        장내 미생물 군집은 소화 과정, 면역 시스템, 질병 발생 등 숙주의 다양한 면에 광범위한 영향을 주는 것으로 알려져 있으며, 주요 장내 미생물 종은 숙주의 생리 기능에 핵심적인 역할을 수행한다고 발표된 바 있다. 곤충의 장내 미생물 군집에 관한 연구가 최근 활발히 이루어지고 있으며, 이들 연구는 주로 장내 미생물 군집과 기생충, 병원체 간의 상호작용, 종간의 신호 전달 네트워크, 먹이의 소화 과정 등을 중심으로 이루어지고 있다. 이러한 연구들은 대부분 Illumina MiSeq을 활용하여 16S rRNA 유전자의 V1부터 V9 영역 중 선택된 특정 부분을 대상으로 짧은 서열 정보를 대상으로 진행되었다. 그러나, 최근에는 PacBio HiFi 기술이 상용화되면서 16S rRNA의 전장 분석이 가능할 수 있게 되었다. 이번 연구는 장수말벌(Vespa mandarinia)의 해부를 통해 gut과 carcass 부분을 분리한 뒤, 각 샘플을 Illumina MiSeq과 PacBio HiFi 기술을 활용하여 미생물 군집 간의 차이점을 확인하기 위하여 수행되었다.

        Lyophyllum decastes has been used for culinary purpose. The present study was conducted to evaluate the antioxidant and anti-inflammatory effects from methanol, acetone, and hot water extracts of L. decastes fruiting bodies. The acetone and methanol extracts showed the higher 1,1-diphenyl-2-picryl-hydrazy radical scavenging activities than that of the hot water extract at 0.5–2.0 mg/mL and was comparable to the BHT, the positive control. The ferrous ion chelating effects of the mushroom extracts at 0.5–2.0 mg/mL were significantly higher than that of BHT. The reducing power of acetone extract (2.12) was significantly lower than that of BHT (2.73) at 2.0 mg/ mL. The mushroom extracts also showed inhibitory effects on production of nitric oxide (NO), and expression of inducible nitric oxide synthase (iNOS) in lipopolysaccharide-induced murine macrophage cells in a concentration dependent manner. In vivo anti-inflammatory experiment on carrageenan-induced hind-paw edema of rat model, the acetone extract of the mushroom significantly suppressed the carrageenan-induced rat hind paw edema of rats in a dose dependently. The results suggest that the fruiting bodies of Lyophyllum decastes are a good natural resource of antioxidant and anti-inflammation.

        White rot fungus Cerrena unicolor IUM 5400 produced ethanol from diverse sugars, including glucose, mannose, galactose, and cellobiose at 0.38, 0.28, 0.08, and 0.27 g of ethanol per g of sugar consumed, respectively. The fungus produced relatively high amounts of ethanol from xylose (0.28 g of ethanol per g of sugar consumed); however, the ethanol conversion rate of arabinose was relatively low (at 0.08 g of ethanol per g sugar consumed). When cultured in a basal medium containing 20 g/L rice straw or corn stalks, C. unicolor IUM 5400 produced 0.18 g and 0.18 g of ethanol per g of rice straw and corn stalks, respectively. The results suggest that C. unicolor IUM 5400 is a white rot fungus that can effectively hydrolyze cellulose or hemicellulose to sugars and simultaneously convert them to ethanol.

        Pleurotus eryngii, a white rot fungus, produces two extracellular lignin-degrading enzymes, laccase and manganese peroxidase (MnP). Owing to these enzymes, P. eryngii efficiently degrades synthetic chemicals such as azo, phthalocyanine, and triphenyl methane dyes. In this study, we investigated the degradation processes of four aromatic dyes, congo red (CR), methylene blue (MB), crystal violet (CV), and malachite green (MG), by P. eryngii under solid and liquid culture conditions. CR and MG were the most quickly degraded under solid and liquid culture conditions, respectively. However, compared to CR, CV, and MG, MB was not degraded well under both culture conditions. The activities of ligninolytic enzymes (laccase and MnP) were also investigated. Laccase was identified to be the major enzyme for dye degradation. A positive relationship between decolorization and enzyme activity was observed for CR, MB, and CV degradation. In contrast, decolorization of MG ensued after high enzyme activity. These results indicate that the degradation process differs between MG and the other aromatic dyes. Therefore, P. eryngii could be a potential tool for the bioremediation of synthetic aromatic dye effluent.

        A novel brown rot fungus Phaeolus schweinitzii IUM 5048 was firstly used for ethanol production. It was found that this fungus produced ethanol with various sugars, such as glucose, mannose, galactose and cellobiose at 0.28, 0.22, 0.06, and 0.22 g of ethanol per g of sugar consumed, respectively. This fungus showed relatively good ethanol production from xylose at 0.23 g of ethanol per g of sugar consumed. However, the ethanol conversion rate of arabinose was relatively low (at 0.08 g of ethanol per g sugar). P. schweinitzii was capable of producing ethanol directly from rice straw and corn stalks at 0.11 g and 0.13 g of ethanol per g of substrates, respectively, when the fungus was cultured in a basal medium supplemented with 20 g/L rice straw or corn stalks. These results suggest that P. schweinitzii can hydrolyze cellulose or hemicellulose to fermentable sugars and convert them to ethanol simultaneously under oxygen limited condition

        한약재로 사용되는 더덕과 도라지에 표고의 유용성분을 함유한 소재를 개발하고자 더덕표고균사발효물(FCLM)과 더덕도라지균사발효물(FPLM)을 제조하였다. 두 가지 발효물의 일반성분, 유리당, 유기산, β-glucan, ergothioneine, ergosterol, vitamin D2 함량 및 3T3-L1 지방전구세포의 분화에 미치는 영향을 연구하였다. 일반성분 분석결과, FCLM에서 조섬유와 조지방함량이 FPLM에 비하여 높게 나타났으며, FPLM은 조단백질과 가용성무질소물의 함량 이 FCLM에 비하여 높게 나타났다. 유리당은 FCLM과 FPLM 모두 arabinose, glucose, sucrose 3종이 검출되었으며, 총 유리당 함량은 FPLM에서 높게 나타났다. 유기 산 함량은 발효 전 더덕과 도라지에 비하여 발효 후 FCLM과 FPLM에서 낮게 나타났다. β-Glucan 함량은 FCLM과 FPLM 두 발효물에서 대조구로 사용한 표고보다 높은 함량을 보였다. 항산화 물질로 알려진 ergothioneine의 함량은 FCLM이 FPLM보다 높게 나타났다. Ergosterol 함량은 대조구로 사용한 표고에서 가장 높게 나타났으며, 발효물들은 비슷한 함량을 나타내었다. Vitamin D2는 발효물들에서만 검출되었으며, FPLM (0.58±0.01 mg%)가 FCLM (0.47±0.01 mg%)보다 높은 함량을 보였다. 더덕과 도라지 표고균사발효물인 FCLM과 FPLM는 3T3-L1 지방전구세포에 대하여 발효 전 더덕과 도라지에 비하여 높은 수준의 세포 생존율을 나타내었다. 또한 FCLM과 FPLM은 3T3-L1 지방전구세포 분화를 발효 전 더덕과 도라지에 비하여 높은 수준으로 억제하여, 비만 억제 효과 가 있는 것으로 기대된다.

        Anti-melanogenesis and skin anti-wrinkle effects of methanol (ME) and hot water (HE) extracts from the fruiting bodies of Ganoderma applanatum were investigated in this study. The total phenolic contents of the ME and HE of the mushroom were 11.68 and 3.15 μg GAEs/mg, respectively, whereas the total flavonoid contents of the ME and HE were 21.82 and 2.69 μg QEs/mg, respectively. The survival rate of B16-F10 murine melanoma cells treated with 750 μg ME and HE were 83.46% and 85.54%, respectively, thereby suggesting that mushroom extracts were slightly cytotoxic at the tested concentration. The in vitro tyrosinase inhibition by ME (83.15%) and HE (83.44%) was significantly lower than that of kojic acid (99.61%), the positive control, at 2.0 mg/mL. Although the inhibition of cellular melanin synthesis in B16-F10 melanoma cells by 2.0 mg/mL of ME (50.24%) and HE (51.24%) was lower than that of arbutin (64.84%), the inhibition by both ME and HE was higher than 50%. Collagenase inhibition by HE was comparable to 2.0 mg/mL epigallocatechin (EGCG), the positive control; however, elastase inhibition by ME and HE was lower than that of EGCG at the concentration tested. The results showed that the fruiting bodies of G. applanatum had good anti-tyrosinase, good anti-collagenase, and moderate anti-elastase activities, which might be useful for developing novel skin-whitening and anti-wrinkle agents.

        본 연구에서는 장수진흙버섯 자실체로부터 메탄올과 열수를 이용해 추출한 물질의 항당뇨, 항콜린에스테라아제, 항염증 효과를 탐색하였다. 항당뇨 실험에서 α-amylase 효소에 대한 메탄올과 열수 추출물의 저해 효과는 1.0~2.0 mg/ml의 농도에서 각각 50.67~61.33%와 52.67~ 65.00%를 보여 양성대조군인 acarbose의 83.67~96.33% 에 비해 크게 낮았으나 α-glucosidase에 대한 메탄올과 열수 추출물의 1.0~2.0 mg/ml의 농도에서의 저해 효과는 각각 74.33~89.67%와 75.67~91.00%를 보여 53.67%와 84.67%를 보인 양성대조군 acarbose에 비해 유의하게 높아 탄수화물 분해효소인 α-glucosidase에 대한 저해 효과가 특이적으로 높았다. 메탄올과 열수 추출물의 아세틸콜린 에스테라아제에 대한 저해 효과는 1.0 mg/ml의 농도에서 각각 96.05%와 94.58%를 보여 양성대조군인 galanthamine (97.80%)과 유사하였으나 butyrylcholinesterase에 대한 저해 효과는 1.0 mg/ml의 농도에서 각각 91.05%와 82.27%를 보여 galanthamine의 저해 효과 81.12%에 비해 유의하게 높았다. 염증 저해 효과 실험에서 RAW 264.7 대식세포 가 배양되고 있는 배지에 메탄올과 열수 추출물을 각각 전처리 후 염증 매개 물질인 LPS를 1 μg/ml 처리하여 24 시간 배양 후 NO 생성의 저해 효과를 조사한 결과 각각의 추출물 농도가 증가함에 따라 생성된 NO의 양도 점차 유의하게 감소하는 경향을 나타내었다. Carrageenan의 주사에 의해 흰쥐의 뒷발에 유도된 부종 저해실험에서는 투여한 메탄올 추출물의 농도가 증가함에 따라 뒷발에 유발 된 부종의 용적도 농도 의존적으로 점차 감소하는 추세를 나타냈다. 따라서 장수진흙버섯의 자실체에 함유된 물질은 항당뇨, 항콜린에스테라아제 및 항염증 효과를 지니고 있어서 천연 건강식품으로의 개발 가능성이 높은 것으로 사료 된다.

        본 연구에서는 말뚝버섯의 자실체 메탄올 추출물의 항산화, cholinesterase 저해 및 항염증 효과를 탐색하였다. DPPH 라디칼 소거능, hydroxyl radical 소거능, 철 이온 제거능 및 환원력 등의 항산화 효과를 측정한 결과 DPPH 라디칼 소거능, hydroxyl radical 소거능 및 환원력은 양성대조군으로 사용한 BHT에 비해 낮았으나 실험에 시용한 2.0 mg/ml의 농도에서 50% 이상의 저해효과를 나타내었고 철 이온 제거능은 BHT에 비해 높게 나타나서 다른 종류의 식의약용 버섯에 비해 항산화 효과가 우수하였다. 치매환자의 기억력 감퇴와 관련된 acetylcholinesterase와 butyrylcholinesterase의 저해실험에서 말뚝버섯 자실체의 메탄올 추출물은 실험에 사용한 전 농도 범위에서 양성대 조군인 galanthamine에 비해 유의하게 낮았지만 1.0 mg/ ml의 농도에서 50% 이상의 저해 효과를 나타냈다. Invitro 항염증 실험에서 RAW 264.7 대식세포에 서로 다른 농도의 메탄올 추출물을 처리한 후 염증 유발물질인 LPS 를 처리하여 RAW 264.7 세포가 생성한 NO의 양을 측정한 결과 추출물을 투여한 실험군의 NO 농도가 LPS만 단 독으로 처리한 양성대조군에 비해 유의하게 낮았고 처리한 메탄올 추출물의 농도가 증가함에 따라 생성된 NO의 양은 유의하게 감소하는 경향을 나타났다. 또한 in vivo 항 염증 실험에서 먼저 각기 다른 농도의 메탄올 추출물을 생쥐의 뒷발에 주사한 후 추가로 기염제인 carrageenan을 주사하여 흰쥐 뒷발에 유도된 부종 (edema)이 추출물에 의해 저해되는 정도와 염증 치료제로 처방되는 indomethacin을 양성대조군으로 하여 실험을 진행하였다. 실험 결과 흰쥐에 주사한 말뚝버섯 자실체의 메탄올 추출물의 농도가 증가함에 따라 흰쥐 뒷발에 유도된 부종의 용적도 농도 의 존적으로 유의하게 감소하는 것이 관찰되어 말뚝버섯자 실체에는 염증을 저해하는 성분이 함유되어 있는 것으로 사료되었다. 따라서 말뚝버섯 자실체의 메탄올 추출물에는 항산화, acetylcholinesterase과 butyrylcholinesterase의 저해 및 항염증 효과를 나타내는 유용 성분이 함유되어 있어 앞으로 의약용 기초 소재로서의 이용 연구가 필요하다고 사료된다.

        Oyster mushrooms are popular and physiologically beneficial to human health. This study was conducted to investigate the effect of dietary supplementation of oyster mushrooms such as Pleurotus ostreatus, P. pulmonarius, and P. citrinopileatus on high cholesterol fed rats. Five-week old female Sprague-Dawley albino rats were divided into three groups of 8 rats each. Feeding a diet containing a 5% fruiting body powder of Pleurotus species to high cholesterol fed rats reduced plasma total cholesterol (TG), triglyceride (TG), low-density lipoprotein (LDL). Feeding each oyster mushroom also significantly reduced body weight in high cholesterol fed rats. However, it had no adverse effects on plasma albumin, creatinine, blood urea nitrogen, uric acid, glucose, total protein or enzyme profiles such as glutamate oxalate transaminase (GOT), glutamate pyruvate transaminase (GPT), and alkaline phosphatase (ALP) which are related to kidney function. Feeding each mushroom increased total lipid and cholesterol excretion in feces. A histological study of hepatic cells by conventional red O staining showed normal findings in mushroom-fed hyperlipidemic rats. These results suggested that diet supplementation of 5% fruiting body power of Pleurotus species provided health benefits by acting on the atherogenic lipid profile in high cholesterol fed rats.

        본 연구는 한국에서 분리한 송곳니구름버섯 (Irpex consors) 균사체의 에탄올 생산 가능성을 탐색하기 위해 수행되었다. 송곳니구름버섯의 균사체를 당이 함유된 배지에 접종하여 에탄올 생산량을 측정하였다. 포도당, 만노오스, 자일로스 등 단당류와 셀로비오스 등의 이당류가 각각 1 g 함유된 발효배지에 송곳니구름버섯의 균사체를 접종하고 배양한 결과 각각의 이들 당류에서 0.23, 0.19. 0.21, 0.17 g의 에탄올이 생산되었다. 또한 볏짚을 열수, 3% 가성소다, 3% 황산용액으로 각각 전 처리한 후 송곳니구름버섯의 균사체를 접종하고 배양한 결과 1 g의 볏짚은 각각 0.12, 0.15, 0.19 g의 에탄올로 전환되었다. 본 실험을 통해 송곳니구름버섯의 균사체는 여러 종류의 환원당을 이용해 에탄올을 생산할 수 있는 것은 물론 열수, 가성소다 및 황산으로 전 처리한 볏짚을 에탄올로 전환하는 것도 가능한 것으로 나타났다. 따라서 송곳니구름버섯 균사체의 에탄올 생산 수율을 본 실험의 결과 보다 높일 수 있다면 당류뿐 만아니라 볏짚을 비롯한 리그닌셀룰로오스의 바이오매스를 이용해 바이오에탄올을 효율적으로 생산해 우리나라 에너지 수요를 자급하는데 큰 기여를 할 수 있을 것으로 사료된다.

        본 연구에서는 메탄올과 열수를 이용해 갈색먹물버섯의 자실체로부터 추출한 물질의 항산화, 항당뇨, 항콜린에스 테라아제, 항티로시나제와 항염증 효과를 탐색하 였다. 고 속액체크로마토그래피를 이용해 추출물의 페놀성 화합물 을 분석한 결과 procatechuic acid, chlorogenic acid, (-)- epicatechin, naringin 등 총 4종류의 페놀성 화합물이 확 인되었다. 항산화 효과 실험에서 DPPH radical 소거능은 양성대조군으로 사용한 BHT에 비해 낮았지만 효과가 비 교적 우수하였고, 철 이온을 제거하는 항산화 효과는 메 탄올과 열수 추출물이 양성대조군인 BHT에 비해 30% 높게 나타났으나 환원력은 양성대조군에 비해 43% 정도 낮은 것으로 나타났다. 항당뇨 실험에서 α-amylase와 α- glucosidase에 대한 메탄올과 열수추출물의 저해효과는 2.0 mg/ml의 농도에서 각각 62.26%와 67.59%를 보여 양 성대조군인 acarbose의 81.81%에 비해 낮았다. 아세틸콜 린에스테라아제에 대한 메탄올과 열수추출물의 저해효과 는 1.0 mg/ml의 농도에서 각각 94.64%와 74.19%를 보여 양성대조군인 galanthamine의 97.80%에 비해 낮았다. 티 로시나아제에 대한 메탄올과 열수추출물의 저해효과는 2.0 mg/ml의 농도에서 각각 91.33%와 91.99%를 나타내 양성대조군인 kojic acid의 99.61%와 매우 유사한 효과를 얻었다. 염증저해 효과 실험에서는 RAW 264.7 대식세포 가 배양되고 있는 배지에 갈색먹물버섯 자실체의 메탄올 과 열수추출물을 각각 전 처리 한 후 염증매개 물질인 LPS를 투여하여 메탄올과 열수 추출물의 NO 생성 저해 효과를 조사한 결과 추출물의 농도가 증가함에 따라 생성 된 NO의 양이 감소하는 경향을 나타내었다. 따라서 갈색 먹물버섯의 자실체에는 항산화, 항당뇨, 항아세틸콜린에 스터라제 항티로시나아제 및 항염증 효과를 나타내는 물 질이 함유되어 있어서 천연 건강식품으로 이용이 가능할 것으로 사료된다.

        Dictyophora indusiata (Vent.) Desv., “Queen of the mushroom”, is a mushroom in family Phallaceae of Basidiomycota, which is commonly used as edible and medicinal mushroom in China and Korea. This study initiated to evaluate the anti-cholinesterase, skin anti-wrinkle and melanogenesis inhibitory of 80% methanol extract from fruiting body of D.indusiata. In the anti-cholinesterase experiment, acetyl-cholinesterase and butyryl-cholinesterase inhibitory activities were performed. The extract were inhibited acetyl-cholinesterase and butyryl-cholinesterase 44.09% and 49.14% at the concentration 1 mg/mL, respectively. The skin anti-wrinkle effect of the extract were determined by measuring anti-collagenase and anti-elastase activities. While melanogenesis inhibitory activities were performed by tyrosinase, DOPA inhibitory and melanin synthesis inhibitory activities. The tyrosinase inhibitory activity of the extract were from 37.60~68.96%, while DOPA inhibitory activity were from 15.43 ~ 29.58% at the concentration ranged from 0.125~2.0 mg/mL. In addition, cellular tyrosinase activity was tested with result of the enzyme activity reduced from 99.09% to 72.91% against 25~500 μg/mL of the extract. The methanol extract of D.indusiata was inhibited melanin synthesis activity 41.86% at the concentration 500 μg/mL. The collagenase inhibitory activity of the extract from D.indusiata were 34.87%, which was comparable with the positive control, EGCG ( 45.31%). While the extract showed good inhibition of elastase enzyme (46.64%). The experiment results suggested that fruiting body of Dictyophora indusiata could be used as natural anti-cholinesterase and skin care agents.

        Calocybe indica (Purkayastha, 1974), Milky mushroom, is a relatively new to the world of mushroom industry, which is belong to Lyophyllaceae of Basidiomycota, and commonly used as edible mushroom in India and Southern Asia country. This study was conducted to investigate the free radical scavenging, skin whitening and anti-collagenase activities of methanol extract from fruiting body of C.indica cultivated in Bangladesh. In addition, the polyphenol compounds of the extract were analyzed by HPLC. The mushroom extract showed good activity in lipid peroxidation which ranged from 29.35% to 55.39% at the concentration 0.125~2.0 mg/mL. The scavenging activity of the extract on 1,1-diphenyl-2-picrylhydrazy radicals were from 20.22~83.93% at the same tested concentration, which were comparable with the positive control BHT. The hydroxyl radical scavenging activity of the extract was 76.24~93.92%. The skin whitening effect of the mushroom extract was performed with UV light protecting, tyrosinase and DOPA inhibitory activities. The methanol extract absorbed UV-B wavelength with maximum level of 0.356 at 280 nm. The tyrosinase and DOPA inhibitory activities of the extract were ranged from 30.36 ~ 66.25% and 7.13 ~ 30.25% at the concentration 0.125~2.0 mg/mL, respectively. Furthermore, anti-collagenase activity were determined by measuring collagenase and elastase inhibitory activity. The collagenase and elastase inhibitory activity of the extract were 42.77% and 51.08% at the concentration 1.0 mg/mL, respectively. The experiment results suggested that fruiting body of Calocybe indica could be used as natural skin care and antioxidant agents.

        Russula compacta, a wild mushroom, belongs to Russulaceae, Russulales of Basidiomycota. This study was conducted to evaluate the free radical scavenging, anti-inflammatory, anticholinesterase and anti-α-glucosidase effects from fruiting bodies of R. compacta extracted with methanol and hot water. In 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging effects, the methanol and hot water extracts showed good scavenging effects comparable with positive control, BHT. The chelating effect of methanol and hot water extracts of the mushroom were significantly higher than the positive control, BHT. The reducing power of the methanol and hot water extracts of the mushroom were lower than the positive control at the concentrations tested. In the HPLC anaysis of phenolic acids profile of the mushroom extract, 7 phenolic acids such as gallic acid, vanillin, rutin hydrate, resveratol, quercetin formononetin, and biochanin-A were detected. Nitric oxide (NO) production in lipopolysaccahride (LPS) activated RAW 264.7 cells was inhibited by 1.5-fold with the treatment of methanol extract when compared with the control. In the anti-cholinesterase activity assay, the methanol extract inhibited the acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) effects by 73.9% and 81.05% at the 1.0 mg/mL concentration, whereas galanthamine, the standard drug, inhibited the AChE and BChE activities by 97.80% and 81.12%, respectively at the same concentration. The methanol and hot water extracts of the mushroom inhibited the α-glucosidase activity by 55.44% and 62.00%, respectively at the 2.0 mg/mL concentration, while acarbose, the positive control inhibited the α-glucosidase activity by 81.81% at the 2.0 mg/mL concentration. From the experimental results, the fruiting bodies of R. compacta contained natural antioxidant, anti-inflammatory, anti-cholinesterase, and anti-diabetic substances, which might be used for health foods.

        Coprinellus miaceus, belongs to Coprinaceae of Agaricales, Basidiomycota, has been used for an edible purposes in asian countries. This experiment was initiated to evaluate the free radical scavenging, free radical scavenging, anti-acetylcholinesterase, anti-inflammatory and α-glucosidase inhibitory activities of C. micaceus fruiting bodies extracted with methanol and hot water. In 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, the scavenging activities of methanol and hot water extracts were lower than that of positive control, BHT. The chelating effects of methanol and hot water extracts were significantly higher than positive control, BHT at the concentrations of 0.125-2.0 mg/mL. In the reducing power assay, methanol and hot water extracts exhibited the lower activities than the positive control, BHT at the 0.125-0.2 mg/mL concentration. In the HPLC analysis of phenolic acids profile of the mushroom fruiting bodies, 4 phenolic compounds including procatechuic acid, chlorogenic acid, (-)-epicatechin, and naringin were detected. The tyrosinase inhibitory activities of methanol and hot water extracts were 91.33% and 91.99% at 2.0 mg/mL concentration, while the inhibitory activity of kojic acid, the positive control, was 99.61%. Nitric oxide (NO) production in lipopolysaccahride (LPS) activated RAW 264.7 cells were inhibited by the methanol extract in a concentration dependent manner. In the acetylcholinesterase (AChE) inhibitory activity assay, methanol and hot water extracts of the mushroom inhibited the AChE by 94.64% and 74.19%, respectively at the 1.0 mg/mL concentration, whereas galanthamine, the standard drug inhibited the AChE activity by 97.80% at the same concentration. The methanol and hot water extracts of the mushroom inhibited the α-glucosidase activity by 62.26% and 67.59%, respectively at the 2.0 mg/mL concentration, while acarbose, the positive control inhibited the α-glucosidase activity by 81.81% at the same concentration. Therefore, it is concluded that fruiting bodies of C. micaceus contained natural antioxidant, anti-inflammatory, anti-acetylcholinesterase and α-glucosidase inhibitory substances which might be used for promoting human health.

        Lentinus giganteus is a edible mushroom cultivated in Asian countries. The present study was initiated to evaluate the anti-inflammatory, anti-acetylcholinesterase, anti-α-glucosidase, and free radical scavenging activities from fruiting bodies of L. giganteus extracted with methanol and hot water. The free radical scavenging activities of methanol and hot water extracts on 1,1-diphenyl-2-picrylhydrazyl (DPPH) were 92.26% and 90.17% at 2.0 mg/mL concentration, respectively and comparable with positive control, BHT. The chelating activities of methanol and hot water extracts were significantly higher than the positive control tested. The reducing power of methanol and hot water extracts showed lower activities compared to positive control, BHT. The phenolic and flavonoid contents of hot water extract were 1.56 μg/mg and 24.35 μg/mg, respectively. Nitric oxide (NO) produced by lipopolysaccahride (LPS) activated RAW 264.7 cells were significantly inhibited by treatment of methanol and hot water extracts. The methanol extract inhibited the acetylcholinesterase (AChE) activity by 91.19% at the 2.0 mg/mL concentration, whereas galanthamine, the standard drug, inhibited the AChE activity by 97.80%. The hot water extracts inhibited the α-amylase and α-glucosidase activities by 78.86% and 80.78%, respectively at the 2.0 mg/mL concentration, while acarbose, the positive control, inhibited the activities by 89.91% and 81.81%, respectively at the same concentration. Therefore, it is concluded that fruiting bodies of L. giganteus contain antioxidant, anti-inflammatory, anti-cholinesterase, and anti-diabetic substances, which can be used for natural health food for promoting human health.
