The forty-nine cases of psittacine birds were submitted for disease diagnosis to the Avian Disease Division of the Animal and Plant Quarantine Agency (APQA) of Korea from 2012 to 2021. On the basis of clinical manifestations and the presence of gross lesions, necropsy, bacteriological culture, virus detection and histopathological examination were performed according to an APQA diagnostic protocol. Bacterial diseases like chlamydiosis, colibacilosis, viral disease like proventricular dilatation disease, budgerigar fledgling disease (BFD), psittacine beak and feather disease and fungal infection and parasitic diseases like blackhead disease and coccidiosis were confirmed in forty-nine cases. We present here five cases report diagnosed fungal pneumonia, blackhead disease, BFD-fungal infection, BFD and psittacosis-bacterial enteritis in psittacine. Since the pathogen can be detected even in healthy parrots, histopathological diagnosis is necessary to determine the cause of death, and clinical symptoms and gross lesions must be fully considered. In addition, zoos and aviaries where large quantities of parrots are reared, these diseases can be reduced through sanitary management of the breeding environment.

Foot-and-mouth disease (FMD) is a highly contagious vesicular disease that affects cloven-hoofed animals, causing substantial economic losses to the livestock industry. The causative FMD virus (FMDV) comprises four structural proteins (VP1, VP2, VP3, and VP4) and several non-structural proteins. Among the capsid proteins, VP4 is the most conserved, making it an attractive target as a diagnostic and vaccine antigen, regardless of FMDV serotype. In this study, we attempted to express the VP4 protein N-terminally fused to a glutathione S-transferase (GST) tag in Escherichia coli. Whereas VP0 and VP2 proteins were expressed in the soluble fraction, we failed to detect VP4, even in the insoluble fraction. To investigate the effect of VP4 C-terminal amino acid residues on protein expression, we constructed three VP4 mutants fused to GST, among which the mutant in which the C-terminal 15 amino acid residues had been deleted showed the highest level of protein expression. Furthermore, protein expression was observed even in the mutant in which three amino acid residues (DKK) had been fused to the C terminus. However, unlike the other two mutants, the wild-type VP4 mutant was poorly expressed, thereby indicating that the C-terminal amino acid residues could play a pivotal role in determining expression of the VP4 protein in E. coli.

This study is intended to determine the antimicrobial resistance properties and class 1 integrons of 30 Citrobacter freundii strains isolated from pet turtles in order to determine their threat to human health. Citrobacter freundii isolates were isolated and identified by employing biochemical tests and 16SrRNA gene sequencing. Disc diffusion test and PCR amplification were employed to detect antimicrobial resistance patterns and genes, respectively. Ampicillin, amoxicillin, cephalothin and nalidixic acid resistance were observed among all isolates. Rifampicin and cefoxitin resistance was the second most prevalent and observed in 97% of the isolates, respectively. All isolates scored multiple antimicrobial resistance (MAR) indices ≥ 0.2. Aminoglycoside resistant genes were the most prevalent. aac(3')-IIa was detected in 80% of the isolates followed by aphAI-IAB and strA-strB genes in 33% and 50% of the isolates, respectively. β-lactamase encoding blaTEM, blaCTX and blaSHV genes were detected in 53%, 43% and 17% of the isolates, respectively. tetA and tetB genes were the only tetracycline resistance genes detected in 17% and 10% of the isolates, respectively. Class 1 integron integrase encoding intI1 gene (47%) and dfrA17-aadA5 gene cassette array (7%) were also detected. Pet turtle-borne multidrug-resistant C. freundii is a reservoir of antimicrobial resistance determinants in the domestic environment which poses a risk of infection.

This study aimed to investigate whether bacterial ghosts (BGs), empty cell envelopes of a gram-negative bacterium, delivering envelope protein domain III (EDIII) of dengue virus (DENV) serotype 2 could induce protective immune responses against dengue infection. In this study, we constructed Salmonella Typhimurium BGs expressing and delivering EDIII (BG-EDIII) and evaluated these ghosts for their immunogenicity studies in C57BL/6 mice. Our results demonstrated that the mice vaccinated once orally with BG-EDIII followed by an intramuscular boosting with a recombinant EDIII protein elicited significantly higher humoral and cell-mediated immune responses compared to the BGs alone vaccinated group (p<0.001). Upon challenge with DENV2, significantly lower viral load and liver damage was observed in BG-EDIII vaccinated group than BGs alone control group (p<0.05). The outcomes of this study revealed the ability of BG- EDIII to stimulate immune response with no observable damage to the vital organs.

This study aims to establish a modified analytical method with sensitivity and reliability for streptomycin (STP) and dihydrostreptomycin (DHS) of residues level in pig tissues, plasma and urine by LC-MS/MS on the basis of previous studies. The mass parameters of quantitative and qualitative ions for STP and DHS were optimized using multiple reaction monitoring in positive mode. The separation of compounds was conducted using BEH Amide column according to material’s characteristics. The analytes in plasma were extracted with only organic solvents. In muscle and kidney, KH2PO4 buffer solution containing 2% CCl3COOH and EDTA-Na was used as extraction solvent. The WCX cartridege was selected as SPE cartridge in considering high recoveries for STP and DHS. The analytes in urine were extracted by organic solvents with acid and addition of EDTA. The limits of detection (LODs) in STP and DHS ranged 0.45~3.66 μg/kg and 0.22~0.78 μg/kg, respectively. The limits of quantification (LOQs) were 1.35~11.10 μg/kg in STP and 0.66~2.36 μg/kg in DHS. The recoveries (%) were 94.29~104.5% in STP and 92.32~108.45% in DHS except for plasma with lower values (61.45/68.5%, respectively). In the precision evaluation, the coefficient of variation (CV, %) of STP showed <10.50% on intra-day and <18.04% on inter-day. The CV (%) of DHS showed <8.42% on intra-day, whereas <17.98% on inter-day. The modified method is reliable for continuous residual monitoring in pig to ensure food safety for consumer’s health. In addition, this method could be used in study relation to residue depletion and pharmacokinetics of veterinary drug.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike (S) protein mediates virus entry by binding to the host cell receptor, human angiotensin converting enzyme 2 (hACE2), and catalyzing virus–host membrane fusion. The S protein also mediates cell–cell fusion, potentially allowing the virus to spread virion-independently. Here, we compared the fusogenicity of SARS-CoV-2 variant S proteins using a cell–cell fusion assay. In cells overexpressing hACE2, cell–cell fusion ability of all tested SARS-CoV-2 variants was similar to that of the Wuhan-Hu-1 strain. However, in cells with endogenous hACE2, SARS-CoV-2 variants, especially the Delta variant, stimulated significantly greater cell–cell fusion than the original strain. Our results showed that the Delta variant S protein is highly fusogenic and can spread rapidly by utilizing small amounts of hACE2.

The slaughter of livestock is inevitably required to obtain meat products from livestock. Since slaughter means pain and death for animals, reducing the pain and distress of animals during slaughter is very important from a perspective of animal welfare based on the principle of respect for life. Generally, two stunning methods, CO2 stunning and electrical stunning, are used for slaughter. When the carotid arteries of the lungs are cut off for bleeding, the bronchial tubes are also severed. To determine the degree of blood inflow into the lungs through the severed bronchial tubes, the prevalence rates of pulmonary diseases and pulmonary congestion in slaughtered pigs were analyzed. In this study, the prevalence of pneumonia was 24.9% in Slaughterhouse A using the gas method, but it was decreased by about 10% to 15.7% and 12.6%, respectively, in Slaughterhouses B and C using the electric method. On the other hand, the prevalence of pulmonary congestion in Slaughterhouses A, B, and C was 4.24%, 14.10% and 16.40%, respectively. In other words, the prevalence of pulmonary congestion was higher by about 10% in the groups of pigs slaughtered by the electric method (Slaughterhouses B and C) than in the group of pigs slaughtered by the gas method (Slaughterhouse A). These results indicate that blood inflow into the pulmonary alveoli led to a diagnosis of pulmonary congestion instead of pneumonia in some pigs with pneumonia. In short, it was found that about 10% of pigs stunned by the electric method were not in a state of complete unconsciousness but in a partially conscious state during slaughter. It is suggested that slaughterhouses should be equipped with gas stunning equipment instead of applying the electric method due to lower costs.

Chlorine dioxide (ClO2) has recently emerged as an ideal disinfectant and has shown a wide range of antimicrobial activities in various pathogenic microorganisms. In this study, the virucidal effect of ClO2 at low concentration (0.02 ppm) and higher concentration (0.06 – 0.09 ppm) against Adenovirus and Herpesvirus was evaluated based on the NF T 72-281 and ASTM 1053-11 standard methods at different exposure times. The virus suspension was dried onto the carrier and then exposed to gaseous ClO2 (gClO2) at 22 ± 2∘C. For Adenovirus, exposure at a low concentration of ClO2 at the middle height resulted in the average log10 reduction of 0.95, 2.65, and 5.30 after 1, 3, and 6 h post-exposure (pe), respectively. Moreover, more than 4-log10 reduction was achieved at 4 and 6 h pe with higher concentrations of ClO2. On the other hand, the antiviral activity of gClO2 at the middle height was also effective against Herpesvirus. In particular, at 1 h pe, a less than 4-log10 reduction was observed at all examined concentrations of ClO2, whereas exposure for 3 and 6 h (with low concentration) or 2 h (with higher concentration) inactivated completely viruses attached to the carrier. These results suggested that ClO2 fumigation is a potential alternative method for disinfecting healthcare facilities, high-containment laboratories, and households with a safe concentration for human health.

African swine fever (ASF) is a hemorrhagic viral disease of pigs requiring laboratory diagnosis for confirmation. Though tissue and blood samples are considered optimal for ASF diagnosis, collection of these samples can be laborious, time-consuming, and pose a risk of contaminating the environment. Here, we suggest an alternative non-invasive sampling method, hair plucking, for ASF diagnosis. ASF virus was detected in plucked hair samples from experimentally infected pigs. Although the sensitivity was inferior to whole blood, the results suggest that hair plucking can be an alternative method that can also improve animal welfare.

This study evaluated the effect of a combination of acetaminophen (AAP), vitamin C (VC) and thioctic acid (TA) on reducing serum cortisol concentrations in mice with foot-and-mouth disease (FMD) vaccination. For 5 days from 3 days before FMD vaccination to 2 days after vaccination, mice were orally administered with AAP 600 mg/kg feed (n=25, AAPT), combination of AAP 600 mg and VC 200 mg per kg feed (n=25, AVCT) and combination of AAP 600 mg, VC 200 mg and TA 20 mg per kg feed (n=25, AVTT), respectively. From day 1 to day 7 after FMD vaccination, the cortisol concentration of all groups treated with the drug was significantly decreased compared to that of the positive control group with FMD vaccination (p<0.05). In conclusion, the combined treatment of AAP, VC and TA was the most effective in relieving stress from FMD vaccination compared to the single treatment.

Avian colibacillosis is severe respiratory and systemic disease caused by avian pathogenic E. coli (APEC). The disease results in economic loss because of early mortality and reduced productivity in chickens. Several antimicrobials have been used extensively for reducing the losses due to colibacillosis. β-lactams, including penicillin derivatives, cephalosporins, and carbapenems, are one of the most commonly used, clinically important antibiotics. In this study, the phenotypic and genotypic characteristics of 14 CTX-M-producing APEC were investigated. Two types of CTX-M genes were detected, namely, CTX-M-1 gene (8 isolates) and CTX-M-15 gene (6 isolates), and all CTX-M-producing APEC were multidrug resistant. Moreover, 10 of the 14 CTX-M-producing E. coli isolates carried the TEM-1 gene, and 8 of the 14 isolates contained class 1 integrons. The following three different gene cassette arrangements were identified in six isolates: aadA1 (n=2), dfrA1 (n=2), and aadA1+dfrA1 (n=2). All CTX-M-producing APEC isolates harbored IncF plasmids, including F, FII, FIA, and FIB. CTX-M-1 gene was located on I1, F, and FIB, and CTX-M-15 gene on FII, FIA, and FIB. The findings from this study indicate that E. coli carrying CTX-M gene are circulating in the broiler poultry industry. Comprehensive surveillance and persistent monitoring of the integrated broiler operations are warranted to prevent the dissemination of resistant isolates.
