        수발아 평가와 GI 평가는 정의 상관을 나타내었고, GI 평가는 년차간 변이를 나타내지 않아서 향후 수발아 저항성 계통 평가에 활용이 가능하지만 품종과 연차간 상호작용에서 변이가 나타나기 대문에 향후 지속적인 평가를 통하여 활용여부를 결정해야 할 것으로 생각한다. 수강밀과 우리밀은 수발아율과 GI 모두 낮게 나타나서 수발아 저항성 자원으로 활용될 수 있으며, 국내 밀 품종에는 없지만 수발아 저항성 외국 자원에서 나타나는 Vp-1Bb와 Vp-1Bf는 수발아 저항성 증진을 위해 도입을 고려해야 할 것이다. 염색체 4AL에 존재하며 외국 밀 품종에서 효과적으로 활용되고 있는 SSR 마커인 ZXQ118와 Xhbe03를 이용하여 국내 밀 품종을 평가한 결과, 국내 밀 품종의 변이 폭이 다양하지 않아 이들 표지 인자를 이용하여 수발아 저항성 품종을 구별하기는 어려웠기 때문에 앞으로 국내 밀에서 수발아 저항성 계통 선발에 활용이 가능한 마커 개발에 대한 연구가 필요하다.

        Fusarium head blight (FHB), caused by Fusarium graminearum is a major disease problem on wheat and barley in Korea and around the world. We screened for Type II resistance in the greenhouse using single floret inoculation and for Type I resistance in the field using spray inoculation. Sumai 3 was used the FHB resistant check. Three hundred and seventy lines were evaluated for resistance to spread of symptoms within spike (type II). The 2012 field screening with 300 wheat lines was located in Kimjae-si Joeonbuk Korea. All plots were inoculated twice. The first inoculation was applied at anthesis for wheat. The second inoculation was applied three days after the initial inoculation (dai) for each plot. The inoculum was F. graminearum (GZ3639) prepared at a concentration of 100,000 macroconidai/ml with Tween 20 added as a wetting agent. Mist-irrigation was applied from the first inoculation on May 7 till June 7 to facilitate FHB development. FHB severity was assessed visually 21 days after inoculation on 20 arbitrarily selected spikes per plot. FHB severity was determined as the percentage of symptomatic spikelets from the total of all spikelets observed in these 20 spikes. Based on the field test, we could observe four categories of FHB severity: resistant (R: 0-20%), moderately resistant (MR: 21-40%), moderately susceptible (MS: 41-60%), and susceptible (S: 61-100%). The results showed that forty four lines showed the resistant category on FHB severity between 2.7% and 19.8%. In addition, ten lines showed similar FHB severity compared to Sumai 3 (9.9%).

        A New six-rowed Naked Barley Cultivar “Nurichal” with good coking quality was developed by cross with HB88154-B-25-1 line and Hinchalssalbory by the Winter Cereal and Forage Crop Research Div. NICS, RDA in 2010. HB15305-B-B-31-2 was selected in 2006 and designated as ‘Iksan 95’. It showed good agronomic performance in the regional yield trials (RYT) from 2008 to 2010 and was released with the name of ‘Nurichal’ possessing good water absorption and expansion. The average heading and maturing dates of ‘Nurichal’ were April 24 and May 30 in paddy field, which were two day and one day later than those of the check cultivar ‘Saechalssalbori’ respectively. The new cultivar, ‘Nurichal’, had 82cm of culm length that was 5cm shorter than that of ‘Saechalssalbori’, and 4.0cm of spike length. It showed 774 spikes per m2, 63 grains per spike, 25.0 g of 1,000-grain weight, and 793 g of test weight. ‘Nurichal’ showed a little weaker winter hardiness than those of the check cultivar, ‘Saechalssalbori’ and showed similar lodging resistance with check cultivar. It sowed stronger resistance to barley yellow mosaic virus (BaYMV) compare to check cultivar. Grain shape was small and round. Protein content was 10.3% and β-glucan content was 6.8%. Its average yield of the pearled grain in the regional yield trial was 3.84 MT/ha in paddy field, which were higher 2% than those of the check cultivar. Its water absorption was similar and expansion was higher than ‘Saechalssalbori’. This cultivar would be suitable for the area above the daily minimum temperature of -6℃ in January in Korean peninsula.

        Whole grain wheat bread and bakery products is highly considered as a functional food due to the high amount of vitamins, minerals, and dietary fiber in whole wheat flour. However, as the lower end-use properties of whole wheat products limit consumer selection, it is required to measure the functionality of whole wheat flour. Thus, it is observed the fermentation properties of bread dough from the composite flour of whole grain and white wheat flour. Whole grain cv. Chokyeong developed in the NICS was ground using a hammer mill with 0.5 mm sieve. The composite flour was prepared by substituting wheat flour with whole grain flour at 15, 30, 45% levels. Breads were prepared using the 100 g composite flour according to a basic straight-dough mehtod (AACC 10-10A) with slight modification. A rheofermentometer was used to measure the maximum dough height (Hm) and the maximum gas formation (H’m) during fermentation. Also, total CO2 production was recorded. The Hm decreased from 38.8 to 27.5 (mm) with increasing whole wheat flour from 15 to 45%. Whereas, the H’m was increased from 61.7 to 60.8 (mm) with whole wheat flour increment. Total CO2 production was also decreased with increasing whole wheat flour ranging 1312∼1164 (ml). The fermentation results could be useful to partly predict the effect of substituting whole wheat flour on the end-use quality of pan-breads.

        Improvement of winter hardiness, diseases resistance and good quality have been recently received more attention by covered barley(Hordeum vulgare L.) breeders than ever in Korea. “Hyegang”, a new covered barley cultivar with similar maturing and high yield was developed by the National Institute of Crop Science, RDA in 2011. It was derived from the cross between “Milyang87/NWX-GB-G 2” and “Milyang87/Chalbori” with good quality. The initial cross was done in 2000 and the selected line, “Iksan445” (YB4625-B-B-6-2), showed high yield and good quality characteristics under yield trial test in 2008. It was cultivated three times for three years (2009~2011) in the four locations of regional yield trial (RYT) in korea and was investigated about agronomic and quality characteristics and released as “Hyegang”. The heading date was May 4 in upland and April 26 in paddy field which was 1 and 0 day lateness respectively than that of check cultivar, “Olbori”. The culm length was 77 cm which was 6 cm shorter than that with check cultivar. It showed the spike length of 4.7 cm, 800 spikes / m2, 64 grains / spike and 25.1g for 1,000 grains weight. It showed similar maturing and stronger resistance to barley yellow mosaic virus (BaYMV) compare to check cultivar. It showed higher diastatic power 216DP than that of check cultivar, ‘Olbori’. Average yield of “Hyegang” in the RYT was 4.61 MT/ha in upland and 4.38 MT/ha in paddy field, espectively.

        “Joongmo2008”, a winter wheat (Triticum aestivum L.) cultivar was developed by the National Institute of Crop Science, RDA. It is an awned, semi-dwarf and hard winter wheat. The heading and maturing date of “Joongmo2008” were similar to “Keumkang”. Culm and spike length of “Joongmo2008” were 87 cm and 8.6 cm. It had similar test weight (800g/L) and higher 1,000-grain weight (46.7g) than “Keumkang”. It showed resistance to winter hardiness and moderate to pre-harvest sprouting, which lower rate of pre-harvest sprouting (9.8%) than “Keumkang”. “Joongmo2008” had higher flour yield (73.5%) and higher ash content (0.49%) than “Keumkang”. It showed similar lightness (90.01) of flour color than “Keumkang”. It showed higher protein content (16.4%), gluten content (13.4%), SDS-sedimentation volume(59㎖) and mixing time of mixograph than “Keumkang”. “Joongmo2008” had lower amylose contetn (24.5%) and higher viscosity (153BU) than “Keumkang”. It showed same compositions in HMW-GS compositions(5+10) and Puroindolines composition compared to “Keumkang”. “Joongmo2008” showed lower firmness (0.74N) of and higher bread volume of baked pan-bread to “Keumkang”. Average yield of “Joongmo2008” in the regional adaptation yield trial was 4.54 ton/ha in upland and 3.67 ton/ha in paddy field. “Joongmo2008” would be suitable for the area above -10℃ of daily minimum temperature in January in Korean peninsula.

        Specialty barley extracts were prepared and investigated for its antioxidant activity and biological activity. Hunter L* values of the Iksan 86 extracts had higher than that of the Iksan 87 and Zasoojeongchal extracts. The extraction yields of Iksan 86, Iksan 87, and Zasoojeongchal was 8.08, 6.62, and 7.30%, respectively. The contents of total phenolic compounds of the Iksan 86, Iksan 87, and Zasoojeongchal extracts were 16.24, 15.51, and 13.95 GAE mg/g of sample, respectively. The DPPH radical scavenging activity of Iksan 86, Iksan 87, and Zasoojeongchal extracts were 50.00, 33.27, and 7.56% at a 500 ppm, respectively. The samples showed an inhibition of xanthin oxidase. ACE inhibition effect of specialty barley extracts, Iksan 86, Iksan 87, and Zasoojeongchal, was 39.81, 41.06, and 27.78%, respectively. Tyrosinase inhibition rates (%) of Iksan 86, Iksan 87, and Zasoojeongchal extracts were 26.21, 24.57, and 20.00%, respectively. Results indicated that specialty barley extracts possesses various biological activities including antioxidative capacity, xanthin oxidase inhibition activity, angiotensin converting enzyme inhibition activity, and tyrosinase inhibition activity.

        Pearling, an important primary process on food barley utilization, refers to the gradual removal of grain tissues starting from the outer grain tissues/layers, bran, and germ. The removal of barley bran through pearling yields a bright white kernel that is ideal for various food applications. The removed grain layers are usually sold as feed. However, its use for producing various innovative food products such as high-fiber functional pasta. Here we investigated composition of pearled barley. Barley grains from two hull-less varieties, Saechalssal and Hinchalssal, were pearled to various degrees(10-60%). The composition (protein, β-glucan, fat, ash, total phenol and proanthocyanidin) of pearled barley fractions flour(PF) was determined. Protein, fat, ash, total phenol and proanthocyanidin content was decreased in PF according to increasing pearling degree. Hunter L value and whiteness was increased. β-glucan content also was increased from 3.35, 2.73% to 8.9, 9.3% in Sachalssal and Hinchlssal individually. PF of Sachalssal and Hinchalssal showed different content in all components. Protein content was the highest in 90~80% flour fraction. Total phenol, proanthocyanidin and crude fat content was the highest in outer grain layer, bran(100~90% fraction) than other fractions.

        barley grain and malt is highly related to beer quality, especially hordein is known to be a more significant factor in malting process than albumin. In this study, we proposed selection criteria for high quality malting barley with aid of grain and malt quality parameter scores and storage protein subunit profile informations. Albumin and hordein were extracted and denatured protein subunits were evaluated with malt and grain quality parameters. Total 13 local adaptability test (LAT) lines were planted in four locations (Naju, Iksan, Jeju, and Jinju) and evaluated for malt and beer making qualities. Seventeen germplasms (world collections for high or low seed storage protein content) were also evaluated for biochemical genetic marker. Denatured seed storage protein subunits of albumin and hordein of all tested lines and germplasms were evaluated using 12% 1D SDS-PAGE. Scored data of protein subunit's presence or absence was applied to Agglomerative Hierarchical Clustering (AHC) for statistical analysis. Subunits fractionated within specific molecular weight ranges (97.4-31.0, 66.2-31.0, and 45.0-31.0 kDa) were highly correlated with agricultural characteristics. Several LAT lines showing good performance in agricultural characteristics were clustered in dendrogram constructed by biochemical-genetic assay using XLSTAT. Specific band pattern showed in good performance LAT lines were also observed in some germplasms of world collection having low protein contents which are known to have superior quality in malting. The results would provide selection criteria for high quality malting barley in the malting barley breeding program.

        A new whole crop silage barley cultivar, “oungyang” was developed from the cross between “unong and Milyang55 at the Yeongnam Agricultural Research Institute (YARI) in 2002. An elite line, YB3882-3B-17-1-3-1, was selected in 1999 and designated as “ilyang111” It showed good agronomic performance in the regional adaptation yield trials (RYT) from 2000 to 2002 and was released by the name of “oungyang”with higher yield and better animal feed quality than that of Olbori. The average heading and maturing dates of Youngyang were May 2 and June 6, which were late by 2 & 1 days than those of Olbori, respectively, in Suwon. Youngyang had longer culm length, 83cm but it had lesser spikes per m2 637 than that of Olbori, respectively, in RYT from 2000 to 2002. The rate of leaf sheath/whole length (culm+leaf) and the rate of the grains/ whole crop weight was similar to Olbori. It showed resistance to BaYMV at the regions of Naju, Jinju, Milyang andt Iksan. however, the response of Youngyang to other environmental stresses (tolerance of cold and wet) was similar to Olbori. The dried whole crop yield potential of Youngyang in the RYT was about 11.67 MT/ha in paddy fields which was about 18% higher than that of Olbori. The grain yield potential was 6.32 MT/ha in paddy fields which was about 5% higher than that of Olbori. Youngyang had lower rate of shattering, higher content of protein and NDF, lower ADF and TDN, and better quality of silage than those of Olbori.

        A new covered barley variety, “eungangbori”was developed from the cross between Milyang 55 which have lodging tolerance and easy brittleness and Suweon 260 with good quality by barley breeding team in the Yeongnam Agricultural Research Institute (YARI) in 2002. A promising line, YMB3855-3B-14-1-1-1, was selected in 1999. It was designated as the name of Milyang 110. It was prominent and had good result from regional adaptation yield trials (RAT) for three years from 2000 to 2002 and released as the name of “eungangbori” Geungangbori is resistant to barley yellow mosaic virus and moderately resistant to powdery mildow. The average maturing date was same with Olbori on paddy field in regional adaptation yield trials for 2000-2002. Its culm length is 17 cm shorter than that of Olbori and the spike length is 4.4cm, it’ longer than olbori. The 1,000 grain weight of Geungangbori was 34 g, same as Olbori, but the number of spikes per m2 and test weight ware lower than those of Olbori. The yield potential of Geungangbori was 4.22 MT/ha on paddy in regional adaptation yield trials for 2000-2002. which was 7% higher than that of Olbori. The cooking quality of Geungangbori were similar to Olbori such as water absorption rate and expansion rate. But the crude protein content is lower than Olbori. This variety is suitable for double cropping system with rice in the southern part of the Korean Peninsula.

        Recently, barley is gaining renewed interest for food due to nutritional and functional characteristics. and many breeder developed new barley varieties containing nutritional and functional components. Anthocyanin have been well characterized for its bioactive properties, anti-oxidant, anti-cancer, anti-proliferative and anti-inflammatory properties. Some colored varieties with purple lemma that is a kind of anthocyanin were developed. We have to find out the harvesting time containing maximum anthocyanin content. In this study, we analyzed anthocyanin content at intervals of 4-6days after heading date. We analyzed Delphinidin-3-glucoside(Del3g), Cyanidin-3-glucoside (Cya3g), Pelargonidin-3-glucoside(Pel3g), Peonidin(Peo3g), Malvidin-3-glucoside(Mal3g) using the HPLC with Xterra C18 column and UV Detector. Anthocyanin was extracted with 80% MeOH solution containing 1% HCl for 24hr. Mobil phase composition of HPLC was 45% Acetonitrile solution with 10% Formic acid. Anthocyanin content was increased rapidly from 30day after heading date to 35day. At 35-37day, anthocyanin content was maximum. Thereafter, it was decreased rapidly to 40day.

        The purple and blue grain of barley have anthocyanins that is good sources of antioxidant. Thus we investigated the malt and wort characteristic of purple and blue barley for brewing. During steeping period, anthocyanins content reduced both materials. However the lost content was different from 80% and 20% in purple and blue barley, respectively. Also the anyhocyanin content was different both malt comparing to raw grain from 0.4~4.2% in purple but 50% in blue grain. The wort and beer color(EBC unit) and content of polyphenol and soluble nitrogen from malt of purple and blue were higher than control malt(Hopum). There was no anthocyanins in wort and beer of malt in purple and blue. In these results, it was identified that the anthocyanins from raw grain lost during malting and brewing processing with conventional method.

        Seed storage proteins of different solubility were extracted and denatured subunits of each protein were evaluated with malting barley quality parameters. Its been known that each subunit of seed storage protein encoded by each gene and subunit profiles were highly related to end-use quality in cereals. The purpose of this study is to provide selection criteria for high quality malting barleys with aid of bichemical-genetic information. Total 13 regional test lines and three locations (Naju, Jinju, and Jeju) were incorporated in this study. Albumin and hordein were extracted, denatured, and separated in 12% SDS-PAGE. Presence and absence of subunits of each protein were scored. Dendrogram (using XLSTAT program) was constructed to evaluated the relatedness of lines. The correlation between band profiles and quality test were assessed through Agglomerative Hierarchical Clustering (AHC) for statistics analysis. Hordein subunits can be classified into four groups, A, B, C, and D group. In general, hordein fractions contribute higher than albumine to determine malting quality. Specific molecular weight ranges (97.4-31.0, 66.2-31.0, and 45.0-31.0 kDa) of subunits were highly correlated with malting barley quality parameters. The subunit information would be directly incorporated in providing selection criteria for high quality malting barley in the malting barley breeding program.
