        Heterogeneous nuclear ribonucleoprotein A2/B1 (hnRNPA2/B1) is an N6-methyladenosine (m6A) RNA modification regulator and a key determinant of premRNA processing, mRNA metabolism and transportation in cells. Currently, m6A reader proteins such as hnRNPA2/B1 and YTHDF2 has functional roles in mice embryo. However, the role of hnRNPA2/B1 in porcine embryogenic development are unclear. Here, we investigated the developmental competence and mRNA expression levels in porcine parthenogenetic embryos after hnRNPA2/B1 knock-down. HhnRNPA2/B1 was localized in the nucleus during subsequent embryonic development since zygote stage. After hnRNPA2/B1 knock-down using double stranded RNA injection, blastocyst formation rate decreased than that in the control group. Moreover, hnRNPA2/B1 knock-down embryos show developmental delay after compaction. In blastocyste stage, total cell number was decreased. Interestingly, gene expression patterns revealed that transcription of Pou5f1, Sox2, TRFP2C, Cdx2 and PARD6B decreased without changing the junction protein, ZO1, OCLN, and CDH1. Thus, hnRNPA2/B1 is necessary for porcine early embryo development by regulating gene expression through epigenetic RNA modification.

        In knowledge information society entering global unlimited competition era, our middle and small sized companies are the base of national and enterprise competitiveness and core base of national economy as an origin of jobs. The competitiveness of middle and small sized companies is so important to decide the level of the competitiveness of the whole industries, but the gap from big sized company is much low in comparison with advanced countries from angles of important factors of enterprise growth, productivity and technological competitiveness, so, it's urgent to improve the competitiveness through productivity enhancement. The purpose of this research is to do a positive study on effects of learning organization management outcomes of middle and small sized company on enterprise productivity among various types of outcome brought for strengthening of enterprise competitiveness. It confirmed learning organization has functions and usefulness as a management tool bringing positive effects on enterprise productivity through the Productivity improvement.

        Recently, service quality must reflect several demands of customers who show rapid and various changes so as to be compared with the past. So, objective and rapid methods for them are necessary more. For them, first of all, service company must calculate their standard of service quality accurately by measuring service quality exactly. To measure service quality accurately, this researcher collected and analyzed data by survey for customers who are customers of logistics services, grasped potential satisfaction standard(P) by 5 point Likert scale and one survey for accurate classification of quality attributes through weighted customer satisfaction coefficient changing quality attributes by developing the study on Kano model and Timko's customer satisfaction coefficient, and suggested Potential Customer Satisfaction Improvement index(PCSI) for examining the improvement of customer satisfaction so as to utilize them as an index of differentiated and concrete measurement of service quality.

        Physicochemical properties of buckwheat roasted at specified temperatures (175, 200, 225 and 250°C) and times (5 and 10 min) were investigated. Appearance of roasted buckwheat showed that non-enzymatic browning reaction occurred mainly at the embryo due to the presence of sugars and proteins. Microstructure of roasted buckwheat showed the formation of air cells inside the endosperm indicating the expansion of grain during roasting. Rapid Visco Analyzer profiles revealed that the extent of starch gelatinization was depending preferentially upon the roasting time rather than temperature. Total polyphenol and flavonoid content in roasted buckwheat were not significantly different from those of unroasted grains, although polyphenols and flavonoids in ethanol extracts increased slightly after roasting. Antioxidant activities of buckwheat increased after roasting.

        CDK2 inhibition plays a central role in DNA damage–induced cell cycle arrest and DNA repair. However, whether CDK2 also influences early porcine embryo development is unknown. In this study, we examined whether CDK2 is involved in the regulation of oocyte meiosis and early embryonic development of porcine. We found that disrupting CDK2 activity with RNAi or an inhibitor did not affect meiotic resumption or MII arrest. However, CDK2 inhibitor-treated embryos showed delayed cleavage and ceased development before the blastocyst stage. Disrupting CDK2 activity is able to induce sustained DNA damage as demonstrated by the formation of distinct γH2AX foci in nuclei of day 3- and day 5-embryos. Inhibiting CDK2 triggers a DNA damage checkpoint by activating of the ATM-P53-P21 pathway. However, the mRNA expression of genes involved in non-homologous end-joining (NHEJ) or homologous recombination (HR) pathways for double strand break (DSB) repair reduced after administering CDK2 inhibitor to 5-day-old embryos. Furthermore, CDK2 inhibition caused apoptosis in day 7 blastocysts. Thus, our results indicate that an ATM-P53-P21 DNA damage checkpoint is intact in the absence of CDK2; however, CDK2 is important for proper repair of the damaged DNA by either directly or indirectly influencing DNA repair-related gene expression.

        Zinc (Zn2+) is one of essential factors during mammalian oocyte maturation and fertilization. Previous studies showed that depletion of cellular Zn by metalion chelator impair asymmetric division of oocyte. But the detailed mechanism of these phenomena is unclear. We found that depletions of zinc by cell-permeable heavy metal chelator N,N,N',N'-tetrakis(2-pyridylmethyl)ethane-1,2-diamine (TPEN) caused the decrease of cytoplasmic actin mesh level. Spire2-GFP is co-localized with zinc at the cortex and intracellular vesicle. By the treatment of TPEN, number of Spire2-GFP decorated vesicle is drastically decreased, indicating that Zn2+is essential for the localization of the spire in mouse oocyte. Two putative zinc-binding regions were located in the C-terminal part of Spire2. Mutations of zinc binding site on spire abolish its localization at the intracellular vesicle. Over expression of C-terminal region containing zinc binding site of spire impair oocyte maturations and decrease cytoplasmic actin mesh. Taken together, these results suggest that intracellular zinc is crucial for the proper localizations of spire in the mouse oocyte, and unraveling the novel regulatory mode of actin nucleator spire by Zn2+.

        We analyzed the recent characteristics of Particulate Matter (PM) including PM10 (PM with diameter of less than 10μm) and PM2.5 (PM with diameter of less than 2.5μm) observed in Busan metropolitan area, and compared them with those measured in Seoul metropolitan area. This analysis includes the monthly, seasonal, and annual variations and differences, in emissions and chemical compositions observed in both Busan and Seoul areas. Synoptic meteorological conditions were investigated at the time when high PM concentrations occurred in each of the two areas. The results showed clearly decreasing trends of annual mean concentrations with strong seasonal variations: lower in summer and higher in winter in both areas. In comparison with Seoul, the seasonal variation in Busan demonstrated relatively lower, but showed greater summer fluctuations than in Seoul metropolitan area. This is implying the importance of secondary generation of PM in summer via active photochemical reaction in Busan area. In high concentration days, Busan’s chemical composition of sulfate was higher than that of nitrate in summer, whereas nitrate was higher than sulfate in Seoul. The ratios of NO3 - to SO4 2-(N/S ratio) showed lower in Busan approximately by a factor of 1/2(half of N/S ratio) in Busan compared with that in Seoul. Others such as synoptic characteristics and emission differences were also discussed in this study.

        The research concerned of the regeneration of plants from embryos obtained from anther cultures of ginseng (Panax ginseng C. A. Meyer). The aim was to determine the influence of the regeneration medium on the efficiency of the regeneration process. We conducted to determine the optimum conditions such as cold pretreatment, plant growth regulators and carbon sources on anther culture of P. ginseng. Highest callus formation rate was obtained when flower buds pretreated at 4℃ for 1 day. Among the treated growth regulators with various degrees of concentration in Murashige and Skoog's (MS) medium, 4.53 μm of 2.4-dichlorophenoxyacetic acid and 4.44 μm of 6-benzylaminopurine gives the most responsive callus with the frequency of 73.89% and 129.53 g of fresh weight. When we used 3-9% of sucrose and maltose among the different kinds and various concentrations of carbohydrates, callus was formed highest 67.29% in the medium with 3% of sucrose. Shoots induced from callus supplemented with 28.9 μm of gibberellic acid and rooted in Gamborg's B5 medium supplemented with 14.7 μm of indole-3-butyric acid.
