 ± 2.0%) of nicotinic acid than other groups (0 mM, 52.1 ± 2.3%; 20 mM, 47.8 ± 5.1%, P<0.05). The live sperm with acrosome reaction was significantly higher in 5 and 10 mM of nicotinic acid (26.1 ± 1.8%, 24.9 ± 1.5%) than other groups (0 mM, 35.3 ± 0.8%; 20 mM, 36.5 ± 1.9%, P<0.05). The live sperm with mitochondrial integrity was significantly higher in 5 and 10 mM (84.2 ± 3.6%, 88.4 ± 2.3%) of nicotinic acid than other groups (0 mM, 77.3 ± 4.4%; 20 mM, 73.3 ± 3.6%, P<0.05). Blastocyst rate of in vitro development was significantly higher in 10 mM (17.0 ± 1.3%) of nicotinic acid than other groups (0 mM, 9.4 ± 0.5%; 5mM, 12.6 ± 0.8%; 20 mM, 5.0 ± 1.0%, P<0.05). Moreover, total cell number was higher in 5 and 10 mM (53.6 ± 2.9%, 57.9 ± 2.8%) of nicotinic acid than other groups (0 mM, 41.0 ± 1.4%; 20 mM, 23.2 ± 2.8%, P<0.05). Hydrogen peroxide in embryos was lower in 5 mM nicotinic acid (0.7 ± 0.1%) than other groups (0 mM, 1.0 ± 0.1%; 10mM, 0.9 ± 0.0%; 20 mM, 1.4 ± 1.0%, P<0.05). In conclusion, nicotinic acid-treated semen improves cryosurvival and quality of spermatozoa. Also, the fertilized oocytes with nicotinic acid improve quality of embryo and blastocyst formation.
We investigated the inhibitory effects of solvent extracts from Ailanthus altissima in A549 human lung cancer cell. A. altissima has been recognized as a traditional healthy food due to its various biological activities against hypertension, strokes, fever, pain, neuralgia, inflammation, and cancer effects. Recently, it has been reported that the extracts of various wild vegetables show strong anti-cancer properties by induction of apoptosis. However, the mechanisms of their cytotoxicity in human lung cancer cells have been poorly understood. The present study was investigated the effects of solvent extracts from A. altissima on cell growth and apoptosis on A549 human lung cancer cells. A treatment of A. altissima inhibited the growth of A549 cells in a dose-dependent manner by inducing apoptosis. Especially, the chloroform fraction showed the highest anti-cancer effect among five kinds of fractions. And also, induction of apoptosis by chloroform fraction were associated with down-regulation of Bcl-2, and up-regulation of pro-apoptotic Bax expression. From these results, A. altissima may have a therapeutic potential in human lung cancer cells and as a functional food.
This study was conducted to analyze the relationship between planted plants and immigration plants by monitoring the wildflower garden constructed at Seoul Samgaksan Elementary School. The results of this study are as follows:
First, plants planted in the garden 32 taxa, except for weakened only 1 taxa (Clematis patens), the remaining plants maintained or increased their populations. Among maintained Plants, that did not volume growth were 6 taxa and volume growth on the spot were 5 taxa. 15 taxa were spread around the planting area and 5 taxa spread sporadically to unpredictable areas other than the planting area. Although most of the planted plants show good growth, no special management is required, but Lonicera japonica needs the density management that cuts off the stem in the early spring and 5 sporadically spreading taxa, the planting intent of garden is attenuated, so remove the root when new leaves come out in spring.
Second, the total number of plants transferred after 2 years of wildflower garden constructed were 83 taxa total, which is 2.6 times that of planted plants. The seed disseminule form was analyzed as 52% barochory, 27% anemochory&hydrochory, 16% autochory and 5% zoochory. Also about 43% of the transferred plants were estimated to been imported from soil seed bank.
Third, we classified immigration plants into largely native plants and naturalized plants. The native plants were transferred to more than naturalized plants: 65 native plants, 1 cultivated plants and 17 (22%) naturalized plants. The results of the monitoring of the rooftop garden ‘Choroktteul’ in Seoul City Hall showed that there were differences in the type and number of the immigration plants depending on the distance and quality of the surrounding greenery.
Fourth, the management method of immigration plants is divided into three categories: elimination, transplantation, and utilization. The 39 taxa were selected for the elimination: Invasive alien plants among naturalized plants, fast spreading plants, plants that expand sideways with rhizomes, and plants that interfere with the growth of planted plants: grow to over 1 meter in height and climbing plant. The elimination period should be dry, clear, windy day, remove the root without any residue. The transplanted plants are 18 taxa, tree, plants not suitable for habitat, and in elementary school textbooks. It is a group of 26 taxa which are used in the garden: for landscape that plants have ornamental values (flowers, leaves, and autumn colors etc.) but do not attenuate the intent of the garden, and for groundcover that plants have low plant height and fast spread. It is also possible to prevent immigration plants when you are mulching or planting plants densely populated areas when making a garden.
The research concerned of the regeneration of plants from embryos obtained from anther cultures of ginseng (Panax ginseng C. A. Meyer). The aim was to determine the influence of the regeneration medium on the efficiency of the regeneration process. We conducted to determine the optimum conditions such as cold pretreatment, plant growth regulators and carbon sources on anther culture of P. ginseng. Highest callus formation rate was obtained when flower buds pretreated at 4℃ for 1 day. Among the treated growth regulators with various degrees of concentration in Murashige and Skoog's (MS) medium, 4.53 μm of 2.4-dichlorophenoxyacetic acid and 4.44 μm of 6-benzylaminopurine gives the most responsive callus with the frequency of 73.89% and 129.53 g of fresh weight. When we used 3-9% of sucrose and maltose among the different kinds and various concentrations of carbohydrates, callus was formed highest 67.29% in the medium with 3% of sucrose. Shoots induced from callus supplemented with 28.9 μm of gibberellic acid and rooted in Gamborg's B5 medium supplemented with 14.7 μm of indole-3-butyric acid.
A cDNA clone encoding a MDR-like ABC transporter protein was isolated from Brassica rapa seedlings, through rapid amplification of cDNA ends (RACE). This gene (named as Brmdr 1; GenBank accession no.: DQ296184 ) had a total length of 4222 bp with an open reading frame of 3900 bp, and encoded a predicted polypeptide of 1300 amino acids with a molecular weight of 143.1 kDa. The BrMDR1 protein shared 71.0, 62.5, 60.0 and 58.2% identity with other MDR proteins isolated from Arabidopsis thaliana (AAN28720), Coptis japonica (CjMDR), Gossypium hirsutum (GhMDR) and Triticum aestivum (TaMDR) at amino acid level, respectively. Southern blot analysis showed that Brmdr1 was a low-copy gene. Expression pattern analysis revealed that Brmdr1 constitutively expressed in the root, stem petals and stamens, but with lower expression in leaves and open flowers. The domains analysis showed that BrMDR1 protein possessed two transmembrane domains (TMDs) and two nucleotide binding domains (NBDs) arranging in "TMD1-NBD1-TMD2-NBD2" direction, which is consistent with other MDR transporters. Within NBDs three characteristic motifs common to all ABC transporters, "Walker A", "Walker B" and C motif, were found. These results indicate that BrMDR1 is a MDR-like ABC transporter protein that may be involved in the transport and accumulation of secondary metabolites.
A population of 120 double-haploid lines derived from Samgang/Nagdong was used for analyzing the developmental behaviors of plant height and tiller number by conditional mapping via composite interval approaches at five growth stages of rice. The traits were normally distributed with transgressive segregations in both directions. A total of 4 QTLs associated with plant height were identified in 40th/initial, 50th/40th and 60th/50th periods, respectively. The qph7.1 was observed in 50th/40th and 60th/50th periods, with increasing effects from Samgang and Nagdong alleles, respectively. This indicated that an individual gene or genes at the same genomic region might have opposite genetic effects at various growth stages. Five QTLs associated with tiller number were identified in four stages except 60th/50th stage. Four of five increasing effects of QTLs were from Nagdong alleles. Six of nine QTLs for two traits were detected before 60 days. It was evident that the expression of QTLs for plant height and tiller number was more active at early stages than at maturity stage. Furthermore, QTLs identified at early stages showed opposite additive effects on plant height and tiller number. These results provided a possible genetic explanation for the negative correlation between two traits.
Genetic variation and structure of 9 subpopulations of Symplocarpus renifolius Schott ex Tzvelev on Mt. Cheonma, in Korea, were determined via starch-gel electrophoresis. The genetic diversity at 10 loci for 8 isozymes (P99=66%, A=2.26, Ho=0.212, He=0.230) was found to be considerably higher than that seen in other long-lived perennial plants. On the whole, the genotype frequencies were in accordance with Hardy-Weinberg expectations. Approximately 5%(θ=0.049) of the total variability was among subpopulations. The high levels of observed genetic diversity in S. renifolius were attributed to a universal outcrossing system and other specific factors like differences in age classes and widely scattered individuals around the main distribution. Heterozygosity was highest at a mid-range of elevation(450m~600m). The lowest heterozygosity at lower elevation was attributed to the possible origin of seeds transported by water from upstream regions during the monsoon season. Spatial structure in a subpopulation evidenced a strong autocorrelation between closer individuals within 3~4m of distance. This was assumed to be attributable to the restricted seed dispersal characteristics of S. renifolius. In accordance with the findings generated in this study, some implications regarding the conservation of S. renifolius at the Mt. Cheonma were also presented.
