        OBJECTIVES : In this study, microstructural components of crumb rubber modified asphalt (CRMA) binder were investigated using environmental scanning electron microscope (ESEM). To clearly understand the elemental composition of the CRMA binder, energy dispersive X-ray spectroscopy (EDX) was employed on the ESEM samples. METHODS: CRMA binders were produced using open blade mixers at 177℃ for 30 min. The binders were artificially aged through a series of accelerated aging processes. Sample preparation was done by making a mold shape on the glass slide. Thereafter, the morphology of the CRMA binder was observed using the ESEM coupled with the EDX. RESULTS : The images captured from the ESEM indicate that the unaged CRMA binder appears to have a single-phase continuous nonuniform structure after the addition of crumb rubber particles, whereas the artificially aged CRMA binder was observed to have two different phases. ESEM coupled with EDX shows detailed internal structure of the modified binders compared to other technologies (i.e., optical microscopy, atomic force microscopy, and conventional scanning electron microscope). CONCLUSIONS: The captured images resemble the internal structures such as the viscous properties of the unaged CRMA binder and the interaction between the rubber particles and the base binder at aged condition. ESEM is a powerful instrument and with the introduction of EDX, it provided more details of the network microstructure of the asphalt binder. ESEM coupled with EDX is recommended for use in future investigation of microstructure of asphalt binders.

        PURPOSES : The performance properties (indirect tensile strength, rutting resistance, and resilient modulus) of recycled aged CRM mixtures and their correlation with Superpave binder properties (viscosity, high failure temperature, G*sinδ, and stiffness) were investigated. METHODS: A series of Superpave binder tests was performed by using a rotational viscometer, DSR, and BBR to evaluate the performance properties. In addition, the CRM mixes were artificially aged through accelerated aging processes, and their properties were evaluated. The correlation between the properties of recycled aged CRM binders and the engineering properties of recycled aged CRM mixtures was experimentally determined. RESULTS : The rut depth values decreased and the ITS values increased with increasing high failure temperature. In general, the resilient modulus properties seemed to be poorly correlated with the high-temperature values, regardless of the aggregate source. CONCLUSIONS: The recycled aged CRM binders and mixtures can lead to satisfactory performance, and the properties of these binders are strongly correlated with the engineering properties of the mixtures.

        PURPOSES: This study set out to investigate the current state of microsurfacing in Texas and compared the results with the current state of the practice nationwide. METHODS: For this study, case studies were extracted from the existing literature and compared with the data obtained both from site visits by the research team and data obtained from a survey by Texas Department of Transportation (TxDOT) personnel. The successes and failures of microsurfacing are detailed and explanations of different issues are outlined. Forensic studies from Texas are included in the descriptions of those microsurfacings that failed when configured as part of the cape seals. RESULTS : Microsurfacing has been shown to be an effective pavement preservation technique when applied to an appropriate road, at an appropriate timing, and as a remedy for certain issues. The failures experienced in Texas can mostly be attributed to cape seals and an inability to recognize structurally faulty pavement. CONCLUSIONS : When applied to an appropriate road, at an appropriate timing, and as a remedy for certain issues, microsurfacing has been shown to perform well in numerous case studies. The majority of microsurfacing failures are the result of poor project selection, usually involving the treatment being applied to structurally unsound pavement.

        In this study, the effect of stacking sequence on the flexural and fracture properties of carbon/basalt/epoxy hybrid composites was investigated. Two types of carbon/basalt/epoxy hybrid composites with a sandwich form were fabricated: basalt skin-carbon core (BSCC) composites and carbon skin-basalt core (CSBC) composites. Fracture tests were conducted and the fracture surfaces of the carbon/basalt/epoxy hybrid composites were then examined using scanning electron microscopy (SEM). The results showed that the flexural strength and flexural modulus of the CSBC specimen respectively were ~32% and ~245% greater than those of the BSCC specimen. However, the interlaminar fracture toughness of the CSBC specimen was ~10% smaller than that of the BSCC specimen. SEM results on the fracture surface showed that matrix cracking is a dominant fracture mechanism for the CSBC specimen while interfacial debonding between fibers and epoxy resin is a dominant fracture process for the BSCC specimen.

        The legume family (Fabaceae) is the third largest family of flowering plants worldwide and often damaged by stink bugs, particularly Riptortus clavatus Thunberg (Heteroptera: Alydidae). This insect selectively feeds and oviposits on these plants, resulting in >50% of damage to soybeans harvested in autumn. The most common control methods for this insect are periodic application of chemical pesticides, pheromone traps, and bait crops, but still they encounters residual problem, insect resistance or low control efficacy. The current management system needs alternative control methods, which possibly combined with the present methods. Some entomopathogenic fungi have high virulence against the nymphs and adults of R. clavatus in a favorable conditions, reaching ∼80% mortality with mycosis in 3∼5 days. The genera of Beauveria and Metarhizium are possible control agents for effective and safe management of the serious pest. Fungal infection in R. clavatus was observed using an egfp-expressing fungal transformant. Thermotolerant entomopathogenic fungal conidia can be produced in cereal substrates, which enables conidia to be stable for long times. Our considerations need to be given to the combinations with other control methods, such as chemical pesticides or pheromone trap.

        We have determined the complete mitochondrial genome of the yellow-spotted long horned beetle, Psacothea hilaris (Coleoptera: Cerambycidae), an endangered insect species in Korea. The 15,856-bp long P. hilaris mitogenome harbors gene content typical of the animal mitogenome and a gene arrangement identical to the most common type found in insect mitogenomes. As with all other sequenced coleopteran species, the 5-bp long TAGTA motif was also detected in the intergenic space sequence located between tRNASer (UCN) and ND1 of P. hilaris. The 1,190-bp long non-coding A+T-rich region harbors an unusual series of seven identical repeat sequences of 57-bp in length and several stretches of sequences with the potential to form stem-and-loop structures. Furthermore, it contains one tRNAArg-like sequence and one tRNALys-likes equence. Phylogenetic analysis among available coleopteran mitogenomes using the concatenated amino acid sequences of PCGs appear to support the sister group relationship of the suborder Polyphaga to all remaining suborders, including Adephaga, Myxophaga, and Archostemata. Among the two available infraorders in Polyphaga, a monophyletic Cucujiformia was confirmed, with the placement of Cleroidea as the basal lineage for Cucujiformia. On the other hand, the infraorder Elateriformia was not identified as monophyletic, thereby indicating that Scirtoidea and Buprestoidea are the basal lineages for Cucujiformia and the remaining Elateriformia.

        Human embryonic stem (ES) cells are derived from the inner cell mass of the preimplantation embryo. Human ES cells have the capacity to differentiate into various types of cells in the body. Human ES cells are indefinite source of cells for cell therapy in various degenerative disorders including neuronal disorders. Directed differentiation of human ES cells is a prerequisite for their clinical application. The objective of this study is to develop the culture condition for the derivation of neural precursor cells from human ES cells. Neural precursor cells were derived from human ES cells in a stepwise culture condition. Neural precursor cells in the form of neural rosette structures developed into neurospheres when cultured in suspension. Suspension culture of neurospheres has been maintained over 4 months. Expressions of nestin, soxl, sox2, pax3 and pax6 transcripts were upregulated during differentiation into neural precursor cells by RT-PCR analysis. In contrast, expression of oct4 was dramatically downregulated in neural precursor cells. Immunocytochemical analyses of neural precursor cells demonstrated expression of nestin and SOX1. When induced to differentiate on an adhesive substrate, neuro-spheres were able to differentiate into three lineages of neural systems, including neurons, astrocytes and oligo-dendrocytes. Transcripts of sox1 and pax6 were downregulated during differentiation of neural precursor cells into neurons. In contrast, expression of map2ab was elevated in the differentiated cells, relative to those in neural precursor cells. Neurons derived from neural precursor cells expressed NCAM, Tuj1, MAP2ab, NeuN and NF200 in immunocytochemical analyses. Presence of astrocytes was confirmed by expression of GFAP immuno-cytochemically. Oligodendrocytes were also observed by positive immuno-reactivities against oligodendrocyte marker O1. Results of this study demonstrate that a stepwise culture condition is developed for the derivation of neural precursor cells from human ES cells.
