Ulcerative colitis (UC) is a chronic inflammatory intestinal disease characterized by an imbalance in immune function and the overexpression of inflammatory cytokines and mediators. Vitamin B2, also known as riboflavin (Libof), is an essential water-soluble vitamin with numerous beneficial properties, including antioxidant, anti-aging, anti-inflammatory, antinociceptive, and anti-cancer effects. In this study, we aimed to investigate the protective effects of Libof on dextran sulfate sodium (DSS)-induced experimental colitis. The C57BL/6 mice were used as the in vivo model of chronic colitis to investigate the anti-inflammatory effects of Libof. RAW 264.7 cells were used for the in vitro investigation of the molecular mechanisms underlying these effects. In vivo, Libof alleviated the DSS-induced disease activity index (DAI), colon length shortening, and colonic pathological damage. In vitro, Libof inhibited lipopolysaccharide (LPS)-induced tumor necrosis factor (TNF)-alpha and interleukin (IL)-6 production in RAW 264.7 cells. Moreover, Libof inhibited LPS-induced nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression in RAW 264.7 cells. In conclusion, these findings indicate that Libof shows potential as an agent for the treatment of UC.
The purpose of this study was to investigate the effect of Cheonggukjang pills with aronia and blueberries on dextran sulfate sodium (DSS)-induced colitis in mice. There have been several reports that Cheonggukjang is effective for intestinal health, but the efficacy of Cheonggukjang containing fruits has not yet been reported. In this study, we showed the effect of cheonggukjang pills with blueberries and aronia (CPBA) on DSS-induced colitis in BALB/c mice. CPBA was obtained from Soonchang Moonokae foods and orally administered once a day for 2 weeks before DSS treatment. Colitis was induced in mice by feeding 5% (w/v) DSS drinking water for 7 days. The results showed that CPBA treatment significantly alleviated DSS-induced disease activity index associated with a decrease in colon length. CPBA improved DSS-induced histological changes and intestinal epithelial barrier integrity in mice colon. In addition, CPBA administration significantly reduced the levels of DSS-mediated interferon-γ and interleukin-6 in serum and tumor necrosis factor-α in colon tissue. Moreover, the gene expression of COX-2 and iNOS, which are factors involved in inflammatory signaling, was significantly reduced by CPBA treatment. These results suggest that CPBA have a protective effect against DSS-induced mice colitis and may be a candidate for colitis treatment.
염증성 장질환은 장관 내 비정상적인 만성 염증이 호전과 재발을 반복하는 질환으로 아직까지 명확한 발병기전은 밝혀져 있지 않다. 그 중 궤양성 대장염은 염증 및 혈변, 설사 심한 경우에는 발열을 동반한다. 현재 치료제로는 설파살라진, 메살라민과 같은 항염 증제, 항생제 등을 사용하고 있지만, 부작용으로 두통, 빈혈 등의 있고 드물게는 간염, 폐렴 등이 유발되기 때문에 보다 안전한 방법이 요구되는 실정이다. 본 실험은 5주령의 수컷 마우스를 일주일간 적응기간 후, CON군 DSS군, 저농도군, 중농도군으로 나누었다. CON군을 제외한 모든 군에 DSS를 일주일간 투여해 장염을 유발한 후 2주간 WK-20을 투여한 후 검사를 한 결과 저농도, 중농도군에서 대장염의 증상이 유의적으로 완화된 것으로 보였다. 따라서, WK-20은 DSS를 통해 유발되는 대장염에서 치료적 효과를 보이지만 MTT 검사를 통해 농도의 조절이 필요한 것으로 사료된다.
Backgound : Inflammatory bowel diseases (IBDs) are chronic disorders that are characterized by intestinal epithelial inflammation and injury. Currently, the most employed therapies are antibiotics and anti-inflammatory drugs; however, the side effects limit long-term effectiveness.
Methods and Results : We evaluated the impact of glucose-lysine Maillard reaction products (Glc-Lys MRPs) on colitis, induced in rats by an administration of 5% dextran sulfate sodium (DSS) in drinking water. Glc-Lys MRPs ameliorate DSS-induced colitis, as determined by a decrease in disease. index activity, colon weight/length ratio, nitric oxide levels in serum, recovery of body weight loss, colon length and serum lysozyme levels. Furthermore, Glc-Lys MRPs increase the glutathione content and the activity of glutathione peroxidase, superoxide dismutase and catalase, and inhibit lipid peroxidation and myeloperoxidase activity in colon tissues. In particular, Glc-Lys MRPs suppress the mRNA level of the inflammatory cytokines and nuclear factor-κB in colon tissues.
Conclusion : This study suggests the potential of Glc-Lys MRPs in preventing or treating IBDs.
This study was conducted to investigate the anti-inflammatory effects of Pruns mume, Schisandra chinensis, Chaenomeles sinensis-- Prunus mume mixtrue (PM) treatment on colitis induced in mice by dextran sodium sulfate (DSS) treatment. A total of 25 male BALB/c mice (average weight 20.7 ± 1.6 g) were divided into 5 treatment groups and fed a commercial diet (A), PM administration (B), commercial diet + induced colitis by DSS (C), PM administration + induced colitis by DSS (D) and sulfasalazine + induced colitis by DSS (E). We found that PM treatment (D) and sulfasalazine (E) decreased the expression of TNF-α and COX-2 compared to the DSS-induced colitis group (C). The expression of IL-4, STAT6, IFN-γ, STAT1 was decreased in group D and group E compared to the colitis group (C), COX-2 and STAT1 were more decreased in group D. The serum IgE levels decreased in the PM treatment groups (C and D) compared to the non-PM treatment groups (A and B) although there was no significant difference between the PM treatment groups. It is notable that a therapeutic application of the PM extracts ameliorated DSS-induced colitis in mice.
