        Quercetin is a natural flavonoid phytochemical that is extracted from various plants. Having an advantages due to its varied biological properties, such as anti-inflammatory, anti-viral, anti-oxidant, and anti-cancer effects, quercetin is used to treat many diseases. Recently, it has been reported that autophagy inhibition may play a key role in anti-cancer therapy. Therefore, in this study, we investigated the molecular mechanisms and anti-cancer effects of quercetin in human osteosarcoma cells via autophagy inhibition. We ascertained that quercetin inhibited cell proliferation and induced cell death, these process is demonstrated that apoptosis via the mitochondrial pathway and the caspase cascade. Quercetin also induced autophagy which was inhibited by 3-MA, autophagy inhibitor and the blockade of autophagy promoted the quercetin-induced apoptosis, confirming that autophagy is a pro-survival process. Thus, these findings demonstrate that quercetin is an effective anti-cancer agent, and the combination of quercetin and an autophagy inhibitor should enhance the effect of anti-cancer therapy.

        The purpose of this multiplex PCR assay is establishment and application for rapid and simultaneous detection of six pathogens related with insect diseases. Five pathogens were chosen based on the insect disease incidence rate in South Korea and specific primers of those pathogen were designed to detect insect diseases and test multiplex PCR for detecting Fungi; Beauveria bassiana(Bb), Metarhizium anisopliae(Ma), Bacteira; Bacillus thuringiensis(Bt), Pseudomonas aeruginosa(Pa), and Serratia marcescens(Sm). This research carried out the results detecting five kinds of insect pathogen of P. b. seulensis by multiplex PCR. Multiplex PCR is effective and save time to detect simultaneously these insect pathogens and multiple infections to prevent insect disease. In our study, using multiplex PCR, we demonstrated that P. b. seulensis was frequently infected with S. marcescens and co-infected with M. anisopliae in more than 80% of cases, indicating that such an analysis can be useful for pathogen identification, especially if different pathogens produce similar symptoms.

        Insect and natural enemy fauna were surveyed at conventional (CV) and organic-farming persimmon orchards (OF) at Jinju, Korea, using sweeping net and naked eyes in 2013 and 2014. Using sweeping net, 49 species at an OF and 18 species at a CV were observed in 2013. In 2014 too, more species were observed at OF (66 species, 33 families, 7 orders) than at a CV (25 species, 15 families, 5 orders). During both the years, dominant species were all hemipteran insects like Nysius plebejus (Lygaeidae) and Cletus punctiger (Coreidae) in both types of orchards, followed by Rhopalus maculatus (Rhopalidae), Riptortus pedestris (Alydidae). Similarly, naked eye inspection also found more species at OF (192 species, 72 families, 11 orders) than at CV (104 species, 52 families, 10 orders) in 2014. Dominant species in naked eye inspection were Lycorma emelianovi (Hemiptera: Fulgoridae), Apis mellifera (Hymenoptera: Apidae), Uroleucon cephalonopli (Hemiptera: Aphididae) at OF, and A. mellifera and R. maculatus at CV in 2014. Using naked eye inspection or sweeping net in 2014, more species of natural enemies were observed at OF (31 and 9 species, respectively) than at CV (18 and 5 species, respectively).

        Shikonin, a major ingredient in the traditional Chinese herb Lithospermumerythrorhizon, exhibits multiple biological functions including antimicrobial, anti-inflammatory, and antitumor effects. It has recently been reported that shikonin displays antitumor properties in many cancers. This study was aimed to investigate whether shikonin could inhibit oral squamous carcinoma cell (OSCC) growth via mechanisms of apoptosis and cell cycle arrest. The effects of shikonin on the viability and growth of OSCC cell line, SCC25 cells were assessed by MTT assay and clonogenic assays, respectively. Hoechst staining and DNA electrophoresis indicated that the shikonin-treated SCC25 cells were undergoing apoptosis. Western blotting, immunocytochemistry, confocal microscopy, flow cytometry, MMP activity, and proteasome activity also supported the finding that shikonin induces apoptosis. Shikonin treatment of SCC25 cells resulted in a time- and dose-dependent decrease in cell viability, inhibition of cell growth, and increase in apoptotic cell death. The treated SCC25 cells showed several lines of apoptotic manifestation as follows: nuclear condensation; DNA fragmentation; reduced MMP and proteasome activity; decrease in DNA contents; release of cytochrome c into cytosol; translocation of AIF and DFF40 (CAD) onto the nuclei; a significant shift in Bax/Bcl-2 ratio; and activation of caspase-9, -7, -6, and -3, as well as PARP, lamin A/C, and DFF45 (ICAD). Shikonin treatment also resulted in down-regulation of the G1 cell cycle-related proteins and up-regulation of p27KIP1. Taken together, our present findings demonstrate that shikonin strongly inhibits cell proliferation by modulating the expression of the G1 cell cycle-related proteins, and that it induces apoptosis via the proteasome, mitochondria, and caspase cascades in SCC25 cells.

        The boar sperm has more lipid droplets and specialty of seminal plasma compared with other species, causing difficulties of freezing sperm and decreases for the utilization of frozen semen into the artificial insemination. However, several studies reported significant results for the recovery of sperm motility and reproductive by addition of cryoprotectants and seminal plasma after thawing. This study was designed to investigate the effects of supplementation of trehalose or glycerol in the LEY (lactose and egg yolk in BTS) solution for the conventional freezing and vitrification process. Two boars aged 16 months were used to collect semen for 2 times in a week. The samples were allotted to 3 freezing solutions (LEY + glycerol 10.5% + OEP 1.5%, LEY + trehalose 1M + OEP 1.5%, and sucrose 1.5M + trehalose 1 M + OEP 1.5%) after centrifugation at 800 g for 10 minutes. Semen was equilibrated in freezing solutions for 10 minutes and injected into plastic straws with 2∼3 air bubbles to minimize freezing damages. Vitrification was performed to locate sperm in 5 cm above LN2 for 5 minutes, and the conventional freezing was conducted with an automatic freezer. Motility and survival rates were measured by CASA (Computer assisted sperm an alyzing system) and FITC (Fluorescein isothiocyanate), respectively after thawing semen at 50℃ for 12 seconds. The results were analyzed by ANOVA with STATVIEW statistical program. The vitrificatioin solution (LEY + 10.5% glycerol + 1.5% OEP) presented higher motility (20.9%) than other solutions while the solution (LEY + 1M trehalose + 1.5% OEP) showed the lowest (motility : 5.2%). However, survival rates of vitrified sperms detected by FITC showed 1~4% live sperms in almost of dead sperms at all vitrification solutions’ groups, but survival rate of freezing solution of LEY + 1M trehalose + 1.5% OEP LEY and LEY + 10.5% glycerol + 1.5% OEP were showed 49%, and 79%, respectively. There were differences (P<0.05) survival rate of conventional freezing in LEY + 10.5% glycerol + 1.5% OEP and LEY + 1M trehalose + 1.5% OEP and the remaining showed no differences. The results suggested that vitrified boar semen was not enough to be utilized for the artificial insemination, but it showed possibility to utilize for ICSI and conventional freezing with glycerol would be useful method for artificial insemination in pig while we choose the outstanding semen against tolerance to freezing damages.

        To compensate for the critical shortage of human organs for allotransplantation, xenotransplantation studies using genetically modified pigs are being performed in Korea. Two types of pigs that are used are α1,3-galactosyltransferase gene knockout (GalT KO) pigs and GalT KO+hCD46 (human complement regulatory protein) pigs. The present study measured the gestation time, birth weight, daily growth rate, and heart weight of both kinds of transgenic minipigs. The gestation period for both types of pigs was 117∼119 days. There was no difference in the body weight of GalT KO (—/+) and GalT KO (—/—) piglets, but GalT KO+hCD46 (—hCD46+/+) pigs were significantly heavier at birth than were GalT KO+hCD46 (—hCD46+/—hCD46+) pigs. During the first 10 weeks of life, the daily weight gain of GalT KO+hCD46 (—hCD46+/—CD46+) piglets, which are considered the optimal type for xenotransplantation, was 0.19 kg. The weight of hearts from GalT KO piglets up to two months of age was affected more by body weight than by age. Transgenic pigs showed no differences in gestation period or reproductive ability compared with normal pigs. These results comprise basic data that may be used in xenotransplantation studies and transgenic animal production in Korea.

        We develop a parallel cosmological hydrodynamic simulation code designed for the study of formation and evolution of cosmological structures. The gravitational force is calculated using the TreePM method and the hydrodynamics is implemented based on the smoothed particle hydrodynamics. The ini- tial displacement and velocity of simulation particles are calculated according to second-order Lagrangian perturbation theory using the power spectra of dark matter and baryonic matter. The initial background temperature is given by Recfast and the temperature uctuations at the initial particle position are as- signed according to the adiabatic model. We use a time-limiter scheme over the individual time steps to capture shock-fronts and to ease the time-step tension between the shock and preshock particles. We also include the astrophysical gas processes of radiative heating/cooling, star formation, metal enrichment, and supernova feedback. We test the code in several standard cases such as one-dimensional Riemann problems, Kelvin-Helmholtz, and Sedov blast wave instability. Star formation on the galactic disk is investigated to check whether the Schmidt-Kennicutt relation is properly recovered. We also study global star formation history at different simulation resolutions and compare them with observations.

        Severe fever with thrombocytopenia syndrome (SFTS) is an emerging disease, causing high fever, thromobocytopenia, leukopenia, gastrointestinal simptoms (vomiting, diarrhea), hemorrhage and multiorgan dysfunction. It was known that Haemaphysalis longicornis (H. longicornis), a major tick in Korea, is the principal vector for severe fever with thrombocytopenia syndrome virus (SFTSV). Firstly, to investigate the existence and prevalence of SFTSV in the country, we collected the ticks in 9 provinces in South Korea and detected SFTSV from H. longicornis ticks from 2011 to 2012. And we also investigated SFTSV infection in ticks collected from healthy humans without SFTS symptom and their residential areas. From the former study, we collected 13,053 ticks, and H. longicornis (90.8%, 11,856/13,053) was the most abundant among them. The minimum infection rate (MIR) of SFTSV in H. longicornis was 0.5% (59 pools). From the later study, totally 422 ticks of 8 species were collected from humans and at their neighborhood throughout the nation from May to October, 2013 and SFTSVs were detected from 12 pools (5.7% MIR) by RT-PCR. Finally, we diagnosed 35 patients by detecting SFTSV gene with their sera, and isolated 26 SFTSVs among them in 2013. These findings indicate that SFTSVs are prevalent and SFTS is probably endemic in Korea.

        The objective of this study was to compare the effect of semen extenders on the sperm motility, viability, acrosome integrity and functional integrity of plasma membrane (HOST: hypo-osmotic swelling test) during liquid preservation of Korean Native boar semen. In this experiment, semen was diluted in Androhep plus, Beltsville Thawing Solution (BTS), ModenaTM, Seminark and Vitasem LD. Sperm-rich fractions were collected from three Korean Native boars and sub-samples were diluted (30×106 spermatozoa/ml) in different semen extenders. Semen samples were stored at 17℃ for 96 hours. On everyday (0, 24, 48, 72, 96 h) after storage, the sperm characteristics relevant for fertility, such as sperm motility, viability, acrosome integrity and HOST positive were evaluated. The motility of spermatozoa stored in different extenders was no significantly different among other extenders (P>0.05). Also, no difference was observed among samples processed with different extenders in the percentage of sperm viability, acrosome integrity and HOST positive. All extenders maintained a high percentage (70%) of sperm motility, viability and acrosome integrity through 96 h of storage. The result of this study show that there was no significant differences among extenders in their capacity to preserve motility, viability and membrane integrity of spermatozoa from normal, fertile Korean Native boars for 96 h of liquid preservation at 17℃.

        This study was conducted to investigate the changes of hormone levels of serum progesterone (P4) and estradiol-17 β (E2) in sows of Landrace (L), Yorkshire (Y) and F1 (L × Y) (respectively n=3) with excellent ability, and to provide a baseline data for improving reproductive performance. In this experiment, the sows at the age of 12 months or more were used. The sows were fed by two way methods, one is conventional methods and the other is 3 days-flushing feed before estrus. Each pig’s blood was collected in 3, 6, 9, 12, 15 and 18 days after the estrus for the analyses of P4 and E2. Serum was separated by centrifugation for 15 min. with 3,000 rpm. Progesterone and estradiol-17β were measured by immunochemical assay (ELIZA test). In conventional feeding, serum progesterone levels were significantly (p<0.01) higher in F1 than in L and Y. No significant differences in P4 concentrations were seen between the L and Y of sows. Serum E2 levels were similar the serum progesterone levels. In the case of flushing feed, the tendency of hormonal changes were similar to conventional methods. But almost of hormonal levels were a little higher than that of conventional methods. P4 level of L and Y in flushing feed were significantly different (p<0.01). Serum E2 level of Y in flushing feed was significantly different among the breeds (p<0.01). These results were similar to the tendency of hormonal changes in general sows and moreover, flushing feed is known to develop the swine production, these results proved the fact of the methods. And these results suggested that more studies about hormonal changes in sows according to seasonal and nutritional factors should be needed.

        Propionibacterium acnes (P. acnes) cause an inflammatory acne that plays an important role in the pathogenesis of acne by inducing inflammatory mediators. Bee venom therapy has been used in oriental medicine for the relief of pain and the treatment of inflammatory diseases. However, a direct effect of bee venom in skin inflammation has not been established. The purpose of this study was to investigate anti-inflammatory properties of bee venom in skin inflammation stimulated by heat-killed P. acnes using human keratinocytes and monocytes cell line. P. acnes stimulates the production of proinflammatory cytokines such as interleukins-1β, -8, interferon-γ and tumor necrosis factor-α in HaCaT and THP-1 cells. Bee venom effectively inhibits the secretion of IL-1β, IL-8, IFN-γ, and TNF-α. P. acnes treatment activates the expression of TLR2, which results in IL-8 expression. However, bee venom treatment reduces the expression of TLR2 and IL-8. Based on these results, bee venom has effects on anti-inflammatory activity against P. acnes in HaCaT and THP-1 cells.

        Transgenic chickens have been spotlighted as an highly potent bioreactor for their fecundity, short generation time, and eggs associated with mass production of protein. In this study, we generated transgenic chickens exhibiting oviduct specific expression of human growth hormone fused to human transferrin for oral administration. Gene of the modified growth hormone located at downstream ovalbumin promoter (∼3.6 kb) was introduced to stage X blastodermal cell employing retrovirus vector system. Several transgenic chickens were successfully generated. However, genomic analyses showed unexpected deletion within the transgene. The modification of the transgene seemed to occur during germ cell formation because the deletion was detected only from the sperm DNA of the G0 founder animal. There was no evidence of deletion in the somatic cell DNA samples of the same chicken. Consequently, same pattern of the deletion was confirmed in both somatic and germ cells of the G1 progeny.
