In this study, we isolated and identified an aggregation-sex pheromone from Monochamus saltuarius, the major insectvector of the pine wood nematode in Korea. Adult male of M. saltuarius produces 2-undecyloxy-1-ethanol, which is knownto be an aggregation-sex pheromone in other Monochamus species. We performed field experiments to determine the attractivenessof the pheromone and other synergists. More M. saltuarius adult beetles were attracted to traps baited with the pheromonethan to unbaited traps. Ethanol and (-)-α-pinene interacted synergistically with the pheromone. Traps baited with pheromone+(-)-α-pinene+ethanol were more attractive to M. saltuarius adults than traps baited with pheromone, (-)-α-pinene, or ethanol alone.Ipsenol, ipsdienol, and limonene were also identified as synergists of the aggregation-sex pheromone for M. saltuarius adults.

Insect growth regulators (IGRs) are attractive pest control agents due to their high target specificity and relative safety to the environment. Recently, plants have been shown to synthesize IGRs that affect the insect juvenile hormone (JH) as a part of their defense mechanisms. We identified several JH agonists (JHAs) and antagonists (JHANs) from plant essential oil compounds using a yeast two-hybrid system transformed with the Aedes aegypti JH receptor as a reporter system. They showed high mosquitocidal activities with relatively low LC50 values and caused retardation of ovarian development in female mosquitoes. While the JHAs increased the expression of JH-induced gene, the JHANs caused reduction in the expression of the same gene. The compounds identified in this study could provide insights on the plant-insect interactions and may be useful for the development of novel IGR insecticides.

Insect growth regulators (IGRs) are compounds that induce developmental anomalies such as premature molting or supernumerary larval stages. IGRs can be divided into 3 groups based on their mode of action, one of which is known as the juvenile hormone agonists (JHAs). Previously, we have developed a novel screening method which can identify juvenile hormone antagonists (JHANs) using a yeast-two hybrid system incorporating Aedes aegypti juvenile hormone receptor complex proteins (MET and FISC). It has been reported that plants may use IGRs in their defenses against insect herbivores. In this study, 171 plant essential oil samples were screened using the yeast-two hybrid system in order to identify JHAs and/or JHANs. Eight out of 171 samples, 4 JHA and 4 JHAN candidates, were selected and their insecticidal activities were investigated against A. aegypti larvae. They showed high levels of insecticidal activities, with LC50 values ranging from 11.2~21.4 ug/ml. These results indicate that plants do use JHAs and/or JHANs as a part of their defenses against insect herbivores. Also, JHAs and JHANs identified in this study could be exploited as effective biological control agents.

Essential oils or phytochemicals that are naturally synthesized by plants have received much attention as alternative pest control strategy. They are composed of highly volatile compounds which are less toxic to human but highly toxic to pests such as insects, microorganisms and nematodes. Until now, many researchers have evaluated the toxicities of essential oils and phytochemicals in Korea. Among them some of essential oils are formulated as repellents against mosquitoes. However, even such a broad research, the mode of action of essential oils is still unclear. To develop more effective and environmental friendly natural pesticides and to help understand the mode of action of essential oils, we have investigated the structureactivity relationship (SAR) between the chemical components of bio-active essential oils and nematode as well as insect pests. SAR of short-chain aliphatic compounds and ester analogues and pine wood nematode, Brusaphelenchus xylophilus; and salicylate analogues and adzuki been weevil, Callosobruchus chinensis are discussed here.

Oak wilt disease caused by Raffaelea quercus-mongolicae was firstly reported at Sungnam city, Gyeonggi province, Korea, in 2004, and spread to several area of Korean peninsula. Raffaelea quercus-monglicae was transferred by ambrosia beetle, Platypus koryoensis, and caused oak wilt disease. Because oak wilt disease is mediated by ambrosia beetle, Platypus koryoensis, control of this disease concentrates on managing the ambrosia beetle population. Until now, several control methods such as felling and fumigation of dead oak trees with metham sodium, sticky-roll trap for catching emerged or attacking beetles, and oak tree logs for attraction of ambrosia beetle have been mainly used to control oak wilt disease in Korea. Although effective, current control methods have many side effects. Felling of oak tree is very difficult and dangerous to workers. To reduce the side effects of conventional control methods, new and safe types of control method for managing the ambrosia beetle populations are essential. In this study, we investigated the number of caught beetles, effect of height and direction of collection bottles by using newly developed mass capturing device for P. koryoensis to evaluate the effectiveness of this device as practical use in field.

Medicinal plant extracts from 27 plant species in 20 families were tested for their larvicidal activity against house fly, Musca domestica (L.). Responses varied with plant material and concentration. Among plant species tested, Phryma leptostachya var. asiatica showed 100% larvicidal activity against M. domestica at 10 mg/g concentration. Larvicidal activities of Atractylodes japonica, Saussurea lappa, Asiasarum sieboldi, and Gleditsia japonica var. koraiensis were 89.3, 85.3, 93.3 and 96.6% at 10 mg/g concentration, respectively. Extracts of Prunus persica, Curcuma longa and Paeonia moutan produced moderate activity. Larvicidal activity of other plant extracts was less than 50%. Among test plant species, Phryma leptostachya var. asiatica showed the most potent larvicidal activity. The active constituent of P. leptostachya var. asiatica roots was identified as the leptostachyol acetate by spectroscopic analysis. The LC50valuesofleptostachyolacetateagainstM. domestica larvae was 0.039 mg/g. Naturally occurring medicinal plant extracts and Phryma leptostachya var. asiatica root–derived compounds merit further study as potential house fly larval control agents or lead compounds.

Monochamus saltuarius, which mediates Bursaphelenchus xylophilus is one of the important vector of pine wilt disease recently. M. saltuarius showed difference with weight, growth, and oviposition according to species of trees. The most feeding amount and feeding area were determined in P. koraiensis. However, M. saltuarius almost did not feed and longevity was observed very short time only for 10 day in Sciadopitys verticillata. Weight, length and head width were measured the highest growth of M. saltuarius in P. koraiensis. Female of M. saltuarius was determined more growing than male of M. saltuarius in all species of trees. Tree preference for oviposition of M. saltuarius was the highest choice in P. densiflora. However, lots of scar for oviposition have observed but M. saltuarius laid less eggs in P. rigida. This result is related to bark thickness of trees. Ovipositional frequency has been consistently maintained to P. densiflora and P. koraiensis at 20 day intervals. In this study, physiological and ecological characteristic of M. saltuarius to preference of trees were researched could be helpful for managing of M. saltuarius.

We evaluated the larvicidal activity of 20 plant essential oils and components from ajowan (Trachyspermum ammi) and Peru balsam (Myroxylon pereira) oils against Aedes aegypti. Of the 20 plant essential oils, ajowan and Peru balsam oils at 0.1 mg/mL exhibited 100% and 97.5% larval mortality, respectively. At this same concentration, the individual constituents, (+)-camphene, benzoic acid, thymol, carvacrol, benzyl benzonate and benzyl trans-cinnamate caused 100% mortality. The toxicity of blends of constituents identified in 2 active oils indicated that thymol and benzyl benzoate were major contributors to the larvicidal activity of the artificial blend. We also tested the acute toxicity of these 2 active oils and their major constituents against the water flea, Daphnia magna. Peru balsam oil and benzyl trans-cinnamate were the most toxic to D. magna. Two days after the treatment, residues of ajowan and Peru balsalm oils in water were 36.2% and 85.1%, respectively. Less than 50% of benzyl trans-cinnamate and thymol were detected in the water at 2 days after treatment. Our results show that the essential oils of ajowan and Peru balsam and some of their constituents have potential as botanical insecticides against Ae. aegypti mosquito larvae.

Mass mortality of oak trees has been reported in Korea since 2004. It seemed to be occurred by a possible pathogenic fungus Raffaelea quercus-mongolicae and spread by an ambrosia beetle, Platypus koryoensis (Coleoptera: Platypodidae) which was a vector of that fungus. P. koryoensis attacked healthy or stressed living Quercus mongolica. The numbers of damaged and/or died Q. mongolica by P. koryoensis has increased every year since its first report. The damages caused by Platypus spp. were reported world widely. Pinehole borers P. caviceps, P. apicalis and P. gracilis damaged and killed living southern beech (Nothofagus spp.) and/or Kamawi (Weinmannia racemosa) in New Zealand. The relationship between pinhole bores, the fungal pathogen Sporothrix and their host beech is well established. P. subgranosus was responsible to death of myrtle beech (N. cunninghamii) in Tasmania, Australia. In southern Europe, P. cylindrus attacked cork oak (Quercus suber), and Phytophthora cinnamoni was known as a pathogen. P. mutatus was a primary pest to poplars, especially Populus deltoides in South America. Alfaro et al. warned on the threat of P. mutatus to world poplar resources since its introduction to Italy on 2002. P. quercivorus, a vector of pathogenic fungus Raffealea quercivora was responsible for Japanese Oak wilt disease on Q. crispula and Q. serrata. In this study, we will introduce the current status of oak wilt disease in Korea and integrated control method of this disease

The pinewood nematode, Bursaphelenchus xylophilus is a serious forest pathogen in many countries including Japan, China and Korea. To minimize the environmental problems caused by synthetic chemicals broadly utilized in the control of B. xylophilus, we estimated the nematicidal potency of 63 aliphatic compounds against B. xylophilus by measuring their inhibition activity against acetylcholinesterases (ACE, EC 3.1.1.7) of B. xylophilus (BxACEs). In the primary inhibition assay using B. xylophilus crude protein, C6, C9, C10 2E-alkenal C12 alkanoic acid were observed the > 45% BxACE inhibition rate and they were subsequently estimated the inhibition rate against three recombinant BxACEs. Whole compounds showed the high inhibition rate against BxACE-1 and BxACE-2. Interestingly, C6 2E-alkenal and C12 alkanoic acid exhibited the high inhibition rate against BxACE-3 which would be insensitive to ACE inhibitors.
