Background : Boxthorn has been cultivated by conventional fertilization and irrigation methods that can be characterized by applying a great deal of fertilizers and a small amount of water compared to the other crops. With increasing rain shelter greenhouses, there were significant needs for developing new fertigation and irrigation methods. In this study, several prescribed fertigation methods for balanced fertilizer application and a few irrigation start methods based on soil moisture tension has been tested to confirm the fruit quality and productivity for Boxthorn growers. Methods and Results : In the experiment, ‘Cheong-un’ cultivar was grown in the rain shelter greenhouses for the tests. Plots were set by randomized block design. According to the growth investigation on the ‘Cheong-un’ cultivar which is supplied with four different formulated fertigation, the formulation N 15 me/ℓ increased the size and weight of fruit leading to 17% increased yield than the others. Additionally, on the amount of annual supply of fertigation with the concentration of N 15 me/ℓ, the treatment applied with 10kg/10a during the cultivation period facilitated overall growth of ‘Cheong-un’ cultivar and increased the yield as well. In the irrigation experiment ‘Cheong-un’ cultivar was grown in the four differently planned treatments that start irrigation at –20, -40, -50 and -100kPa soil moisture tensions. According to the investigation, the irrigation treatment starting at -50kPa enhanced the number of fruit bearing and productivity of Boxthorn consistently during two years. Conclusion : Therefore, the newly developed fertigation and irrigation methods could enhance the annual income of Boxthorn growers by increasing growth and yield.

This study was carried out to investigate the effect of sucrose, boric acid, and calcium nitrate in optimum pH and relative humidity on pollen germination and pollen tube growth of boxthorn (Lycium chinense M.). Medium containing 100mg/L of H3BO3 showed maximum pollen germination and pollen tube elongation, 64.8% and 920μm. Additionally supplied calcium nitrate to the medium lowered pollen germination rate and pollen tube elongation. Here we report the optimum conditions for the pollen germination and pollen tube development was observed at 10% sucrose, 1.0% agar, pH 6.0, and 100% relative humidity.

For the development of Boxthorn organic cultivation techniques, we investigated effect of several organic compost as a foundation fertilizer and growing plant fertilizer. And we adopted partly opening rain shelter greenhouse to protect anthracnose. In organic compost applying test yield characteristics of ‘Mixed organic compost’ treatment was the best but in betaine content measurement of dried fruit, ‘Mixed organic compost and nitrogen guano ’ treatment was the best and it’s chlorophyll and 100 fruit weight were also betfer than the other treatments.

This study was undertaken to develop a technique of discrimination using SSR makers in boxthorn cultivars.Forty one boxthorn cultivars, which were collected from Korea and China, were evaluated by 10 SSR markers. Total of 61alleles were detected, ranging from 3 to 13 with an average of 6.1 alleles per locus. The averages of gene diversity and PICvalues were 0.482 and 0.428, with a range from 0.25 (GB-LCM-022 and GB-LCM-087) to 0.83 (GB-LCM-167) and from0.24 (GB-LCM-022 and GB-LCM-087) to 0.81 (GB-LCM-167), respectively. Five markers out of 10 markers, GB-LCM-022, GB-LCM-075, GB-LCM-104, GB-LCM-167 and GB-LCM-217, were selected as key markers for discrimination inboxthorn cultivars. All of boxthorn cultivars were individually distinguished by the combination of five SSR markers.

This study was conducted to investigate the optimal plant growth regulator level for the shoot formation of Lycium chinense Mill. In vitro plant propagation was developed for leaf explants of boxthorn. Leaf explants were cultured on MS medium supplemented with cytokinins (BA and 2-iP) alone. Plants were successfully regenerated through in vitro culture by using leaf explants of boxthorn grown in the field. After 4 weeks of culture, 58% of shoot formation had developed from the leaf explants. The shoot formation rate of 'Jangmyeong' was highest followed by 'Myeongan', ;Cheondae', and 'Bullo'. The use of 0.2mg/L BA was critical for enhanced production of shoot formation and resulted in 58% of the culture producing shoot formations. Regenerated plantlets transplanted to pots were developed and successfully acclimatized to greenhouse.
