        Leptotrombidium pallidum and Leptotrombidium scutellare are the major vector mites for Orientia tsutsugamushi, the causative agent of scrub typhus. Before these organisms can be subjected to whole-genome sequencing, the genome sizes of L. pallidum and L. scutellare were estimated by a method based on quantitative real-time PCR. In addition, k-mer analysis of the genome sequences obtained from Illumina sequencing was conducted to verify the mutual compatibility and reliability of results. The genome sizes estimated by qPCR were 191.3±7 Mb for L. pallidum and 262.1±13 Mb for L. scutellare. The estimated genome sizes based on k-mer analysis were 175.5 Mb for L. pallidum and 286.6 Mb for L. scutellare. The estimates from two independent methods were mutually complementary and in a similar range to those of other Acariform mites. The relatively small genome size would facilitate genome analysis, which could contribute to understanding Arachnida genome evolution and mite vector competence and provide key information for scrub typhus prevention.

        Bemisia tabaci is a serious pest in various horticultural crops in the world. Due to use of chemical pesticide for their management they develop pesticide resistance and environmental contamination. It is necessary to develop alternative bio-pesticides using natural products from plants and natural enemies. Nicotiana benthamiana is a variety of wild tobacco plants and produce acyl sugars from glandular trichomes in the leaves. When adult whiteflies were reared with fresh N. benthamiana leaves, they were completely dead within 84 h. Oral feeding of 20% N. benthamiana extracts using ethanol and water showed complete mortality of whiteflies within 48 hours. Spray of N. benthamiana extracts into the leaves was lethal to eggs but not to nymphs of whiteflies. Further, tomato plants sprayed with N. benthamiana extracts were highly repellent to adult whiteflies. Quantitative real-time PCR indicated that the expression of various genes of B. tabaci was changed by oral feeding of N. benthamiana extract. This study suggests N. benthamiana extract is a useful for the control of whiteflies and can be used as an alternative natural pesticide for the whitefly management.

        The common bed bug, Cimex lectularius L. (Hemiptera: Cimicidae), is an ectoparasitic pest that feeds on humans as well as other mammals. We investigate that point mutations on the voltage-sensitive sodium channel are associated with the resistance to pyrethroids. Two point mutations (V419L and L925I) in the voltage-sensitive sodium channel (VSSC) α-subunit gene have been identified in deltamethrin-resistant bed bugs. L925I, located the intracellular loop between IIS4 and IIS5, has been previously found in a highly pyrethroid-resistant populations of whitefly. V419L, located in the IS6 transmembrane segment, is a novel mutation. To establish a population-based genotyping method as a molecular resistance monitoring tool, a quantitative sequencing (QS) protocol was developed. Frequency prediction equations were generated from the plots by linear regression, and the signal ratios were shown to highly correlate with resistance allele frequencies (r2 > 0.993). In addition to QS, the filter contact vial bioassay (FCVB) method was established and used to determine the baseline susceptibility and resistance of bed bugs to pyrethroids. A pyrethroid-resistant strain showed > 9375- and 6990-fold resistance to deltamethrin and λ-cyhalothrin, respectively. Resistance allele frequencies in different bed bug populations predicted by QS correlated well with the FCVB results, confirming the roles of the two mutations in pyrethroid resistance. Taken together, employment of QS in conjunction with FCVB method should greatly facilitate the detection and monitoring of pyrethroid resistant bed bugs in the field.

        Localized tenosynovial giant cell tumor (TGCT) usually occurs in the hand and foot regions. However, localized TGCT with extensive cartilaginous metaplasia is rare, especially in the tendon sheath of the toe. Here, we report a case of localized TGCT with cartilaginous metaplasia in a 57-year-old man. The tumor presented as a lobular mass measuring 2.2 cm in its greatest dimension and arose in the flexor digitorum tendon sheath of the right 2nd toe. Clinically, the mass was palpable 1 year ago and brought pain during walking. Microscopically, the mass was composed of focal conventional TGCT and cartilaginous components. The conventional TGCT areas consisted of mononuclear cells, multinucleated giant cells, and hemosiderin deposition. The chondroid areas were extensive and comprised more than 90% of the whole tumor. In this case, the mononuclear cells in the conventional TGCT areas showed focal immunohistochemical staining for podoplanin and S100 protein as well as diffuse staining for CD68, which is consistent with the staining pattern of conventional TGCT. The mononuclear cells in the chondroid areas were focal positive for podoplanin and diffuse positive for S100 protein. Chondroid metaplasia in diffuse TGCT has been reported in 10 cases involving the temporomandibular, elbow, and hip joints. However, there has been no report of a localized form of chondroid TGCT involving an extra-articular region.

        차 (Tea)는 네팔을 포함한 남아시아의 주요 농산물 수출품목 중에 하나이다. 네팔의 차 생산은 150년 전에 시작되었지만, 상업적인 생산 규모는 발전 가능성이 있음에도 불구하고초창기 모습 그대로이다. 차 생산에서 우위를 차지하고 있는인도, 스리랑카, 방글라데시와 같이 네팔은 좋은 품질의 차를생산할 수 있는 큰 잠재력을 가지고 있다. 그리고 현재 차 산업의 선도자로 남아시아 국가의 잠재력을 강화하기 위해 남아시아지역협력연합 (South Asian Association for the RegionalCooperation, SAARC)은 각 회원국의 National Tea board가차 산업을 육성할 수 있도록 지원하기 위하여 차 위원회를 설립할 예정이다. 이러한 남아시아지역협력연합의 차 위원회 설립에 맞추어 본 연구는 남아시아 차 산업에서 부각되고 있는네팔의 차 생산과 소비, 그리고 수출 경향을 다른 남아시아국가와 비교하여 분석하였다. 남아시아는 전세계 차 생산량의약 1/3를 생산하고 있으며, 대표적인 차 생산국가는 인도, 스리랑카, 방글라데시, 네팔이다. 남아시아 중에서 네팔은 연간약 1만8천톤의 차를 생산하고 있으며, 최근 20여년간(1994~2012) 차 생산량이 약 100배(196톤→18,309톤) 가까이 급증하고 있다. 네팔의 차 산업은 수출증대를 통해 외화획득과 경제발전에 기여할 뿐만 아니라 차 생산과 가공산업에참여하는 농가와 농촌여성의 고용창출 측면에서 매우 중요한역할을 담당하고 있다.

        In this study, characteristics and immuno-modulatory effects of Weissella cibaria JW15 isolated from Kimchi, traditional Korean fermented food, were examined for investigation of the capacity of potentially probiotic strains. We measured acid, bile, and heat tolerance, adhesive properties to intestinal epithelial cells, and inhibitory activity against pathogens. JW15 could survive at pH 3.0 for 2 hr, but not at pH 2.0. JW15 also showed tolerance to 0.3% oxgall bile salt, and heat tolerance at 70°C and 80°C for 5 min, respectively. Adhesive ability to Caco-2 cells was similar to that of Lactobacillus rhamnosus GG (LGG), a well-known commercial probiotic. JW15 exhibited antimicrobial activities to pathogenic bacteria such as Escherichia coli, Listeria monocytogenes, Staphylococcus aureus, and Salmonella enteritidis. The immuno-modulatory effects of JW15 were compared with those of LGG, a well-known immune enhancer. For analysis, production of nitric oxide (NO), NF-κB (Nuclear factor κB), Interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α) was measured. The concentration of NO induced by JW15 was higher than that by LGG at low concentration (1 × 107 cfu/mL). Low and high (5 × 107 CFU/mL) concentration of JW15 induced statistically higher production of NF-κB, IL-1β, and TNF-α than that produced by LGG, respectively. In conclusion, Weissella cibaria JW15 had ability as a probiotic strain, including acid, bile, and heat tolerance, adhesive properties to intestinal epithelial cells, and inhibitory activity against pathogens. In addition, JW15 showed better immuno-modulatory effects than LGG when NO, NF-κB, IL-1β, and TNF-α were measured. According to these results, the characteristics and immunomodulating activity of Weissella cibaria JW15 are suitable for consideration as a potential probiotic.

        Inflammation mainly mediated by innate immune cells as the first line of host defense against pathogens is an acute response that limits tissue damage and eliminates pathogens in the body. In triggering inflammation, several pattern recognition receptors work together; membrane-associated Toll-like receptors, c-type lectin receptors, retinoic acid-inducible gene-like helicase receptors, absent in melanoma-like receptors, and cytosolic nucleotide-binding domain and leucine-rich repeat receptors. Among them, inflammasome is a newly trigger of inflammation in response to exogenous and endogenous stimuli and its activation leads to the assembly of multiprotein platforms composed of NLRP3 (NOD-like receptor family, pyrin domain containing 3), ASC (apoptosis associated speck-like protein containing a CARD), and procaspase 1. Thus, the activated inflammasome activates caspase 1, resulting in processing and secretion of interleukin (IL)-1β. Recent emerging data suggest that dysregulated metabolites, i.e., amyloids, ceramides, and cholesterol crystals, have been classified as inflammasome activators. In addition, IL-1β may play a critical role in the pathogenesis of chronic inflammation-induced disorders such as Alzheimer’s diseases, type 2 diabetes, and atheriosclerosis. This review introduces the basic concept of inflammasome activation and auto-inflammatory diseases. In addition, it discusses the updated signaling models of inflammasome activation that link metabolic dysfunction in order to outline future therapeutic approaches to inflammasome-mediating diseases.

        Muscle satellite cell (SC) is responsible for postnatal muscle growth, repair, and regeneration. Satellite cell is an im-portant source of multi-potent stem cell process and differentiation into adipogenic, myogenic, and osteoblastogenic. The objective of this study was to identify alter of transcriptome during differentiation in porcine satellite cell and to elevated transcriptome at different stages of postnatal development to gain insight into the differences in differ-entiated PSC. We used RNA-seq technique to investigate the transcriptomes during differentiation in pig muscle. Sequence reads were obtained from Illumina HiSeq2000. Differentially expressed genes (DEG) were detected by EdgeR. Gene ontology (GO) terms are powerful tool for unification among representation genes or products. In study of GO biological terms, functional annotation clustering involved in cell cycle, apoptosis, extracellular matrix, phosphoryla- tion, proteolysis, and cell signaling in differences stage. Taken together, these results would be contributed to a better understanding of muscle biology and processes underlying differentiation. Our results suggest that the source of DEGs could be better understanding of the mechanism of muscle differentiation and transdifferentiation.

        Satellite cells were derived from muscular tissue in postnatal pig. Satellite cell is an important to growth and development in animal tissues or organs. However, the progress underlying induced differentiation is not clear. The aim of this study was to evaluate the morphologic and the transcriptome changes in porcine satellite cell (PSC) treated with insulin, rosiglitazone, or dexamethasone respectively. PSC was obtained from postnatal muscle tissue. In study 1, for study the effect of insulin and FBS on the differentiated satellite cells, cells were cultured at absence or presence of insulin treated with FBS. Total RNA was extracted for determining the expression levels of myo-genic PAX3, PAX7, Myf5, MyoD, and myogenin genes by real-time PCR. Myogenic genes decreased expression levels of mRNA in treated with insulin. In study 2, in order to clarify the relationship between rosiglitazone and lipid in differentiated satellite cells, we further examined the effect of FBS on lipid accumulation in the presence or absence of the rosiglitazone and lipid. Significant differences were observed between rosiglitazone and lipid by FBS. The mRNA of FABP4 and PPARγ increased in rosiglitazone treatment. In study 3, we examined the effect of dexame-thasone on osteogenic differentiation in PSC. The mRNA was increased osteoblasotgenic ALP and ON genes treated with dexamethasone in 2% FBS. Dexamethasone induces osteoblastogenesis in differentiated PSC. Taken together, in differentiated PSCs, FABP4 and PPARγ increased to rosiglitazone. Whereas, no differences to FBS and lipid. These results were not comparable with previous reports. Our results suggest that adipogenic, myogenic, and osteoblasto-genic could be isolated from porcine skeletal muscle, and identify culture conditions which optimize proliferation and differentiation formation of PSC.

        Muscular satellite cell (SC), which is stem cell of postnatal pig, is an important for study of differentiation into adipogenesis, myogenesis, and osteoblastogenesis. In this study, we isolated and examined from pig muscle tissue to determine capacity in proliferate, differentiate, and expression of various genes. Porcine satellite cells (PSC) were isolated from semimembranosus (SM) muscles of 90∼100 days old pigs according to standard conditions. The cell proliferation increased in multi-potent cell by Masson’s, oil red O, and Alizarin red staining respectively. We per-formed the expression levels of differentiation related genes using real-time PCR. We found that the differentiation into adipocyte increased expression levels of both fatty acid binding protein 4 (FABP4) and peroxisome proliferator- acti-vated receptor gamma (PPARγ) genes (p<0.01). Myocyte increased the expression levels of the myosin heavy chain (MHC), myogenic factor 5 (Myf5), myogenic regulatory factor (MyoD), and Myogenic factor 4 (myogenin) (p<0.01). Osteo-blast increased the expression levels of alkaline phosphatase (ALP) (p<0.01). Finally, porcine satellite cells were indu-ced to differentiate towards adipogenic, myogenic, and osteoblastogenic lineages. Our results suggest that muscle satellite cell in porcine may influence cell fate. Understanding the progression of PSC may lead to improved strat-egies for augmenting meat quality.

        Polyporus umbellatus (Syn. Grifola umbellata) is a sclerotium forming mushroom belongs to family Poly-poraceae of Polyphorales, Basidiomycota. The sclerotia of P. umbellatus have long been used for traditional medicinesin China, Korea and Japan. This study was initiated to obtain the basic data for artificial sclerotial production of P. umbel-latus. Here, we investigated the favorable conditions for mycelial growth of P. umbellatus and its symbiotic fungus Armill-aria mellea. We also evaluate the favorable carbon and nitrogen sources for sclerotial formation in dual culture betweenP. umbellatus and A. mellea. The favorable conditions for mycelial growth of P. umbellatus were 20°C and pH 4, whileoptimal conditions for mycelial growth of A. mellea were 25°C and pH 6. The carbon sources for optimal mycelial growthof P. umbellatus were fructose and glucose, while carbon sources for favorable mycelial growth of A. mellea were alsofructose and glucose. The nitrogen sources for favorable mycelial growth P. umbellatus were peptone and yeast extract,while optimal mycelial growth of A. mellea were obtained in peptone and yeast extract. When P. umbellatus and A. melleawere dual cultured on carbon sources, sclerotia were induced on basal media supplemented with glucose, fructose andmaltose at pH 4~6, while nitrogen sources inducing sclerotia were basal media supplemented with peptone and yeastextract for 60 days at 20°C under dark condition.

        Recently, there had been reports on ethanol fermentation from mono-saccharide and disaccharide by mushroom mycelia. This experiment was conducted to study ethanol production from xylose by mycelila of mushrooms isolated from Korea. The cultures used in this study were obtained from Culture Collection and DNA Bank of Mushrooms in the Division of Life Sciences, Incheon National University. The results showed that Neolentinus lepideus, Trametes hirsuta and Cerrena unicolor produced ethanol from xylose contained media. The ethanol concentration produced in the xylose contained media ranged from 2.5∼3.8%. The highest ethanol concentration(3.8%) was obtained from fermentation of xylose by Neolentinus lepideus mycelia. All of the mushroom mycelia used in this study showed a good ability of ethanol fermentation from glucose, fructose, mannose, cellobiose and maltose.

        Ganoderma applanatum is a medicinal mushroom belongs to Ganodermataceae of Polyporales, Basidiomycota. This study was conducted to evaluate the antioxidant, anti-inflammatory and anti-xanthine oxidase activities of G. applanatum fruiting bodies extracted with methanol. The antioxidant activities were performed on reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging, and ferrous ion chelating activities. In addition to this, polyphenol and flavonoid contents were aslo analyzed. The methanol extract showed the good reducing power of 3.5 at the concentration of 4.0 mg/ml. The scavenging activity of methanol extract of G. applanatum 1,1-diphenyl-2-picrylhydrazy radicals was better than those of positive control, BHT and tocopherol. The ferrous ion chelating effect of methanol extract was moe effective than those of positive control, BHT and tocopherol. The antioxidant activities of the G. applanatum were increased with the increasing concentration of the extracts. The xanthine oxidase inhibitory activity of methanol extract of G. applanatum was good as positive control, allopurinol. The anti-inflammatory activity of mushroom extract was measured by carrageenan-induced hind paw edema of albino rat. The injection of 50 mg/kg of methanol and hot water extracts significantly reduced the carrageenan-induced paw edema compared with the positive control, indomethacin. The results indicated that fruiting bodies of G. applanatum could be used for natural medicine for anti-inflammatory and anti-xanthine oxidase.

        Recently, there had been many reports on ethanol fermentation by mushrooms. This study was initiated to screening of ethanol fermentation by mushroom mycelilal cultures preserved in Culture Collection and DNA Bank of Mushrooms in the Division of Life Sciences, Incheon National University. The experimental results showed that ethanol concentration produced by Cerrena unicolor, Trametes pubescens and Daedalea dickinsii, Microporus vernicipes and Perenniporia fraxinea in the glucose medium ranged from 2.3∼4.7%. The highest ethanol concentration was obtained from fermentation of glucose by Cerrena unicolor (4.7%). Some of the mushrooms used in this study have a good ability to efficiently ferment arabinose, fructose, mannose, cellobiose, maltose and sucrose . The highest ethanol concentration was obtained under semi-aerobic condition compared with aerobic and anaerobic conditions. The media used for ethanol fermentation by T. pubescens and P. fraxinea. contained small amounts of β -D-glucan, which is known to have anti-tumor activity.

        The chigger mite, Leptotrombidium pallidum, is widely distributed throughout South Korea and is a major vector for Orientia tsutsugamushi, the causative agent of scrub typhus. In this study, the genome size of the chigger mite was estimated to determine the necessary coverage level prior to whole genome sequencing. Cloning of EF1α and RpS3 as putative single copy reference genes were conducted and their partial sequences were determined. Using the serially diluted reference genes with known amount as standard templates, the weight of a single copy of the genome was predicted by a method based on quantitative real time PCR. The average genome length estimated from the weight using two methods was 191 ± 7 Mb. When the genome size of other arthropods (Drosophila melanogster, Apis mellifera and Tetranychus urticae), with their genome analysis completed, were estimated using the same method and compared with actual values, the estimation accuracy was 79.8-98.9%, suggesting our current estimation of L. pallidum genome size is reliable. The estimated L. pallidum genome size is in a similar range to other Acariform mites, such as the dust mite and scabie mite, but appoximately 10-fold smaller compared to the deer tick, which belongs to Parasitiform. Our finding provides key information for further genome sequencing and understanding of mite genome evolution.

        As a part of disease vector and possible overseas inflow caused by global warming monitoring around airport and port area, the distribution of domestic mosquitoes was investigated using BG-sentinel trap. Mosquitoes were collected during 5-days per each area in early September, using 10 traps in Cheongju (airport) and 15 traps in Pyeongtaek (port). The numbers of mosquitoes collected in Cheongju and Pyeongtaek area were 1,762 including 10 species and 1,042 including 11 species respectively. In Cheongju, Aedes albopictus (657 individuals, 37.3%) and Culex pipiens complex (415 individuals, 23.6%) were dominant species; while in Pyeongtaek, Cx. pipiens complex (806 individuals, 77.4%) was dominant followed by Anopheles spp. (90 individuals, 8.6%) and Ochlerotatus koreicus (79 individuals, 7.6%). Similar species were collected in both Choengju and Pyeongtaek area. Although most species collected in both Cheongju and Pyeongtaek area were the same, Cx. inatomii was only collected in Pyeongtaek. In case of the Genus Aedes, more number of Ae. albopictus was collected compared to Oc. koreicus in Cheongju; while Oc. koreicus was more in Pyeongtaek. In case of the genus Culex, the individuals belonging to the vishinui group were collected such as Cx. bitaeniorhynchus, Cx. tritaeniorhynchus and Cx. orientalis. This study may provide basic information for efficient prevention of vector mosquito and potential overseas inflow of diseases, also we would try to expand different area in Korea.

        Anoplophora isa moderate-sized genus with 36 species of woodboring cerambycid beetles that occur throughout Asia, with the highest diversity in the tropical and subtropical region. Two species, A. chinensis (Forster) and A. glabripennis (Motchulsky) are known in the Korean peninsula. The latter is an infamous invasive species, commonly called the Asian longhorned beetle, in the U.S. causing economic damage on hard woods. For the present study, we carried out field surveys from 2010 to 2013 and literature review on the host plants of Korean Anoplophora species. A. chinensis is associated with 14 host plant species in 4 families and A. glabripennis 17 species of 11 families. Most importantly, the Manchurian striped maple, Acer tegmentosum Maxim. (Aceraceae), is newly recognized as a host of the Asian longhorned beetle. In the present study, a revised host plant checklist of Korean Anoplophora species is provided, with correction of scientific and Korean names of the host plants.

Due to the development of pesticide resistance and environmental contamination, however, it is necessary to develop alternative biopesticides using natural products from plants and natural enemies. Nicotiana benthamiana is a variety of wild tobacco plants and produce acyl sugars from glandular trichomes in the leaves. Acyl sugars are known to be highly toxic to various plant sapping insects such as whiteflies, aphids and thrips. Here, we extracted acyl sugars in two different ways from the leaves. At first, collected leaves were simply washed with water. Otherwise, collected leaves firstly dried and homogenized into the powder, then extracted with ethanol. Spray of 10% water-extracted solution into adult whiteflies showed 80% mortality. Otherwise, spray of 10% ethanol-extracted solutions showed complete mortality at 48 h after treatment and also strong repellency of adult whiteflies into the treated tomato plants. Our results suggest N. benthamiana is a useful for the control of whiteflies and can be used as an alternative natural pesticide for the whitefly management.
