        Previously, we reported that the osmolarity conditions in the satellite region were affected CpG DNA methylation status while Pre-1 sequence was not affected CpG DNA methylation in pNT blastocyst stage. This study was conducted to investigate the DNA methylation status of repeat sequences in pig nuclear transfer (pNT) embryos produced under different osmolarity culture conditions. Control group of pNT embryos was cultured in PZM-3 for six days. Other two treatment groups of pNT embryos were cultured in modified PZM-3 with 138 mM NaCl or 0.05 M sucrose (mPZM-3, 320 mOsmol) for two days, and then cultured in PZM-3 (270 mOsmol) for four days. The DNA methylation status of the Pre-1 sequences in blastocysts was characterized using a bisulfite-sequencing method. Intriguingly, in the present study, we found the unique DNA methylation at several non-CpG sequences at the Pre-1 sequences in all groups. The non-CpG methylation was hypermethylated in all three groups, including in vivo group (86.90% of PZM- 3; 83.87% of NaCl; 84.82% of sucrose; 90.94% of in vivo embryos). To determine whether certain non-CpG methylated sites were preferentially methylated, we also investigated the methylation degree of CpA, CpT and CpC. Excepting in vivo group, preference of methylation was CpT>CpC>CpA in all three groups investigated. These results indicate that DNA methylation of Pre-1 sequences was hypermethylated in CpG as well as non-CpG site, regardless modification of osmolarity in a culture media.

        A diagnostic tool has been proposed to convert the observed surface distribution of hydrogen recombination line intensities into the radial distributions of the electron temperature and the density in HII regions. The observed line intensity is given by an integral of the volume emission coefficient along the line of sight, which comprises the Abel type integral equation for the volume emission coefficient. As the emission coefficient at a position is determined by the temperature and density of electrons at the position, the local emission coefficient resulted from the solution of the Abel equation gives the radial distribution of the temperature and the density. A test has been done on the feasibility of our diagnostic approach to probing of HII regions. From model calculations of an HII region of pure hydrogen, we have theoretically generated the observed surface brightness of hydrogen recombination line intensities and analyzed them by our diagnostic tool. The resulting temperatures and densities are then compared with the model values. For this case of uniform density, errors in the derived density are not large at all the positions. For the electron temperature, however, the largest errors appear at the central part of the HII region. The errors in the derived temperature decrease with the radial distance, and become negligible in the outer part of the model HII region.

        This study was performed to identify the differentially methylated region (DMR) and to examine the mRNA expression of the imprinted H19 gene in day 35 of SCNT pig fetuses. The fetus and placenta at day 35 of gestation fetuses after natural mating (Control) or of cloned pig by somatic cell nuclear transfer (SCNT) were isolated from a uterus. To investigate the mRNA expression and methylation patterns of H19 gene, tissues from fetal liver and placenta including endometrial and extraembryonic tissues were collected. The mRNA expression was evaluated by real-time PCR and methylation pattern was analyzed by bisulfite sequencing method. Bisulfite analyses demonstrated that the differentially methylated region (DMR) was located between -1694 bp to -1338 bp upstream from translation start site of the H19 gene. H19 DMR (-1694 bp to -1338 bp) exhibits a normal mono allelic methylation pattern, and heavily methylated in sperm, but not in oocyte. In contrast to these finding, the analysis of the endometrium and/or extraembryonic tissues from SCNT embryos revealed a complex methylation pattern. The DNA methylation status of DMR Region In porcine H19 gene upstream was hypo methylated in SCNT tissues but hypermethylated in control tissues. Furthermore, the mRNA expression of H19 gene in liver, endometrium, and extraembryonic tissues was significantly higher in SCNT than those of control (p<0.05). These results suggest that the aberrant mRNA expression and the abnormal methylation pattern of imprinted H19 gene might be closely related to the inadequate fetal development of a cloned fetus, contributing to the low efficiency of genomic reprogramming.

        본 연구는 동아시아 영역에 속해있는 5개 SKYNET 관측소(서울, 지바, 엣추 지마, 후쿠에 지마, 헤도 미사키)에서 6년 간 관측한 AOT 자료를 활용하여 MODIS에서 산출된 AOT를 검증하였고, 아울러 에어로솔 기후장 분석도 함께 수행하였다. 검증연구를 위해 관측소 25 km 이내의 MODIS AOT를 평균하였고, MODIS 관측시각 30분 전후 SKYNET AOT를 평균하여 시공간 일치 자료를 생산하였다. 시공간 일치 자료의 비교 결과 MODIS AOT의 정확도는 에어로솔 종류에 크게 영향을 받지 않았으나, MODIS AOT 산출과정 중 거대 입자 모형의 사용비율이 높아질수록 SKYNET AOT에 비해서 비교적 큰 값을 산출하는 경향을 보였다. 또한 AOT가 낮은 대기에서 MODIS AOT는 과대 추정하는 경향을 보였고 그 경향성은 AOT가 높아질수록 줄어들었다. MODIS-SKYNET AOT간의 회귀분석 결과 기울기는 0.86, Y절편은 0.16으로 나타났고 결정계수(R2)는 0.61로 나타났다. 이러한 통계적 결과로 미루어 볼 때 동아시아 영역에서 산출된 MODIS AOT는 지상 관측에 견줄 만큼 정확하다고 볼 수 있다.

        In the region of the intermediate open cluster M35 (NGC 2168), the time-series of V CCD images was taken for 12 nights from December 18, 2007 to September 25, 2010. From this observation, we detected 22 variable stars including 15 new ones. They are 6 δ Scuti, a Cepheid, an RR Lyrae, 9 eclipsing binaries and 5 semi-long periodic and/or slow irregular type variable stars. For the V photometry of the δ2 Scuti stars, the multi-frequency analysis was performed using the Discrete Fourier Transform and the linear least-square fitting.

        선박의 충돌회피동작의 전 단계로서 위험영역을 설정하고, 이 영역을 기준으로 위험영역과 안전영역을 경계지우기 위한 많은 연구가 이루어져 왔다. 본 연구는 가변 위험영역을 설정하기 위하여 시간의 함수로서 주어진 접근 파라미터 분석을 통하여 적절한 충돌회피 동작을 위 한 필요한 목적함수를 제시하는데 목적이 있다. 종래의 연구방법들이 가질 수 있는 DCPA 및 TCPA에 기반을 둔 위험영역의 설정 및 충돌회피 의 설정법이 가질 수 있는 문제점의 보완은 실 환경에 적용 가능한 충돌회피 알고리즘 개발의 토대를 이룬다. 단순한 DCPA 및 TCPA에 따른 충돌회피 동작의 판단은 주어진 항해상황에 적정하지 못 할 수 있다.

        The purpose of this study is to investigate learning desire based on the interests, application of clothing and textiles in Technology․Home Economics curriculum of middle school. For this research, a questionnaire survey was conducted to 404 middle school students living in Seoul and Kyunggi areas. The data was analyzed by frequency, descriptive statistics, Cronbach's α coefficient, one-way ANOVA, and Duncan-test using SPSS 14.0. The conclusions of this study are as follows: First, the perception of middle school student for Technology․Home Economics was low. Students were interested not in the clothing and textile education but in getting dressed specially, and purchasing of clothes. Second, the more interested in the area of clothing and textiles, the higher is the application of apparels coordination, apparel purchase, and apparel management. And the more interested in the area of clothing and textiles, the higher is the desire in class contents. Therefore, teaching-learning methods should be developed to improve middle school students' interests, and applications in the clothing and textiles.

        Osmolarity of culture media is one of the most important factors affecting in vitro development. This study was conducted to investigate the DNA methylation status of Pre-1 and satellite sequence in pig nuclear transfer (pNT) embryos produced under different osmolarity culture conditions. Control group of pNT embryos was cultured in PZM-3 for six days. Other two treatment groups of pNT embryos were cultured in modified PZM-3 with 138 mM NaCl or 0.05M sucrose (mPZM-3, 320 mOsmol) for two days, and then cultured in PZM-3 (270 mOsmol) for four days. Previous our studies have reported that pNT embryos cultured in both hypertonic media showed significantly higher blastocyst formation rate than that of control. The DNA methylation status of the satellite sequences in blastocyst was characterized using bisulfite-sequencing technology. The satellite region had a similar methylation pattern of in vivo blastocyst among two culture groups excepting the control group. Each level of methylation is that the satellite DNA moderately methylated (43.10% of PZM-3; 56.12% of NaCl; 55.06% of sucrose; 60.00% of in vivo embryos). As a result of the sequence of PRE-1, CpG methylation pattern was similar to three groups, including in vivo group. In case of the satellite DNA region, the osmolarity conditions were affected CpG DNA methylation status while PRE-1 sequence was not affected CpG DNA methylation in pNT blastocyst stage. These results indicate that the modification of osmolarity in a culture media may influence to spatially change of DNA methylation of repetitive sequence for pNT embryo development.

        본 연구의 목적은 중학교 1학년 과학 기본과정 지구과학영역의 자율학습용 콘텐츠에 대하여 분석 및 평가를 하는 것이다. 이를 위해 한국교육학술정보원(2008)의 '초 중등교육 e-러닝 품질관리 가이드라인(Ver.2.0)'에서 제시한 '사이버가정학습 콘텐츠 품질관리도구'를 적용하였다. 콘텐츠 분석에 대한 결과는 다음과 같다. 첫째, 학습안내는 학생들이 한 차시에 학습하게 될 내용들을 간략히 소개하고 있는 것으로 나타났다. 그리고 선수학습에서는 진단평가가 이루어지지 않은 것으로 분석되었다. 둘째, 본 학습에서는 학습자의 수준에 맞게 콘텐츠를 선택하여 학습할 수 있었고, 본 학습 시간은 15분 내외로 구성되어 있는 것으로 나타났다. 셋째, 학습평가에서는 학습자가 오답에 대한 피드백은 이루어지지 않고, 틀린 문항 수만 제시되어 있었다. 그리고 학습정리는 그 차시에 배운 중요한 내용에 대해서는 다시 요약 정리하여 제시하고 있다. 콘텐츠 평가에 대한 결과는 다음과 같다. 첫째, 요구분석, 교수학습전략, 상호작용에 대해서는 각 차시마다 큰 차이를 보이지 않았다. 그리고 윤리성에 대한 평가는 적합하지 못한 단어나 문장을 포함하고 있지 않은 것으로 나타났다. 저작권의 평가는 콘텐츠 내의 저작물에 대한 국제저작권표시방법을 준수하여 표시한 것으로 분석되었다. 둘째, 교수설계의 평가는 단순 그림 위주의 설명을 제시하였고, 플래시 등의 시각자료가 미흡한 것으로 나타났다. 그리고 지원체계의 평가에서는 학습자가 콘텐츠를 자유롭게 이용할 수 있도록 설치되어 있는 것으로 나타났다. 그러나 콘텐츠 학습 중 학습자가 메모할 수 있는 메모기능은 없는 것으로 분석되었다. 또한 평가에 대한 평가는 서술 내용에 대한 명확한 평가기준을 제시하고 있지는 않았다. 따라서 각각의 차시마다 조금씩 차이를 보였다. 셋째, 학습내용의 평가분석은 수정 보완하지 않은 탓에 최신의 정보를 포함하고 있지 않아 각 차시마다 큰 차이가 있는 것으로 나타났다.

        Next to Paper I (Jeon 2009a), time-series BV CCD images of the open cluster M38 were taken for 4 nights on December, 2009. The observations have been carried out for total 27 nights. In addition to the 20 variable stars in the Paper I, the discovery of 44 new variable stars has been presented in this paper: 6δ Scuti stars, 2γ Doradus stars, 18 eclipsing binaries and 18 semi-long periodic and/or slow irregular type variable stars. For the V photometry of the δ Scuti and γ Doradus stars, multi-frequency analysis was performed using the Discrete Fourier Transform and linear least-square fitting. The period search for the eclipsing binaries and the semi-long periodic and/or slow irregular type variable stars was performed by phase fitting method. As a result, the periods for 23 variable stars among the 44 ones were defined.
