        본 연구는 산마늘 개체군 자생지의 소산 식물상을 밝히기 위하여, 2009년 3월부터 2010년 6월까지 총 4차례의 현지조사를 통하여 조사한 결과, 관속식물은 3문 2강 32목 64과 142속 181종 2아종 30변종 1품종으로 총 214분류군 (taxa)으로 파악되었다. 특정식물종 중 I 등급 식물이 15과 16속 15종 2변종 17분류, II 등급 식물이 13과 15속 14종 2변종 16분류군, III 등급 식물이 16과 19속 18종 2변종 20분류군, IV 등급 식물이 20과 23속 19종 4변종 23분류군, V 등급 식물이 2과 2속 2종 2분류군으로 총 78분류군이었다. 희귀식물은 9과 11속 10종 2변종 12분류군이었으며, 특산식물은 17과 19속 18종 3변종의 21분류군으로 조사되었다. 조사지역 내 멸종위기식물은 출현하지 않았고, 귀화식물과 생태계교란식물이 나타나지 않은 것으로 보아 생태계가 안정되고, 생태적 위해성의 우려는 없는 것으로 판단되었다. 또한 산마늘 개체군 자생지에는 개다래, 층층나무, 만병초, 마가목 등 계곡성 및 고산성식물종이 주를 이루는 생육환경을 나타내고 있었다.

        본 연구는 토마토의 토양 수분결핍조건에서의 생장과 생리적인 반응을 근본적으로 조사하기 위하여 수행되었다. 토양에 두가지 수분조건, 심한 수분결핍(-100kPa)과, 대조구인 약한 수분결핍 처리(-25kPa)는 실시간 토양수분함량을 모니터링을 할 수 있는 토양센서와 관수 모듈을 갖춘 micro-irrigation 시스템을 고안, 온실에서 유지되었다. 토양수분함량은 30일동안 변동되었으며, -25kPa로 맞춰진 처리구는 평균 -47kPa, -100kPa 처리구는 평균 -119kPa로 차이를 나타냈다. 이 두 가지 다른 토양수분상태에서 자란 식물체 사이의 생육을 비교해 본 결과 수분결핍상태(-100kPa)에서 자란 식물체가 대조구인 약한 수분결핍(-25kPa) 처리구에 비해 절간수의 차이없이 신장이 유의하게 감소하였으며 건물중의 축적은 더 높게 나타났다. 또한 건물중 당 엽면적의 차이 없이, 엽면적과 엽건중이 수분 결핍이 약한 처리구에 비해 수분결핍이 심한 처리구가 더 높게 나타났다. 이러한 생육상의 차이는 심한 수분스트레스가 엽두께의 변화없이 생체중의 증가와 엽면적 확보를 통해 토마토의 수분스트레스에 적응을 야기시킬 수 있음을 제시했다. 수분결핍에 따른 토마토 생육기간동안, 생리적변화를 조사한 결과, -100kPa 처리구에서 자란 토마토가 대조구인 -25kPa 처리구에 비해 엽의 상대수분함량의 증가와 잎의 삼투압이 낮게 나타났다. 이는 수분스트레스아래서 토마토의 더 나은 수분상태를 유지하기 위한 생리적인 적응을 설명해준다. 아울러 심한 수분스트레스는 대조구에 비해 PSII 활성과 수분활용도를 증가되었으며, 낮은 기공저항도를 나타내었다. 처리간의 광합성의 차이는 없었으며, 토마토 과실의 수와 생육량의 차이는 없었다. 이러한 결과는 토마토 'Picco'가 엽형태의 변형과 삼투압, 수분활용도와 PSII의 활성을 통해 수분결핍상태에서 적응할 수 있게 만들 능력을 보여준다. 본 연구결과에서 나타난 토마토의 수분스트레스 적응 메커니즘은 토마토의 가뭄저항성 스크린에 있어서 고려되어져야 할 것으로 보인다.

        느타리버섯재배에서 심각한 피해를 주고 있는 푸른곰팡이병을 억제하고 버섯균사의 생장을 촉진하여 버섯의 안정생산을 도모하기 위한 탄산칼슘의 처리효과를 실내 및 실외시험을 실시한 결과. PDA배지에서의 CaCO3의 처리는 버섯균의 균사생장을 촉진하고, 병원균은 억제되었으며, 볏짚배지상의 CaCO3의 처리에서는 버섯균은 무처리에 비하여 균사생장이 증가되었으며, 병원균 대체적으로 0.6%의 처리까지는 증가되나 0.8%처리부터는 감소되는 경향을 나타냈다. 컬럼 내의 볏짚배지에서의 Trichoderma longibrachiatum의 접종구에서는 CaCO3의 처리효과가 있었으나 T. virens의 접종구에서는 CaCO3의 효과가 없었다. CaCO3의 처리에 따른 느타리버섯 품종간의 균사생장의 차이는 없었다. CaCO3는 처리에 따라 느타리버섯의 균사생장은 촉진하고 푸른곰팡이병원균과 붉은빵곰팡이균에 대해 약간의 억제 능력은 인정되었다. 포장시험 결과 수량성은 무처리구에 비하여 높으며, 발병율은 낮은 경향이나 절대적인 방제효과는 없었다.

        Background : The major active components of ginseng are ginsenosides and their pharmacological effects include anticanser, anti-stress, anti-fatigue, antioxidant and aging inhibitory effects. These ginsenosides components is higher in leaves than roots. Therefore, consumers are increasingly interested in using ginseng sprouts. Methods and Results : Ginseng sprouts were cultivated during 60 days from June to late July in greenhouse. After 60 days of cultivation, the stem was hardened, and the cultivation was done until 60th because of the tendency that the value of ginseng sprouts was lowered. The content of the three indicator components were analysed by high performance liquid chromatography. Total ginsenoside content increased 1.07 times in leaves and decreased 0.67 times in roots according to cultivated period. The contents of ginsenoside Rg1, Re, Rb1, Rc F3 and F4 of leaves were increased and ginsenoside Rg1, Re, Rb1, Rb2 and Rf of roots were decreased on cultivated 60 days, especially. Total free sugar content increased 1.29 times in leaves and decreased 0.68 times in roots according to cultivated period. The total phenolic acid contents of leaves decreased slightly until 40 days and then increased. The major components of ginsenoside, free sugar and phenolic acid in leaves were Re, sucrose and sinapic acid, respectively. Conclusion : From the above results, ginseng sprouts cultivated for 60 days is more effective ingredient than roots, so it can be said that it is good for consumers to use.

        Background : Recently, the quality of ginseng is degrading while the yield decreases by 15 - 20% due to the high-temperature injury, the physiological disorder and the diseases caused by the climate warming so that development of disaster tolerant varieties strong against high temperatures, salts and diseases are demanded. For such reason, three lineages showing excellent overground and underground growing were selected from the gene resources collected from 1997 to 2000. And the productivity test has been conducted with these lineages having Cheonpung and Yeonpung, the existing varieties as the reference varieties. For the lineages excellent in growing, the variety registration will be performed after the regional adaptation test. Methods and Results : The results of preliminary yield trial on the 3 lines (GS98-3-4, GS00-58 and GS97-69) show that the above-ground and underground growth of the 4-years-old ginseng group selected as superior lines were superior to that of Chunpoong and Yunpoong. eference varieties. Especially, GS98-3-4 shows excellent root weight and root diameter, while GS97-69 shows excellent leaf length and leaf widths. Replicated yield trial on the 3 lines (GS97-62, GS98-1-5, GS00-44) show that the above-ground and underground growth of three lines were superior to that of Chunpoong and Yunpoong. In particular, the underground root weight of GS00-44 was the heaviest. Conclusion : According to the productivity tests on the 3 lines selected for excellent growing, the selected lineages are excellent in both above-ground and underground growing compared to the reference varieties of Cheonpung and Yeonpung. On the 4-years-old ginseng, GS00-44 & GS98-3-4 show excellent underground growing while GS98-1-5 & GS97-62 show excellent overground growing.

        Background : The fruit of Lycii fructus (Gugija) has beeen used as a tonic medicine and a long-term healthy food without side effect in Asia. An increase in the demand for natural healthy food, Gugija has been thought as a source of healthy foods, and then the extracts of dried Gugija have been frequently used as food ingredients. But, its extracts with Gugija alone has a slightly bad taste. Physiological characteristics of Gugija extracts were investigated by adding white ginseng (WG) and red ginseng (RG) to enhance taste. Methods and Results : Gugija extract were prepared by adding 0, 20, 40, 60, 80 and 100% of ginseng (WG and RG) to the weight of Gugija, respectively. Antioxidant activities of Gugija extracts were investigated DPPH and ABTS radical scavenging activity, SOD-like activity, FRAP, activity. As the amount of WG added increased, DPPH, ABTS radical scavenging activity, and FRAP activity of Gugija extract decreased. On the other hand, the physiological activity of Gugija extract increased with increasing amount of RG. The SOD-like activity of Gugija extract added up to 60% of WG and to 40% of RG was continuously lowered, in the addition amount thereafter, the activity increased as the addition amount increased. In components, As the amount of WG and RG added increased, the ginsenosides content of Gugija extract increased, the content of ginsenosides was higher in extract added RG than WG, but betaine and total flavonoid content decreased. Conclusion : From the above results, we may suggest that Gugija extracts by adding RG increased the antioxidant activities, SOD-like activity, and the preference degree.

        Background : Korea ginseng root has been traditionally used as a tonic as it is stated to have the capacity to normalize body functions and strengthen systems that are caused by various stresses. But, white ginseng (WG) has lower antioxidant activity than other medicinal crops. Omija (Schisandra chinensis Baillon) has various physiological functionalities such as anti-cancer, anti-inflammatory, and antioxidant activities, which have the effective components of Omija are lignans (schizandrins and gomisins), and this components were contented mostly in seed part on Omija. Physiological characteristics of WG extracts were investigated by adding Omija to enhance functionality. Methods and Results : WG extract were prepared by adding 0, 20, 40, 60, 80 and 100% of Omija to the weight of WG. Physiological characteristics of WG extracts were investigated DPPH radical scavenging activity, FRAP, and nitrite scavenging activity. As the amount of Omija added increased, DPPH radical scavenging activity of WG extract increased proportionally. IC50 of 10 times water extracts were 0.455, 0.028 and 0.041 ml/ml on WG (100WG), WG added 50% Omija (50OM50WG), and Omija (100OM0WG), respectively. On the other hand, FRAP of extracts decreased slightly with increasing amount of Omija. Nitrite scavenging activity of Omija extract were significantly more active than WG extract, exhibited to 88.89% and 72.65% on WG extract added with 40% Omija (40OM60WG) and non added (100WG), respectively, pH 1.2. and then, to 33.45% and 1.38% with 40OM60WG and 100WG extract, respectively. Conclusion : From the above results, we may suggest that WG extracts by adding Omija increased the DPPH radical scavenging activity, nitrite scavenging activity and the preference degree on taste.

        Background : Currently sprout ginsengs are produced in extensive variations of cultivation times, methods and specifications that are different in each farm in Korea. Also, regardless the effective ingredient content of ginseng, overgrown sprout ginseng are distributed so that the quality standardization of sprout ginseng is urgently required. The sprout ginseng market is only a segment of the Korean ginseng market still. However, If development of processed products utilizing the functionalities is connected, the market may exponentially grow. Also, if the use of sprout ginseng is extended to productions of drinks, cosmetics, facial masks and others in the cosmetics and health functional food industries, the export competitiveness to the global market is expected to increase. Therefore, this study aims the quality standardization of sprout ginseng to secure the market competitiveness in Korea and overseas by examining the property change following the cultivation phases. Methods and Results : For the overground growing in green houses, the sprout ginsengs planted in early April and early June show to be better in growing than the ones planted in early August. For the underground growing, while no significance is found among the cultivation periods, the sprout ginsengs planted in early June are excellent in terms of the root length and the weight of overground portion. Also, according to the examination on the stem properties of 2-year grown sprout ginsengs in each cultivation time, a trend that, as the growing period is longer from 25 days to 35 days, 45 days and 95 days, the hardness, gumminess, chewingness and springiness of stem are higher. And, on the hardness, gumminess, chewingness and springiness of leaf, while no statistically significance is found in the growing periods from 25 days to 45 days, the 95-day sprout ginsengs show high figures referring that the leaves becomes tougher. Conclusion : In case of cultivating sprout ginsengs in a green house, cultivating in April - June is advantageous for the overground and underground growing. And, it is determined that cultivating sprout ginsengs for 35 - 45 days produces leaves and stems with the most excellent chewing textures felt by consumers when chewing.

        This experiment was carried out in order to collect the basic data on the standardization of the manufacturing process of Rehmannia glutinosa Libosch. var. purpurea Makino drying. By the drying methods of R. glutinosa, the content of water, inorganic components, reducing sugar, catalpol and benzo[α]pyrene were investigated. The water content was 15.6~17.2% when R. glutinosa was dried by cold-warm air moisture absorption drying method (CAMAD) at 60℃ during 6 days. Among of the inorganic components of R. glutinosa the K content was the most followed by P, Na, Ca and Mg. The reducing sugar content of R. glutinosa by the hot air drying method (HAD) was much more than that by the CAMAD. The catalpol content of R. glutinosa was not different by the drying temperature when it was dried by the CAMAD. The catalpol content of the large size tuber (about 50.0 g/unit) showed a tendency to increase from 60℃ until 70℃ drying temperature, but that of the small size tuber(about 4.0 g/unit) was decreased as being a trend as the drying temperature high when R. glutinosa was dried by the HAD, But the catalpol content R. glutinosa had a tendency to drop significantly at drying temperature above 80℃. The benzo[α]pyrene content was little detected when R. glutinosa was dried by both the SLD and the CAMAD, and the sampling by the HAD indicated within the scope of 5 μg/kg which was the scope to regulate by Korean food and drug administration. In conclusion, it seemed that an appropriate drying temperature of R. glutinosa by the CAMAD and the HAD was about 60℃ and about 70℃, respectively, when we consider the catalpol content and benzo[α]pyrene detection in the manufacturing process of drying R. glutinosa.

        The objective of this study was to evaluate antioxidant activities of Rehmannia glutinosa (Raw Jihwang) by traditional method. The total phenol content of Rehmannia radix Preparata (the final cycle of Jihwang) was increased to 205%, compared with Rehmannia glutinosa. Antioxidant activities, determined by ferric-reducing antioxidant potential (FRAP), 2,2'-azinobis(3 ethybenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, 1,1-diphenyl-2-picrydrazyl (DPPH) and hydroxyl radical scavenging activities, increased remarkably as the number of steaming-drying cycles increased. Especially, FRAP value increased 285%. Also, IC50 values for DPPH and hydroxyl radical scavenging activities of the final 9th-cycling product, decreased 48.4% and 76%, respectively, compared with those of Rehmannia glutinosa. Our result was suggested that antioxidant activities of Rehmannia radix Preparata improve according to the increasing number of steaming-drying cycles.

        산마늘 잎 에탄올추출물을 Steptococcus thermophilus 를 이용하여 발효시킨 후 항산화활성 및 세포독성을 검정한 결과, 총 폴리페놀 함량은 발효 전에는 2.63 mg/g, 발효 후에는 1.65 mg/g이었으며 발효 후 함량이 감소하였고, 플라보노이드 함량은 발효 전에는 57.77 mg/g, 발효 후에는 62.27 mg/g으로 발효 후에 증가하였다. 전자공여능은 발효 전에는 82.29%, 발효 후에는 81.40%로 vitamin C (97.71%) 보다 낮았으나 우수한 활성을 나타냈으며, 아질산염소거능은 발효전 후 추출물 모두 pH 2.5에서는 BHT 보다는 낮지만 활성이 우수하였고, pH 4.2에서는 발효하지 않았을 때 BHT보다 더 좋은 활성을 나타냈으며. SOD 유사활성능은 15% 정도로 낮은 효과를 나타냈다. NO 생성능은 발효 전에 오히려 더 많은 NO를 생성하였고 발효 후에는 생성이 억제되었다. 산마늘 추출물이 LPO 생성에 미치는 영향은 H2O2 처리군에 비하여 발효 여부와 관련 없이 산마늘 추출물처리에 따라 농도가 증가할수록 MDA 함량이 감소하여 LPO 생성을 억제하는 것으로 나타났으며, 섬유아세포의 생존율은 산마늘 추출물 처리 농도의 증가에 따라 약간 감소하는 경향이나 대조구와 차이가 거의 없었다.

        The extent of growth L. plantarum (LP), L. delbrueckii subsp. bulgaricus (LD), L. fermentum (LF), S. thermophilus (ST), B. longum (BI) and S. cerevisiae (SA) was generally good with the lower concentration of the ginseng extract. Total sapogenin content was slightly different with kinds of a fermentation microorganism and the time of fermentation process, and generally reduced compare to before fermentation. The content of ginsenoside Rb1, Rb2, Rb3, Re and Rf were decreased with the fermentation but ginsenoside Rd was increased by the E, LF and SA fermented extract. The content of compound K increased in the order of not-fermented extrac 〈 enzyme fermented extract 〈 enzyme and microorganism fermented extract, and as the fermented time get longer, the content of compound K was sightly increased. Especially, the content of compound K of the SA fermented extract was the most increased, also it of the BI, LD and LF fermented extract was increased, so these extract were considered a high valuable. Polyphenol content of the BI, LD, LP and ST fermented extract indicated 9.18±0.39~15.68±0.54 mg/10 g which was lower than it of a not-fermented extract (11.92±0.26~28.41±0.39 mg/10 g). Flavonoid content of a ginseng fermented extract indicated 26.93±0.17~156.45±1.29 mg/10 g, it was higher than a not-fermented extract (18.06±0.90 mg/10 g). As the fermented time get longer, the flavonoid content tendency to increase. DPPH radical scavenging activity of a fermented ginseng extract was 24.11±1.41~55.62±0.33%, it was slightly lower compared to a natural antioxidant, vitamin C. But it of the LF and ST fermented extract was similar to a natural antioxidant, vitamin C. It has not a concerned in a fermentation. Nitrite scavenging ability of a 24 hr fermented extract was above 80% at pH 2.5 and 4.2, it was similar to an artificial antioxidant, BHT (84.76±0.13%; pH2.5, 84.98±0.11%; pH 4.2). It has not a concerned in a fermentation. SOD-like activity of a fermented extract was lower than that of a not-fermented extract (19.22±0.51%), but it of the E and LP-fermented extract was a very highly notable value. As the fermented time get longer, the SOD-like activity tendency to increase.

        류마티스성 관절염의 동물 모델인 CIA(collagen induced arthritisd) 생쥐에 CRN을 복강 투여하여 CIA의 발병률 및 관절염지수 등에 대한 효과를 조사한 결과는 다음과 같다. RF(rhumatoid factor) 인자인 IgG와 IgM 생산량, 그리고 염증 사이토카인인 IL-6와 TNF-α의 생산량이 감소하였다. 사이토카인 분석결과, 비장세포에서 Th1과 Th2 세포에서 분비되는 IFN-γ와 IL-4를 측정하여 CRN 투여로 Th1 세포에서 Th2 세포로 면역반응이 전환됨을 시사하는 것은 CRN의 치료효과로 사료된다. 혈청 중에서 RA 인자인 IgG와 IgM, 염증 사이토카인인 TNF-α와 IL-6 및 type II bovine collagen antibody 생산량 이 대조군에 비하여 CRN 투여군이 감소하여 CRN이 관절염의염증 사이토카인을 억제하는 것으로 확인된다.

        본 연구에서는 택칠에서 분리된 CRN(corilagin)이 류마티스성 관절염 치료제로 사용될 수 있는가능성을 조사하기 위하여 류마티스성 관절염의 동물 모델인 CIA(collagen induced arthritis) 생쥐에 CRN을 복강 투여하여 CIA의 발병률 및 관절염 지수 등에 대한 효과를 관찰하여 다음과 같은 결과를 얻었다. 택칠에서 분리한 corilagin을 관절염 유발 생쥐에게 투여한 결과, 관절염의 발병진행이 억제되고 관절염 지수가 감소되는 것을 확인하였다. 세포 조직학적 관찰을 통하여 염증성 세포들이 현저하게 줄어들고, 관절강이 잘 확보되었으며, pannus의 형성이 보이지않고 연골 또한 잘 보호되어져 있음을 확인하였다. 관절염 유발 생쥐의 형광유세포 분석결과 각 조직에 침윤되는 염증세포가 감소하였다.

        인삼, 목과 혼합추출액이 βA로 유도된 AD 병태 모델에 미치는 영향을 관찰한 결과, 다음과 같은 결론을 얻었다. 1. 인삼, 목과 혼합추출액은 AD 병변 뇌조직의 허혈(虛血) 상태를 유의성 있게 개선하였고 허혈(虛血)로 인한 뇌조직 손상을 억제하였다. 2. 인삼, 목과 혼합추출액은 AD 병변 뇌조직의 면역조직화학 염색법으로 Tau 단백질, GFAP 단백질, presenilin 1/presenilin 2 단백질의 발현 억제를 확인하였다. 이상의 결과로 미루어 보아 인삼, 목과 추출액은 AD의 예방과 치료에 사용될 수 있을 것으로 판단되며 정확한 기전에 대한 연구와 AD 치료에 있어서 인삼, 목과 혼합추출액의 임상적 활용에 대한 연구가 향후 지속적으로 이루어져야 할 것으로 사료된다.

        C. destructans는 인삼에서 가장 문제가 되고 있는 뿌리섞음병을 유발하는 매우 중요한 미생물이다. 현재까지 정상적인 인삼포장이나 폐포지에서도 이 병원균의 농도를 조사할 만한 방법이 없어 이를 쉽게 조사함으로서 인삼 예정지 관린시 도움을 줄 수 있는 새로운 방법이 절실이 요구되고 있다. 본 연구에서는 nested PCR이란 분자생물학적 방법을 이용하여 효과적으로 매우 낮은 농도의 C. destructans을 검출할 수 있는 방법을 개발하였다. 2개의 universal ITS primers(ITS5F와 ITS4R)을 사 용 하 여 Cylindrocarpon spp.의 rDNA로부터 ITS영역을 증폭하였다. 이어 C. destructans의 specific primer(Nest 1 과 Nest 2)을 사용하여 최적의 PCR조건으로 재증폭시켜 밴드를 확인하였다. 또한 이런 2번의 과정을 4개의 primer를 동시에 사용함으로서 한번에 확인할 수 있는 방법을 개발하였으며 이에 따른 PCR조건도 확립하였다. 따라서 본 방법에 의해서 인삼포장의 토양에서 채취된 매우 낮은 농도의 wild type C. destructans spore로부터 성공적으로 positive band을 확인함으로써 추후 인삼포장의 선정 및 4년생에서 6년까지(홍삼포) 재배기간등의 예측에 활용 될 것으로 생각된다.
