Soybean [Glycine max (L.) Merr.] seed is an important dietary source of protein, oil, carbohydrates, isoflavones, and other nutrients for humans and animals. Raffinose and stachyose are the main antinutritional factors in soybean seed. They are carbohydrates belonging to the raffinose family of oligosaccharides, which are not readily digested in humans and cause flatulence or diarrhea. The genetic reduction of the raffinose and stachyose contents in mature soybean seeds will improve the nutritional value of soybean. The objective of this research was to evaluate agronomic traits with 10 F6 strains selected from breeding populations derived from a cross among seven parents. The contents of raffinose and stachyose in mature seeds were detected by high-performance liquid chromatography. Agronomic traits such as flower color, flowering date, harvesting date, lodging, plant height, seed coat color, hilum color, 100 seed weight, and yield were evaluated. Ten intermediate parents showed low raffinose and stachyose contents. The intermediate parent 883-1 had a small seed size, six intermediate parents (15A1, 15D1, RS-5, RS-33, RS-64, and RS-70) had a medium seed size, and two intermediate parents (14G20 and RS-21) had a large seed size. The intermediate parent RS-21 had a black seed coat and a green cotyledon. Four intermediate parents (883-1, 14G20, RS-5, and RS-21) had elite agronomic traits. The new intermediate parents developed through this study will be used to develop improved soybean cultivars with low contents of raffinose and stachyose.
The consumption of waxy corn is steadily increasing in Korea. Waxy corn is harvested before it reaches full maturity and consumed immediately or follwing cold storage. Glutinous and sweet kernels are preferred due to their high palatability. The kernel properties can change rapidly following harvest, and, therefore, optimal conditions to maintain the kernel quality of corn should be identified. In addition, the timing of harvest of waxy corn ear should be also determined for optimal marketable corn production. From 10 days after silking (DAS) to25 DAS. fresh ear weight and single kernel weight rapidly increased from 78.3 g and 1.13 g, respectively, to 224.9 and 3.61 g, respectively. However, by 30 DAS both fresh and single kernel weight decreased by 10.6% and 6.1%, respectively. Kernel hardness significantly increased up to 25 DAS, and a further slight increase in kernel hardness was observed at 30 DAS. Total sugar content in kernel decreased from 12.5% at 10 DAS to 3.5% at 35 DAS, which was the result of the conversion of sugars to starch during ear development. Crude protein content in kernel did not vary significantly in comparison to kernel hardness. During storage of ear, kernel hardness increased from 726 g cm-2 at harvest to 1,894 g cm-2 following 28 days of storage at a low temperature (0°C). Kernel hardness increased 2.5 fold from 15 DAS to 30 DAS. Soluble protein level in kernel increased until 10 DAS, following which a slight decrease was observed. The soluble protein content decreased from 1.85% at 5 DAS to 1.45% at 35 DAS. Total sugar content in kernel decreased regardless of storage temperature; however, the rate of reduction was lower at 0°C than that observed following storage at 4°C and 10°C. The rate of reduction in kernel moisture content was also lower at 0°C than that observed at 4°C and 10°C.
In monsoon climates, persistent shade is a troublesome weather condition with an impact on the growth and yield of corn (Zea mays L.). We imposed 7, 14, 21, and 28 days of consecutive shade (CS) on Gwangpyeongok and P3394 corn hybrids at the beginning of the kernel filling stages. Shade had little impact on leaf area and dry matter accumulation in the stem and leaves. However, dry matter accumulation in the ear was severely reduced by approximately 28% and 53% after 14 and 28 days of CS, respectively. For the components of grain yield, 7 and 14 days of shade did irreparable damage to the number of filled kernels, the kernel number per ear row, and the percent of filled kernels, but did little damage or reversible damage after removal of the shade to the 100-grain weight and the row number per ear. Shade significantly reduced the relative growth rate (RGR) due to a decrease in the net assimilation rate (NAR). These results suggest that source activity limitation by shade during the kernel filling stages leads to the inhibition of sink activity and size. The yield of biomass, ear, and grain logistically declined as the length of CS increased. Probit analysis revealed that the number of days of CS needed to cause 25% and 50% reductions in grain yield were 3.7 and 23.1, respectively. These results suggest that the plant yield loss induced by shade at the beginning of the kernel filling stages is mainly achieved within the first 7 days of consecutive shade.
Soybean proteins are widely used for human and animal feeds worldwide. The use of soybean protein has been expanded in the food industry due to their excellent nutritional benefits. But, antinutritional and allergenic factors are present in the raw mature soybean. P34 protein, referred as Gly m Bd 30K, has been identified as a predominant immunodominant allergen. The objective of this research is to identify the genetic mode of P34 protein for the improvement of soybean cultivar with a very low level of P34 protein. Two F2 populations were developed from the cross of "Pungsannamulkong" x PI567476 and "Gaechuck2ho" x PI567476 (very low level of P34 protein). Relative amount of P34 protein was observed by Western blot analysis. The observed data for the progeny of "Pungsannamulkong" and PI567476 were 133 seeds with normal content of P34 protein and 35 seeds with very low level of P34 protein (X2=1.157, P=0.20-0.30). For the progeny of "Gaechuck#1" and PI567476, the observed data were 177 seeds with normal content of P34 protein and 73 seeds with very low level of P34 protein (X2=2.353, P=0.10-0.20). From pooled data, observed data were 310 seeds with normal content of P34 protein and 108 seeds with very low level of P34 protein (X2=0.156, P=0.50-0.70). The segregation ratio (3:1) and the Chi-square value obtained from the two populations suggested that P34 protein in mature soybean seed is controlled by a single major gene. Single gene inheritance of P34 protein was confirmed in 32 F2 derived lines in F3 seeds, which were germinated from the low level of P34 protein obtained from the cross of "Pungsannamulkong" and PI567476. These results may provide valuable information to breed for new soybean line with low level of P34 protein and identification of molecular markers linked to P34 locus.
Chinese milk vetch (AstragaIus sinicus L.) is a biennial leguminous plant. Although this species is becoming a commongreen manure croP, Chinese milk vetch seed has a low germination rate due to low water permeability of the seed coat which islimiting fact
Overwintering capacity, closely related to winter hardiness, of Chinese milk vetch planted with different sowing times and sowing practices was investigated to improve the incorporation into cropping system in Korea. The tolerance to low temperature was evaluated with LT50 using leaf disc leaching method. Dry weight of CMV was reduced remarkably with delayed planting from Sep. 5 to Oct. 20. The differences in tolerance to freezing temperature were not conspicuous among CMV genotypes, however, the differences between genotype (collections at different regions) were due to the plant architecture, mainly to the leaf angle. The crouching genotype collected at central region of Korean peninsula, which showed excellent freezing tolerant, has planophile leaves. The feature of internal constituents of CMV genotypes did not show any noticeable differences with respect to the freezing tolerance which evaluated by leaf disc leaching experiment. To overcome the poor overwintering capacity, tolerant genotype should be developed by selection with considering the plant architecture. The reduction of CMV growth during overwintering period was ameliorated with furrow-sowing under late-sown condition, therefore, when the CMV is inevitably sown late after recommended time, the seeds should be sown on furrow to overcome the cold stress.
Changes in the level of metabolites in leaves and pods were examined with respect to the seed chemical composition in black soybean. There was no further increase in pod length after 42 days after flowering (DAF). Pod weight, however, persistently increase until 73 DAF, thereafter the weight was slightly lowered. The seed storage protein, however, increased drastically as the increasing rate of pod weight was lessened at 61 DAF. The accumulation of seed storage proteins was occurred conspicuously as the increasing rate of pod weight was slowed down. The chlorophyll content both in leaves and pods was drastically decreased after 50 DAF. The beginning of drastic reduction in chlorophyll content was occurred concomitantly with the reduction of soluble protein content in leaves. The sugar content in leaves showed similar tendency with chlorophyll and soluble protein content. The starch level in leaves, however, showed different changing pattern during seed development. The starch content in leaves was increased persistently until 66 DAF, thereafter the content was decreased drastically to about 55~% of maximal value at 66 DAF. Total phenolics content in leaves and the anthocyanins content in seeds were stable without noticeable increase until 66 DAF. The contents were increased dramatically after 66 DAF showing the synchronized pattern with the decrease in starch level in leaves. The levels of the selected metabolites in leaf and seed suggested that the accumulation of chemical components of black soybean seed is launched actively at 66 DAF. The profile of storage proteins was nearly completed at 61 DAF because there was no large difference in densitometric intensity among protein subunits after 61 DAF. In soybean, chemical maturation of seed begins around 61 to 66 DAF at which most metabolites in vegetative parts are decreased and remobilized into maturing seeds.
Germination and emergence habits of Chinese milk vetch (CMV) were examined to obtain the basic information for the effect of environmental conditions and cultural practices on the seedling density. Seed germination tests with different water potentials, temperatures, and soil and water depths showed the environmental effects on the characters related to seed germination in CMV. Imbibition under different temperatures reflects that initial velocity was rapid at higher temperature, however, the times to full imbibition were not different between 15 and 25~circ . The optimal germination temperature for CMV germination was ranged from 15 to 20 and the germination was highly affected by water potential of media at relatively high temperature above 20~circ . When the seeds were sown in flooded condition the germination was not proportionally affected by water depth. In addition, there was no correlation between water depth and oxygen concentration. The germination of seeds flooded by 2cm water depth were poorly germinated compared to other depths. Results indicated that the germination of submerged seeds was more highly influenced by flooding depth than the temperature, it was also affected more strongly at 10 than 20~circ . Emergence of CMV depending on the thickness of covered soil was poor when the soil layer was greater than 5cm. In the experiment with seeds collected between 22 days after flowering (OAF) and 52 DAF, the highest germination ability of CMV seeds was observed at 39 DAF and germinability was decreased subsequently as seeds became mature. The lower germinability may be due to the enhanced seed dormancy.
Seeds of burcucumber were treated with accelerated aging, cold-stratification, and light quality illuminated during desiccation to enhance their germination and seedling emergence. The germination was increased by aging and cold-stratification although the latter treatment showed greater effectiveness than the former one. In the combined treatment of aging 6 days at 45~circC and cold-stratification, the germination was promoted under longer period of cold-stratification to reach nearly 100% in 3 week cold-stratification on the ninth day from sowing. In the sequentially combined treatment of aging, cold-stratification, and light quality during 24 hour desiccation at 35~circC , no-stratified seeds showed the highest rate in red light treatment but the lowest in far-red light. This implies that the phytochrome action run during the desiccation of imbibed seeds. The red light exposure during drying for the cold-stratified seeds after aging accelerated the germination even more than the dark treatment and germinated 100% on the next day of sowing. It is concluded that the sequential treatment of aging, cold-stratification, and red light illumination during desiccation can highly promote percentage and speed of burcucumber seed germination.
Introgression from genetically modified plants (GMPs) may be dependent on the genetic similarity to wild relative plants. In Korea, many wild plant species are botanically related to the cultivated plants that have a potential to be genetically transformed. The controversy for hazards of GMPs is continuing because the studies on gene flow or introgression are little. Based on the systematic criteria, we have surveyed Korean wild plant species that showed the similarity to cultivating crops. The consideration for feasibility of genetic pollution (introgression of transgene) is necessary for the successful accomplishment in the practical use of GMPs. Although the detrimental effects of GMPs on wild relatives have not been clearly verified, Korean wild plant species related to crop plant (potential GMP) have to be investigated with respect to the introgression. Korean flora consists of ca. 5,500 species. Among them, 1,448 species are classified as weed species (966 native, 325 naturalized, and 167 escaped ones), which is vulnerable to GMPs in term of introgression. We suggested the principal Korean wild plants related to major crops that might be affected by GMPs via introgression. The investigated species herein are selected based on the morphological and phenological relationship. It is necessary to verify the genetic relationship between cultivated plants and wild relatives sing more precise molecular techniques, which provide the information of likelihood for the introgression of transgene
Soybean [Glycine max (L.) Merrill] crops, grown in a rice soybean rotation, can suffer when grown in soil with excessive moisture. The objective of this work were to determine the reduction in growth and yield, responses of vegetative and reproductive growth of soybean to excessive soil moisture achieved by prolonged irrigation. Responses of different cultivars were determined at growth stages from V6 to R8 to clarify the sensitive growth stages or characteristics to excessive soil moisture. Cultivar differences in response to excessive soil moisture condition were conspicuous in seed dry weight and harvest index (HI) but not in the response of seed number or pod number per plant. The timing of irrigation causing the condition of excessive soil moisture influenced the vegetative or reproductive traits. Soybean plants were more affected by irrigation commencing at the pre-flowering than at the post-flowering stage. Post-flowering irrigation did not reduce growth of vegetative organs significantly; in fact the growth of stems and leaves was facilitated by the prolonged irrigation commencing at flowering. Differences between cultivar response to prolonged irrigation were assumed to relate to the reduced amount of assimilates translocated to the reproductive organ
