Brucellosis is an important and re-emerging zoonotic disease worldwide. The prevention of human infection is achieved predominantly through the control of brucellosis in agricultural animals, which in turn depends on accurate diagnosis and vaccination. However, conventional serological diagnosis of brucellosis has several limitations, and currently available vaccines for animals have several drawbacks, including the ability to cause infection in humans. Phosphoglycerate kinase (Pgk) is one of the specific proteins reactive with mouse sera in the early stage of Brucella infection, and deletion of the pgk gene in B. abortus strain 2308 resulted in extreme attenuation of this strain in vitro and in vivo. Furthermore, the B. abortus pgk mutant has been used as a live vaccine, and in challenge experiments, it induced protection that was superior to that conferred by commercial strains. In this study, the pgk gene from Brucella abortus 544 was successfully amplified and cloned into a maltose binding protein fusion protein expression vector (pMAL). The recombinant protein was expressed in Escherichia coli DH5α and purified. The immunogenicity of purified recombinant B. abortus 544 Pgk (rPgk) was evaluated by western blot analysis using Brucella-positive mouse sera. rPgk could be used as an antigenic component for future serological tests and potential vaccine development.

The average prevalence rate (PR) for foodborne disease outbreaks (FBDOs) in the first half (F-H) and latter half (L-H) of the decade from 2001 to 2010 in Korea was 14.0 and 16.9 per 100,000 population, respectively. The number of patients per outbreak of foodborne diseases (FBDs) in the F-H of that decade was 57.6 and in the L-H was 25.4 (p < 0.05). A comparison of the 2 periods covered in this study indicates that FBDOs in the L-H most frequently involved restaurants or delis (51.5% of total cases), and in the F-H, this involvement was noted in 34.0% of total cases (p < 0.01). The epidemic patterns of microbial FBDOs show that the overall incidence of outbreaks resulting from 5 of 8 key pathogens including Bacillus cereus (+1.6%), Campylobacter jejuni (+1.5%), Clostridium spp. (+0.7%), pathogenic Escherichia coli (+8.3%), and Norovirus (+14.4%) tended to be higher in the L-H than in the F-H of the decade from 2001 to 2010. Conversely, those caused by the other 3 key agents, including Salmonella spp. ( 9.2%), Staphylococcus aureus ( 2.8%), and Vibrio parahaemolyticus ( 6.4%) were significantly lower in the L-H than in the F-H of the decade. Moreover, in the decade between 2001 and 2010, the number of patients (n) and the PR for microbial FBD increased from the F-H (n = 13,346, PR: 5.6) to the L-H (n = 33,732, PR: 13.8) (p < 0.01).

Salmonellosis is a significant public health concern and a major cause of bacterial foodborne poisoning in Korea. Approximately 95% of salmonellosis cases in humans are related to the intake of contaminated food products, which mostly originate from livestock such as poultry, pigs, and cattle. To successfully develop strategies to reduce the spread of Salmonella, it is important to investigate the prevalence of Salmonella spp. isolated from food animals and meat. In Korea, the prevalence of Salmonella spp. in cattle was 1.2 2.0%, whereas in pigs it was 1.5 21%. Of all livestock and meat products analyzed, the Salmonella isolation rate was the highest in chickens (18.8 30.3%) and their meat. Antibiotic resistance is an important social issue. The frequent use of antibiotics in the livestock industry has led to increased antibiotic resistance in Salmonella isolates that are extracted from livestock, which raises serious public health concerns. To prevent the spread of antibiotic resistance, it is essential to collect data related to antibiotic-resistant Salmonella strains isolated from food animals and meat. Antibiotic resistance tests revealed that Salmonella isolated from cattle, pigs, and poultry were highly resistant to tetracycline and streptomycin. Recent studies show that the overuse of antibiotics in food animals can also affect antibiotic resistance in humans. Therefore, it is important to establish an appropriate management strategy to successfully prevent misuse of antibiotics within the Korean livestock industry.
