아토피 피부염은 만성 염증을 동반한 알레르기성 질환 이며, 일반적으로 Type 2 염증 반응과 관련이 깊다고 알 려져 있다. Corchorus olitorius (C. olitorius)의 아토피 피 부염 치료 효과를 확인하기 위하여 2,4-dinitrochlorbenzene 로 아토피 피부염 모델을 유도한 BALB/c 마우스를 대상 으로 피부 염증 개선도를 육안으로 확인하였으며, 질환 관 련 IgE와 사이토카인(IL-4, IFN-γ and TNF-α)의 수준을 측정 하였다. 7일간의 투여기간동안 양성대조약물인 Dexamethasone 투여군의 체중은 감소하였던 반면에 모든 C. olitorius 투 여군의 체중은 꾸준히 증가하였다. 이와 같은 결과는 C. olitorius이 동물에 위해한 영향이 없었다는 것을 의미한다. C. olitorius 200 mg/kg/day 투여군의 치료 효과가 가장 뛰 어났으며, 모든 C. olitorius 투여군의 염증 관련 면역글로 불린과 사이토카인의 수준은 감소하였다. 따라서, C. olitorius 는 아토피 치료제로 충분히 활용될 수 있다고 판단된다.
The occurrence of allergic disease has increased harmfully in the last few decades. Atopic dermatitis (AD) is an allergic inflammation disorder characterized by itchy, red, swollen, cracked skin. Although the pathogenesis of AD is not fully understood, it is assumed that deregulation of T-helper 1 (Th1) and T-helper 2 (Th2) immune responses, a predominance of allergen-specific IgE, and interrupted epidermal barrier function are keys to the pathogenic mechanism. Activated T helper 2 (Th2) immune function is hallmark of various allergic diseases. Oxidative stress implicated in cutaneous damage in various inflammatory skin diseases.
We investigated the effect of fermented soybean (SCGB1) on the improvement of AD. Soybean fermentation was carried out using B. amyloliquefaciense SCGB1 (KCCM11964P), which is known to produce of natural antibiotics. And then, we experiment of SCGB1 and soybean powder (NC) in DNCB-induced AD model. Mice were respectively oral administration of variety dose for 14 days. As a results, it was confirmed that serum Immunoglobulin E (IgE) expression was dose-dependently decreased in SCGB1 and NC compared to negative control, and it was reduced the skin pruritus inducing factor that Interleukin-31 (IL-31) mRNA level. In addition, the inflammatory cells were infiltration in skin for histological analysis. As a result, it reduced that epidermal hyperplasia, cancellation and aveolarization compared to negative control. These results suggest that SCGB1 may be effective for prevention and treatment of AD.
아토피성 피부염은 만성 재발성 염증성 피부질환으로 피부장벽기능의 이상과 환경유발인자에 대한 피부과민성과 연관되어 있다. 이전의 연구에서는 아토피성피부염에 효과적인 약용식물 추출물을 발굴하기 위하여 노회, 자회지정, 석류, 석곡 추출물들의 세포독성, 항산화, 항염, 항알레르기 효과를 검토하였다. 본 연구에서는 지질다당류로 활성화시킨 대식세포 RAW26.7 에 대한 약용식물 추출물들의 항염작용을 보다 상세하게 검토하여 항염작용의 근본적인 분자기전을 확인하고자 하였다. 역전사중합효소연쇄반응분석(reverse transcription polymerase chain reaction analysis) 결과, 석류, 석곡, 노회는 염증성 사이토카인인 IL-6 와 IL-1β 유전자발현을 현저하게 억제시켰으며 자화지정은 영향이 없었다. 형질주입과 발광효소분석(transfection and luciferase analysis) 결과, 약용식물 모두가 전사 핵인자 카파비(NF-kB)의 활성화를 억제시켰다. 웨스턴 블럿 분석(western blot analysis) 결과, 노회는 JNK MAP 인산화효소의 활성화를 차단하였지만 p38 MAP 인산화효소의 활성화는 차단하지 못하였다. 반면에 자화지정, 석류, 석곡은 JNK MAP 인산화효소뿐만 아니라 p38 MAP 인산화효소의 활성화도 차단하였다. 이들 실험결과들은 노회, 자화지정, 석류, 석곡은 항염효능을 가지고 있으며 따라서 아토피성 피부염의 증상을 경감 또는 완화시키는 잠재력이 있음을 보여 준다.
Atopic dermatitis is a chronic inflammatory skin disease associated with cutaneous hyper-reactivity to environmental triggers. In order to develop effective therapeutic herbal extracts for atopic dermatitis, cytotoxicity, antioxidant, anti-inflammatory and anti-allergic activities were investigated for various herbal extracts. Among candidate extracts, we selected Aloe vera L. (AV), Viola mandshurica W. Becker (VM), Punica granatum L. (PG), Dendrobium nobile L. (DN) and mixture of the above extracts (MX) for further investigations. All of them did not show cytotoxic activities to macrophage RAW264.7 cells below the concentration of 100 ppm. All showed antioxidant, anti-inflammatory and anti-allergic effects, although to various extents. In antioxidant effects, AV showed the highest effect, followed by PG and VM, while DN did the lowest. In evaluation for anti-inflammatory activities in macrophage RAW264.7 cells, AV and DN inhibited almost completely the LPS-induced production of nitric oxide, while AV, DN and VM showed strong inhibitory activities on the LPS-induced production of TNF-α. In anti-allergy effect in mast cell HMC-1, DN showed the highest effect, followed by AV and PG, while VM did the lowest. In the topical allergy reaction induced by compound 48/80 in Sprague-Dawley rat, DN exhibited significant anti-allergic effect, while PG, VM and AV did slight effect. These results suggest that AV, VM, PG and DN have antioxidant, anti-inflammatory and anti-allergic activities, and thus have the potential to reduce and alleviate the symptoms of atopic dermatitis.
Background : Atopic dermatitis (AD) is a common allergic, inflammatory skin disease. Here, we focused on the anti-allergic and anti-inflammatory effects of Gardenia jasminoides Ellis fructus extracts (GFE) in DNCB treated NC/Nga mice.
Methods and Results : In an in vivo study, GFE treatment significantly decreased immunoglobulin E (IgE) levels and cytokines expression in spleen and serum of treatment groups mice, compared with the negative controls (administered 0.2% DNCB only). Mice treated with GFE had fewer inflammatory cell infiltrates in the dermis and hypodermis compared to the negative controls. In addition, we also studied β-Hexosaminidase release assay and possible anti-inflammatory mechanism of GFE in RBL-2H3 cells. GFE suppressed the expression of COX-2 and TNF-α, the translocation of NF-κβ, and the phosphorylation of Syk, p38, JNK, and Erk1/2 in stimulated RBL-2H3 cells.
Conclusion : These results strongly suggest that GFE inhibits the allergic response by reducing mast cell activation and may have therapeutic potential as an anti-atopic dermatitis agent.
