The objectives of this study are to examine the genetic variation and the origin elucidation in oriental fruit flies (Diptera:Tephritidae) using DNA barcode, in prepartion for their future introductions into Korea. About 1,600 specimens of B.dorsalis sp. complex and B. correcta were collected from 10 countries, the Indochina peninsula, the Philippines, Taiwanand South China. A total of 182 cytochrome c oxidase I (COI) sequences were obtained and aligned from these regionalspecimens. Three chinese sequences from the GenBank reference were also included. Six hundreds base pair fragmentswere aligned and trimmed and used for a barcode. The phylogenetic tree was generated using the neighbor-joining methodwith 1,000 bootstrap replicates. There were two distinct groups in the phylogenetic tree, Bactrocera dorsalis sp. complexand B. correcta. Three specimen, intercepted in the hand-carried mango at the airport inspection and collected in the confiscatedmango, smuggled from Vietnam were included for a test. The DNA sequences from the airport were 100% identical toone of various Vietnam specimens, and that from the confiscated mango was mostly similar to those of Vietnam, suggestingthe usefulness of the barcode for elucidating the origin of oriental fruit fly.

Bactrocera zonata (Peach fruit flies) is a polyphagous fruit pest native to South and South-East area and now foundin more than 20 countries. This species is not only a serious pest of peach but also is capable of infesting various speciesof 14 plant families. In Korea, they are potential pests of a number of economically important crops including peach, apricot, apple, and even Citrus spp. In northern India, B. zonata is currently considered to be more important fruit pestthan the notorious oriental fruit flies (B. dorsalis). Due to the high risk of potential introduction to southern part of Korea,we put together a national action plan against the peach fruit fly in order to quickly and completely suppress invadingpopulation of this pest species.

The oriental fruit fly, Bactrocera dorsalis (Hendel), has potential of serious damage to various vegetables or fruits, especially genus of Citrus, such as mandarin orange and tangerine in Korea, where its larvae do damages. Animal and Plant Quarantin Agency of Korea and Animsl Systematics Laboratory of Kunans National University have collected the samples of oriental fruit fly in East Asian countries, Vietnam, Philippines, Myanmar, China, Cambodia, India and Nepal. To confirm genetic differences and structure of B. dorsalis population samples collected from those countries, we analyzed 192 individuals from 28 locations over 7 countries using 15 microsatellite loci. In total samples, number of different alleles, number of effective alleles and Shannon's Information Index were 6.421±0.364 (standard error, SE), 3.664±0.137 (SE) and 0.902±0.030 (SE), respectively. Grand means (±SE) of observed and expected heterozygosity over all loci and populations were 0.534 (±0.018) and 0.597 (±0.017), respectively. Among all populations, Fst values ranged from 0.016 to 0.705 with averaging 0.194 (±0.026).

For building up the Tephritid Tree of Life, we have so far accumulated fresh samples of 570 species of 220 tephritid genera, covering 11% and 44% of World species and genera, respectively. Based on the molecular systematics approach, the followings are examples of our findings: (1) two monophyletic groups within the superfamily Tephritoidea were clearly recognized (compatible with Willi Hennig’s Pallopteroidea and Otitoidea that are not currently used); (2) expanded concept of the family Tephritidae including parasitic taxa (Tachiniscinae, Ctenostylidae and Pyrgotidae); (3) reconfirmation of monophyletic Trypetini, Carpomyini, Tephritinae, and Dacinae; and (4) the first Tephritoidea might have evolved around the middle of Paleocene Epoch (~59 mya) and the family Tephritidae around the late Eocene (~36 mya).

The experiments were conducted from April, 2010 to July, 2011 to explore the species richness of fruit fly and to observe the incidence of Bactrocera species by using Methyl Eugenol trap. In the first experiment, 11 species such as Bactrocera arecae, B. carambolae, B. dorsali, B. kandiensis, B. latilineola, B. malaysiensis, B. neocognata, B. raiensis, B. verbascifoliae, B. correcta and Carpomya pardalina were recorded. All species except Carpomya pardalina were observed in mango. Eight species in guava and five species in jujube were recorded. Bactrocera dorsalis was found to be the most abundant in mango and guava, and B. correcta in jujube. During the second experimental period, the experimental area can be recorded as an infested area because index of fruit fly population captured in traps, FTD (flies/trap/day), was seven. The highest population (437 flies/trap/week) was observed on May 26 in mango orchard. The number of fruit flies was observed to be significant positive correlation with weekly rainfall (R2 = 0.67 in mango, R2 = 0. 34 in guava and R2 = 0.43 in jujube) and relative humidity (R2 = 0.65 in mango), and negative correlation with mean temperature (R2 = 0.72 in mango) and sunshine hour (R2 = 0.61 in mango and R2 = 0.33 in jujube).

,  , The striped fruit fly, Bactrocera scutellata, damages pumpkin and other cucurbitaceous plants. The developmental period of each stage was measured at seven constant temperatures (15, 18, 21, 24, 27, 30, and 33±1.0℃). The developmental time of eggs ranged from 4.2 days at 15℃ to 0.9 days at 33℃. The developmental period of larvae was 4.2 days at 15℃, and slowed in temperatures above 27℃. The developmental period of pupa was 21.5 days at 15℃ and 7.6 days at 33℃. The mortality of eggs was 17.1% at 15℃ and 22.9% at 33℃, Larval mortalities (1st, 2nd, 3rd) were 24.1, 27.3 and 18.2%, respectively, at 15℃, Pupal mortalities were 18.2% at 15℃ and 23.1% at 33℃. The relationship between developmental rate and temperature fit both a linear model and a nonlinear model. The lower threshold temperatures of eggs, larvae, and pupae were 12.5, 10.7, and 6.3℃, respectively, and threshold temperature of the total immature period was 8.5℃. The thermal constants required to complete the egg, larval, and pupal stages were 33.2, 118.3, and 181.2DD, respectively. The distribution of each development stages was described by a 3-parameter Weibull function.

The striped fruit fly, Bactrocera scutellata (Diptera: Tephritidae), is one of the most important pests in Cucurbitaceae plants. Among 6 attractants for B. scutellata used, Cue-lure was shown better effect than the others. Newly emerged adult of B. scutellata had been occurred early June of the year tested. There were two peaks of occurrences of B. scutellata, one was from mid-July to early August and the other was early September suggesting that B. scutellata has two generation a year. The larva of B. scutellata damaging the female flower and stem of the pumpkin was found for the frist time on Korea. The blossoms were damaged and fallen by B. scutellata which were 53.8% of male flower and 30.7% of female flower in the 6 pumpkins. The pumpkin flowers were damaged 51.9% by B. scutellata. Especially the pumpkin flowers damaged were shown 72.7% (1015/1397 individuals) from August to September. The larva of B. scutellata was found from the Trichosanthes kirilowii on 24 June 2009 in Jinan-gun Jeollabuk-do.
