        소형 다화 분지성의 품종을 육성하기 위하여 호접란 ‘KSVivien’은 2004년 강산난원에서 Dtps. ‘Sogo Vivien’과‘Brother Little Hatter’을 각각을 모부본으로 교배하여 육성한 F1개체 중에서 선발한 소형 다화 분지성의 신품종이다. 2005~2006년에 걸쳐 실생개체 중에서 영양생장과 개화특성이 우수한 개체 KS-507-9를 선발하였다. 2007년과 2009년에 각각 1차, 2차 특성검정을 통하여 품종의 안정성과 균일성을 확인하고 ‘KS Vivien’으로 명명하였다.‘KS Vivien’은 화색이 연분홍(2N81B)에 줄무니가 있는스트라이프계통이며, 화형은 앉아피기로 꽃의 길이와 폭은 각각 4.0, 4.3cm로 소형이다. 화서의 형태는 분지하는복총상화서이다. 잎의 길이와 폭은 각각 9.9, 6.4cm이며엽형은 좁은 도난형의 반직립이다. 기내증식율이 높고 식물체가 강건하고 생육이 빠르기 때문에 재배가 용이한 품종이다. 2010년 11월 11일 품종등록(등록번호 제3377호)하여 종자산업법에 의해 보호받고 있다.

        Lilies are of great economic important floweringplant that belongs to the genus Lilium can be grown underdiverse climatic conditions. During the last decade the lilyhas gained popularity worldwide among cut flowers and pot-ted flowering plants. Seeing the great recognition of lilies ininternational flower trade, several breeding approacheshave been adopted on different Lilium sections/groups andmore than 10,000 lily cultivars have been bred. Innovativebreeding strategies and advancement in molecular and bio-technology techniques have made the assortment of liliesdramatically. Different DNA marker approaches have greatpotential to increase the precision and efficiency of conven-tional lily breeding via marker-assisted selection (MAS) anddifferent breeders used it for Lilium crop improvement. Inthis review, different approaches and techniques that lilybreeders have employed to develop novel cultivars are dis-cussed. It is imperative to recognize that there must be anenhanced integration and synchronization in different lilyobjective oriented breeding programs, so that current issues,barriers etc. can be well identified with their appropriatesolutions.

        Chrysanthemum is one of the most popular and economically important ornamental plants due to its huge diversity in growing habits, wide range of colors, and different patterns of flower. In the present study, we conducted the karyotype analysis in four naturally occurring genotypes of Chrysanthemum boreale. Karyotype studies based on the fluorescence in situ hybridization (FISH) using 5S and 45S rDNAs. Four chrysanthemum genoteyps showed an aneuploid chromosome number of 2n=18+2 (111016 and 111021) or a diploid of 2n=18 (121001 and 121002). All the genoteyps had the same karyotype formula of 14 metacentrics and 4 submetacentrics. In 111016, the chromosome length during somatic metaphase ranged from 3.11 ± 0.26 μm (shortest) to 3.94 ± 0.20 μm (longest), with a total length of 32.94 μm. The chromosome length at the mitotic metaphase ranged from 3.11 to 6.46 μm, with a total length of 32.94 μm in 111016 and 51.05, 32.81, and 46.00 μm in 111021, 121001, and 121002, respectively. The 5S rDNA and 45S rDNA signals recorded different in all four wildly grown genotypes of C. boreale. This information can be useful in cultivar improvement, as well as in elucidation of the evolution of the chrysanthemum.

        Potting media are an integral part of potted plants production system, formulated by a mixture of various raw materials to act as a substrate for plant support and source of nutrition. There is a wide range of potting media available, which are used for quality production of high-value plants worldwide. In this study, quality assessment of Dracaena plants grown in different potting media like: mushroom compost, peat, sand, farmyard manure, coconut coir, and leaf compost in different combinations were carried out. A mixture of sand, mushroom compost, coconut coir, and peat proved to be the best, by promoting root and shoot growth, leaf area, fresh and dry weights of root and shoot, and also improved overall quality of the plant. Media containing garden soil alone or a mixture of sand and leaf compost produced the lowest quality of plants with considerable reduction in most of the growth parameters. Anatomical parameters showed weak relation to media composition as compared to other morphological features. The best medium was made with sand, mushroom compost, coconut coir, peat, that produced plants with thick epidermis and larger pith cells, but thin endodermis and smaller metaxylem vessels in their roots. As moisture availability was sufficient enough in this medium combination, the development of water storing tissue may not contribute significantly. However, epidermis can play a decisive role in protecting roots from external environmental hazards.

        Polyploidy analysis being one of the important mechanisms to study the genetic resources and plant diversity which plays significant role in crop evolution, breeding programs, and crop improvement was conducted in this study. The root tips of nine Cymbidium genotypes were used as materials to determine their chromosome number and flow cytometric analysis of chromosomal ploidies. The chromosome number (2n=40) has been observed in all the nine genotypes. Results showed that among nine cymbidium genotypes, six were tetraploid (2n=4x=80), two diploid (2n=2x=40), and one triploid (2n=3x=60). This study provides useful chromosome landmarks and evidence about genome evolution, heteromorphic chromosomes based heterozygosity, basic chromosome number and ploidy level in the genus Cymbidium. These results can be used as theoretical base line for crossbreeding in the genus Cymbidium.

        본 연구에서는 다양한 색상의 염료가 절화 장미 ‘Akito’ 품종의 염색에 미치는 효과를 알아보기 위해 시행되었다. 12가지 색상의 염료를 이용한 염색 결과, 다채로운 색상을 구현해 내고 관상 가치를 높일 수 있었지만 절화수명은 대조구보다 1-3일 단축되었다. 4종류의 blue 염색염료를 비교한 결과 Fantasy blue (Palace Chemical Co., Japan)와 Blue No.1(Vidhi Dyestuff Mfg LTD, India)의 색상이 좋았다. 고농도 (7.5g •L-1와 10.0g •L-1)의 blue 염색의 경우 잎의 착 색이 심하고 불균일 하였으며 꽃잎의 가장자리가 짙어 지는 현상이 나타났다.

        새로운 종간교잡계통의 백합 품종을 육성하기 위해 화주절단수분법으로 종간친화성과 배배양을 통한 종간 교잡을 실시하였다. Oriental hybrid ‘Siberia’를 교배 모본으로하고 L. amabile, L. callosum, L. hansonii, L. tsingtauense, ‘Lorina’를 부본으로 사용하였다. 개 화당일 채취한 화분의 발아율은 Martagon group인 L. hansonii가 50%로 가장 높았으며, ‘Lorina’가 22%로 가장 낮았다. 자방 성숙시기는 ‘Siberia’ × ‘Lorina’(OL) 가 109일로 가장 길었으며, ‘Siberia’ × L. tsingtauense( OM)는 70일, OA 교배조합인 ‘Siberia’ × L. amabile(OA)는 82일, ‘Siberia’ × L. callosum(OA)이 68일로 관찰되어 OL 교배조합이 다른 교배조합에 비 해 자방의 성숙시기가 길었다. ‘Siberia’와 ‘Lorina’의 종간교배 중 3개의 자방에서 35개의 배주를 배양하였 으며 그 중 32개의 식물체를 얻어 가장 높은 재분화 율(91.4%)을 나타내었다. ‘Siberia’ × L. tsingtauense의 조합에서는 7개의 자방을 얻어 4개의 식물체를 획득하 였으며, ‘Siberia’ × L. amabile, ‘Siberia × L. callosum 의 교배조합에서는 각각 75개, 37개의 배주를 얻었으 나 발아하지 않아 식물체를 획득하지 못했다.

        오리엔탈 백합의 생육 특성조사를 통해 백합 육종효 율을 증진시키기 위해 본 연구를 실시하였다. 23 품종 중 상향 개화성 품종이 13품종으로 나타났으며, 반점 이 없는 품종은 9품종으로 조사되었다. ‘Caruso’는 초 장이 36.7 cm로 가장 짧아 분화용 품종육성에 적합할 것으로 판단되고, ‘Medusa’는 100 cm로 가장 길어 절화용 품종 육성에 유리할 것으로 판단된다. 개화소요 일은 ‘Viviana Zantriana’가 79일로 가장 짧았으며, ‘Sorbonne’이 115일로 가장 길었다. 임성율은 대부분 70% 이상으로 높았으나 ‘Red Reflex’는 2%로 가장 낮았다. 발아율은 ‘Eton’이 70%로 가장 높았고 ‘Red Reflex’ 외 6품종이 20% 미만으로 나타났다. 발아율이 낮은 품종은 부본보다는 모본으로 사용하는 것이 육종에 효율적일 것으로 판단된다.

        Dual-color fluorescence in situ hybridization karyotype analysis was created using repetitive sequences including two types of rDNA repeats (45S and 5S rDNAs) and Arabidopsis-type telomere sequence repeats. The somatic metaphase cells of Carthamus tinctorius were observed as diploids (2n=2x=24). A symmetrical or slightly asymmetrical karyotype with seven pairs of metacentric and five pairs of submetacentric chromosomes was observed. The lengths of the somatic metaphase chromosomes ranged from 4.18 to 6.53 ㎛, with a total length of 60.71 ㎛. One locus of 45S rDNA was located on the pericentromeric regions of three pairs of chromosomes and the other pair was situated on the terminal regions of the short arms of a single pair of chromosomes. One locus of 5S rDNA was detected on the interstitial regions of the short arms of two pairs of chromosomes. Arabidopsis-type telomeric repeats were detected on the terminal regions of all pairs of chromosomes. Co-localization of loci between telomeric repeats and 45S rDNA was observed in a single pair of chromosomes. The results provide additional information for the existing physical mapping project of C. tinctorius and will also serve as a benchmark to a more intricate cytogenetic investigation of C. tinctorius and its related species.

        In the genus Chrysanthemum, repetitive DNA sequences, the dominant part of a genome, are still to be elucidated. To explore the matter, the present study applied fluorescent in situ hybridization (FISH) to the mitotic metaphase chromosome of Chrysanthemum boreale with C0t DNA as probes. Based on DNA re-assotiation kinetics, three kinds of C0t DNA exhibiting different degrees of repetitive nature were fractionated and used as FISH probes to map the repetitive sequences. Signals from all C0t DNAs were successfully observed but their coverage on the chromosomes was different among C0t-1, C0t-10, and C0t-100. C0t-1 FISH signals resulted to have its intensity on the telomeric region and were also dispersed on both chromosome arms except for some distal regions. In C0t-10, signals were observed in all parts of the chromosome with greater intensity around pericentromeric regions. FISH with C0t-100 DNA was observed in bright signals all over the chromosome. Signals of C0t FISH found in this study covered the regions where ribosomal DNAs and telomeric repeats of C. boreale have been distributed (previous report), thus signifying their repetitive attributes. The present results could enhance the efficiency of studying genomes, chromosomes and repetitive sequences of C. boreale and subsequently hasten the realization of the genetic scheme of Chrysanthemum.

        Chrysanthemums (Asteraceae) are important ornamental crops in worldwide that are well known as commercial valuable cultivars for cut flowers, potted plants, and garden flowering plants. Genus chrysanthemum consisted of 41 species that are mostly distributed in East Asia. Chrysanthemum has diverse ploidy levels with the basic chromosome number of x=9 from 2n=2x=18 (diploid) to 2n=10x=90 (decaploid). Fluorescence in situ hybridization (FISH) is a useful tool for studying the distribution of ribosomal DNAs. In this study, we have confirmed ploidy level by chromosome counting method. The somatic metaphase chromosome numbers were observed 2n=2x=18 in Chrysanthemum boreale, and 2n=6x=54 in C. indicum and C. zawadskii. More detailed Karyotype was constructed based on FISH method using 5S and 45S rDNA probes. Two (2) loci of 5S rDNA signals were detected in interstitial region of long arm chromosome in C. boreale and six (6) loci were in C. indicum and C. zawadskii. All of 45S rDNAs were located in terminal region of short arm chromosome which were visualize in six (6) loci in C. boreale and C. indicum and twelve(12) loci in C. zawadskii. In this study, it was the main topic to perform physical mapping of the location of 5S and 45S rDNA. Three of wild chrysanthemum showed variations in number of ribosomal DNAs. In the present investigation will help to further study of genome sequencing project in chrysanthemum.

        Fluorescence in situ hybridization (FISH) is a powerful tool for the detection of DNA sequences in the specific region of the chromosomes. As well as for the integrated physical mapping, FISH karyotype analysis has to be preceded. The detailed karyotypes of two onion cultivars, which are resources for onion genome sequencing project (‘Eumginara’ and ‘Sinsunhwang’), were constructed based on triple color fluorescence in situ hybridization (FISH) using 5S rDNA, 45S rDNA, and tandem repeat sequence. All used our materials showed 2n=2x=16 with x=8 as basic chromosome number. 5S rDNAs were located on 4 loci in one pair of interstitial region of short arm chromosome in both onion cultivars. Two pairs of 45S rDNAs were positioned in distal region of short arm chromosome in ‘Eumginara’. Otherwise 5 loci of 45S rDNAs were located in distal region of two pairs of short arm chromosome in ‘Sinsunhwang’. Among them, two signals of 45S rDNAs were co-localized in distal part of short arm and long arm chromosome, respectively. In case of tandem repeat sequence was detected on telomeric region of 8 pairs of chromosomes except on 45S ribosomal DNA sites. These results will provide a valuable background for physical mapping and help to further more understand the genome sequencing project in Allium cepa.

        Radish, Raphanus sativus L., is an annual vegetable of the family Cruciferae. Radish has RR genome with 18 somatic chromosome numbers (2n=2x=18). Until now, detailed karyotypic analysis is not only constructed only by conventional staining techniques but also other method. Fluorescence in situ hybridization (FISH) is a powerful molecular cytogenetic technique using chromosomal markers that reveal the positions of specific genes, such as ribosomal DNAs, thereby making it easy to identify individual chromosomes. We have constructed detailed karyotypes of four different local and wild varieties of radish, based on the chromosome arm length and fluorescence in situ hybridization (FISH) with the 45S rDNA and 5S rDNA as probes. As for the karyotype of radish, 9 pairs of chromosomes were extremely small in size with about 1 to 3 um in length at mitotic metaphase having metacentrics or submetacentrics. Three pairs of 45S rDNA signals and two pairs of 5S rDNA signals were observed in four radish species. One pair of 45S rDNA signal was located on terminal region of short arm chromosome, while two pairs were in interstitial region. Two pairs of 5S rDNA signals were located on interstitial region of chromosome. In conclusion, it was feasible to identify the radish by karyotype and physical mapping analyzed using ribosomal DNA.

        We established a practical method for rapid and large-scale production of Valeriana fauriei var. dasycarpa Hara roots by bioreactor culture and confirmed valerenic acids and valepotriates production. We also compared valerenic acids and valepotriates production patterns according to various media conditions. Among the media tested, B5 medium gave the maximum biomass production of 101 g fresh weight, which was a 5.03-fold multiplication rate obtained 4 weeks after inoculation of 20 g of fresh weight. The best production of total valerenic acids (7.86 mg/l) and valepotriates (8.96 mg/l) was B5 medium.
