It is essential to select materials with excellent mechanical properties to prevent chemical and mechanical damage to the surfaces of materials used in machines and structures and to extend their lifespan. Co-based stellite alloy, which has wear resistance, heat resistance, and corrosion resistance, is essential for products used in harsh environments. However, due to the problem of enormous costs, research on hard facing, which uses a stellite coating layer only on the contact surface, is urgently required. Currently, high-facing research on Stellite coating layers is focused on powder, and GMAW research using wire is relatively lacking. In this study, welding experiments were performed to form stable weld beads using stellite 6 welding wire, and the correlation between parameters and weld beads was analyzed. A CMT welder was used to minimize the heat effect on the base material.
Laser cladding a surface treatment process that grants superior characteristics such as toughness, hardness, and corrosion resistance to the surface, and rebuilds cracked molds; as such, it can be a strong tool to prolong service life of mold steel. Furthermore, compared with the other similar coating processes – thermal spray, etc., laser cladding provides superior bonding strength and precision coating on a local area. In this study, surface characteristics are studied after laser cladding of low carbon steel using 18%Cr-2.5%Ni-Fe powder (Rockit404), known for its high hardness and excellent corrosion resistance. A diode laser with wavelength of 900-1070 nm is adopted as laser source under argon atmosphere; electrical power for the laser cladding process is 5, 6, and 10 kW. Fundamental surface characteristics such as crossectional microstructure and hardness profile are observed and measured, and special evaluation, such as a soldering test with molten ALDC12 alloy, is conducted to investigate the corrosion resistance characteristics. As a result of the die-soldering test by immersion of low carbon alloy steel in ALDC12 molten metal, the clad layer's soldering thickness decreases.
This paper provides a study on the application and proposals of safety culture, new public management and social amplification of risk framework via ship accidents in Korea. This document analyzes what are the concept of safety culture, new public management as well as social amplification and risk framework and describes how 3 issues act, harmonize, interrelate through M/V Sewol accident. Korean government is needed to apply social amplification of risk framework to the in order to promote the safety culture in the maritime administration. Hence, this paper proposes safety framework in order to prevent and resolve future unexpected accident especially for maritime field.
본 연구에서는 피로 회복 또는 원기 회복에 효능이 있는 것으로 알려진 홍경천과 홍삼을 이용하여 홍경천-홍삼 복합 발효물의 산화적 손상 억제 효과를 평가하고자 H2O2로 산화적 스트레스를 유도시킨 C2C12 근육세포에 홍경천-홍삼 복합 발효물의 처리한 후, 세포의 morphology, cell viability 및 항산화 효소들의 유전자 발현 양상을 비교, 분석하였다. 홍경천-홍삼 복합 발효물은 C2C12 근육세포의 cell viability를 유의적으로 증가시켰으며, Cu/Zn-SOD, Mn-SOD 및 GPx 등과 같은 세포내 항산화 효소의 발현을 증가시킬 뿐만 아니라, 근육세포 분화의 주요 전사인자인 Myo D의 발현 또한 증가시키는 것으로 나타났다. 이상의 결과로, 홍경천-홍삼 복합 발효물은 세포내 항산화 효소 시스템을 증가시켜 외부로부터의 산화적 손상에 대한 방어효능을 갖는 것으로 나타났으며, 향후 in vivo 시스템 이용한 추가적인 연구가 수행된다면, 홍경천-홍삼 복합 발효물을 이용한 항피로 건강기능식품의 소재개발이 가능할 것으로 판단된다.
The study was conducted to investigate the condition for mixed fermentation of Rhodilola sachalinensis with red ginseng using Lactobacillus acidophillus 128 and the changes of physicochemical properties and antioxidant activities before and after the lactic acid fermentation was examined. In the single fermentation of Rhodiola sachalinensis extract, the pH and titratable acidity rarely changed, and the number of lactic acid bacteria decreased greatly. On the other hand, in the lactic acid fermentation of Rhodiola sachalinensis-red ginseng mixed extract of 50% red ginseng content, the pH decreased, whereas the titratable acidity and the number of lactic acid bacteria increased. The solid content of optimal mixed extract for lactic acid fermentation was 0.5%. Sugar content decreased during fermentation, but total phenolic compounds tended to increase during fermentation. The salidroside and p-tyrosol content of the initial Rhodiola sachalinensis-red ginseng mixed extract was 419.5 ㎎% and 60.1 ㎎%, respectively; after fermentation, the salidroside content after lactic acid fermentation decreased greatly to 81.8 ㎎%, and the amount of p-tyrosol increased greatly to 324.9 ㎎%. The DPPH scavenging activity of Rhodiola sachalinensis-red ginseng mixed fermentate was 78.1% at 0.1% concentration, showing a tendency to increase as compared to 50.3% of Rhodiola sachalinensis-red ginseng mixed extract before the fermentation (p<0.05); it was a higher antioxidant activity as compared to the single fermentation of Rhodiola sachalinensis or red ginseng.
This study was conducted to develop HTHP ginseng (high temperature and high pressure ginseng) with improved antioxidative activity and phenolic acid composition by high temperature and high pressure process. The HTHP ginseng extract was analyzed for the total phenol content, DPPH radical scavenging activity and phenolic acid composition. The total phenol content was increased in HTHP ginseng (14.76 ㎎/g) compared to raw ginseng (3.59 ㎎/g) and red ginseng (3.93 ㎎/g). DPPH radical scavenging activities of HTHP ginseng, raw ginseng and red ginseng extracts were 4.8~78.4%, 1~47.4% and 1.8~56.5% at 1~100 ㎎/㎖ concentration. Also ABTS radical scavenging activities of HTHP ginseng, raw ginseng and red ginseng extracts were 8.9~99.8%, 3.4~96% and 1.2~96.5% at 1~100 ㎎/㎖ concentration. In HPLC analysis, amounts of measured phenolic acid of HTHP ginseng greatly increased than raw ginseng and red ginseng, but salicylic acid was not detected in HTHP ginseng. In addition, DPPH radical scavenging activity of phenolic acid from HTHP ginseng was increased. Consequently, we believe high temperature and high pressure process is better method than existing method to increase the bioactivity of ginseng.
Reactive oxygen species (ROS) are produced by oxidative stresses which cause various chronic diseases such as diabetes and obesity. Ginseng (Panax ginseng C.A. Mayer) has been reported to contain various biological activities such as anti-cancer, anti-diabetic, neuroprotective, radioprotective, anti-amnestic and anti-aging effects. In this study, we investigated the effects of Panax ginseng, treated with high temperatures and high pressures, on oxidative stress in C2C12 myoblasts and 3T3-L1 adipocytes. Oxidative stress was induced in the C2C12 cells through the introduction of H2O2 (1 mM), and cells were then treated with various ginseng preparations: dried white ginseng (DG), steamed ginseng (SG) and high temperature and high pressure treated ginseng (HG). In addition, 3T3-L1 preadipocytes were treated with various ginsengs for up to 8 days following standard induction of differentiation. Our results show that HG treatment significantly protected oxidative stress in both cell lines and enhanced gene expression of antioxidant enzymes. Therefore, in this study, we investigated the protective effects of ginseng on the oxidative stress of adipocytes and muscle cells.
Rhodiola sachalinensis fermentates by lactic acid bacteria were prepared using the adsorption process, and were investigated for changes of the main compounds and anti-oxidative activities during the adsorption and fermentation process. While the R. sachalinensis extract (RSE), which did not go through the adsorption process, showed little change in pH during fermentation and a significant reduction in the number of lactic acid bacteria, the pre-preparatory adsorption process was found to be helpful for promoting fermentation and for maintenance of bacterial numbers. The contents of total phenolic compounds mostly decreased during the adsorption process, but showed an increasing tendency to rebound during the fermentation process. The contents of salidroside and p-tyrosol in the RSE were 1153.3 ㎎% and 185.0 ㎎% respectively, and they did not significantly change after treatment with acid clay or bentonite as adsorbents, which were 1093.0 and 190.5 ㎎% by acid clay, and 882.2 and 157.3 ㎎% by bentonite. When the extract was fermented after treatment with acid clay or bentonite, the salidroside contents were decreased by 282.7 and 505.0 ㎎% respectively, but the p-tyrosol contents were increased by 714.0 and 522.4 ㎎% respectively. Compared to the DPPH radical scavenging activity of the RSE (66.8%) at the conc. of 0.1%, that of the fermented RSE, which went through adsorption process with acid clay or bentonite, was significantly increased to 79.4 and 72.7% respectively at the same concentration (p<0.05). Though fermentation by lactic acid bacteria was suppressed in the RSE, the results suggested that the adsorption process may promote fermentation without any change in the content of major active compounds. It is expected that fermentation by lactic acid bacteria could improve the antioxidant activity and various associated functionalities of R. sachalinensis.
In this study, we examined immuno-modulatory activities of crude polysaccharides from wild ginseng adventitious roots (WGAR). The crude polysaccharide (WGAR-CP) was isolated from WGAR by hot water extraction, ethanol precipitation, and dialysis. The major constituents in WGAR-CP were neutral sugar (64.77%), and uronic acid (34.32%). WGAR-CP demonstrated anti-complementary activity dose-dependently. The immuno-modulatory effects of WGAR-CP were also analyzed by measuring nitric oxide and cytokines in the supernatants of mouse peritoneal macrophages. Mouse peritoneal macrophages stimulated with WGAR-CP produced nitric oxide and various cytokines such as interleukin (IL)-6 and IL-12 in a dosedependent manner. In conclusion, WGAR-CP may have immuno-modulatory activities by activating a complementary system and macrophages, which produces cytokines.
Rice blast caused by the fungal pathogen, Magnaporthe oryzae, is a serious disease affecting yield loss and decreasing its quality in rice production. Rice breeders in Korea have developed many japonica varieties showing resistance to blast. However, the blast resistance in most japonica varieties has broken down within a few years after they were released to farmers because of the spread of new virulent races of M. oryzae. There is the most effectiveness to look for novel resistant gene(s) that can express the resistance to broad-spectrum races in diverse environmental conditions. We identified a major QTL, qLB4.1 linked tightly to RM6352 and RM3643 in 52.6 cM region on chromosome 4 related to the resistance for isolate inoculation and nursery test, and neck blast from a Korean weedy rice, Geumleungaengmi33. This QTL explained 26.1∼28.6% and 45.3∼53.1% of total phenotypic variation by the allele of GL33 for isolate inoculation and nursery test, respectively. A line SR30058(52)-1-1 (Suweon545) that containing the QTL qLB4.1 was developed from Ilpum*4/GL33 by marker-assisted backcross method. This line showed resistant reactions to blast nursery test across regions and years, and resistance to neck blast at the hot-spot field in Jecheon. Suweon545 showed also durable resistance of lower 10% of diseased leaf areas (DLAs) in sequential planting method. This line screened by graphical mapping using 136 SSR markers that evenly distributed on 12 chomosomes. Suweon545 contained GL33 alleles of donor parent in a total of 12 loci (8.8%) including QTL region on chromosome 4. In future, Suweon545 would be useful to develop the broad-spectrum resistance variety in japonica rice breeding program.
Rice is the most important crop as the staple food and two priorities in rice production are high yield and good quality in Korea. Far more improvements in grain quality, especially eating quality are required to meet the demand of consumers in rice producing areas. In the study presented here, a recombinant inbred lines (RILs) population derived from a cross between a temperate japonica and a tropical japonica and its genetic linkage map were employed to locate the QTL locus underlying six parameters of RVA profiles. Out of six RVA profile parameters, two characteristics, breakdown viscosity (BDV) and setback viscosity (SBV) are more correlated to eating quality of cooled rice. A total of four QTLs for two RVA profile parameters were identified. Two QTLs, qBDV-6 and qBDV-9 for BDV were detected on chromosomes 6 and 9. These QTLs increased the BDV by 26.2 and 16.4 from Ilpumbyeo allele, respectively. A QTL, qBDV-6 in the interval RM540-RM587 of the wx gene on chromosome 6 was reacted as a major QTL which could explain 26.2% of the total phenotypic variation. A QTL, qBDV-9 in the interval RM5688-RM444 explained 7% of the total variation as a minor QTL. Two QTLs, qSBV-6 and qSBV-9 for SBV also identified at the same region with QTLs of BDV on chromosomes 6 and 9. A QTL, qSBV-6 in the interval of the wx gene on chromosome 6 could explain 24.4% of the total phenotypic variation. A QTL, qSBV-9 in the interval RM5688-RM444 explained 8.2% of the total variation. These QTL region on chromosomes 6 and 9 would be use as useful marker to select elite lines of good eating quality in early generations in japonica rice breeding.
Field resistance is defined as the resistance that allows effective control of a parasite under natural field condition and is durable when exposed to new races of that parasite. To identify the genes for field resistance against rice blast, quantitative trait loci (QTLs) conferring field resistance in japonica rice cultivars were detected and mapped using SSR markers. QTL analysis was carried out in 190 RILs population from the cross between Suweon365 (moderately resistance) and Chucheong (highly susceptible). Fourteen QTLs for nine blast races inoculated were detected on chromosomes 1, 2, 4, 5, 6, 7, 11 and 12. They explained from 6.4 to 39.7% of total phenotypic variation. Eight QTLs for blast nursery screening in 4 regions for three years were detected on chromosomes 1, 2, 4, 5, 11 and 12. The phenotypic variation was explained by each QTL ranged from 5.9 to 38.0%. Three BC2F5 backcrossed progeny lines were developed to transfer the QTLs into the susceptible cultivar Chucheong as a recurrent parent. A NIL4 containing two QTLs Qbl6.2 and Qbl7 for blast races showed the reaction 6 to 7 in blast nursery in 2007 and 2008, respectively. Two lines NIL143 and NIL93 containing Qbl11.2 and Qbl12.1 for QTLs related with field resistance, respectively, were 3 to 4 reactions in blast nursery.
