Embryoid bodies (EBs) generated from human embryonic stem cells (hESCs) include spontaneously induced endodermal lineage cells (ELCs). Activin-A plays important roles in the endoderm differentiation of hESCs. Despite studies on the generation of ELCs from hESCs with treatment of Actvin-A, it was unclear for localization and pattern of ELCs by Activin-A during differentiation of hESCs. Accordingly in this study, we knew that Actvin-A increased the cystic EBs formation, including the highly enriched AFP (endoderm lineage specific marker)-expressing cells in the surface of cystic EBs. To induce the EBs formation from undifferentiated hESCs, cells were transferred onto petri-dish and cultured in suspension condition without bFGF removed hESC media (EB media) for 3 days. Next to investigate the effect of Activin-A, EBs were subsequently cultured in EB media supplement with 100 ng/ml Activin-A for 3 days. After 5～7 days of Activin-A treatment, cystic EBs began to appear which increased in numbers reaching ～60% of initially formed EBs over 5 days. Endoderm lineage marker, AFP were highly expressed and specifically localized at the surface region of cystic EBs comparison with normal EBs. We next attached the cystic EBs onto gelatin-coated plates and cultured for 5 days. In the results of real-time PCR and immunocytochemistry analysis, AFP-expressing cells migrated and localized at the outgrowth region of attached cystic EBs. To obtain the AFP-expressing cells of the outgrowth region, we manually isolated by using micro- dissection and cultured them. These cells strongly express AFP over 70% of isolated cells post re-plating. Here, we first showed an expression pattern of specifically localized ELCs by Activin-A during differentiation of hESCs. From this observation, we could highly purified ELCs from undifferentiated hESCs. Taken together, our system will provide a novel and efficient option to generate ELCs from hESCs.

본 연구는 그룹코칭에 대한 연구가 절대적으로 부족한 상황에서 문헌 연구를 통해 그룹코칭을 팀코칭과 비교함으로써 그 정의를 보다 명확히 하고 그룹코칭과 유사한 영역으로 언급되고 있는 퍼실리테이팅과 트레이닝과의 차이를 정리해보았다. 또한 그룹코칭을 비구조화, 준구조화, 구조화 그룹코칭으로 구분하고 각각의 구조에서 요구되는 것을 명확히 함으로써 그룹코칭에 대한 혼란을 최소화 하고자 하였다. 현재 국내에서 활발히 활동하고 있는 그룹코칭 전문코치 15인을 대상으로 그룹코칭의 성공에 영향을 미치는 요인에 대한 설문을 실시한 결과 전문코치와 관련된 요인(그룹코칭 진행기간, 전문코칭경험, 전문코칭교육)과 목표설정이 그룹코칭을 성공적으로 이끄는데 매우 중요한 것으로 나타났다. 본 연구자는 문헌연구와 설문 결과를 근거로 효과적인 그룹코칭 프로세스를 제안하였다. 사전, 코칭진행, 사후 프로세스로 전개하고 있으며 각각의 프로세스에서 중요하게 다루어야 하는 요인들을 언급하였다. 조직(기업) 그룹코칭에서는 그룹코칭의 참가자들뿐만 아니라 조직도 코치(코칭펌)의 고객으로서 프로세스에 포함시켜야 한다. 또한 참가자들의 직속상사를 그룹코칭 프로세스에 포함시킴으로써 참가자들이 보다 적극적으로 그룹코칭에 참여하고 현장에 적용할 수 있는 환경을 조성하도록 한다. 다양한 형태로 진행되고 있는 그룹코칭이라도 ‘코칭’이 정확하게 적용되어야 그룹코칭으로서의 생명력을 갖게 된다. 본 연구는 다양한 형태의 그룹코칭을 분류하는 기준을 제시함으로써 향후 그룹코칭 연구의 출발점을 보다 명확히 하였다는 점에서 그 의미가 있다고 할 수 있다.

본 연구는 IT프로젝트를 진행 함에 있어 프로젝트의 시작부터 완료까지, 각 단계의 진행에 따라 나타 날 수 있는 IT프로젝트의 특징 중 하나인 프로젝트 진행 단계에서의 비가시성을 고려하여 고객의 다양하고 복잡하게 변화 하는 요구 사항에 대하여 IT프로젝트의 진행 과정에 보다 유연하게 대응하고 고객의 요구사항에 맞춘 IT프로젝트를 완료하기 위하여 요구공학 관점에서 요구 관리 프로세스의 중요한 요소들을 연구하고 성공적인 IT프로젝트를 위한 효과적인 요구사항 관리 모델을 제시하고자 한다.

Bacillus thuringiensis (Bt) is effective to control the diamondback moth, Plutella xylostella. However, its relative slow and unstable control efficacy limits its wide use by farmers. To facilitate pathogenic rate of Bt, a bacterial mixture technique has been developed in this study. Two entomopathogenic bacteria, Xenorhabdus nematophila (Xn) and Photorhabdus temperata temperata (Ptt), possess high immunosuppressive activity against several lepidopteran insects. The mixture treatments using Bt + Xn or Bt + Ptt significantly enhanced Bt pathogenicity in median lethal concentration and time. Though live Xn and Ptt bacterial cells gave significant effect on the pathogenicity, their 48 h culture broth after removing the bacterialcells still possessed the synergistic effect on the Bt pathogenicity. The larvae fed with the bacterial culture broth suffered significant immunosuppression in response bacterial to infection

최근 철도차량은 안전성과 신뢰성 향상을 위해 점차 복잡하게 설계 제작되고, 품질에 대한 기대와 요구수준이 점차 높아짐에 따라 운영기관에서는 과학적이고 체계적인 예방 정비를 통한 안전성과 가용성 향상을 위해 노력하고 있다. 이러한 목적을 달성하기 위하여 여러 방안들이 연구되고 있으며, 대표적으로 신뢰성 기반 유지보수(RCM; Reliability Centered Maintenance)가 철도분야에 지속적으로 도입되고 있는 추세이다. 본 연구에서는 새로운 예방정비 기술로 대두되고 있는 RCM의 기본이론에 대한 고찰과 RCM의 일반적 실시 절차를 소개하고, RCM의 국제규격인 IEC 60300-3-11, NAVAIR 00-25-403, MIL-STD-2173을 비교 분석하여 이를 바탕으로 철도차량에 RCM 도입 시 효과적이고 적합한 절차 및 방안을 제시하고자 한다.

온라인 게임의 품질 관리는 게임의 완성도를 결정하는 중요한 단계 중 하나이다. 본 논문에서는 대학 수업을 통해 게임의 품질 관리를 효과적으로 운영하기 위한 포커스 그룹 테스트(Focus Group Test) 프로세스를 제안한다. 이를 위해 게임 테스팅 업체에서 시행하는 테스트 항목들을 검토하여 포커스 그룹 테스트를 통해 게임 품질 관리를 효과적으로 하기 위한 테스트 그룹과, 그룹별 세부 테스트 내용을 제안한다. 다음으로 대학 수업을 통해 포커스 그룹 테스트를 효 과적으로 운영하기 위한 운영 시스템을 제안한다. 마지막으로 개발 중인 상용 온라인 게임을 의뢰받아 포커스 그룹 테스트를 수행한 사례를 소개함으로서 제안한 내용이 효과적으로 운영될 수 있음을 검증한다. 제안한 방식의 수업 운 영을 통해 실무 지향적으로 수업이 가능하며 개발사와 대학 간의 효과적인 산학 협력이 가능할 것으로 판단된다

중간처우시설은 사회로 나아갈 범죄자들에게 일시적으로 주거의 문제를 해결해 주고 사회 적응력 향상과 재범예방 프로그램을 제공하여 사회복귀라는 현대 교정행정의 목적을 효과적으로 수행할 수 있는 시스템이다. 우리나라에서도 이와 같은 점을 인식하여 법무부 교정본부에서 안양교도소 구외에 사회내 중간 처우제도라고 할 수 있는 중간처우시설(‘소망의 집’)을 최초로 설립하였으며, 이를 4개 지방교정청별로 확대 운영하고 있으며 그 수요는 더 증가할 전망이다. 그런데 중간처우시설에서 획일적인 프로그램만을 제한적인 대상에게 일괄적으로 적용한다면 이는 수용자의 필요에 부합하지 않을 뿐만 아니라 예산과 인력의 낭비라는 비판을 면하기 어려울 것이다. 이에 따라 이 연구에서는 중간처우시설의 설립 여부에 대한 이전의 논의를 벗어나서 중간처우시설에서 어떠한 유형의 프로그램이 누구를 대상으로 실시되어야 가장 효과적일 수 있는가, 즉 수용자의 필요와 특성에 부합하는 중간처우시설 프로그램을 운영하기 위한 방안을 모색 하고자 하였다. 이를 위하여 미국과 캐나다에서 실시된 중간처우시설 프로그램 운영실태와 평가에 대한 경험적 연구를 통하여 시사점을 도출하고 이와 관련하여 수용자 선정을 위한 사정 도구의 개발, 프로그램의 다양화 및 개별화, 프로그램의 평가수단 개발 등의 방안을 제시하였다.

But nowadays, the needs of self management of these topics is rising up for the business continuity especially to the multi-national corporate. To prove their soundness they issued the management status report periodically. Internal audit will works not only the self confirming tool but also the best supporting tool for the policy managements in such circumstances. So, an effective internal audit will lead the business continuity management to the future.

건설업은 공사수행을 포함한 대부분의 기업 활동이 인력에 대한 의존도가 높은 산업으로서 원가구성 측면에서도 외주비와 재료비를 제외하고 가장 큰 원가항목이 인건비다. 따라서, 건설업의 인력관리 강화는 효율적인 인력 운용과 원가관리를 위해서도 가장 중요한 관리항목이다. 본 연구에서는 건설업에서 체계적인 인력 및 시간관리를 통해 인력운용의 효율을 높일 수 있고, 업무시간 실적 정보에 의해 업무에 대한 성과관리의 정확성을 높일 수 있는 제도운영 및 시간관리 시스템 (TMS: Time Management System) 구축 방안을 소개하였다. 이러한 시간관리는 높은 수준의 인력 및 업무 관리를 가능하게 하고 업무에 대한 우선순위 결정과 적정 시간 분석으로 관습적이고 비효율적인 업무요소를 파악하여 효율적인 인적자원 관리를 하고자하는 건설사뿐만 아니라 프로젝트 산업 군에 속하는 여러 기업들에게 도움을 줄 것으로 기대한다.

The most effective methods against pine wilt disease (Bursaphelenchus xylophilus, PWD), black pine bast scale (Matsucoccus thunbergianae), pine needle gall midge (Thecodiplosis japonensis) is the trunk injection of insecticides. The period of trunk injection that was considered with ecology and physiology of pests such as pine wilt disease, black pine bast scale and pine needle gall midge, was applied from Dec. to next Feb. and June, Dec. respectively. And there are differences in quantities of being injected chemicals to pine trees by the period of trunk injection. Thus, we investigated to prevent the effect of insecticides, to estimate the effect under the trunk injection of insecticides for pine trees against pests by the injecting period and time, boring site, opening and shutting of injection site. On Pinuns thunbergii, it was examined to figure out the effect of insecticides by the injecting period, twice a month on a sunny day around 10 AM from January to September. Injecting of insecticides was tested at a dose of 5ml per cm dbh of a pine tree after boring with hand drill at 50 cm high from the ground. As a result, abmectin 1.8% EC and phospamidon 50% SL were injected over 90% of mortalities from the end of January to the middle of March but as time goes by, injecting insecticides tended to decrease. To compare the volume of injecting insecticides in a day, between 10 AM and 2 PM on February 23rd, abamectin 1.8% EC was injected at a dose of 5ml per pine trees with micro pipette after boring with a drill machine at 50 cm high from the ground. As a result, injecting insecticide of a pine which was injected around 10 AM was completely injected and about 90% of the insecticide was injected in case of a pine that was injected around 2 PM. Trunk injection of insecticides was examined to figure out differences of the volume of injected insecticides before and after rain. There was no difference not only the rainfall but also the bored direction.

Woolly apple aphid, Eriosoma lanigerum, overwintered as adult or nymph stage on rootstocks, and crown- and root sucker in the soil. In an unsprayed apple orchard, the number of E. lanigerum colony started to increase from mid-April, showed the 1st peak between late June and early July, thereafter decreased followed by the 2nd peak in late July, and then again peaked in late September as the size in the 1st peak. In this orchard, the number of E. lanigerum colonies per tree did not exceed 3.5 colonies during the peak occurrence period, and was maintained around 2 colonies throughout seasons. In all seasons, parasitism of Aphelinus mali on E. lanigerum was much lower on root colonies than on aerial colonies that located on shoots and tree trunks above the ground. The parasitism of E. lanigerum was high in most orchards examined, showing parasitism of > 70% in maximum in most cases. In the laboratory bioassay for the mortality effects of several insecticides on E. lanigerum, fenitrothion, dichlorphos, machine oil, methidathion, thiacloprid, and imidacloprid showed 97.8, 96.8, 95.4, 91.5, 26.7, and 7.8% morality, respectively. Also, the adult emergence rates from A. mali mummies were 51.2, 72.6, 14.2, 3.5, 72.2, and 85.4% in the treatment of the above insecticides, respectively. Insecticides belong to neonicotinoid, which are newly developed to control aphids, showed low mortality against E. lanigerum. Fenitrothion and dichlorphos were effective on E. lanigerum control and had a low toxic to A. mali. Consequently, the insecticides should be useful in integrated pest management system for E. lanigerum in apple orchards.

본 연구는 PIT tag (passive integrated transponder)과 무선추적법(radio telemetry)을 오대산국립공원과 월악산국립공원에 서식하고 있는 파충류에 적용한 후, 이를 평가하여 효과적으로 국립공원 내 파충류 자원관리 방안을 모색하기 위하여 수행되었다. PIT tag은 현재까지 알려진 파충류 개체표시법 중에서 가장 효과적인 방법으로 2006년 3월부터 2008년 10월까지 오대산국립공원과 월악산국립공원의 조사지역에 출현한

The objective of this study was to evaluate the effective inoculation position of Neoseiulus californicus for control of Tetranychus urticae on apple branch. This study was conducted under green house conditions. N. californicus was inoculated at different position (Top, Middle and Bottom) of apple branch with a 20:1 ratio (T. urticae:N. californicus). Overall, N. californicus significantly reduced T. urticae numbers in the treatment branches than in the control branch. At 10th day, the T. urticae population was most significantly reduced in the treatment ‘Top’ in which N. californicus were inoculated on the leaf positioned in the top of a branch than in other treatments. At 20th day, most of T. urticae were exterminated in all treatment branches. The daily movement of N. californicus and T. urticae,and their coexistence on apple branch was monitored. Adult N. californicus disappeared from the branch as soon as T. urticae were exterminated in the treatment branches. The result indicates that N. californicus disperse downwards more than upwards in the tree, and the most effective inoculation position for N. californicus for control of T. urticae is the leaves positioned in the top of a branch.

It is hot issue to get the competitiveness of product through reliability-growth as well as availability and safety in almost high-technology industry including military, chemistry, nuclear, telecommunication, transportation service and so on. In advanced countries, they experienced remarkable growth based on reliability technology in materials&parts industry as well as high-tech industry. It has become a symbol of national competitiveness and created a higher value added. This paper is taking on effectively developing ways to deploy IEC 60300 dependability management system and the base technology that are needed to improve competitiveness of industries in Korea.

In reality, distribution planning for a supply chain is established using a certain probabilistic distribution estimated by forecasting. However, in general, the demands used for an actual distribution planning are of deterministic value, a single value for each of periods. Because of this reason the final result of a planning has to be a single value for each period. Unfortunately, it is very difficult to estimate a single value due to the inherent uncertainty in the probabilistic distribution of customer demand. The issue addressed in this paper is the selection of single demand value among of the distributed demand estimations for a period to be used in the distribution planning. This paper proposes an efficient demand selection scheme for minimizing total inventory costs while satisfying target service level under the various experimental conditions.

In this study we tried to construct a more efficient tetracycline-inducible gene expression system by replacing previous key elements with more advance ones. At the beginning, we substituted PGK (phophoglycerate kinase) promoter for CMV (cytomegalovirus) promoter to control “rtTA2sM2” which has been known for high induction efficiency in response to tetracycline. With this modification, expression of the EGFP marker gene under the induction condition was significantly increased. Next, we replaced “TRE” fragment with a modified version named “TRE- tight” which has been reported to have higher affinity and specificity to the transactivator by minor base change of the “TRE” DNA fragment sequence. Use of “TRE-tight” instead of “TRE” resulted in more than 10 fold increment in terms of induction efficiency and significant decrement of background expression in non-inducible condition. By combining PGK promoter and “TRE-tight” fragment, we could upgrade previous tetracycline-inducible system to show more stringent turn on/off gene switch ability and stronger expression of the gene of our interest. Use of this newly developed system must be very helpful to the studies of gene expression, especially to the transgenic animal study in which non-controllable constitutive expression of the transgene has been one of the urgent problems to be solved.

최근 선박건조기술이 발전했음에도 불구하고 교통량의 증가와 해양레저의 활성화로 사고위험은 더욱 증가하고 있다. 이 논문에서는 그동안 발생했던 해양사고를 분석하여 사고발생을 최소화할 수 있는 여러 가지 방안과 사고발생시 신속하고 구조할 수 있는 체계적인 구조시스템에 대하여 논의한다. 선박이 해상에서 안전하게 항행하기 위해서는 선박기술, 항해관련 장비, 우수한 인적자원, 교통관제 등과 같은 구성요소들이 제도적으로는 구축되어야 하고, 각각의 항행요소들이 유기적으로 작동되어야 한다. 또한 사고발생시 사고수습이 신속하게 진행될 수 있도록 체계적으로 정부조직이 구성되어야 하고 그 조직들이 긴밀한 협력관계를 유지해야 한다. 민간부분에서의 협력지원도 현재보다 더욱 강화되어 국가의 관리영역에서 벗어난 해역에서는 민간부분에서 구조할 수 있도록 민간 구조대가 활성화되어야 한다.
