        Background : Jiwhang (Rehmannia glutinosa L.) cultivation area of Chungnam province was 62ha (219 farms, 53.9% of domestic cultivation area, MAFRA. 20142)**). Recently, jiwhang is rapidly spread to 150ha (Geumsan administration statistics, 2015) as taking place of income crop in Chungnam. High quality jiwhang GAP production and improvement of the distribution system including magnus customers, producer association fosterage and conventional market vitalization were the reason of rapid cultivation area increase of Jiwhang in Geumsan. But low emergence rate caused by spring drought should be solved. This study was carried out to obtain basic data for emergence rate improvement in jiwhang. Materials and Methods ○ Experiment variety : Korea jiwhang ○ Treatment : ①Control ② 1 time irrigation at planting time ③1 time irrigation at planting time + transparent vinyl mulching ○ Planting date : April 30, 2015 ○ Planting distance : 30×15cm ○ Experiment place : Ginseng and Medicinal Plant Research Institute (Geumsan-gun, Chungcheongnam-do) Conclusion : There was no significant difference in the aboveground growth among treatments. The seedling emergence rate was higher in 1 time irrigation at planting time + transparent vinyl mulching(1ITVM) than control or 1 time irrigation at planting time(1I). Aerial part weight was significantly havier in 11ITVM than control or 1I. The roots yield was also more in 1ITVM than control or 1I. According to economic analysis, total income in 1ITVM compared to 1I and control was 125.1% and 54.0% higher respectively.

        Tree canopy is a valuable component consisting of urban ecosystem. The purpose of this study was to classify urban tree canopy (UTC) by using high resolution imagery and object-oriented classification (OOC), which was used to classify the different land cover types. With an urban canopy mapping system based on OOC and Decision Tree Classification (DTC), a site mapping was carried out by merging spectral data of high resolution imagery. This methodological approach showed high classification accuracy to distinguish small patches and continuous UTC boundaries on the high resolution imagery. For shadow removal, decision tree classification with various environmental variables such as brightness channel and band combination could effectively work. Our proposed methodology can be successfully used for the assessment and restoration of fragmented urban ecosystem and offer an opportunity to obtain high classification accuracy for the distinction of UTC components in urban landscape areas.

        The transposable element is a DNA sequence that can be changed its position within the genome, sometimes it can create or reverse mutations and altering the cell's genome size. Target region amplification polymorphism (TRAP) is a rapid and efficient PCR-based marker technique, which uses bioinformatics tools and expressed sequence tag (EST) database information to generate polymorphic markers around targeted candidate gene sequences. TE-TRAP is a new marker system which used terminal inverted repeat (TIR) instead of targeted candidate gene sequences. Sorghum holds a good potential plant organism for transposon tagging due to its small genome size, low amount of repetitive DNA and co-linearity with other cereal genomes, which allows the use of information derived from sorghum in other cereal grasses. IS2868 of sorghum accession was treated Gamma irradiation on seed. To define availability and utilization of TE-TRAP, twenty-one accessions were used to evaluate the genetic diversity and underlying relationships. One-thousand thirty-three TE-TRAP markers were amplified by thirty-one primer combination. Altogether, 712 (62.8%) markers were observed polymorphic segregation, whereas 421 (37.2%) showed monomorphic patterns. To estimate genetic differentiation of population by various gamma radiation doses, the analysis of molecular variance (AMOVA) was performed using 4 to 5 different radiation doses population of M1 sorghum individuals. This study and marker system will provide valuable information to assist radiation mutation breeding.

        The transcriptomes of four ginseng accessions such as Cheonryang (Korean ginseng cultivar), Yunpoong (Korean ginseng cultivar), G03080 (breeding line of Korean ginseng), and P. quinquefolius (American ginseng) was characterized. As a result of sequencing, total lengths of the reads in each sample were 156.42 Mb (Cheonryang cultivar), 161.95 Mb (Yunpoong cultivar), 165.07 Mb (G03080 breeding line), and 166.48 Mb (P. quinquefolius). Using a BLAST search against the Phytozome databases with an arbitrary expectation value of 1E-10, over 20,000 unigenes were functionally annotated and classified using DAVID software, and were found in response to external stress in the G03080 breeding line, as well as in the Cheonryang cultivar, which was associated with the ion binding term. Finally, unigenes related to transmembrane transporter activity were observed in Cheonryang and P. quinquefolius, which involves controlling osmotic pressure and turgor pressure within the cell. The expression patterns were analyzed to identify dehydrin family genes that were abundantly detected in the Cheonryang cultivar and the G03080 breeding line. In addition, the Yunpoong cultivar and P. quinquefolius accession had higher expression of heat shock proteins expressed in Ricinus communis. These results will be a valuable resource for understanding the structure and function of the ginseng transcriptomes.

        This study was conducted to determine the optimal dose of gamma-ray for mutation breeding in sorghum (Sorghum bicolor M.). Gamma-rays irradiated to dry seeds with various doses (0 to 1000 Gy) at Korea Atomic Energy Research Institute. Lethal dosage (LD50) was approximately 256 Gy. Significant decreases in growth characteristics (plant height, tiller number and fresh weight) were observed by dose of increased over 300 Gy. Reduction doses (RD50) was approximately 363 Gy in plant height. We also conducted comet assay to observe nucleus DNA damage due to gamma irradiation. In comet assay, a clear difference was identified over 100 Gy treatments. With increasing doses of gamma-ray in the range of 100 to 400 Gy, the rate of head DNA was decreased significantly from 93.52% to 67.57%. The most of the Sorghum cells were severely damaged in the integrity of DNA by gamma-ray. These data provide valuable information when the optimal dose should be chosen for purpose of mutation breeding program of Sorghum.

        Kaposi's sarcoma is a lympho-angioproliferative disease that primarily affects the skin and subcutaneous tissues. Most importantly, Kaposi’s sarcoma occurs in the immunocompromised, especially AIDS patients and organ transplant recipients being treated with immunosuppressants. The occurrence of classic Kaposi’s sarcoma after the diagnosis of any primary solid tumor has rarely been reported. We experienced a 70-year-old man, who had received surgery and chemoradiotherapy for the treatment of primary gallbladder cancer, that developed multiple tender deep purple papules on his trunk, back and feet which were proven to be pathologic symptoms of Kaposi’s sarcoma.

        The sex ratio (F:M) in the same population of oyster, Crassostrea gigas at the commencement of the study (2007) was 1:1.0, but changed to 1:2.8 by the end of the study (2008). The sex reversal rate in two-year-old oysters was 40.2%. Specifically, female to male sex reversal rate was 66.1%, which is higher than the male to female sex reversal rate of 21.1%. The sex reversal pattern of C. gigas appears to go from male female male, and as such is determined to be rhythmical hermaphroditism.

        In this study we established the high throughput screening system of high functional soybean cultivars using PLS modeling from FT-IR spectral data of soybean(Glycine max L) seeds. Crude extract of 20% methanol from soybean seed powders (153 lines) were used for FT-IR spectroscopy. Total fatty acid, carotenoids, flavonoids and phenolic compounds contents from soybean seed powders were analyzed using UV-spectrum and GC analysis respectively. PCA analysis showed that 153 soybean lines formed a single clusters with a few outlier. PC score 1 and 2 represented 39.5, 16.4% of total variation, respectively. And than showed change patten from the middle to outside for PCA plot. We conducted PLS regression analysis between FT-IR spectral data and fatty acids data. Palmitic acid showed the highest regression coefficient (R=0.78). This result implied that the content of palmitic acid could be predicted from FT-IR spectral data from soybean seed powders with relatively high fidelity. PLS modeling of total carotenoids also showed regression coefficient of 0.69. Regression coefficient of total flavnoids and phenolic compounds were 0.44, 0.39, respectively. At present, we are trying to confirm the accuracy of PLS prediction modeling using targeted metabolite analysis (GC-MS, LC-MS) from predicted soybean lines. To increase the accuracy of PLS modeling, we also trying to standardization of spectroscopy and spectral data processing. Furthermore we are going to develop PLS modeling from GC-MS, LC-MS data. The PLS prediction modeling established in this study could be applied for high throughput screening of other leguminous plant.

        The production of sweet (su) and super sweet corns (sh2) has been economically feasible in Korea in recent years. Major factors limiting super sweet corn production are low germination and low seedling vigor. Since seed quality is closely related to seed maturity, the optimum harvest time for the seed production of sweet and super sweet corns was studied and the quality of seeds with varying maturities was investigated in 2001 and 2002 cropping seasons. The parents of the sweet corn seeds were Hybrid Early Sunglow and 'Golden Cross Bantam 70' and those of super sweet corn were Xtrasweet 82 and 'For­tune'. Seeds were harvested at 21, 28, 35, 42, 49, and 56 days after silking (DAS). As the seeds developed, seed weight of sweet corn increased and the seed moisture content decreased faster than that of super sweet corn. Germination rates of sweet corn seeds harvested 21 and 28 DAS at 25~circC and emergence rates in the cold soil test were significantly lower than those of seeds harvested after 42 DAS in both years. Although the germination rates of super sweet corn seeds with varying maturities showed similar patterns as sweet corn seeds at 25~circC , the emergence rate of super sweet corn seeds in cold soil test continuously increased with seed maturity. This suggests that seed quality of super sweet corn should be tested in a cold soil test to estimate field emergence. As the seeds developed, leakage of total sugars and electrolytes from the both sweet and super sweet corn seeds decreased up to 42 or 49 DAS. The α-amylase activities of both sweet and super sweet corn seeds increased with seed maturity from 21 to 35 or 49 DAS depending on genotype and year. The optimum harvest time for the seed production of sweet corn was 42 DAS and 49 DAS for super sweet corn considering emergence rate and plumule dry weight in the cold soil test, leakage of sugars and electrolytes from the seeds, and α-amylase activity.
