The development of advanced nuclear facilities is progressing rapidly around the world. Newly designed facilities have differences in structure and operation from existing nuclear facilities, so Safeguards by Design (SBD), which applies safeguards at the design stage, is important. To this end, designers should consider the safeguardability of nuclear facilities when designing the system. Safeguardability represents a measure of the ease of safeguards, and representative evaluation methodologies are Facility Safeguardability Analysis (FSA) and Safeguardability Check-List (SCL). Those two have limitations in the quantification of safeguardability. Accordingly, in this study, the Safeguardability Evaluation Method (SEM), which has clear evaluation criteria based on engineering formulas, was developed. Nuclear Material Accountancy (NMA), a key element of Safeguards, requires the Material Balance Area (MBA) of the target facility and performs Material Balance Evaluation (MBE) based on the quantitative evaluation of nuclear materials entering or leaving the MBA. In this study, about 10 factors related to NMA were developed, including MBA, Key Measurement Point (KMP), Uncertainty of a detector, Radiation signatures, and MUF (Material Unaccounted For). For example, one of the factors, MUF is used in MBA to determine diversion through analysis of unquantified nuclear materials and refers to the difference between Book Inventory and Physical Inventory, as well as errors occurring during the process in bulk facilities, errors in measurement, or intentional use of nuclear materials. This occurs in situations such as attempted diversion, and accurate MUF evaluation is essential for solid Safeguards implementation. MUF can be evaluated using the following formula (MUF=(PB+X-Y)-PE). The IAEA’s Safeguards achievement conditions (MUF < SQ) should be met. Considering this, MUF-related factors were developed as follows. (􀜵􀜧􀜯 = 1 − 􀯆􀯎􀮿 􀯌􀯊 ) In this way, about 10 factors were developed and described in the text. This factors is expected to serve as an important factor in evaluating the safeguardability of NMA, and in the future, safeguardability factors related to Containment & Surveillance (C&S) and Design Information Verification (DIV) will be additionally developed to conduct a comprehensive safeguardability evaluation of the target facility. This methodology can significantly enhance safeguardability during the design stage of nuclear facilities.

This study was aimed to isolate bacterial inoculants producing chitinase and evaluate their application effects on corn silage. Four corn silages were collected from four beef cattle farms to serve as the sources of bacterial inoculants. All isolates were tested against Fusarium graminearum head blight fungus MHGNU F132 to confirm their antifungal effects. The enzyme activities (carboxylesterase and chitinase) were also measured to isolate the bacterial inoculant. Based on the activities of anti-head blight fungus, carboxylesterase, and chitinase, L. buchneri L11-1 and L. paracasei L9-3 were subjected to silage production. Corn forage (cv. Gwangpyeongok) was ensiled into a 10 L mini silo (5 kg) in quadruplication for 90 days. A 2 × 2 factorial design consists of F. graminearum contamination at 1.0104 cfu/g (UCT (no contamination) vs. CT (contamination)) and inoculant application at 2.1 × 105 cfu/g (CON (no inoculant) vs. INO (inoculant)) used in this study. After 90 days of ensiling, the contents of CP, NDF, and ADF increased (p<0.05) by F. graminearum contamination, while IVDMD, acetate, and aerobic stability decreased (p<0.05). Meanwhile, aerobic stability decreased (p<0.05) by inoculant application. There were interaction effects (p<0.05) on IVNDFD, NH3-N, LAB, and yeast, which were highest in UCT-INO, UCT-CON, CT-INO, and CT-CON & INO, respectively. In conclusion, this study found that mold contamination could negatively impact silage quality, but isolated inoculants had limited effects on IVNDFD and yeast.

Nuclear fusion energy is considered as a future energy source due to its higher power density and no emission of greenhouse gas. Therefore, various researches on nuclear fusion is being conducted. One of the key materials for the nuclear fusion research is tritium because the D-T reaction is the main reaction in the nuclear fusion system. However, that tritium can also be used for non-peaceful purposes such as hydrogen bombs. Therefore, it is necessary to establish the safeguards system for tritium. In that regards, this study analyzed the possibility of applying safeguards to tritium. To achieve this objective, the tritium production capacity through the light water reactor was analyzed. Tritium Production Burnable Absorber Rod (TPBAR) was modeled through the MCNP code, and tritium production was analyzed. The TPBAR is composed of a cylindrical tube with a double coating of aluminum and zirconium, filled with a sintered lithium aluminate (LiAlO2) pellet to prevent the release of tritium. Tritium is produced by the reaction of Li-6 in the TPBAR with neutrons, and it is extracted and stored at the Tritium Extraction Facility (TEF). As a result, the tritium production increased as the burnup and Li-6 mass increased. In addition, when the tritium produced in this way was transferred to TEF and extracted through the process, the application of safeguards measures was analyzed. To this end, various safeguards measures were devised, such as setting an Material Balance Area (MBA) for TEF and analyzing Material Balance Period (MBP). As there is no designated Significant Quantity (SQ) for tritium, cases were classified based on the type and form of nuclear weapons to estimate the Sigma MUF (Material Unaccounted For) of the TEF. Finally, the comprehensive application of safeguards to tritium was discussed. This research is expected to contribute to the establishment of IAEA safeguards standards related to tritium by applying the findings to actual facilities.

Roller Compacted Concrete Pavement (RCCP) is a pavement placed and compacted using an asphalt paver and a compaction roller by applying a small amount of concrete mixture and shows excellent structural performance as a result of hydration reaction of cement and interlocking of aggregates by roller compaction. It also provides economic advantages over conventional concrete pavements by reducing unit cement content and construction period, simplifying construction process, and decreasing traffic closure time (Wayne, 2006). However, given that it tends to show lower IRI levels than common concrete pavements since its low unit water content and binder weight ratios make uniform quality control difficult and roller compaction after paving makes the surface irregular and rough, with rough profile at the bottom of the pavement being reflected on the surface, RCCP is used mainly in port and industrial roads for low speed (60km/h or less) traffic (Dale Harringtion, 2010; Gregory, 2009). In order to apply RCCP to high-speed roadways, diamond grinding (DG) or asphalt overlay that is highly effective in improving roughness is needed (Fares Abdo, 2014; Gregory, 2009). Applying DG over RCCP leads to excellent skid resistance and noise reduction effects as a great percentage of aggregates makes the pavement surface rough, enhancing durability of concrete and the life of DG functionality. In addition, RCCP can be used as a high performance base layer of composite pavements, as it can reduce reflecting cracking at joints and cracked sections thanks to early strength development and low drying shrinkage of concrete. In this study, we assessed longitudinal roughness improvement effects by roughness-affecting factor by applying DG methods and asphalt overlays to three RCCP sites with a variety of sub-structural conditions and analyzed the effects on roughness of existing RCC pavements depending on surfacing method (DG, APOverlay).

Porcine epidemic diarrhea virus (PEDV) causes acute diarrhea and dehydration in pigs with high mortality. PEDV is belong to Coronavirus, enveloped, single-stranded, positive-sense RNA virus. PEDV particles were composed of four structure proteins such as a glycosylated peplomer (spike, S) protein, envelope (E), glycosylated membrane (M) protein, and unglycosylated RNA-binding nucleocapsid (N) protein. Many of previous studies talk about this four structure proteins have a great potential to diagnosis and prevent PEDV. In this study we investigated expression of these structure proteins using the bacterial and baculovirus expression system. In bacterial expression system, our results showed that structure proteins fused polyhedrin and intein gene were expressed higher than non-fusion structure proteins. The expressed fusion proteins were used to immune mice for generating a polyclonal antibodies. In baculovirus expression system, co-infection of insect cells with these four recombinant baculoviruses led to self-assembly of virus-like particles as demonstrated by Transmission electron microscopy. They were confirmed by western blot analysis using pre-made polyclonal antibodies. Finding in this study may provide important information for vaccine and diagnostic development.

The population of managed honey bees has been dramatically declining the recent past in worldwide. The one of most common disease of bees is nosemosis, the nosemosis is caused by microsporidia in the genus Nosema. Nosema apis and N. ceranae have been described as honeybee pathogens. These microsporidia are highly evoloved fungi with an obligately intracellular parasitic lifstyle. The disease causes significant detriment to honey production and results in economic losses. In our knowledge, Fumagillin is the only antibiotic approved for control of nosemosis in honey bees, however this antibiotic may have unintended effects on the honey bee host, ultimately contributing to increased prevalence and pathogenicity of Nosema. Therefore, we screened anti-Nosema substances from entomopathogenic fungal culture filtrates using in vitro polar tube germination assay. These fungal metabolites are employed as antibiotic agents. As results, Total 3 samples (23% of 13 total samples) showing the germinating inhibition against N. ceranae. This screening method may be useful for the detection of anti-Nosema substances from various samples and selected samples in this study may be a good feature to be used in the development of a new biocontrol method of nosemosis.

Porcine parvovirus (PPV), a member of the genus Parvovirus, family Parvoviridae, is a significant causative agent in porcine reproductive failure, causing serious economic losses in the swine industry. PPV is a non-enveloped virus and its capsid is assembled from three viral proteins (VP1, VP2, and VP3). The major capsid protein, VP2 is the main target for neutralizing antibodies in PPV. When VP2 was expressed in large amounts, it assembled into virus-like particles (VLPs) similar in size and morphology to the original virions. In this study, we generated the recombinant Bombyx mori nucleopolyhedrovirus (BmNPV) to express the VP2 protein. Expression of the VP2 protein was analyzed by SDS-PAGE and Western blot. The recombinant VP2 protein of approximately 64 kDa was detected by both analyses. The formation of VLP by recombinant VP2 was confirmed through transmission electron microscopy examination. The purified VP2 protein assembled into spherical particles with diameters ranging from 20 to 22 nm.

Insect pollinators of the endanger orchid Cypripedium japonicum were surveyed and identified during two years, as a part of a conservation project of the orchid at Jukyeup-san and Hwaak-san (Mt.), South Korea. In total 40 individuals of 16 species in 4 families were identified. The dominant family was Halictidae, and Lasioglossum exiliceps Vachal visited the most frequently C. japonicum during the surveys. The average visiting frequency was 2.5 individuals per hour and the highest 4.3, from 12:00 – 13:00 in a day. After 15:00 insects did not visit the flowers at all. However, all of the visiting insects were found to not carry a pollinium or pollens of the orchard on their bodies; pollen carryover by any of the visiting insects did not occur at all. The orchid seems to require certain pollinators in particular body thickness due to its unique pollination mechanism. The orchid has two exit route openings, around 1 cm in diametre, where the entrapped insects can exit and an anther is situated just in front of each opening. It was inferred that a pollen carrier should be around 1 cm in body thickness. Therefore, the candidate species as the proper pollen carriers can be Tetralonia nipponensis Perez, Xylocopa appendiculata circumvolans Smith and Bombus consobrinus Dahlbom among the surveyed visitors.

Insects constitute the largest and most diverse group of animals on Earth. They also serve as the hosts or nutrient sources for an immense assemblage of parasites, pathogens, and predators, ranging from bacteria and fungi to plant and animals. More than 700 known fungal species from 100 genera have adopted an entomopathogenic lifestyle. These fungi are widely distributed, occurring in aquatic, forest, and agricultural habitats, and are often used as active ingredients in microbial insect pest control agents. Their mode of action against insects involves the attachment of conidia to the insect cuticle, followed by germination, cuticle penetration, and internal dissemination throughout the insect. During this process, secreted enzymes, protein toxins, and secondary metabolites can be used by the fungus to overcome the host immune system, modify host behavior, and defend host resources against competing pathogens and saprophytes. In some cases, the host insect relationship has been found to be associated with bioactive fungal metabolites. These metabolites exhibit a wide variety of insecticidal, antibacterial, antifungal, anticancer, antioxidant, and antiviral activities. Using molecular techniques and phylogenetic analyses, both the asexual (anamorphic) stages and sexual (teleomorphic) stages of entomopathogenic fungi have recently been shown as linked together. Therefore, entomopathogenic fungi, especially in complex with the host insect, might be a promising source of bioactive molecules of pharmaceutical and various industrial interests. Here, we evaluated the antimicrobial activity of entomopathogenic fungi metabolites against plant pathogenic bacteria and fungi for the use in agriculture. The radicals scavenging activity and the anticancer activity were also evaluated for pharmaceutical interests.
