        Currently, Hanul NPP packages glass fiber classified as particulate waste in plastic packaging bags and stores them in 200 L drums. KORAD’s Waste Acceptance Criteria (WAC) presents that very low-level soil can be immobilized by loading it in a soft bag and then packaging it in a 200 L or 320 L steel drum. As currently accepted method of packaging with soft bag applies to only very low-level soils among the wastes with a risk of dispersion, it is necessary to develop a non-dispersible treatment suitable for the characteristics of other particulate waste in the future. Therefore, in order for Hanul packaging pack to be approved as an alternative method for immobilization of dispersible substances, it is necessary to verify the suitability of the packaging bag. In this paper, whether the glass fiber packaging bag used in Hanul NPP satisfies the characteristic of the soft bag presented in the WAC and the possibility of being considered as a non-dispersible measure for particulate are examined. The soft bag must meet the following requirements: material and structure, shape, drop test, and immersion test. The results of the review are as follows. First, since the glass fiber is already packaged in the drum, only the role of the inner layer, made of polyethylene, having a watertight function may be required. Second, when packaging a drum, the packaging bag is compressed into a shaped frame having an inner size of a 200 L drum, so it is packaged with little empty space in the drum. Third, as a result of a drop test of a packaging pack containing 20 kg of contents from a height of 1.2 m, it was confirmed that there was no leakage of contents. Fourth, the packaging bag was immersed in a 1-m depth water tank for 30-minutes, and the performance corresponding to the IPX7 was satisfied. As a result of reviewing the soft bag characteristic of Hanul glass fiber packaging bag, it is considered that the bag can be used as one of the non-dispersible measures because it meets almost the characteristics required by the WAC. In addition, the acceptance criteria of overseas disposal sites present various secure packaging methods in place of immobilization as a non-dispersible measure for waste containing particulate matter. It is necessary to reflect these overseas cases in the establishment of non-dispersible measures for domestic waste acceptance in the future.

        Mosquitoes are transmit many dangerous disease such as malaria, yellow fever and dengue fever. So far, chemical insecticides such as DEET have been mainly used to control mosquitoes, but there are many side effects. This study used ultrasonic sounds as an alternative to chemical insecticides. We found that Culex pipiens, which are common in Korea, exhibit avoidance behavior in a specific ultrasonic frequency. Through electrophysiological recording, we have inferred that avoidance behavior is caused by different from each other mechanisms depending on the ultrasonic frequency. Using immunohistochemical staining, we analyzed the expression pattern of auditory related genes in the chordotonal organ. Quantitative real time-PCR was used to compare the expression levels of auditory related gene depending on the time of exposure to ultrasonic sounds.

        The silkworm (Bombyx mori), as an industrial insect, possesses a high economic value. Casual discrimination and accumulated genetic information of silkworm varieties are essential ground for the practical utilization and long-term conservation. In this study, nine available microsatellite loci were successfully genotyped from ~50 silkworm strains preserved in Korea. According to genotyping analysis, we obtained 3 ~ 16 alleles per locus, with an average of 7.4, the observed heterozygosity ranging from 0.04 to 0.98, and the polymorphic information content (PIC) ranging from 0.06 to 0.88, revealing that some loci are highly variable. Among 54 strains 13 strains were casually identified by the presence of 17 strain-specific apomorphic alleles. Furthermore, 30 among remaining strains contained strain-specific allele combinations that are also apomorphic to each strain, allowing us to discriminate each of these from other strains by genotyping of multiple loci. These results collectively suggest that the silkworm microsatellite DNA is actually and potentially important molecular marker for the discrimination of the silkworm strains that are preserved as hundreds in Korea, as more loci are genotyped.

        Background : This study was conducted to serve as a basis for the production of high quality organic cultivation Platycodin grandiflorum Ridix roots by analyzing the content of saponins according to organic fertilizer for organic farming. Methods and Results : Platycodin grandiflorum Ridix was treated by with biodegradable film, rice husks, and sawdust, non-mulching (hand weeding). A root of Platycodon grandiflorum were analyzed saponins using HPLC harvested on October. Content of Plantycodin D3 was an average of 68.5 ㎎/100g, which appeared as 111.2 ㎎/100g highest content in the sawdust treated. Deapioplatycodin D showed the highest 48.1 ㎎/100g in the sawdust treated. The content of Platycodin D was shown to 246.2∼373.8 ㎎/100g, the highest values were observed in non-mulching, it showed the lowest value in the rice husks treated. The content of Polygalacin D showed up to 156.0 ㎎/100g in the non-mulching as the biggest difference, the lowest value were 30.1 and 32.2 ㎎/100g in sawdust and rice husks treated. Large differences between fertilizers was not showed, but the overall saponin content of Platycodin grandiflorum Ridix were low in value compared to the control than organic fertilizers. Conclusion : Large differences between mulching materials was showed, thus the overall saponin content of Platycodin grandiflorum Ridix was highest in value compared to non-mulching than sawdust treatment. Mulching Platycodin grandiflorum Ridix with sawdust was an effective method for improving saponin contents.

        Background : This study was performed to know the effect of mulching materials on the growth and root characteristics of Platycodin grandiflorum Ridix. Methods and Results : Platycodin grandiflorum Ridix was treated by with biodegradable film, rice husks, and sawdust, non-mulching (hand weeding). To measure the fresh weight and length, thickness and the number of fine root of the Platycodin grandiflorum Ridix roots harvested in October.         Background : The root of Platycodin grandiflorum has been widely used as a crude drug or a dependent on the cultivation area and cultivation method. Therefore, this study was conducted to know the effect of organic fertilizer on the changes of antioxidents and antioxidant activity of Platycodin grandiflorum Ridix. Methods and Results : As basal fertilizer were treated before transplanting Platycodin grandiflorum Ridix with chemical fertilizer(Super21, 21-17-17, Namhae Chemical), mixed organic fertilizer, bacterial culture, fermented oil cake and manure in each trial of nitrogen based on 3㎏/10a criteria. The total polyphenol content was highest values were 15.5mg/g appear in the mixed organic fertilizers, the lowest were 9.1mg/g appeared in the manure and control. Flavonoid content showed the highest 15.3 mg/g in the mixed organic fertilizer, and investigated at the lowest 5.0 mg/g in the fermented oil cake. ABTS radical scavenging activity was the highest in was 65.7 mgTE/g in the chemical fertilizer, showed the lowest value in the control to 13.0 mgTE/g. Therefore, both flavonoid and polyphenol content showed a high value on organic fertilizers. Conclusion : Therefore, both flavonoid and polyphenol content showed a high value on organic fertilizers. It is determined that to have researches for a variety of antioxidants by the organic fertilizers made in the future.

Content of Plantycodin D3 was an average of 40.6 ㎎ /100g, which appeared as 48.1 ㎎/100g highest content in the bacteria cultures treated. Deapioplatycodin D showed the highest 70.9 ㎎/100g in the control, the rest was investigated 28.1∼54 ㎎/100g. The content of Platycodin D was shown to 327.4∼373.8 ㎎/100g, the highest values were observed in mixed organic fertilizer, it showed the lowest value in the control. The content of Polygalacin D showed up to 336.7 ㎎/100g in the manure as the biggest difference, the lowest value were 54.5 and 74.2 ㎎/100g in bacterial cultures and control. Large differences between fertilizers was not showed, but the overall saponin content of Platycodin grandiflorum Ridix were low in value compared to the control than organic fertilizers. Conclusion : For the increasing the yield that Pharmacological effect, organic fertilizer is effective to increase the number of fine-root high which high contented with saponins.

        Background : Platycodon grandiflorum radix used as a therapeutic agent for lung and respiratory diseases in traditional Korean medicine. The saponin and sugar content in P. grandiflorum root is dependent on the drying temperature and method. In this study, we investigated saponins and antioxidant contents in dried P. grandiflorum roots either not cut or cut to different lengths. Methods and Results : P. grandiflorum roots were either cut to 0.5, 1.0 or 2.0 ㎝ in length or left uncut. They were dried at 45℃ and the effect of antioxidant substances, antioxidant ability, and saponin content were investigated. P. grandiflorum roots rapidly dried out when cut to short lengths. The saponin contents (platycodin D, polygalacin D, and deapioplatycodin D) was highest in the uncut P. grandiflorum roots. When the cut lengths were long, the levels of polyphenolic compound increased. However, 2,2'-azinobis- 3-ethylbenzo-thiazoline-6-sulfonic acid (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were higher in the samples cut to shorter lengths. Conclusions : These results suggested that the saponin and antioxidant content of P. grandiflorum roots could be maximized by controlling the drying rate of the roots via adjustments to the root cutting length.

        Cytoplasmic male sterility (CMS) and fertility restoration have been utilized as valuable tools for F_1-hybrid seed production in many crops despite laborious breeding processes. Molecular markers for the selection of CMS-related genes help reduce the expenses and breeding times. A previously reported genomic region containing the Ppr-B gene, which is responsible for restoration of fertility and corresponds to the Rfo locus, was used to develop gene-based or so-called "functional" markers for allelic selection of the restorer-of-fertility gene (Rfo) in F_1-hybrid breeding of radish (Raphanus sativus L.) Polymorphic sequences among Rfo alleles of diverse breeding lines of radish were examined by sequencing the Ppr-B alleles. However, presence of Ppr-B homolog, designated as Ppr-D, interferes on specific PCR amplification of Ppr-B in certain breeding lines. The organization of Ppr-D, resolved by genome walking, revealed extended homology with Ppr-B even in the promoter region. Interestingly, PCR amplification of Ppr-D was repeatedly unsuccessful in certain breeding lines implying the lack of Ppr-D in these radishes. Ppr-B could only be successfully amplified for analysis through designing primers based on the sequences unique to Ppr-B that exclude interference from Ppr-D gene. Four variants of Rfo alleles were identified from 20 breeding lines. A combination of three molecular markers was developed in order to genotype the Rfo locus based on polymorphisms among four different variants. These markers will be useful in facilitating F_1-hybrid cultivar development in radish.
