        Recently an outbreaked pest belongs to Hemiptera: Recaniidae, Ricanula sp. is greatly concerned about the outspreading throughout the South of Korea by wide range of host, including Cornelian cherry, Jujube, and peach trees. In Chungbuk province, this pest was first occurred at Jincheon and Okcheon in 2012, Cheongju in 2013 and now found out at Eumseong and Goesan in 2015. Ricanula sp. was oviposited directly into one-year twig, did damages on fruit-bearing formation and finally withered the host. This study was performed to understand the ovipositional characteristics and to develop the standardized forecasting method. Oviposition by Ricanula sp. was abundant in tree than in bush, adult laid eggs on new inner twigs and then covered with wax compound. Total no. of oviposited egg-mass was 10 to 318, and that of on new twig was 5 to 185 per tree, with different to host trees. Thickness of oviposited twigs were done within 2 ~ 5.5 mm and the height was mostly founded with range of 1 to 2 m, founded with highest height over 3 m. Oviposited no. of egg-mass within 30 cm twig was appeared differently from 2 to 7 every host. From based on this investigated result, we provide this for standardized forecasting method. This pest will need to control when egg-mass will occur over 2 at new twig, within 30 cm from the tip, set as total 25 point/ 5 plants (5 point per plant).

        The species composition of the parasitoid complex and the degree of parasitism by each species were analyzed for Ivela auripes Butler (Lepidoptera, Lymantridae) populations in the increasing phase. Total of 2,177 eggs, larvae and pupae were collected and reared from the Gwangneung forest Pocheon-si, Korea. The most abundant parasitoids from each developmental stage were: a trichogrammatid wasp (Trichogramma kurosuae) from eggs, a braconid wasp (Cotesia melanoscela) from larvae, an ichneumonid wasp (Pimpla disparis) and a tachinid fly (Tachinidae sp.1), each from pupae. 19 other parasitoids, mostly hyperparasitoids, also occurred including Tachinidae sp.2, Pteromalidae sp.1, Ichneumonidae sp.3, and Apechthis capulifera. Illustrations and a list of the species parasitizing of I. auripes are provided.

        The aim of this study was to carry out the comparative analysis of the brushing force following various brushing techniques bytoothbrush mounted pressure sensing unit. The study group consisted of 10 dental hygienist participants. The brushing forces (on buccal area of each first molar) were monitored on 8 different kinds of brushing techniques; Fones, Bass, Rolling, Scrub, Charters, stillman, Modified bass and Modified stillman. In Bass, Charters, Fones and Scrub method, force distributions showed a small gap of maximum and minimum value (Max/Min) while a big difference was noted in Modified bass, Modified stillman, Rolling and Stillman methods. Especially, the biggest difference of Max/Min value was observed in the area of lower left first molar. In conclusion, highly delicate manual skill is needed in showing big error range of force distribution. It means that careful force needs to be focused during toothbrushing instruction when a delicate manual skill was carried out.

        The use of bee venom (Apis mellifera L., BV) occasionally causes side effects such as inflammation and allergic reactions in the recipients. Several case reports also suggested the treatment of BV has some limitations in its clinical uses, due to the occurrence of dermal necrosis and anaphylatic reactions. It is generally understood that bee venom allergy is mainly the result of its allergic component, phospholipase A2 (PLA2). The present study was aimed to generate PLA2-free bee venom (PBV) and evaluate its efficacy as skin care and cosmetic preparation, comparing with original bee venom (BV). Our results showed that both BV and PBV exhibited significant protective effects in UVB-irradiated human keratinocyte (HaCaT) and human dermal fibroblast (HDF) cells and they also induced type I collagen synthesis in UVB-irradiated HDF cells except BV at 3 μg/ml. Furthermore, BV and PBV showed the inhibition of UVB-stimulated matrix metalloproteinase-1 (MMP-1), a major collagen degrading enzyme in skin. However, BV, unlike PBV, exhibited strong cytotoxicities in skin cells (both HaCaT and HDF) at its working concentrations of anti-wrinkle effect. The underlying cell signaling mechanisms of anti-wrinkle effects of BV and PBV were demonstrated by the activation of ERK1/2, and p38. Conclusively, PBV appears to be the bee venom of choice with less cytotoxicity and higher efficacy on UVB-irradiated skin cells in comparison with original bee venom (BV). Therefore, PBV can better be used as a cosmetic ingredient exhibiting excellent anti-wrinkle effect against photoaging than original BV.

        In this study, carbonized fibers were prepared by using acidically cross-linked LDPE fibers. The surface morphologies of the carbonized fibers were observed by SEM. The effects of cross-linking process temperatures were studied using thermal analyses such as DSC and TGA. The melting and heating enthalpy of the fibers decreased as the cross-linking temperature increased. The cross-linked fibers had a carbonization yield of over 50%. From SEM results the highest yield of carbonized LDPE-based fibers was obtained by cross-linking at a sulfate temperature (170oC). As a result, carbonation yield of the carbonized fibers was found to depend on the functions of the cross-linking ratio of the LDPE precursors.

        Carbonate-type organic electrolytes were prepared using propylene carbonate (PC) and dimethyl carbonate (DMC) as a solvent, quaternary ammonium salts, and by adding different contents of 1-ethyl-3-methyl imidazolium tetrafluoroborate (EMImBF4). Cyclic voltammetry and linear sweep voltammetry were performed to analyze conducting behaviors. The surface characterizations were analyzed by scanning electron microscopy method and X-ray photoelectron spectroscopy. From the experimental results, increasing the EMImBF4 content increased the ionic conductivity and reduced bulk resistance and interfacial resistance. In particular, after adding 15 vol% EMImBF4 in 0.2 M SBPBF4 PC/DMC electrolyte, the organic electrolyte showed superior capacitance and interfacial resistance. However, when EMImBF4 content exceeded 15 vol%, the capacitance was saturated and the voltage range decreased.

        Multiple starters consisting of two Bacillus amyloliquefaciens strains (MJ1-4 and EMD17), Pichiafarinosa SY80, and Rhizopus oryzae were used for Doenjang making. Bacillus strains were selected based on their abilities to inhibit toxinogenic fungi and Bacillus cereus, fibrinolytic activity, and their ability to confer good flavor to Cheonggukjang. P. farinosa SY80 and R. oryzae, previously isolated from soy sauce, were selected because they were not inhibited by two bacilli. Doenjang was prepared by inoculation of multiple starters (A1 Doenjang). Control Doenjang was prepared by inoculation of B. subtilis KACC 16750 (Natto strain) and Aspergillus oryzae KCCM 60166 (A2 Doenjang). Another control (A3 Doenjang) was prepared by inoculation of microorganisms present in rice straw. Doenjang samples were fermented for 70 days at 20℃. pH of 3 samples decreased from the initial value of 6.4 to 5.8~6.0 and titratable acidity (TA) increased from 0.6 to 1.1~1.3. The amount of amino-type nitrogen increased during fermentation. There were slight differences in moisture, crude-protein, and crude-fat contents after 70 days. Contamination of fungi was observed only in A3 Doenjang and B. cereus was not detected from all 3 samples. A1 Doenjang showed the highest fibrinolytic activity and A2 Doenjang the second. These results indicate that Doenjang made with carefully selected starters was functionally improved and microbially more safe.

        In the present study, lichen(Parmelia sp.) extract showed insecticidal activity against Aedes albopictus, and the effect of growth inhibiting activity was investigated. Acetone and methanol extracts of the lichen against larvae of Aedes albopictus were showed high insecticidal activities in low lethal concentration. 50% lethal concentration of the acetone extractof the lichen is 0.13% and the 50% lethal concentration of methanol extract of lichen is 0.15% respectively. This experiment that used acetone and methanol extracts of the lichen were observed for 24, 48 and 72 hours. As a result, the higher concentration and the longer exposure time is increased mortality against Aedes albopictus. Pupation time took more time as the higher concentration of acetone extract of lichen. Consequently, the lichen extract is effect in inhibiting the growth of Aedes albopictus larvae. In this experiment indicates that lichen extract has activity against Aedes albopictus and is available as the natural insecticide.

        The spotted-wing drosophila (SWD), Drosophila suzukii (Diptera: Drosophilidae), was originally observed in a few Asian countries, but is now found even in North America and Europe. Genetic information on geographic variation and relationship may broaden our understanding of origin and migration. As a first step, in this study, a portion of mitochondrial COI gene was sequenced to understand genetic relationship and diversity in Korea. Sequencing of 104 individuals provided 57 haplotypes, with the maximum sequence divergence of 1.5%, suggesting high haplotype diversity and moderate sequence divergence. Comparison to GenBankregistered D. suzukii haplotypes (possibly from Spain, Portugal, USA) has shown 100% sequence identity to most of the haplotypes found in this study, but two USA sequences were found to be independent haplotypes, with the sequence divergence ranging from 0.5% ~ 1.4% from our samples in the 553-bp comparison. Phylogenetically, no separable group was found, but, population genetically, the only Chinese population, Sandong, was significantly differentiated (p < 0.05) from all Korean populations, without sharing any haplotype. Among 28 pairwise comparisons of Korean populations only two comparisons showed a significant genetic differentiation, indicating that no population in Korea is completely isolated. Geographically, one haplotype (SWDBA06) was relatively widespread (five among nine localities) and a few haplotypes were found in more than one locality, but most haplotypes were restricted in a locality as a single individual. Overall, high rate of per generation female migration (Nm = 0.75 ~ infinite) and low level of geographic separation (FST=0~0.40) among localities were characteristic. Current data is limited mainly to Korean localities, thus, an expanded study may provide further scrutinized analysis for the fly.

        Insect pollinators of the endanger orchid Cypripedium japonicum were surveyed and identified during two years, as a part of a conservation project of the orchid at Jukyeup-san and Hwaak-san (Mt.), South Korea. In total 40 individuals of 16 species in 4 families were identified. The dominant family was Halictidae, and Lasioglossum exiliceps Vachal visited the most frequently C. japonicum during the surveys. The average visiting frequency was 2.5 individuals per hour and the highest 4.3, from 12:00 – 13:00 in a day. After 15:00 insects did not visit the flowers at all. However, all of the visiting insects were found to not carry a pollinium or pollens of the orchard on their bodies; pollen carryover by any of the visiting insects did not occur at all. The orchid seems to require certain pollinators in particular body thickness due to its unique pollination mechanism. The orchid has two exit route openings, around 1 cm in diametre, where the entrapped insects can exit and an anther is situated just in front of each opening. It was inferred that a pollen carrier should be around 1 cm in body thickness. Therefore, the candidate species as the proper pollen carriers can be Tetralonia nipponensis Perez, Xylocopa appendiculata circumvolans Smith and Bombus consobrinus Dahlbom among the surveyed visitors.

        A partial sequence of the mitochondrial cytochrome oxidase subunit I (COI) gene is widely used as a molecular marker for species identification in animals, also termed a DNA barcode. However, the presence of more than one sequence type in a single individual, also known as heteroplasmy, is one of the shortcomings of barcode identification. In this study, we examined the extent and divergence of COI heteroplasmy, including nuclear-encoded mitochondrial pseudogenes (NUMTs), at the genomic-DNA level from 13 insect species, including four individuals of orthopteran Anapodisma miramae. Furthermore, a long fragment of mitochondrial DNA (~13.5 kb) and cDNA from A. miramae were used as a template for COI PCR to compare the patterns of heteroplasmy between DNA sources and to investigate a possible way to avoid ambiguity in DNA barcoding. When multiple numbers of clones originated from genomic DNA were sequenced, heteroplasmy was prevalent in all species (3~16 heteroplasmic copies), with a varying degree of maximum sequence divergence (<1% in 7 species, <4% in 3 species, <6% in 2 species and 2.15-8.03% in four A. miramae individuals). In five species, NUMTs also were observed when genomic DNA was used as a template. Long fragment DNA also is a source of heteroplasmic amplification, but the divergent haplotypes and NUMTs obtained in the genomic DNA-based PCR were not detected in A. miramae. On the other hand, cDNA was heteroplasmy-free, without NUMTs when multiple numbers of clones were sequenced. Consistently, one dominant haplotype was always obtained from the genomic DNA-origin clones in all species and also from the long fragment- and cDNA-origin clones of A. miramae. Furthermore, the dominant haplotype was identical in sequence, regardless of the DNA source. Thus, one possible solution to avoid the barcoding problem in relationship to heteroplasmy could be the acquisition of multiple numbers of barcoding sequences to determine a dominant haplotype that can be assigned as barcoding sequence for a given species.

        The sister relationship between Trichoptera and Lepidoptera has often been supported in a diverse study, but mitochondrial genomes (mitogenomes) based lepidopteran phylogenetic studies have never utilized Trichoptera as outgroup mainly due to unavailability. Therefore, the effect of alternatives that were previously used (e.g., Diptera, Hymenoptera, and Orthoptera) or Trichoptera as outgroups on the lepidopteran phylogeny remained unknown. In this study, we sequenced three complete mitogenomes of Trichoptera belonging to two suborders and characterized the genomic features of Trichoptera and tested the outgroup effect for lepidopteran phylogeny. The 15,208 ~ 15,285-bp long caddisfly mitogenomes harbor gene content typical of the animal mitogenomes. The orientation and gene order of the three species belonging to the suborder Integripalpia was identical to that of the most common type that has been hypothesized as ancestral for insects, but Cheumatopsyche brevilineata belonging to another suborder Annulipalpia has rearranged QIM, all encoded in forward direction between the A+T-rich region and ND2, instead of the ancestral IQM, with Q inverted. Further, the annulipalpian species had a typical start codon ATG, instead of CGA that are commonly found in other trichopteran species and majority of Lepidoptera. Phylogenetic analysis with different outgroups (Diptera, Hymenoptera, and Orthoptera, Coleoptera, and Trichoptera) and 115 lepidopteran mitogenomes has shown insensitivity either with Trichoptera, Diptera, or Coleoptera, but artificial grouping and lowered nodal support were found with Hymenoptera. The Trichoptera-based consensus topology were: (((((((Bombycoidea + Noctuoidea) + Pyraloidea) + Papilionoidea) + Cossoidea) + Tortricoidea) + Yponomeutoidea) + Hepialoidea).

        The genus Diadegma is a well known parasitoid group and some are known to have symbiotic virus, PDV. A novel IV was discovered from the calyx of D. fenestrale female. This virus was named as D. fenestrale Ichnovirus (DfIV). The encapsidated DfIV genome contains 65 circular DNA segments with an aggregate size of 247,191 bp. Based on BLAST analysis, a total of 120 ORFs were predicted as follows: rep; 48, cys-motif; 11, vinnexin; 10, vankyrin; 9, PRRP; 3 and other unassigned genes (39). These viral genes were expressed in lepidopteran hosts (Phthorimaea operculella and Plutella xylostella) after parasitization which means DfIV genome segments were integrated into lepidopteran hosts. This study was focused on this result. Based on gene expression profile, candidate promoter and integration motifs were selected and then, fused with eGFP as a reporter gene. Modified DfIV genome segment was ligated to a commercial containing f1 ori and Ampr gene to propagate in E. coli. We have named this fusion vector as pIN. The construction methodology of pIN and its application would be further discussed in detail.
