Lactobacillus is a probiotics that improves various diseases and enhances immunity through the activation of cytokines involved in natural immunity. A variety of studies on immunoregulation have been conducted on various Lactobacillus, but few on Lactobacillus johnsonii (Lj) and especially on immune cells. In this study, we investigated how Lj affects the viability and function of RAW264.7 macrophages. The results indicated that Lj decreased the metabolic activity of RAW 264.7 cells and caused a decrease in the number of cells. In Annexin V and PI staining, the viable cells was significantly decreased at 107 cfu/mL Lj concentration. Measurement of cytokines using enzyme-linked immunosorbent assay (ELISA) showed that Lj increased the production of tumor necrosis factor (TNF)-alpha. Also, Lj increased the expression of CD40 and CD86, confirmed by flow cytometry analysis. Taken together, Lj caused decreased metabolic activity and cell viability, but simultaneously increased the expression of macrophage activation markers and TNF-alpha production. Therefore, when an appropriate dosage of Lj is administered, it is expected that the necessary immunomodulation can be increased while minimizing damage on cells. This study provided evidences for the immunomodulatory effects of Lj on RAW 264.7 cells in vitro and is expected to be a valuable information for Lj research.

This study aimed to address the time, cost, and ethical issues associated with traditional animal experiment-based observational methods by utilizing in silico Physiologically Based Pharmacokinetic modeling to predict veterinary drug residues in livestock products and validate them against observational data. Using PK-sim software, we modeled the physiological conditions of pigs to predict the depletion of ceftiofur and spiramycin. We evaluated the ceftiofur (3 mg, 6 mg) and spiramycin models by comparing them with observational data using residuals, MSE, and R-squared values. Specifically, the R-squared values for the ceftiofur models were all negative, indicating poor predictive power. For Ceftiofur (3 mg), the R-squared value was <0 with MSE of 611.3764, and for Ceftiofur (6 mg), it was <0 with MSE of 2447.982, highlighting significant discrepancies. Similar shortcomings were observed in the spiramycin models, with an R-squared value of <0 . These discrepancies can be attributed to inaccuracies in literature data, limited physicochemical data, inadequate consideration of inter-individual differences, mismatches between experimental and model conditions, and limitations of benchmark observational experiments. This underscores the critical importance of enhancing data quality and refining modeling approaches. Future research should focus on validating in silico techniques across diverse animal models and drugs to broaden their applicability in safety assessments. Ultimately, leveraging in silico techniques is crucial for establishing a scientifically robust safety management system for livestock products, overcoming the constraints of current observational experimental methods.

The Japanese encephalitis virus (JEV) is a zoonotic pathogen that affects the nervous systems of humans, pigs, and horses. It has been classified into five genotypes (G1-G5) based on molecular analysis of the pre-membrane or envelope gene. In the Republic of Korea, the predominant JEV genotype has recently shifted from G3 to G1 and G5, highlighting the need for a rapid and accurate diagnostic method. In this study, we designed specific common and differential primer sets for JEV G1, G3, and G5 to detect the JEV gene. Four specific primer sets for JEV G1, G3, and G5 were used to selectively amplify the target gene. The detection limits of the common primer set for JEV G1, G3, and G5 were 100, 0.1, and 10 TCID50/reaction, respectively. The detection limits of the three differential primer sets were 1, 0.1, and 1 TCID50/reaction, respectively. No cross-reactivity was observed with non-JEV reference viruses. We successfully developed a multiplex reverse transcription polymerase chain reaction (RT-PCR) assay to distinguish the three JEV genotypes. Our multiplex RT-PCR assay is highly sensitive and specific, providing a reliable tool for confirming JEV infection in suspected samples. Additionally, our assay can be applied to suspected mosquito samples and commercial veterinary biological products.

Because intact FMDV particles (146S) are often unstable in vitro, stabilizing foot-and-mouth disease virus (FMDV) antigens remains a key challenge in studying viral charateristics. Therefore, finding optimal condition to stabilize the FMDV is essential. In this study, we investigated formulations and potentials of several stabilizers such as appropriate buffer, excipients, and storage conditions to enhance the stability of 146S. Inactivated FMDV O-Jincheon (O-JC) was dissolved in various buffer formulations, and stored at 4℃ for two months to evaluate quantity of 146S at every 2-week interval. Among phosphate buffered saline (PBS), Tris buffered saline (TBS), HEPES buffered saline (HBS), and MOPS buffered saline (MBS), PBS showed more effective 146S stabilization that showed 1.3-1.6 fold higher 146S fraction than TBS, HBS, and MBS after storage for 2 weeks. However, constant dissociations of 146S were observed in all formulations at 8 weeks. Compared with other FMDVs, A22 Iraq and SAT-1, in PBS, O-JC proved to be the least stable in PBS. A variety of excipients including carbohydrate, sugar alcohol, cryo-protectant were tested for the capability in protecting O-JC from dissociation. By adding 4-8% sucrose, more than 60% of 146S fractions were maintained at 8 weeks, those were at least 1.8 fold higher than the PBS-only control. Addition of 1% β-cyclodextrin showed synergistic enhancement in O-JC stability. As the results of this study, it could be suggested that the PBS-based buffer together with 4-8% sucrose + 2% sorbitol or 2% sucrose + 2% sorbitol + 1% β-cyclodextrin could help the better stability of the O-JC in vaccine preparation.

This study investigates the kinetic compensatory gait changes in a small-sized dog (4.2 kg, 2-year-old spayed female poodle) with experimentally induced lameness. Reversible lameness was induced by applying silicone pads to the dog's forelimbs and hindlimbs. A force plate analysis system was used to measure Peak Vertical Force (PVF) and Symmetry Index (SI) during normal and induced lameness conditions. The results showed significant reductions in PVF in the induced limbs. Specifically, the left forelimb’s PVF decreased from 139.00 ± 9.85% in normal gait to 88.00 ± 19.05% after lameness induction, and the right forelimb’s PVF decreased from 130.33 ± 5.51% to 78.00 ± 18.52%. In contrast, compensatory increases were observed in the contralateral limbs, with the PVF of the contralateral forelimb increasing to 125.33 ± 1.15%. Similar patterns were observed in the hindlimbs, although the changes were less pronounced. The Symmetry Index (SI) values also increased in the induced limbs, particularly in the forelimbs (ILF: 53.10 ± 22.85%, IRF: 72.17 ± 15.08%), indicating greater asymmetry. These results suggest that forelimb lameness in small dogs results in more significant compensatory gait changes than hindlimb lameness.

Baicalin, a flavonoid isolated from Scutellaria baicalensis, has anti-inflammatory, antioxidant, and neuroprotective effects. Glutamate is a major neurotransmitter that plays an important role in brain function, but excessive release of glutamate causes excitotoxicity and damages cells. We investigated the neuroprotective effects of baicalin in glutamate-exposed neurons. The mouse hippocampal neuronal cell line (HT22) was cultured in a general manner, glutamate and/or baicalin were treated on the cells. Baicalin was administered 1 hr before glutamate treatment. Cells were collected 24 hr after glutamate, and cell viability was measured using MTT assay. Reactive oxygen species (ROS) and lipid peroxidation (LPO) assays were performed to measure oxidative stress. Glutamate reduced cell viability in a dose- and time-dependent manner. MTT assay showed that baicalin treatment ameliorated the decrease in cell viability due to glutamate toxicity. The effect of baicalin is dose-dependent. Glutamate caused severe nerve damage, including condensation of the cell shape, loss of dendrites and axons. However, baicalin treatment attenuated these morphological changes, and the effect of baicalin was dose-dependent. ROS and LPO analyses showed that glutamate increases oxidative stress, and baicalin attenuates this change due to glutamate toxicity. The effect of baicalin on these results was dose-dependent. We confirmed that baicalin performs an antioxidant function against glutamate toxicity in neurons. In conclusion, these results suggest that baicalin exerts neuroprotective effects on damaged neurons through antioxidant activity.

Afoxolaner is an insecticide and acaricide that belongs to the isoxazoline chemical compound group. it has been used as an active pharmaceutical ingrdient in veterinary medicine to treat fleas and ticks in dogs. When patents expire between 2026 and 2066, it is expected that many products will be applied for approval as generic products, and reserch to establish accurate quality control methods must be conducted and managed. HPLC method was developed for the quantitative and qualitative of afoxolaner in veterinary medicinal products. The separation of active constituents for afoxolaner was achieved on a RP18 (4.6 x 150 mm, 5 μm) column using Water : Acetonitrile : MeOH (25:30:45 v/v/v) as mobile phase, with UV detection at 245 nm. The method was validated for specificity, linearity, accuracy and precision. All calibration curves showed good linearity (R2 of 0.999) within the concentration ranges (12.5 to 400 μg/mL). For accuracy, the recovery rate was calculated by spiking three concentrations of standard into the sample blank. The recovery rate was calculated to be 99.70~100.58%. Precision was measured 9 times repeatedly through intra-day, inter-day tests using standard. It showed excellent precision by satisfying the relative standard deviation of less than 2% both intra-day and inter-day. Limit of detection (LOD) and limit of quantitation (LOQ) were 2.0 μg/mL and 6.1 μg/mL, respectively. This method was successfully applied to analyzing afoxolaner drugs distributed in Korea. The HPLC method described in this study is accurate and reproducible and could be applied for the analysis of veterinary drugs of afoxolaner.

For centuries, humans have leveraged the health-promoting properties of plants for our well-being. While research has been conducted on numerous medicinal plants, the specific benefits of many species remain underexplored. Eupatorium Japonicum (EJ), a member of the Asteraceae family, has historically been consumed in Japan, South Korea, China, and Vietnam for its traditional use in soothing digestive issues. This study aimed to explore the radical scavenging and antiinflammatory efficacy of EJ extract using RAW 264.7 cells. The radical-scavenging effects were assessed using the DPPH and ABTS assays, where an anti-oxidative molecule in the test sample will react with a stable free radical in DPPH and ABTS causing discoloration. The anti-inflammatory efficacy was assessed using the nitric oxide (NO) assay in LPS-induced RAW 264.7 cells, where the amount of NO produced in response to infection was measured using Griess reagent. Reversetranscriptase polymerase chain reaction (RT-PCR) and real-time PCR were executed to confirm the anti-inflammatory activity by measuring the RNA levels of pro-inflammatory cytokines. The DPPH and ABTS assays revealed that EJ extract decreased oxidation in a concentration-dependent manner (7.8-1,000 μg/mL) compared to ascorbic acid and Trolox respectively. EJ extract significantly reduced NO production concentration independently. Furthermore, EJ extract showed no cytotoxic effects as determined through the MTT assay. RT-PCR and real-time PCR analyses revealed inhibition of mRNA expression of pro-inflammatory cytokines (iNOS, COX-2, TNF-α, and IL-6). Western blotting demonstrated EJ’s anti-inflammatory activity by reducing protein levels of iNOS, COX-2, TNF-α, and IL-6. These findings suggest that EJ extract exhibits anti-inflammatory activities and can be further evaluated in the future.

Clove (Syzygium aromaticum) is a highly valued medicinal plant native to Aisa. Widely used as a spice, renowned for its medicinal properties, particularly in Ayurveda and traditional Chinese medicine. In this study, clove bud extract (CBE) was prepared at different ethanol concentrations of 50%, 80%, and 90%, respectively. The antioxidant activity of the CBE was evaluated through DPPH, polyphenol, and reducing power assays, revealing its strong antioxidant potential, with 90% ethanol being the most effective extract. HPLC analysis identified eugenol (8.7 mg/g) as the major active compound, known to possess anti-inflammatory and antioxidant properties. Given the role of oxidative stress and inflammation in atopic dermatitis (AD), the therapeutic potential of CBE was explored using a 1-chloro-2, 4-dinitrobenzene (DNCB)-induced AD mouse model. Five-week-old BALB/c mice were induced with AD by topical application of DNCB. CBE was administered topically to the affected skin (back and ear) areas for 4 weeks. The treatment of CBE significantly reduced the severity of clinical dermatitis, decreased epidermal thickness, and lowered mast cell and eosinophil infiltration in skin tissue, as observed through hematoxylin eosin staining and toluidine blue staining. The results demonstrated CBE as a promising therapeutic agent for managing AD through its regulation of skin inflammation and oxidative stress, making it a potential candidate for future treatments of inflammatory skin disorders.

In this study, an simultaneous LC-MS/MS multi-residue analytical method was developed and validated for the residues of six neonicotinoid insecticides (acetamiprid, clothianidin, dinotefuran, imidacloprid, thiacloprid, and thiamethoxam) in honey. Sample preparation included a combination of QuEChERS extraction kit and liquid-liquid extraction method to effectively extract pesticide components from the honey matrix and optimized analytical conditions to achieve high sensitivity and selectivity. The limits of detection (LOD) and the limits of quantitation (LOQ) were set in the range of 6-15 ng/mL and 19-44 ng/mL, respectively and the correlation coefficient (R²) was greater than 0.99, confirming good linearity. In addition, the intra-day recoveries for each pesticide were 75-104%, and the coefficient of variation (CV) was less than 20%, which met the guideline recommended by the Ministry of Food and Drug Safety. The LC-MS/MS method developed in this study is expected to be used as a multi-residue analysis method for 6 neonicotinoid pesticides in honey.

Probiotics have been evaluated as therapeutic agents for cancer treatment in an increasing number of studies. This study investigated the inhibitory and cytotoxic effects of specific Lactobacillus strains on a human colorectal adenocarcinoma cell line (HT-29). The strains assessed were Limosilactobacillus (L.) reuteri VA 102, Ligilactobacillus (L.) animalis VA 105, and Limosilactobacillus (L.) reuteri KCTC 3594 (ATCC 23272). The viability of HT-29 cells was evaluated using the MTT assay. The findings revealed that cell-free supernatants (CFS) exhibited significant anticancer effects. Heat-inactivated L. reuteri VA 105 and L. reuteri KCTC 3594 induced a pronounced reduction in cell viability. Furthermore, live cultures of L. reuteri VA 105 and L. reuteri VA 102 also showed reduced cell viability compared to the control group. These results suggest that CFS and heat-inactivated cells may be more suitable for therapeutic applications than live bacteria owing to their improved safety profiles and reduced potential for adverse effects. Our findings also emphasize the potential anticancer benefits of these LAB strains.

Malaria remains a significant public health issue, particularly in regions such as the Korean Demilitarized Zone (DMZ). Effective malaria control and prevention require precise prediction of mosquito density across both monitored and unmonitored areas. This study aimed to develop predictive models to estimate the abundance of malaria vector mosquitoes by integrating meteorological and geographical data. Data from mosquito surveillance sites and NASA MODIS land cover datasets acquired between 2009 and 2022 were utilized. Two predictive models, the Gradient Boosted Model (GBM) and Principal Component Regression (PCR), were employed and evaluated. Model performance was assessed using the coefficient of determination (R²). Results showed that PCR outperformed GBM in predictive accuracy, suggesting that PCR is more robust in handling multicollinearity among variables. However, both models did not show practically-usable level of prediction performance. This study provides a preliminary but foundational framework for extending predictive modeling to broader regions, thereby supporting malaria prevention efforts through improved risk mapping.

Moringa oleifera, a versatile plant, has been traditionally used to treat various ailments and is gaining scientific attention due to its potential as a medicine. Native to the Indian subcontinent, it is widely grown in tropical and subtropical regions, thriving in Asia, Africa, and South America, especially in arid climates. This study explores the antioxidant potential of Moringa oleifera leaf extract (MOLE), employing a comprehensive screening approach with various solvents to identify the most effective extraction method. Initial experiments assessed antioxidant efficacy and yield using distilled water (D.W.), 95% ethanol, and 95% methanol. Among these, 95% ethanol extract demonstrated superior antioxidant activity, confirmed through assays such as 2,2-diphenyl-1-14 picrylhydrazyl (DPPH) radical scavenging assay, total polyphenol content analysis, and reducing power assay. In addition, with the 95% ethanol MOLE, a higher extraction efficiency was yielded compared to other solvents, making it the most effective for large-scale preparation. HPLC analysis revealed the presence of key bioactive compounds, including ellagic acid, rutin, Q-3-O, quercetin, and kaempferol. Results revealed that MOLE, prepared using 95% ethanol, exhibited remarkable antioxidant properties, attributed to its rich polyphenolic content. This research underscores the therapeutic potential of MOLE as a natural antioxidant source and highlights the importance of solvent optimization in phytochemical extractions.

An 8-month-old, neutered male, Korean short hair cat was referred for a closed locked jaw. Physical examination and computed tomography(CT) revealed an old trauma fracture with the absence of synovium at the left temporomandibular joint suggestive of left temporomandibular joint ankylosis. Gap arthroplasty successfully eliminated clinical signs, aided by the 3D printed model of the patient’s skull and piezosurgical unit. The major complication, temporal ipsilateral facial paralysis and reankylosis, were auspiciously managed through constant ocular lubrication and early rehabilitation.

This case report discusses a 15-year-old female poodle with multifocal degenerative myelopathy, who achieved significant clinical improvement through intensive acupuncture and rehabilitation therapy. The patient presented with ataxia in both hind limbs, and imaging studies revealed multiple regions of disc protrusion. Rather than surgical intervention, conservative treatment was chosen, which included acupuncture, medication, and laser therapy. Following treatment, notable improvements in neurological symptoms and pain reduction were observed. This case highlights the potential effectiveness of non-surgical approaches in managing degenerative myelopathy.

A red-crowned crane (Grus japonensis) from Seoul Zoo died with mild anorexia and sudden respiratory distress. A white globoid mass in the abdominal cavity and multifocal nodules in the kidneys were identified on the necropsy. Histologic examination showed anaplastic, solid patterned neoplastic cells in the kidneys and the mass. Immunohistochemistry for pancytokeratin and PAX8 was positive, while heterogeneous antigen expression was present in poorly differentiated neoplastic cells. Overall, the neoplastic changes in the kidneys and the mass were consistent with a renal origin. This case highlights the possibility of further discoveries of metastatic renal cell carcinomas in cranes.

Ectopic ureter refers to a congenital anomaly in which one or both ureters do not connect to the urinary bladder at the correct anatomical site. This case report discusses the case of a 6-year-old female mixed-breed dog diagnosed with chronic urinary incontinence, systemic hypertension, pancreatitis, and sepsis resulting from an ectopic ureter. Treatment involved an initial nephro-ureterectomy to address severe pyonephrosis, followed by ureteroneocystostomy for the remaining functional kidney. Post-surgical outcomes showed notable improvements in clinical symptoms, laboratory findings, and blood pressure. This report emphasizes the need for early diagnosis and appropriate surgical treatment in cases of ectopic ureter. Additionally, it aims to present the clinical symptoms and conditions resulting from prolonged disease progression, as well as the corresponding treatment methods and prognosis.
