저항성 기작은 AChE 저해, sodium channel modulator, glutamate gated chloride channels allosteric, mite growth inhibitors, inhibitors of acetyl CoA carboxylase, mitochondrial ATP synthase 저 해, mitochondrial complex I, II, III, 그리고 electron transport 저해 등이 있으며 약제에 따라 작용기작이 다르므로 약제살포 시 확인이 필요하 다.
The Varroa mite, Varroa destructor is an ectoparasite that parasitizes honey bees. The widespread usage of acaricides, particularly fluvalinate, has resulted in the emergence of resistance in Varroa mite populations all over the world. The goal of this study is to track fluvalinate resistance in Varroa mite field populations in Korea using both bioassay and molecular markers. To accomplish this, a residual contact vial (RCV) bioassay for on-site resistance monitoring was developed. Early mortality evaluation in the RCV bioassay was effective for reliably separating mites with the knockdown resistance (kdr) genotype, but late mortality evaluation was useful for distinguishing mites with additional resistance factors. The RCV bioassay of 14 field mite populations collected in 2021 revealed potential resistance development in four populations. Quantitative sequencing was used as an alternate method to examine the frequency of the L925I/M mutation in the voltage-gated sodium channel (vgsc), which is related with the fluvalinate kdr phenotype. While the mutation was not present in Varroa mite populations in 2020, it appeared in 2021, rose in frequency in 2022, and was practically ubiquitous across the country by 2023. This recent emergence and rapid spread of fluvalinate resistance within a span of three years demonstrate the Varroa mite's significant potential for developing resistance. This situation emphasizes the critical necessity to replace fluvalinate with alternate acaricides, such as fenpyroximate, coumaphos, and amitraz. A few novel vgsc mutations potentially involved in resistance were identified. Potential factors driving the rapid expansion of resistance were further discussed.
The present study identified several antecedents that create perceived resistance to supply chain management (SCM) change. This work particularly emphasizes SCM change, which is notable given its central role in reacting market orientation and varied environmental and managerial conditions. A careful case study on a small and medium sized food production company in South Korea leads to the formulation of our framework including one second-order construct of ‘resistance’ as well as eight first-order constructs of ‘implementing the payment terms’, ‘balancing of business process’, ‘fear of responsibility’, ‘business sustainability transparency’, ‘past experience of failure’, ‘competence of work personnel’, ‘cooperation with third parties’ and ‘sharing personnel information with partner’. The hypothesized relation of first- and second-order construct was validated using survey sample data collected from 350 respondents who completed their questionnaire instrument. Results from confirmatory factor analysis revealed that nomological validity was established at statistical significance level by identifying six first order constructs of ‘implementing the payment terms’, ‘fear of responsibility’, ‘business sustainability transparency’, ‘past experience of failure’, a conbined construct of 'competence of work personnel/ cooperation with third parties’ and ‘sharing personnel information with partner’. The findings from our work are expected to provide important insights to the strategy for SCM risk management for small and medium sized company.
Growing resistance to insecticides, especially pyrethroids, poses an increased risk for insect control world-wide. Thus, alternative compounds are urgently needed. Accordingly, my laboratory has been investigating synthetic materials, as well as natural products, which are an attractive source of new chemistries. We have examined the insecticidal and synergistic activity of new compounds using Drosophila melanogaster and the mosquitoes Aedes aegypti and Anopheles gambiae as model insects. In addition, we have also been investigating the modes of action of experimental molecules in various physiological preparations.
Natural product research included thymol, which was toxic to Ae. aegypti adults at doses (3-50 μg/mg) similar to published toxicities for the cabbage looper larvae, Trichoplusia ni. On homomultimeric Anopheles gambiae GABA receptor-chloride channel complex expressed in Xenopus laevis oocytes, thymol acts as a positive allosteric modulator, increasing the potency and maximal effectiveness of applied GABA, consistent with the sluggish paralysis it elicits in exposed insects. Other studies investigated matrine; a bioactive component extracted from Sophora flavescens that is used as the main ingredient in Chinese bio-pesticidal products. Matrine caused flaccid paralysis in headless fourth instar larvae of Ae. aegypti (50% paralysis in 5 hours at 8 ppm) and was toxic to adult females by contact (topical LD50 = 258 ng/mg). Adult toxicity was increased about 2-fold by pretreatment with the mono-oxygenase inhibitor, piperonyl butoxide. Interestingly, this compound was much less effective on D. melanogaster in either glass contact or feeding bioassays. Adult mosquito knockdown was rapid, with little or no expression of hyperactivity or hyperexcitability. Matrine (ca. 1 mM) had effects similar to thymol on expressed An. gambiae GABA receptors, and reduced EPSP amplitude at the Musca domestica neuromuscular junction, without any evidence of neuroexcitation or membrane depolarization. These physiological actions are sufficient to explain the whole animal intoxication by matrine, but require relatively high concentrations to manifest themselves.
Other studies tested a series of potassium channel blockers, including 1-((2-chlorophenyl)diphenylmethyl)-1H-pyrazole (TRAM-34), 11-dansylaminoundecanoic acid (DAUDA), and 5-hydroxydecanoic acid (5-HDC) as insecticides and synergists of pyrethroids. We also evaluated the action of these compounds in patch clamp recordings of engineered HEK cells expressing Anopheles gambiae Kv2.1 channels. Patch clamp studies revealed that fatty acid compounds without functional groups in the alkyl chain (e.g., decanoate, DAUDA) yielded a more potent blocking action on Kv2.1 currents than substituted fatty acids (e.g., 5-HDC). Also, in comparison to 2-methoxy-N-((1-phenylcyclopentyl)methyl)benzamide (2S-65465), a known Kv2 channel blocker (IC50 = 100 nM), decanoate and DAUDA were 6-fold and 12-fold less active, respectively, as blockers of potassium current. TRAM-34 was the least potent inhibitor tested in patch clamp studies (IC50 = 30 uM). When tested on D. melanogaster CNS, the compounds typically gave an initial increase in firing rate, followed by a decrease, both effects at micromolar concentrations. Various toxicity assays showed the same potency ranking as that obtained through patch-clamp recordings, indicating a possible connection between channel block and whole organism effects. In order to possibly improve the insecticidal activity of the fatty acids, we synthesized derivatized analogs (e.g., methyl esters) presumed to better penetrate the cuticular barrier, but activity did not improve. Finally, in contrast to a published patent, we found that fatty acids did not show much synergism with pyrethroids. The implications of this work for resistance management will be discussed.
The control of crop diseases still largely relies on spray applications of standard synthetic fungicides. As a result, the development of fungicide resistance by pathogens is a major limiting factor for the control of diseases. In this paper, the recent outbreak of practical resistance to fungicides including sterol demethylation inhibitor (DMI), Quinone outside inhibitor (QoI), and succinate dehydrogenase inhibitor (SDHI) fungicides as well as the progress of related research will be discussed. To combat resistance development, the Research Committee on Fungicide Resistance, the Phytopathological Society of Japan, was established in 1991 and has been conducting several activities. The Committee made a guideline indicating how to use at-risk fungicides. The occurrence of fungicide resistance may rather increase in the future as the choice of fungicides is often difficult due to the lack of effective alternatives. Development and integration of disease management tools such as disease-resistant or -tolerant cultivars and non-fungitoxic disease resistance inducers need to be accelerated more rapidly not only to dissolve the problem of fungicide resistance but also to reply public concerns about agrochemicals.
Dow AgroSciences has a long history of proactive insecticide resistance management efforts. In our experience, the key to managing resistance is to reduce selection pressure on any one mode-of-action by convincing farmers to rotate among effective insecticide products with different modes of action and to use only the number of insecticide applications required for effective Integrated Pest Management (IPM). To accomplish this, farmers may need to use non-chemical control methods and to rotate to insecticide products that are effective but may not provide the highest levels of control. Integrating rotation of effective insecticides with other IPM techniques can provide high quality and quantity of the harvested crop.
We will share our experience managing spinosyn resistance with a series of best management practices for western flower thrips (Frankliniella occidentalis Pergande) and our resistance management recommendations for ISOCLASTTM (sulfoxaflor), our new insecticide for control of sap-feeding insect pests.
Dow AgroSciences is an active member of the IRAC (Insecticide Resistance Action Committee) and strongly supports the placement of IRAC mode-of-action group numbers on insecticide product labels. This simple approach greatly facilitates effective product rotation.
Resistance management is critical to maintaining the effectiveness of the current arsenal of conventional insecticides and transgenic insect-resistant crops for as long as possible. Responsibility for resistance management is shared between manufacturers, formulator-distributors, retailers, influencers (universities, government), and farmers. It is up to all of us to make sure all stakeholders, and especially farmers, fully understand the benefits associated with insecticide resistance management (IRM) programs and the consequences associated with the resistance development in insect pest populations. ™ Trademark of The Dow Chemical Company ('DOW') or an affiliated company of Dow.
Highland agriculture is a kind of specified agricultural term based on altitude and meteorological characteristics and their main crops are seed potato and vegetables. These crops are only cultivated in summer season when insect pests are most dynamically attacking such as diamondback moth (DBM, Plutella xylostella) and aphids son on. Aphids, insect virus vector, are really annoying factor, particularly in seed potato. Furthermore, these insect pests formed small sub-population or colony based on small field area. For instance, green peach aphid (GPA, Myzus persicae) populations’ genetic backgrounds and insecticide susceptibilities were different even in the same Pyeongchang area. Therefore, we suggested that highland agriculture should adopted IRM strategy for the control of insecticide-resistant aphids. First. Monitoring of annual fluctuation of insect population using traps for more effective monitoring. Second. Performing the insecticide resistant monitoring patch by patch or field using high throughput molecular diagnosis for more accurate monitoring. Third. Establishing the insect pest management program based on these results. We will deeply discussed practical monitoring results and IRM strategy in highland agriculture.
An attempt was made to stimulate future research by providing exemplary information, which would integrate published knowledge to solve specific pest problem caused by resistance. This review was directed to find a way for delaying resistance development with consideration of chemical(s) nature, of mixture, rotation, or mosaics, and of insecticide(s) compatible with the biological agents in integrated pest management (IPM). The application frequency, related to the resistance development, was influenced by insecticide activity from potentiation, residual period, and the vulnerability to resistance development of chemical, with secondary pest. Chemical affected feeding, locomotion, flight, mating, and predator avoidance. Insecticides with negative cross-resistance by the difference of target sites and mode of action would be adapted to mixture, rotation and mosaic. Mixtures for delaying resistance depend on each component killing very high percentage of the insects, considering allele dominance, cross-resistance, and immigration and fitness disadvantage. Potential disadvantages associated with mixtures include disruption of biological control, resistance in secondary pests, selecting very resistant population, and extending cross-resistance range. The rotation would use insecticides in high and low doses, or with different metabolic mechanisms. Mosaic apply insecticides to the different sectors of a grid for highly mobile insects, spray unrelated insecticides to sedentary aphids in different areas, or mix plots of insecticide-treated and untreated rows. On the evolution of pest resistance, selectivity and resistance of parasitoids and predator decreased the number of generations in which pesticide treatment is required and they could be complementary to refuges from pesticides To enhance the viability of parasitoids, the terms on the insecticides selectivity and factors affecting to the selectivity in field were examined. For establishment of resistant parasitoid, migration, survivorship, refuge, alternative pesticides were considered. To use parasitoids under the pressure of pesticides, resistant or tolerant parasitoids were tested, collected, and/or selected. A parasitoid parasitized more successfully in the susceptible host than the resistant. Factors affecting to selective toxicity of predator are mixing mineral oil, application method, insecticide contaminated prey, trait of individual insecticide, sub-lethal doses, and the developmental stage of predators. To improve the predator/prey ratio in field, application time, method, and formulation of pesticide, reducing dose rate, using mulches and weeds, multicropping and managing of surroundings are suggested. Plant resistance, predator activity, selective insect growth regulator, and alternative prey positively contributed to the increase of the ratio. Using selective insecticides or insecticide resistant predator controlled its phytophagous prey mites, kept them below an economic level, increased yield, and reduced the spray number and fruits damaged.
